Tunable Oscillations in the Purkinje Neuron
In this paper, we study the dynamics of slow oscillations in Purkinje neurons
in vitro, and derive a strong association with a forced parametric oscillator
model. We demonstrate the precise rhythmicity of the oscillations in Purkinje
neurons, as well as a dynamic tunability of this oscillation using a
photo-switchable compound. We show that this slow oscillation can be induced in
every Purkinje neuron, having periods ranging between 10-25 seconds. Starting
from a Hodgkin-Huxley model, we also demonstrate that this oscillation can be
externally modulated, and that the neurons will return to their intrinsic
firing frequency after the forced oscillation is concluded. These results
signify an additional functional role of tunable oscillations within the
cerebellum, as well as a dynamic control of a time scale in the brain in the

Photoswitchable diacylglycerols enable optical control of protein kinase C.
Increased levels of the second messenger lipid diacylglycerol (DAG) induce downstream signaling events including the translocation of C1-domain-containing proteins toward the plasma membrane. Here, we introduce three light-sensitive DAGs, termed PhoDAGs, which feature a photoswitchable acyl chain. The PhoDAGs are inactive in the dark and promote the translocation of proteins that feature C1 domains toward the plasma membrane upon a flash of UV-A light. This effect is quickly reversed after the termination of photostimulation or by irradiation with blue light, permitting the generation of oscillation patterns. Both protein kinase C and Munc13 can thus be put under optical control. PhoDAGs control vesicle release in excitable cells, such as mouse pancreatic islets and hippocampal neurons, and modulate synaptic transmission in Caenorhabditis elegans. As such, the PhoDAGs afford an unprecedented degree of spatiotemporal control and are broadly applicable tools to study DAG signaling
In the shadow of fortress Europe? Impacts of European migration governance on Slovenia, Croatia and Macedonia
This article analyses European integration's effects on migration and border security governance in Slovenia, Croatia and Macedonia in the context of ‘governed interdependence’. We show how transgovernmental networks comprising national and EU actors, plus a range of other participants, blur the distinction between the domestic and international to enable interactions between domestic and international policy elites that transmit EU priorities into national policy. Governments are shown to be ‘willing pupils’ and ‘policy takers’, adapting to EU policy as a pre-condition for membership. This strengthened rather than weakened central state actors, particularly interior ministries. Thus, in a quintessentially ‘national’ policy area, there has been a re-scaling and re-constitution of migration and border security policy. To support this analysis, social network analysis is used to outline the composition of governance networks and analyse interactions and power relations therein
Decreased expression of breast cancer resistance protein in the duodenum in patients with obstructive cholestasis
Background/Aims: The expression of transporters involved in bile acid homeostasis is differentially regulated during obstructive cholestasis. Since the drug efflux transporter breast cancer resistance protein (BCRP) is known to transport bile acids, we investigated whether duodenal BCRP expression could be altered during cholestasis. Methods: Using real-time RT-PCR analysis we determined mRNA expression levels in duodenal tissue of 19 cholestatic patients. Expression levels were compared to 14 healthy subjects. BCRP protein staining was determined in biopsies of 6 cholestatic and 6 healthy subjects by immunohistochemistry. Results: We found that in patients with obstructive cholestasis mean duodenal BCRP mRNA levels were significantly reduced to 53% and mean protein staining was reduced to 57%. Conclusions: BCRP, a transporter for bile acids and numerous drugs, appears to be down-regulated in the human duodenum during cholestasis. Incorporating the effect of weather in construction scheduling and management with sine wave curves: application in the United Kingdom
The impact of (adverse) weather is a common cause of delays, legal claims and economic losses in construction projects. Research has recently been carried out aimed at incorporating the effect of weather in project planning; but these studies have focussed on either a narrow set of weather variables, or a very limited range of construction activities or projects. A method for processing a country’s historical weather data into a set of weather delay maps for some representative standard construction activities is proposed. Namely, sine curves are used to associate daily combinations of weather variables to delay and provide coefficients for expected productivity losses. A complete case study comprising the construction of these maps and the associated sine waves for the UK is presented along with an example of their use in building construction planning. Findings of this study indicate that UK weather extends project durations by an average
