    Reflection Moveout Inversion In Azimuthally Anisotropic Media: Accuracy, Limitation, And Acquisition

    Parameter estimation from elliptical variations in the normal-moveout (NMO) velocity in azimuthally anisotropic media is sensitive to the angular separation between the survey lines in 2D, or equivalently source-to-receiver azimuth in 3D, and to the set of azimuths used in the inversion procedure. The accuracy in estimating the orientation of the NMO ellipse, the parameter cr, in particular, is also sensitive to the strength of anisotropy. To invert for the parameters the NMO ellipse, at least three NMO-velocity measurements along distinct azimuth directions are needed. In order to maximize the accuracy and stability in parameter estimation, it is best to have the azimuths for the three source-to-receiver directions 60° apart. Having more than three distinct source-to-receiver azimuths (e.g., full azimuthal coverage) provides a useful data redundancy that enhances the quality of the estimates. In orthorhombic media, inverting for the semi-axes of the NMO-ellipse allows the computation of the difference in the anisotropic parameters δ[superscript (1)] and δ[superscript (2)]. Additional information such as well data, is necessary in order to determine δ[superscript (1)] and δ[superscript (2)]. Furthermore, the accuracy in estimating the semi-axes of the NMO-velocity ellipse is about the same for any strength of anisotropy. To maximize quality in the inversion process, it is recommended that at the design stage of seismic data acquisition to have small sector sizes (≤ 10°) with adequate fold and offset distribution. For three azimuth directions, 60° apart, to perform the inversion, an azimuthally anisotropic layer overlain by an azimuthally isotropic overburden (as might happen for fractured reservoirs) should have a time thickness, relative to the total time, of at least the ratio of the error in the NMO (stacking) velocity to the interval anisotropy strength of the fractured layer. Coverage along more than three azimuths, however, improves this limitation, which is imposed by Dix differentiation, by at most 50% depending on the number of observations (NMO Velocities) that enter the inversion procedure.Saudi Aramc

    Importance of far-infrared mapping in a spiral galaxy: AKARI observation of M81

    The importance of the far-infrared (FIR) mapping is demonstrated for a face-on spiral galaxy, M81, by analyzing its imaging data at 65, 90, and 140 {\mu}m taken by AKARI. Basic products are the dust temperature map, the dust optical depth map, and the colour-colour diagram. The main features are as follows. (i) The dust temperature derived from the total fluxes at 90 {\mu}m and 140 {\mu}m reflects the relatively low temperatures seen in the interarm and spiral arms excluding the warm spots, rather than the high temperatures in warm spots and the centre. This indicates that the total FIR luminosity is dominated by the dust heated by the general interstellar radiation field. (ii) The galaxy is more extended at 140 {\mu}m than at the other shorter wavelengths, which reflects the radial dust temperature gradient. (iii) The dust optical depth derived from the FIR mapping is broadly consistent with that estimated from the FIR-to-ultraviolet luminosity ratio. (iv) The FIR colour-colour diagramis useful to identify a 'contamination' of warm dust. The existence of small-scale warm star-forming regions is supported in the bright spots along the spiral arms.     Discordance between the predictors of clinical and imaging remission in patients with early rheumatoid arthritis in clinical practice: implications for the use of ultrasound within a treatment to target strategy

    Objective: To assess the prevalence, relationship between and predictors of clinical and imaging remission in early rheumatoid arthritis (RA), achieved with treat-to-target management in clinical practice. Methods: A prospective observational study was conducted in patients with new-onset RA. The treatment target was remission by disease activity score (DAS28-CRP<2.6). Twelve month outcomes included DAS28-CRP remission, DAS44-CRP remission, ACR/EULAR Boolean remission (BR) and absent or absent/minimal power Doppler activity (PDA) on ultrasound (US) of 26 joints (total PDA score=0 or ≤1, respectively). Logistic regression was conducted to identify baseline predictors of these outcomes. Results: Of 105 patients with complete 12-month data, the rate of DAS28-CRP remission was 43%, DAS44-CRP remission was 39%, BR was 14%, absent PDA was 40% and absent/minimal PDA was 57%. Amongst patients achieving clinical remission defined by DAS28-CRP, DAS44-CRP or BR, absence of PDA was observed in 42%, 44% and 40%, respectively; absent/minimal PDA was detected in 62%, 66% and 67%, respectively. On multivariable analysis, shorter symptom duration, male gender, fewer tender joints and lower disability were associated with the clinical remission definitions. Lack of osteoarthritis predicted absence of PDA and lower total baseline PDA predicted absent/minimal PDA. Conclusion: DAS28-CRP remission and absence of PDA were observed in almost half of patients, but less than a quarter achieved both. Achievement of BR was rare. The low agreement between any of the clinical and imaging outcomes and differences in their predictors highlight the complex interaction between symptoms and synovitis, with implications for treat-to-target management. Long-term follow-up should determine the most appropriate target

