This paper compares two medicinal species of Kalanchoe, which are often used interchangeably by the population, regarding the distribution of anthocyanins under the influence of four luminosity levels for 6 months. For the morphoanatomical analysis, the 6th stem node of each plant was sectioned. Usual histochemical tests revealed the presence of anthocyanins by cross sections of the stems, petioles and leaf blades. The petioles and leaf blades were submitted to the extraction with acidified methanol, and the anthocyanins were quantified by spectrophotometric readings. At the macroscopic level, it was noticed for both species a higher presence of anthocyanins in stems and petioles of plants under full sunlight. The microscopy of K. brasiliensis stems evidenced the deposition of anthocyanins in the subjacent tissue to the epidermis and cortex, which increased with light intensity. In K. pinnata a subepidermal collenchyma was observed, which interfered in the visualization of anthocyanins. In petioles and leaf blades of K. brasiliensis the deposition of anthocyanins was peripheral, and in K. pinnata it was also throughout the cortex. The quantification of anthocyanins in petioles showed in 70% of light higher averages than in 25%, but in leaf blades there were no significant results. This study contributes to the pharmacognosy of Kalanchoe and it is sustained by the description of flavonoids as biological markers of the genus.

This study aimed to assess the anthropogenic activities at Rio Doce Park, Minas Gerais, Brazil, from ethnopharmacological surveys in Timóteo and Marliéria, which are located around the park. Interviews were conducted with previously identified, key informants, 15 in Timóteo and 10 in Marliéria. Two respondents collected medicinal plants in the forest of the park (from a few trees) but kept the same vulnerability of the use of their barks. Among the 141 surveyed botanical terms, we identified 95 species of 44 different botanical families. On the basis of statistical analyses, the 12 most used species were selected by respondents considering their therapeutic purposes and also obtaining the purpose of the use and dosage, among others. The knowledge about the use of medicinal plants has been maintained through generations but away from the formal health system and a sustainable management plan to encourage the preservation of the park

<p>Abstract</p> <p>Background</p> <p>Relaxin is the endogenous ligand of the G-protein coupled receptor RXFP1, previously known as LGR7. In humans relaxin can also activate, but with lower affinity, the closely related receptor for the insulin-like peptide from Leydig cells, RXFP2, previously known as LGR8. The lack of relaxin impairs male fertility but the precise distribution and the function of relaxin receptors in the male reproductive tract is not known. We investigated the distribution of Rxfp1 and Rxfp2 in the reproductive tract of the male rat and the function of relaxin in the vas deferens, a tissue with high expression of both receptors.</p> <p>Methods</p> <p>The presence of mRNA for Rxfp1 and Rxfp2 was investigated in testes, cultured Sertoli cells, epididymis, vas deferens, seminal vesicle, prostate, and spermatozoa by RT-PCR and Southern blot. Protein expression in the testis, vas deferens, primary culture of Sertoli cells, and spermatozoa was assessed by immunohistochemistry and immunofluorescence. The role of relaxin in the vas deferens was evaluated by contractility studies and radioimmunoassay of cAMP production. The effect of relaxin on mRNA levels for metalloproteinase-7 was measured by Northern blot.</p> <p>Results</p> <p>Transcripts for Rxfp1 and Rxfp2 were present in almost all parts of the male reproductive tract, with high levels in testis and vas deferens. Both receptors were immunolocalized in late stage germ cells but not in mature spermatozoa, although mRNAs for both receptors were also present in mature spermatozoa. Rxfp1 but not Rxfp2 was detected in cultured Sertoli cells. Strong immunostaining for Rxfp1 and Rxfp2 was seen in muscular and epithelial layers of the vas deferens and in arteriolar walls. Relaxin did not affect contractility and cyclic AMP production of the vas deferens, but increased the levels of mRNA for metalloproteinase-7.</p> <p>Conclusion</p> <p>Rxfp1 and Rxfp2 are widely and similarly distributed throughout the male reproductive tract. Our results suggest that Rxfp1 on spermatids and Sertoli cells may be important in spermatogenesis. Relaxin in the vas deferens does not affect contractility, but may affect vascular compliance and collagen and matrix remodeling.</p

Extracellular enolase of Candida albicans is involved in colonization of mammalian intestinal epithelium

Enolase is secreted by C. albicans and is present in its biofilms although its extracellular function is unknown. Here we show that extracellular enolase mediates the colonization of small intestine mucosa by C. albicans. Assays using intestinal mucosa disks show that C. albicans adhesion is inhibited, in a dose dependent mode, either by pretreatment of intestinal epithelium mucosa disks with recombinant C. albicans enolase (70% at 0.5 mg/ml enolase) or by pretreatment of C. albicans yeasts with anti-enolase antibodies (48% with 20 µg antiserum). Also using flow cytometry, immunoblots of conditioned media and confocal microscopy we demonstrate that enolase is present in biofilms and that the extracellular enolase is not an artifact due to cell lysis, but must represent functional secretion of a stable form. This is the first direct evidence that C. albicans extracellular enolase mediates colonization on its primary translocation site. Also, because enolase is encoded by a single locus in C. albicans, its dual role peptide, as glycolytic enzyme and extracellular peptide, is a remarkable example of gene sharing in fungi

