    Mass spectrometry-based methods for identifying oxidized proteins in disease:advances and challenges

    Many inflammatory diseases have an oxidative aetiology, which leads to oxidative damage to biomolecules, including proteins. It is now increasingly recognized that oxidative post-translational modifications (oxPTMs) of proteins affect cell signalling and behaviour, and can contribute to pathology. Moreover, oxidized proteins have potential as biomarkers for inflammatory diseases. Although many assays for generic protein oxidation and breakdown products of protein oxidation are available, only advanced tandem mass spectrometry approaches have the power to localize specific oxPTMs in identified proteins. While much work has been carried out using untargeted or discovery mass spectrometry approaches, identification of oxPTMs in disease has benefitted from the development of sophisticated targeted or semi-targeted scanning routines, combined with chemical labeling and enrichment approaches. Nevertheless, many potential pitfalls exist which can result in incorrect identifications. This review explains the limitations, advantages and challenges of all of these approaches to detecting oxidatively modified proteins, and provides an update on recent literature in which they have been used to detect and quantify protein oxidation in disease

    Partial least squares (PLS) in Operations Management research: Insights from a systematic literature review

    [EN] Purpose: The present paper aims to analyze the Operations Management (OM) research between 2014 and 2018 that has made use of Partial Least Squares (PLS) to determine whether the trends shown in previous literature reviews focused on this topic are maintained and whether the analyzed papers comply with the recommendations for reporting Design/methodology/approach: A systematic literature review has been carried out of OM articles that use PLS as an analysis tool. A total of 102 references from 45 journals from 2014 to 2018, published in WOS and Scopus, has been analyzed. Bibliometric analysis and a review of the PLS reporting standards applied in the context of OM have been developed. Findings: PLS is gaining importance and is widely adopted in OM as a statistical analysis method of choice. In general, certain aspects of PLS are correctly applied and adequately reported in the publications. However, some shortcomings continue to be observed in terms of their application and the reporting of results. A detailed comparison has been developed between the current research and previous OM research (as well as previous research on other disciplines) on this topic. Research limitations/implications: OM researchers making use of PLS should be aware of the importance of correctly reasoning and justifying their choice and fully reporting the main parameters in order to provide other researchers with useful information and enable them to reproduce the performed analysis. Originality/value: This article builds a study with results based on a greater number of articles and journals than the two previous literature reviews focused on this topic. Therefore, it provides a richer and more up-to-date evaluation of trends in the use and reporting of PLS.     Supply chain integration scales validation and benchmark values

    Purpose: The clarification of the constructs of the supply chain integration (clients, suppliers, external and internal), the creation of a measurement instrument based on a list of items taken from earlier papers, the validation of these scales and a preliminary benchmark to interpret the scales by percentiles based on a set of control variables (size of the plant, country, sector and degree of vertical integration). Design/methodology/approach: Our empirical analysis is based on the HPM project database (2005-2007 timeframe). The international sample is made up of 266 plants across ten countries: Austria, Canada, Finland, Germany, Italy, Japan, Korea, Spain, Sweden and the USA. In each country. We analized the descriptive statistics, internal consistency testing to purify the items (inter-item correlations, Cronbach’s alpha, squared multiple correlation, corrected item-total correlation), exploratory factor analysis, and finally, a confirmatory factor analysis to check the convergent and discriminant validity of the scales. The analyses will be done with the SPSS and EQS programme using the maximum likelihood parameter estimation method. Findings: The four proposed scales show excellent psychometric properties. Research limitations/implications: with a clearer and more concise designation of the supply chain integration measurement scales more reliable and accurate data could be taken to analyse the relations between these constructs with other variables of interest to the academic l fields. Practical implications: providing scales that are valid as a diagnostic tool for best practices, as well as providing a benchmark with which to compare the score for each individual plant against a collection of industrial companies from the machinery, electronics and transportation sectors. Originality/value: supply chain integration may be a major factor in explaining the performance of companies. The results are nevertheless inconclusive, the vast range of results obtained are due, amongst other things, to the fact that there is no exactness to the group of scales used, no-one has yet published an analysis of the measurement models nor clear benchmarks as to the variety of the scales used.Peer Reviewe