    Imaging spontaneous currents in superconducting arrays of pi-junctions

    Superconductors separated by a thin tunneling barrier exhibit the Josephson effect that allows charge transport at zero voltage, typically with no phase shift between the superconductors in the lowest energy state. Recently, Josephson junctions with ground state phase shifts of pi proposed by theory three decades ago have been demonstrated. In superconducting loops, pi-junctions cause spontaneous circulation of persistent currents in zero magnetic field, analogous to spin-1/2 systems. Here we image the spontaneous zero-field currents in superconducting networks of temperature-controlled pi-junctions with weakly ferromagnetic barriers using a scanning SQUID microscope. We find an onset of spontaneous supercurrents at the 0-pi transition temperature of the junctions Tpi = 3 K. We image the currents in non-uniformly frustrated arrays consisting of cells with even and odd numbers of pi-junctions. Such arrays are attractive model systems for studying the exotic phases of the 2D XY-model and achieving scalable adiabatic quantum computers.Comment: Pre-referee version. Accepted to Nature Physic

    Atypical genomic cortical patterning in autism with poor early language outcome.

    Cortical regionalization develops via genomic patterning along anterior-posterior (A-P) and dorsal-ventral (D-V) gradients. Here, we find that normative A-P and D-V genomic patterning of cortical surface area (SA) and thickness (CT), present in typically developing and autistic toddlers with good early language outcome, is absent in autistic toddlers with poor early language outcome. Autistic toddlers with poor early language outcome are instead specifically characterized by a secondary and independent genomic patterning effect on CT. Genes involved in these effects can be traced back to midgestational A-P and D-V gene expression gradients and different prenatal cell types (e.g., progenitor cells and excitatory neurons), are functionally important for vocal learning and human-specific evolution, and are prominent in prenatal coexpression networks enriched for high-penetrance autism risk genes. Autism with poor early language outcome may be explained by atypical genomic cortical patterning starting in prenatal development, which may detrimentally affect later regional functional specialization and circuit formation

    Humanized mice in studying efficacy and mechanisms of PD-1-targeted cancer immunotherapy.

    Establishment of an in vivo small animal model of human tumor and human immune system interaction would enable preclinical investigations into the mechanisms underlying cancer immunotherapy. To this end, nonobese diabetic (NOD).Cg- PrkdcscidIL2rgtm1Wjl/Sz (null; NSG) mice were transplanted with human (h)CD34+ hematopoietic progenitor and stem cells, which leads to the development of human hematopoietic and immune systems [humanized NSG (HuNSG)]. HuNSG mice received human leukocyte antigen partially matched tumor implants from patient-derived xenografts [PDX; non-small cell lung cancer (NSCLC), sarcoma, bladder cancer, and triple-negative breast cancer (TNBC)] or from a TNBC cell line-derived xenograft (CDX). Tumor growth curves were similar in HuNSG compared with nonhuman immune-engrafted NSG mice. Treatment with pembrolizumab, which targets programmed cell death protein 1, produced significant growth inhibition in both CDX and PDX tumors in HuNSG but not in NSG mice. Finally, inhibition of tumor growth was dependent on hCD8+ T cells, as demonstrated by antibody-mediated depletion. Thus, tumor-bearing HuNSG mice may represent an important, new model for preclinical immunotherapy research. FASEB J 2018 Mar; 32(3):1537-1549

    Differential regulation by AMP and ADP of AMPK complexes containing different γ subunit isoforms

