    Photodynamic Cancer Therapy A Study of the Photolysis Reaction between Porphyrin and 9,10-Diphenylanthracene

    Photodynamic therapy (PDT) is a method of combating cancerous tumors. PDT works when certain molecules called photosensitizers absorb light and transfer that energy to dissolved oxygen in solution. Next, this singlet oxygen will interfere with nearby molecules that are necessary for cancer cells to function. This research focuses on the kinetics (rates) of the reaction with respect to the photosensitizer, light, and the target molecule. A laser set at 514nm was used in photolysis with the photosensitizer 5,10,15,20-Tetrakis(2,3,5,6-tetrafluorophenyl)porphyrin (TPP). To mimic cancer cells and monitor the effectiveness of the chosen photosensitizer, 9,10-Diphenylanthracene (DPA) was used as the target molecule. In knowing the kinetics of the reaction, practical things like the dosage of the photosensitizer can be better determined for PDT. In this research, consistent data was found that allows for the prediction of the decomposition of DPA when the concentrations of TPP and DPA and the intensity of the laser light are known.     Field Guide to Nonindigenous Marine Fishes of Florida

    The purpose of this field guide is to provide information on nonindigenous (i.e., non-native) fishes that have been observed in Florida’s marine waters. Introductions of non-native marine fishes into Florida’s waters could be intentional or unintentional, and are likely from a variety of sources, including aquarium releases, escape from aquaculture, loss due to extreme weather events (e.g., flooding from hurricanes), and possibly transfer with ballast water or hull-fouling. Presently the lionfishes (Pterois volitans and P. miles) are the only non-native marine fish species known to be established along the coast of Florida. All other marine fishes in this guide (except the euryhaline species, see below) have infrequent occurrences, occur singly or in small groups, and have not yet become self-sustaining populations. Aquarium releases are one of the major pathways whereby nonindigenous fishes gain access to new environments (Ruiz et al. 1997; Fuller et al. 1999). Most of the nonindigenous marine fishes found in Florida’s waters are thought to be aquarium fishes that either were illegally released into the ocean or escaped captivity (e.g., during severe storm/flooding events). Indeed, south Florida is a hotspot for nonindigenous marine aquarium fishes (Semmens et al. 2004). Increased public awareness of the problems caused by released or escaped aquarium fishes may aid in stemming the frequency of releases. For example, HabitattitudeTM (www.habitattitude.net) is a national public awareness and partnership campaign that encourages aquarists and water gardeners to prevent the release of unwanted aquarium plants, fish and other animals. It prompts hobbyists to adopt alternative actions when dealing with these aquatic plants and animals. (PDF file contains 133 pages.

    The problem of retrieving and storing core samples from a hole drilled on the lunar surface is addressed. The total depth of the hole in question is 50 meters with a maximum diameter of 100 millimeters. The core sample itself has a diameter of 60 millimeters and will be two meters in length. It is therefore necessary to retrieve and store 25 core samples per hole. The design utilizes a control system that will stop the mechanism at a certain depth, a cam-linkage system that will fracture the core, and a storage system that will save and catalogue the cores to be extracted. The Rod Changer and Storage Design Group will provide the necessary tooling to get into the hole as well as to the core. The mechanical design for the cam-linkage system as well as the conceptual design of the storage device are described

    Autoradiographic Localization of [3H]-Nisoxetine Binding Sites in the CNS of Male and Female Japanese Quail

    Background In the central nervous system of mammals, transporters localized on the presynaptic nerve terminals regulate the reuptake of neurotransmitters. These transporters are selective for a specific neurotransmitter such as dopamine (DA) and norepinephrine (NE). Specifically in the synapse, the dopamine transporter (DAT) reuptakes DA and the norepinephrine transporter (NET) reuptakes NE. However previous research has found that avian species do not have a gene for DAT, and therefore, birds may be using the NET to clear both NE and DA from the synapse. The current study aimed to extend this finding by localizing NET expression in male and female Japanese quail (Coturnix japonica) brains using [3H]Nisoxetine, a selective NET blocker. Results High densities of binding sites were observed in the olfactory tubercle (OTu), the medial striatum (MSt), and the lateral striatum (LSt). Lower densities of binding sites were detected in the amygdala (AMY) and hypothalamus (Hyp), and low binding was found in the medial preoptic area (mPOA) and the pallium. Conclusion The areas with the highest densities of NET are also areas that previous research has shown to have high levels of DA activity but low levels of NE innervation (e.g. striatum). The distribution of this reuptake transporter is consistent with the theory that NET acts to clear both DA and NE from the synapse

    Biology and ecology of the invasive lionfishes, Pterois miles and Pterois volitans

    The Indo-Pacific lionfishes, Pterois miles and P. volitans, are now established along the U.S. southeast coast, Bermuda, Bahamas, and are becoming established in the Caribbean. While these lionfish are popular in the aquarium trade, their biology and ecology are poorly understood in their native range. Given the rapid establishment and potential adverse impacts of these invaders, comprehensive studies of their biology and ecology are warranted. Here we provide a synopsis of lionfish biology and ecology including invasion chronology, taxonomy, local abundance, reproduction, early life history and dispersal, venomology, feeding ecology, parasitology, potential impacts, and control and management. This information was collected through review of the primary literature and published reports and by summarizing current observations. Suggestions for future research on invasive lionfish in their invaded regions are provided

