    ‘Omaha Blues’ Recalls Lost Childhood

    In 1997, I walked into Joe Lelyveld’s office and handed him a newspaper clipping about his parents. Lelyveld was executive editor of the New York Times, and I was one of his nearly 1,100 news department employees. The clipping, from a World-Herald column, noted that the Lelyvelds had once lived in Omaha; that Arthur, Joe’s father, had been rabbi at Temple Israel in the early 1940s; that his mother, Toby, had taught at the University of Omaha; and that the Lelyvelds had divorced in 1964. The news was that Toby had died in New York at age 85. If I thought that taking him the clipping might give me even the most tenuous personal connection to my boss—I grew up in Nebraska, too—I was wrong. Lelyveld, known to have a somewhat awkward manner socially, looked up as I gave him the clipping but said nothing. He offered only a small, enigmatic smile, and I left. Now, having read “Omaha Blues,” Lelyveld’s quietly powerful memoir of growing up with parents who often seemed to have little interest in him, I realize that in trying to buy a personal moment with Lelyveld, I may have been using the wrong currency

    Performance of a building integrated photovoltaic/thermal (BIPVT) solar collector

    The idea of combining photovoltaic and solar thermal collectors (PVT collectors) to provide electrical and heat energy is an area that has, until recently, received only limited attention. Although PVTs are not as prevalent as solar thermal systems, the integration of photovoltaic and solar thermal collectors into the walls or roofing structure of a building could provide greater opportunity for the use of renewable solar energy technologies. In this study, the design of a novel building integrated photovoltaic/thermal (BIPVT) solar collector was theoretically analysed through the use of a modified Hottel–Whillier model and was validated with experimental data from testing on a prototype BIPVT collector. The results showed that key design parameters such as the fin efficiency, the thermal conductivity between the PV cells and their supporting structure, and the lamination method had a significant influence on both the electrical and thermal efficiency of the BIPVT. Furthermore, it was shown that the BIPVT could be made of lower cost materials, such as pre-coated colour steel, without significant decreases in efficiency. Finally, it was shown that by integrating the BIPVT into the building rather than onto the building could result in a lower cost system. This was illustrated by the finding that insulating the rear of the BIPVT may be unnecessary when it is integrated into a roof above an enclosed air filled attic, as this air space acts as a passive insulating barrier

    An investigation of the degradation of Fluorinated Ethylene Propylene (FEP) copolymer thermal blanketing materials aboard LDEF in the laboratory

    Samples of fluorinated ethylene propylene copolymer thermal blanketing material, recovered from the Long Duration Exposure Facility (LDEF), were investigated to determine the nature and the extent of degradation due to exposure to the low-Earth-orbit environment. Samples recovered from the ram-facing direction of LDEF, which received vacuum-ultraviolet (VUV) radiation and atomic-oxygen impingement, and samples from the trailing edge, which received almost exclusively VUV exposure, were investigated by scanning electron microscopy and atomic force microscopy. The most significant result of this investigation was found on samples that received only VUV exposure. These samples possessed a hard, embrittled surface layer that was absent from the atomic-oxygen exposed sample and from unexposed control samples. This surface layer is believed to be responsible for the 'synergistic' effect between VUV and atomic oxygen. Overall, the investigation revealed dramatically different morphologies for the two samples. The sample receiving both atomic-oxygen and VUV exposure was deeply eroded and had a characteristic 'rolling' surface morphology, while the sample that received only VUV exposure showed mild erosion and a surface morphology characterized by sharp high-frequency peaks. The morphologies observed in the LDEF samples, including the embrittled surface layer, were successfully duplicated in the laboratory

    The Pseudomonas aeruginosa Magnesium Transporter MgtE Inhibits Transcription of the Type III Secretion System

    Pseudomonas aeruginosa is an opportunistic pathogen that causes life-long pneumonia in individuals with cystic fibrosis (CF). These long-term infections are maintained by bacterial biofilm formation in the CF lung. We have recently developed a model of P. aeruginosa biofilm formation on cultured CF airway epithelial cells. Using this model, we discovered that mutation of a putative magnesium transporter gene, called mgtE, led to increased cytotoxicity of P. aeruginosa toward epithelial cells. This altered toxicity appeared to be dependent upon expression of the type III secretion system (T3SS). In this study, we found that mutation of mgtE results in increased T3SS gene transcription. Through epistasis analyses, we discovered that MgtE influences the ExsE-ExsC-ExsD-ExsA gene regulatory system of T3SS by either directly or indirectly inhibiting ExsA activity. While variations in calcium levels modulate T3SS gene expression in P. aeruginosa, we found that addition of exogenous magnesium did not inhibit T3SS activity. Furthermore, mgtE variants that were defective for magnesium transport could still complement the cytotoxicity effect. Thus, the magnesium transport function of MgtE does not fully explain the regulatory effects of MgtE on cytotoxicity. Overall, our results indicate that MgtE modulates expression of T3SS genes