    The γ subunits of heterotrimeric AMPK complexes contain the binding sites for the regulatory adenine nucleotides AMP, ADP and ATP. We addressed whether complexes containing different γ isoforms display different responses to adenine nucleotides by generating cells stably expressing FLAG-tagged versions of the γ 1, γ 2 or γ 3 isoform. When assayed at a physiological ATP concentration (5 mM), γ 1- and γ 2-containing complexes were allosterically activated almost 10-fold by AMP, with EC50 values one to two orders of magnitude lower than the ATP concentration. By contrast, γ 3 complexes were barely activated by AMP under these conditions, although we did observe some activation at lower ATP concentrations. Despite this, all three complexes were activated, due to increased Thr172 phosphorylation, when cells were incubated with mitochondrial inhibitors that increase cellular AMP. With γ 1 complexes, activation and Thr172 phosphorylation induced by the upstream kinase LKB1 [liver kinase B1; but not calmodulin-dependent kinase kinase (CaMKKβ)] in cell-free assays was markedly promoted by AMP and, to a smaller extent and less potently, by ADP. However, effects of AMP or ADP on activation and phosphorylation of the γ 2 and γ 3 complexes were small or insignificant. Binding of AMP or ADP protected all three γ subunit complexes against inactivation by Thr172 dephosphorylation; with γ 2 complexes, ADP had similar potency to AMP, but with γ 1 and γ 3 complexes, ADP was less potent than AMP. Thus, AMPK complexes containing different γ subunit isoforms respond differently to changes in AMP, ADP or ATP. These differences may tune the responses of the isoforms to fit their differing physiological roles

    Origin and Examination of a Leafhopper Facultative Endosymbiont

    Eukaryotes engage in intimate interactions with microbes that range in age and type of association. Although many conspicuous examples of ancient insect associates are studied (e.g., Buchneraaphidicola), fewer examples of younger associations are known. Here, we further characterize a recently evolved bacterial endosymbiont of the leafhopper Euscelidius variegatus (Hemiptera, Cicadellidae), called BEV. We found that BEV, continuously maintained in E. variegatus hosts at UC Berkeley since 1984, is vertically transmitted with high fidelity. Unlike many vertically transmitted, ancient endosymbioses, the BEV–E. variegatus association is not obligate for either partner, and BEV can be cultivated axenically. Sufficient BEV colonies were grown and harvested to estimate its genome size and provide a partial survey of the genome sequence. The BEV chromosome is about 3.8 Mbp, and there is evidence for an extrachromosomal element roughly 53 kb in size (e.g., prophage or plasmid). We sequenced 438 kb of unique short-insert clones, representing about 12% of the BEV genome. Nearly half of the gene fragments were similar to mobile DNA, including 15 distinct types of insertion sequences (IS). Analyses revealed that BEV not only shares virulence genes with plant pathogens, but also is closely related to the plant pathogenic genera Dickeya, Pectobacterium, and Brenneria. However, the slightly reduced genome size, abundance of mobile DNA, fastidious growth in culture, and efficient vertical transmission suggest that symbiosis with E. variegatus has had a significant impact on genome evolution in BEV

    Search for the Θ+\Theta^+ pentaquark in the reaction γdpKK+n\gamma d \to p K^- K^+ n

    A search for the \thp in the reaction γdpKK+n\gamma d \to pK^-K^+n was completed using the CLAS detector at Jefferson Lab. A study of the same reaction, published earlier, reported the observation of a narrow \thp resonance. The present experiment, with more than 30 times the integrated luminosity of our earlier measurement, does not show any evidence for a narrow pentaquark resonance. The angle-integrated upper limit on \thp production in the mass range of 1.52 to 1.56 GeV/c2^2 for the γdpKΘ+\gamma d \to pK^-\Theta^+ reaction is 0.3 nb (95% CL). This upper limit depends on assumptions made for the mass and angular distribution of \thp production. Using \lamstar production as an empirical measure of rescattering in the deuteron, the cross section upper limit for the elementary γnKΘ+\gamma n \to K^-\Theta^+ reaction is estimated to be a factor of 10 higher, {\it i.e.}, 3\sim 3 nb (95% CL).Comment: 5 figures, submitted to PRL, revised for referee comment