    Coherent radar ice thickness measurements over the Greenland ice sheet

    This is the published version, also available here: http://dx.doi.org/10.1029/2001JD900183.We developed two 150-MHz coherent radar depth sounders for ice thickness measurements over the Greenland ice sheet. We developed one of these using connectorized components and the other using radio frequency integrated circuits (RFICs). Both systems are designed to use pulse compression techniques and coherent integration to obtain the high sensitivity required to measure the thickness of more than 4 km of cold ice. We used these systems to collect radar data over the interior and margins of the ice sheet and several outlet glaciers. We operated both radar systems on the NASA P-3B aircraft equipped with GPS receivers. Radar data are tagged with GPS-derived location information and are collected in conjunction with laser altimeter measurements. We have reduced all data collected since 1993 and derived ice thickness along all flight lines flown in support of Program for Regional Climate Assessment (PARCA) investigations and the North Greenland Ice Core Project. Radar echograms and derived ice thickness data are placed on a server at the University of Kansas (http://tornado.rsl.ukans.edu/Greenlanddata.htm) for easy access by the scientific community. We obtained good ice thickness information with an accuracy of ±10 m over 90% of the flight lines flown as a part of the PARCA initiative.     A long period grating optical fiber sensor with nano-assembled porphyrin layers for detecting ammonia gas

    In this study, the ability of a long period grating (LPG) optical fiber sensor coated with a multilayer film of poly(diallyldimethyammonium chloride) (PDDA) and tetrakis(4-sulfophenyl)porphine (TSPP) to detect ammonia gas was demonstrated. The nano-assembled thin film was prepared using a layer-by-layer deposition technique. This combination of an LPG and TSPP could allow highly sensitive optical sensors that specifically bind ammonia to be fabricated. An LPG with a period of 100 μm and a PDDA/TSPP coating produced by depositing fifteen layers afforded a limit of detection of 0.67 ppm for ammonia gas. TSPP molecules in the film acted as ammonia receptors because the TSPP morphologically changed from J- or H-aggregates to free base monomers when it was deprotonated by being exposed to ammonia. Interestingly, HCl vapor could be used to increase the relative amount of J-aggregation in the TSPP and to restore the sensor response. The reversibility of the morphological change in the TSPP allowed reversible changes to occur in the properties of the coating, including the refractive index, film thickness and density, and electrostatic interactions; these influenced the transmission spectrum of the LPG

    Nutritional properties of the invasive lionfish: A delicious and nutritious approach for controlling the invasion

    Lionfish, Pterois volitans and P. miles, are native to the Indo-Pacific and have recently invaded the Western Atlantic Ocean. Strategies for control of this invasion have included limited removal programs and promotion of lionfish consumption at both local and commercial scales. We demonstrate that lionfish meat contains higher levels of healthy n-3 fatty acids than some frequently consumed native marine fish species. Mean lionfish fillet yield was 30.5% of the total body wet weight, a value that is similar to that of some grouper and porgy species. A sensory evaluation indicated that lionfish meet the acceptability threshold of most consumers

    Effect of Levels of Acetate on the Mevalonate Pathway of Borrelia burgdorferi

    Borrelia burgdorferi, the agent of Lyme disease, is a spirochetal pathogen with limited metabolic capabilities that survives under highly disparate host-specific conditions. However, the borrelial genome encodes several proteins of the mevalonate pathway (MP) that utilizes acetyl-CoA as a substrate leading to intermediate metabolites critical for biogenesis of peptidoglycan and post-translational modifications of proteins. In this study, we analyzed the MP and contributions of acetate in modulation of adaptive responses in B. burgdorferi. Reverse-transcription PCR revealed that components of the MP are transcribed as individual open reading frames. Immunoblot analysis using monospecific sera confirmed synthesis of members of the MP in B. burgdorferi. The rate-limiting step of the MP is mediated by HMG-CoA reductase (HMGR) via conversion of HMG-CoA to mevalonate. Recombinant borrelial HMGR exhibited a Km value of 132 µM with a Vmax of 1.94 µmol NADPH oxidized minute−1 (mg protein)−1 and was inhibited by statins. Total protein lysates from two different infectious, clonal isolates of B. burgdorferi grown under conditions that mimicked fed-ticks (pH 6.8/37°C) exhibited increased levels of HMGR while other members of the MP were elevated under unfed-tick (pH 7.6/23°C) conditions. Increased extra-cellular acetate gave rise to elevated levels of MP proteins along with RpoS, CsrABb and their respective regulons responsible for mediating vertebrate host-specific adaptation. Both lactone and acid forms of two different statins inhibited growth of B. burgdorferi strain B31, while overexpression of HMGR was able to partially overcome that inhibition. In summary, these studies on MP and contributions of acetate to host-specific adaptation have helped identify potential metabolic targets that can be manipulated to reduce the incidence of Lyme disease