Eremanthus erythropappus (DC.) MacLeisch (Asteraceae), commonly known as 'candeia-da-serra', is a plant used in folk medicine as wound healing and antimicrobial. The aim of this study was to compare the number of glandular trichomes between the young and the mature leaves, as well as to perform the morpho-anatomical characterization of E. erythropappus leaves. Transverse and paradermal sections of the leaves were prepared according to light and scanning microtechniques for the morpho-anatomical characterization. The quantification of glandular trichomes on the adaxial surface of the epidermis was evaluated in young and mature leaves with camera lucida. The leaves are alternate or fasciculate and the blade has uniseriate epidermis coated with thin and smooth cuticle and dorsiventral mesophyll. There are predominantly anomocytic stomata on both surfaces, as well as glandular trichomes located in epidermal depressions. Various non-glandular trichomes are encountered on the abaxial surface. The palisade parenchyma consists of a single layer of cells and the parenchyma which is faced to the abaxial surface comprehends three to five layers of cells in compact arrangement. The young and mature leaves showed, respectively, 21.78±5.83 e 17.80±6.69 glandular trichomes on the adaxial side. The morpho-anatomical analysis of E. erythropappus leaves has proved to be a practical and rapid method for the identification and quality control of the vegetal species used for medical purposes.Eremanthus erythropappus (DC.)

Echinodorus macrophyllus (Kunth) Micheli – Alismataceae, known as "chapéu-de-couro", "chá-mineiro", "erva-de-pântano", "erva-de-bugre", "congonha-do-brejo", "erva-do-brejo", occurs in Minas Gerais and São Paulo. Although it has therapeutic and commercial importance and is described in the two first editions of the Brazilian Pharmacopoeia (1924, 1959), there are few studies that contemplate it. This present work had the objective to study the leaf and petiole morpho-anatomy of A. macrophyllus, in order to extend its pharmacognosy knowledge, aiming at the quality control. The macroscopic description was carried through disarmed sight, with optical resources and the dimensions with a caliper. The anatomical c haracteristics had been determined, after the harvest, in the fragments of leaves and fixed petioles, freehand sectioned and stained either in glycerin, astra-blue or fuchsine. The morpho-anatomical study of the leaf and petiole of A. macrophyllust contributed to extend and deepen the information contained in the Brazilian Pharmacopoeia (1924, 1959). The designated structural characteristics assist in the identification of the species and supply parameters that could be applied in the pharmacognosy quality control in the future. And, the presence of non-glandular trichomes in the midribs (adaxial and abaxial surface) and the petiole exhibits diaphragm (specialized cells), key structures in the diagnosis of the species, that are not described in the Brazilian Pharmacopoeia (1924, 1959), were inserted in the context of the anatomical description.

The present work investigated the analgesic and anti-inflammatory activities of the methanol extract of the rhizomes of Echinodorus grandiflorus. For this purpose, acetic acid writhing, paw licking induced by formalin, carrageenan-induced rat paw edema and pleurisy tests were performed. The doses of 100, 200 and 400 mg/kg inhibited the contortions 34.30%, 35.84% and 36.07%, respectively. The lick of the paw 1st phase was reduced at the doses of 200 and 400 mg/kg, while the 2nd phase had reduction at 400 mg/kg. All doses inhibited the paw edema, while the volume of the exsudate was reduced 24.61% in the dose of 400 mg/kg. The doses of 200 and 400 mg/kg decreased the leukocytes migration. The results indicate that the methanol extract of E. grandiflorus can constitute target potential for use in therapies of the pain and inflammation.O presente trabalho investigou as atividades analgésica e antiinflamatória do extrato metanólico de rizomas de Echinodorus grandiflorus. We demonstrate the growth of graphene nanocrystals by molecular beam methods that employ a solid carbon source, and that can be used on a diverse class of large area dielectric substrates. Characterization by Raman and Near Edge X-ray Absorption Fine Structure spectroscopies reveal a sp2 hybridized hexagonal carbon lattice in the nanocrystals. Lower growth rates favor the formation of higher quality, larger size multi-layer graphene crystallites on all investigated substrates. The surface morphology is determined by the roughness of the underlying substrate and graphitic monolayer steps are observed by ambient scanning tunneling microscopy.Comment: Accepted in Carbon; Discussion section added; 20 pages, 6 figures (1 updated
