Functionalized erythrocyte-derived optical nanoparticles to target ephrin-B2 ligands.
Over- or under-expression of erythropoietin-production human hepatocellular receptors (Eph) and their ligands are associated with various diseases. Therefore, these molecular biomarkers can potentially be used as binding targets for the delivery of therapeutic and/or imaging agents to cells characterized by such irregular expressions. We have engineered nanoparticles derived from erythrocytes and doped with the near-infrared (NIR) FDA-approved dye, indocyanine green. We refer to these nanoparticles as NIR erythrocyte-derived transducers (NETs). We functionalized the NETs with the ligand-binding domain of a particular Eph receptor, EphB1, to target the genetically modified human dermal microvascular endothelial cells (hDMVECs) with coexpression of EphB1 receptor and its ligand ephrin-B2. This cell model mimics the pathological phenotypes of lesional endothelial cells (ECs) in port wine stains (PWSs). Our quantitative fluorescence imaging results demonstrate that such functionalized NETs bind to the ephrin-B2 ligands on these hDMVECs in a dose-dependent manner that varies sigmoidally with the number density of the particles. These nanoparticles may potentially serve as agents to target PWS lesional ECs and other diseases characterized with over-expression of Eph receptors or their associated ligands to mediate phototherapy

Non-Invasive Photoacoustic Imaging of In Vivo Mice with Erythrocyte Derived Optical Nanoparticles to Detect CAD/MI.
Coronary artery disease (CAD) causes mortality and morbidity worldwide. We used near-infrared erythrocyte-derived transducers (NETs), a contrast agent, in combination with a photoacoustic imaging system to identify the locations of atherosclerotic lesions and occlusion due to myocardial-infarction (MI). NETs (â90ânm diameter) were fabricated from hemoglobin-depleted mice erythrocyte-ghosts and doped with Indocyanine Green (ICG). Ten weeks old male C57BL/6 mice (nâ=â9) underwent left anterior descending (LAD) coronary artery ligation to mimic vulnerable atherosclerotic plaques and their rupture leading to MI. 150â”L of NETs (20â”M ICG,) was IV injected via tail vein 1-hour prior to photoacoustic (PA) and fluorescence in vivo imaging by exciting NETs at 800ânm and 650ânm, respectively. These results were verified with histochemical analysis. We observed â256-fold higher PA signal from the accumulated NETs in the coronary artery above the ligation. Fluorescence signals were detected in LAD coronary, thymus, and liver. Similar signals were observed when the chest was cut open. Atherosclerotic lesions exhibited inflammatory cells. Liver demonstrated normal portal tract, with no parenchymal necrosis, inflammation, fibrosis, or other pathologic changes, suggesting biocompatibility of NETs. Non-invasively detecting atherosclerotic plaques and stenosis using NETs may lay a groundwork for future clinical detection and improving CAD risk assessment
Computational Fluid Dynamic Simulations for Determination of Ventricular Workload in Aortic Arch Obstructions
Objective
The cardiac workload associated with various types of aortic obstruction was determined using computational fluid dynamic simulations. Methods
Computed tomography image data were collected from 4 patients with 4 distinct types of aortic arch obstructions and 4 controls. The categorization of arch hypoplasia corresponded to the âA, B, Câ nomenclature of arch interruption; a type âDâ was added to represent diffuse arch hypoplasia. Measurements of the vessel diameter were compared against the normal measurements to determine the degree of narrowing. Three-dimensional models were created for each patient, and additional models were created for type A and B hypoplasia to represent 25%, 50%, and 75% diameter narrowing. The boundary conditions for the computational simulations were chosen to achieve realistic flow and pressures in the control cases. The simulations were then repeated after changing the boundary conditions to represent a range of cardiac and vascular adaptations. The resulting cardiac workload was compared with the control cases. Results
Of the 4 patients investigated, 1 had aortic coarctation and 3 had aortic hypoplasia. The cardiac workload of the patients with 25% narrowing type A and B hypoplasia was not appreciably different from that of the control. When comparing the different arch obstructions, 75% type A, 50% type B, and 50% type D hypoplasia required a greater workload increase than 75% coarctation. Conclusions
The present study has determined the hemodynamic significance of aortic arch obstruction using computational simulations to calculate the cardiac workload. These results suggest that all types of hypoplasia pose more of a workload challenge than coarctation with an equivalent degree of narrowing

âThey Called Them Communists ThenââŠâWhat D'You Call âEm Now?ââŠâInsurgents?â. Narratives of British Military Expatriates in the Context of the New Imperialism
This paper addresses the question of the extent to which the colonial past provides material for contemporary actors' understanding of difference. The research from which the paper is drawn involved interview and ethnographic work in three largely white working-class estates in an English provincial city. For this paper we focus on ten life-history interviews with older participants who had spent some time abroad in the British military. Our analysis adopts a postcolonial framework because research participants' current constructions of an amorphous 'Other' (labelled variously as black people, immigrants, foreigners, asylum-seekers or Muslims) reveal strong continuities with discourses deployed by the same individuals to narrate their past experiences of living and working as either military expatriates or spouses during British colonial rule. Theoretically, the paper engages with the work of Frantz Fanon and Edward Said. In keeping with a postcolonial approach, we work against essentialised notions of identity based on 'race' or class. Although we establish continuity between white working-class military emigration in the past and contemporary racialised discourses, we argue that the latter are not class-specific, being as much the creations of the middle-class media and political elite

Energy dissipation in soil samples during drained triaxial shearing
The discrete-element method was used to simulate drained triaxial compression of large-scale, polydisperse numerical samples at a range of void ratios while tracing all relevant energy components. The frictional dissipation and boundary work are almost equal regardless of sample density. The volumetric work reaches a steady value at large strain. However, the distortional work increases continually as sample deformation continues post-critical state. There is a preferential orientation for frictional dissipation at around 45° to the major principal stress direction. This matches the orientation at which there is the largest number of sliding contacts. The work equations, which are fundamental in most commonly used constitutive models, are linear when plotted against deviatoric strain. The modified Cam Clay work equation substantially over-predicts the frictional dissipation for dense samples. An alternative, thermodynamically consistent work equation gives a much better description of frictional dissipation and is therefore recommended to ensure accuracy in modelling
âHow people read and write and they don't even noticeâ: everyday lives and literacies on a Midlands council estate
This article presents data from a British Academy-funded study of the everyday literacy practices of three families living on a predominantly white working-class council housing estate on the edge of a Midlands city. The study explored, as one participant succinctly put it, âhow people read and write and they don't even noticeâ. This alludes to the ways in which everyday practices may not be recognised as part of a dominant model of literacy. The study considered too the ways in which these literacy practices are part of a wider policy context that also fails to notice the impact of austerity politics on everyday lives. An emphasis on quantitative measures of disadvantage and public discourse which vilifies those facing economic challenge can overshadow the resilience and resourcefulness of individuals and families in making meaning from their experiences. Drawing together consideration of everyday lives and the everyday literacies which are part of them, this article explores the impact of the current policy context on access to both economic and cultural resources, showing how literacy, as part of this context, should be recognised as a powerful means not only of constricting lives but also of constructing them