    A Triple-A supply chain measurement model: validation and analysis

Design/methodology/approach Following a literature review, Triple-A SC variables (agility, alignment and adaptability) are conceptualized and a list of possible items is created for their measurement. An international 309 plant sample is used to validate the convergent and criterion validities of the composites proposed to measure Triple-A SC. Findings Contributions to the literature: clarification of Triple-A SC variable concepts; identification of key dimensions of Triple-A SC variables; development of a validated Triple-A SC measurement scale for future empirical research and industrial applications. Research limitations/implications A rigorously validated instrument is needed to measure Triple-A SC variables and enable researchers to credibly test theories regarding causal links between capabilities, practices and performance. Practical implications Proposal of a scale for use by managers of different functions to analyze Triple-A SC deployment in the company. Originality/value The only Triple-A SC scale used in the previous literature has serious limitations: scales were not taken from an extended literature review; data were collected from single respondents in a single country.     Evaluación de un secadero solar tendalero túnel: estudio de secado de manzanas.

    En la selección de un secadero solar se debe tener en cuenta las características del producto a secar, la ubicación geográfica del emprendimiento y el factor económico. A nivel nacional no existe una norma para evaluar la performance de un sistema de secado. En el presente trabajo se evalúa un secadero solar tipo túnel con convección forzada Está formado por dos partes: un colector solar y a continuación la cámara de secado. El producto utilizado fue manzana tipo comercial (61,7 kg), cortadas en rodajas, tratadas con metabisulfito de sodio para evitar el pardeado. La circulación del aire se realiza mediante dos ventiladores.     El presente proyecto de investigación, cuyo título fue “Aplicación de la gestión de almacén para aumentar la productividad del despacho en inversiones benjamín cristhoper S.A.C, lima 2022”, se plantea como objetivo general el determinar como la gestión de almacén mejora la productividad del despacho en Inversiones Benjamín Cristhoper S.A.C, Lima 2022. Se busca dar respuesta al problema ¿De qué manera la gestión de almacén aumenta la productividad del despacho en Inversiones Benjamín Cristhoper S.A.C. Lima 2022? El proyecto se elaboró bajo un diseño experimental de tipo aplicada, con un enfoque cuantitativo, la población fue conformada por los despachos diarios y las entradas de productos dentro del área de almacén de congelado, la muestra fue no probabilística. La técnica utilizada fue la observación directa y el análisis documental y como instrumentos se utilizó las fichas de recolección de datos y registros. Se concluyó que: La productividad del pre test, 71.10%, es menor a la productividad post test, 92.60%; por lo que se rechaza la hipótesis nula y se acepta la hipótesis del investigador, siendo esto probado al realizar la prueba Z de Wilcoxon cuya significancia de 0.012 es menor a 0.05

    Estudi dels patrons oscil·latoris de l’escorça prefrontal medial en la gestió de l’estrès