of 21%. However, using climatological data derived from weather observations when planning could lead to average reductions in project durations of 16%, with proportional reductions in indirect and overhead costs
Potential energy function for a photo‐switchable lipid molecule
Photo‐switchable lipids are synthetic lipid molecules used in photo‐pharmacology to alter membrane lateral pressure and thus control opening and closing of mechanosensitive ion channels. The molecular picture of how photo‐switchable lipids interact with membranes or ion channels is poorly understood. To facilitate all‐atom simulations that could provide a molecular picture of membranes with photo‐switchable lipids, we derived force field parameters for atomistic computations of the azobenzene‐based fatty acid FAAzo‐4. We implemented a Phyton‐based algorithm to make the optimization of atomic partial charges more efficient. Overall, the parameters we derived give good description of the equilibrium structure, torsional properties, and non‐bonded interactions for the photo‐switchable lipid in its trans and cis intermediate states, and crystal lattice parameters for trans‐FAAzo‐4. These parameters can be extended to all‐atom descriptions of various photo‐switchable lipids that have an azobenzene moiety
Common psychiatric comorbidities in epilepsy: How big of a problem is it?
Psychiatric illness and epilepsy commonly co-occur in adults and in children and adolescents. Theories of comorbidity are complex, but recurring associations between the conditions suggest overlap that is more than simple co-occurrence. Common underlying pathophysiology may imply that epilepsy itself may constituently include psychiatric symptoms. Conditions such as depression or cognitive difficulties commonly occur and in some cases, are considered to be associated with specific epilepsy characteristics such as localization or seizure type. Regardless of etiologic attributions to psychiatric comorbidity, it is clear today that treatment for epilepsy needs to target psychiatric illness. In many cases, quality-of-life improvements depend more upon addressing psychiatric symptoms than seizures themselves
Towards a nationwide implementation of a standardized nutrition and dietetics terminology in clinical practice: a pre-implementation focus group study including a pretest and using the consolidated framework for implementation research
BACKGROUND & AIMS In order to assure high quality of nutrition and dietetic care as well as research, the implementation of a standardized terminology, such as the World Health Organization (WHO) International Classification of Functioning, Disability and Health for Dietetics (ICF-Dietetics) is indispensable. The aim of this study was to explore the clinical practicability and applicability of the ICF-Dietetics in the field of nutrition and dietetic practice prior to the implementation in order to develop criteria (points to consider) for a targeted implementation strategy. METHODS A focus group study including a pretest of the ICF-Dietetics was conducted. Subsequently, facilitators and barriers for a nationwide implementation of the ICF-Dietetics in clinical nutrition and dietetic practice were identified and linked to interventions (combining theory-based and group-based approach) using the Consolidated Framework of Implementation Research (CFIR) to organize and represent data and summarized in a logic model. RESULTS In the pretest 55 clinical documentations which consisted of 248 different ICF-Dietetics categories were received. In four focus groups with 22 health professionals, 66 relevant higher-level themes and implementation strategy criteria (points to consider) were identified. These themes referred to all five domains of the CFIR, namely intervention characteristics, inner setting, outer setting, characteristics of individuals and implementation process and contained important barriers and facilitators that were linked to six implementation objectives as well as six context requirements and five main actors. CONCLUSIONS This study provides facilitators and barriers to be addressed when implementing the ICF-Dietetics in clinical practice and shows potential interventions based on this analysis. A nationwide implementation was mainly seen as a great advantage for enhancing quality and continuity of care and for providing comparable data. However, it requires further refinements and a multifaceted implementation strategy where the engagement of leadership of institutions plays a crucial role. These results have provided a foundation for a targeted implementation strategy to increase the success, reproducibility and comparability