    Pseudomonas aeruginosa Magnesium Transporter MgtE Inhibits Type III Secretion System Gene Expression by Stimulating rsmYZ Transcription

    Pseudomonas aeruginosa causes numerous acute and chronic opportunistic infections in humans. One of its most formidable weapons is a type III secretion system (T3SS), which injects powerful toxins directly into host cells. The toxins lead to cell dysfunction and, ultimately, cell death. Identification of regulatory pathways that control T3SS gene expression may lead to the discovery of novel therapeutics to treat P. aeruginosa infections. In a previous study, we found that expression of the magnesium transporter gene mgtE inhibits T3SS gene transcription. MgtE-dependent inhibition appeared to interfere with the synthesis or function of the master T3SS transcriptional activator ExsA, although the exact mechanism was unclear. We now demonstrate that mgtE expression acts through the GacAS two-component system to activate rsmY and rsmZ transcription. This event ultimately leads to inhibition of exsA translation. This inhibitory effect is specific to exsA as translation of other genes in the exsCEBA operon is not inhibited by mgtE Moreover, our data reveal that MgtE acts solely through this pathway to regulate T3SS gene transcription. Our study reveals an important mechanism that may allow P. aeruginosa to fine-tune T3SS activity in response to certain environmental stimuli.IMPORTANCE The type III secretion system (T3SS) is a critical virulence factor utilized by numerous Gram-negative bacteria, including Pseudomonas aeruginosa, to intoxicate and kill host cells. Elucidating T3SS regulatory mechanisms may uncover targets for novel anti-P. aeruginosa therapeutics and provide deeper understanding of bacterial pathogenesis. We previously found that the magnesium transporter MgtE inhibits T3SS gene transcription in P. aeruginosa In this study, we describe the mechanism of MgtE-dependent inhibition of the T3SS.     Site of semen deposition and fertility in lactating beef cows synchronized with GnRH and PGF2α

    Our objective was to determine the effect of site of semen deposition on pregnancy rate in beef cows inseminated at a fixed time or after observed estrus. Cows were synchronized with a combination of gonadotropin-releasing hormone (GnRH) prostaglandin-F α (PGF). GnRH was injected 7 days before PGF (day 0; first of breeding season). The trial was conducted at two locations, one in Kansas (147 cows) and one in Colorado (313 cows). At each location, cows were assigned to be inseminated after observed estrus (ESTRUS-AI) or at a fixed time (TIMED-AI). Within these two groups, cows either were inseminated in the uterine body (BODY-bred) or in both uterine horns (HORN-bred). Cows in the ESTRUS-AI group were observed for estrus each morning and evening until day 5 afterPGF and then inseminated 12 hr after first detected estrus. Cows in the TIMED-AI group received a second dose of GnRH on day 2 and were inseminated at that time (48 to 56 hr after PGF). Heat response, AI conception rate, and pregnancy rate were analyzed for BODY-bred and HORN-bred cows within each treatment at each location. No differences in these variables occurred between locations, so the results were combined. Within the ESTRUS-AI group, neither conception rate (70% vs. 73%) nor pregnancy rate (39% vs. 40%) was different between BODY-bred and HORN-bred cows respectively. Pregnancy rate within the TIMED-AI group tended (P=.09) to be greater for BODY-bred (53%) compared to HORN-bred (42%) cows. When BODY-bred and HORN-bred treatments were combined, the pregnancy rate of TIMED-AI cows (48%) tended (P=.07) to be greater than that of ESTRUS-AI cows (39%). Timed-insemination resulted in a greater pregnancy rate than inseminating cows according to estrus. No advantage was seen in conception rates when semen was deposited in the uterine horns compared to the uterine body

    Gynaecomastia in two men on stable antiretroviral therapy who commenced treatment for tuberculosis

    Gynaecomastia is a common clinical presentation that varies from benign presentations in stages of human development to hormonal pathology, mainly due to hepatic  dysfunction, malignancy, and adverse pharmacologic effects. We describe the  development of significant bilateral gynaecomastia after starting treatment for pulmonary tuberculosis (TB) in two males with WHO stage III Human  Immunodeficiency Virus (HIV) infection on stable antiretroviral regimens. Emerging reports suggest that distinct hepatic impairment in efavirenz metabolism modulates oestrogenic activity, which may be potentiated by antituberculosis therapy. Clinical application includes early recognition of efavirenz-induced gynaecomastia, especially after commencing tuberculosis treatment. To avoid decreased adherence resulting from the distressing side effect of gynecomastia, transition to an alternative ART regimen over the course of tuberculosis treatment should be considered