    Aquesta memòria recull el treball realitzat per Na Aina Luque i Garcia al Departament d’Anatomia i Embriologia Humana de la Facultat de Medicina de la Universitat de València sota la direcció del Dr. Vicent Teruel i Martí i la Dra. Joana Martínez i Ricós, entre els anys 2011 i 2016. El primer objectiu d’aquest treball ha estat caracteritzar el patró d’activitat neuronal oscil·latòria de l’escorça prefrontal medial en una situació d’estrès. Per a la qual cosa s’ha estudiat l’activitat poblacional d’aquesta àrea cortical després de portar a terme l’activació del nucli central de l’amígdala. A més, donat que la serotonina està implicada tant en la generació de la resposta d’estrès com en el seu control, hem analitzat el paper que podria jugar el nucli dorsal del rafe en aquest procés. En segon lloc hem estudiat l’efecte que provoca l’aplicació de l’estimulació cerebral profunda del nucli accumbens en l’activitat elèctrica de l’escorça prefrontal medial, per tractar de comparar-lo amb allò observat en la primera part de la Tesi. L’objectiu d’aquesta segona part és avançar en la comprensió dels mecanismes pels quals aquesta tècnica té efecte terapèutic en malalties psiquiàtriques amb components d’estrès. En la primera secció de la tesi s’ofereix una Introducció General en la qual exposem els antecedents sobre l’estrès i el ciruit neural responsable de la seua gestió, així com les dades més recents presents en la bibliografia sobre estimulació cerebral profunda. A continuació, exposem les Hipòtesis del treball, que es concreten en forma d’Objectius. En la secció de Material i Mètodes describim el disseny de cada experiment, així com les tècniques i anàlisis utilitzats. En la secció de Resultats presentem les dades obtingudes en cadascun dels experiments, i a la de Discussió General es discuteixen de manera conjunta aquests resultats en relació la bibliografia existent en aquesta temàtica. Per finalitzar, s’apunten les Conclusions fonamentals derivades de la Tesi, i es detalla la Bibliografia completa utilitzada per a la preparació dels experiments i la discussió dels resultats

    Tumor suppressor ARF regulates tissue microenvironment and tumor growth through modulation of macrophage polarization

    Tumor microenvironment has been described to play a key role in tumor growth, progression, and metastasis. Macrophages are a major cellular constituent of the tumor stroma, and particularly tumor associated macrophages (TAMs or M2-like macrophages) exert important immunosuppressive activity and a pro-tumoral role within the tumor microenvironment. Alternative-reading frame (ARF) gene is widely inactivated in human cancer. We have previously demonstrated that ARF deficiency severely impairs inflammatory response establishing a new role for ARF in the regulation of innate immunity. On the basis of these observations, we hypothesized that ARF may also regulates tumor growth through recruitment and modulation of the macrophage phenotype in the tumor microenvironment. Xenograft assays of B16F10 melanoma cells into ARF-deficient mice resulted in increased tumor growth compared to those implanted in WT control mice. Tumors from ARF-deficient mice exhibited significantly increased number of TAMs as well as microvascular density. Transwell assays showed crosstalk between tumor cells and macrophages. On the one hand, ARF-deficient macrophages modulate migratory ability of the tumor cells. And on the other, tumor cells promote the skewing of ARF-/- macrophages toward a M2-type polarization. In conclusion, these results demonstrate that ARF deficiency facilitates the infiltration of macrophages into the tumor mass and favors their polarization towards a M2 phenotype, thus promoting tumor angiogenesis and tumor growth.     Integration of Transcriptomics and Metabolomics to Reveal the Molecular Mechanisms Underlying Rhodium Nanoparticles-Based Photodynamic Cancer Therapy

    Rhodium nanoparticles have recently been described as promising photosensitizers due to their low toxicity in the absence of near-infrared irradiation, but their high cytotoxicity when irradiated. Irradiation is usually carried out with a laser source, which allows the treatment to be localized in a specific area, thus avoiding undesirable side effects on healthy tissues. In this study, a multi-omics approach based on the combination of microarray-based transcriptomics and mass spectrometry-based untargeted and targeted metabolomics has provided a global picture of the molecular mechanisms underlying the anti-tumoral effect of rhodium nanoparticle-based photodynamic therapy. The results have shown the ability of these nanoparticles to promote apoptosis by suppressing or promoting anti- and pro-apoptotic factors, respectively, and by affecting the energy machinery of tumor cells, mainly blocking the β-oxidation, which is reflected in the accumulation of free fatty acids and in the decrease in ATP, ADP and NAD+ levels.Depto. de Química AnalíticaFac. de Ciencias QuímicasTRUEMinisterio de Ciencia, Innovaciónpu
