A theoretical estimate for nucleotide sugar demand towards Chinese Hamster Ovary cellular glycosylation
Glycosylation greatly influences the safety and efficacy of many of the highest-selling recombinant therapeutic proteins (rTPs). In order to define optimal cell culture feeding strategies that control rTP glycosylation, it is necessary to know how nucleotide sugars (NSs) are consumed towards host cell and rTP glycosylation. Here, we present a theoretical framework that integrates the reported glycoproteome of CHO cells, the number of N-linked and O-GalNAc glycosylation sites on individual host cell proteins (HCPs), and the carbohydrate content of CHO glycosphingolipids to estimate the demand of NSs towards CHO cell glycosylation. We have identified the most abundant N-linked and O-GalNAc CHO glycoproteins, obtained the weighted frequency of N-linked and O-GalNAc glycosites across the CHO cell proteome, and have derived stoichiometric coefficients for NS consumption towards CHO cell glycosylation. By combining the obtained stoichiometric coefficients with previously reported data for specific growth and productivity of CHO cells, we observe that the demand of NSs towards glycosylation is significant and, thus, is required to better understand the burden of glycosylation on cellular metabolism. The estimated demand of NSs towards CHO cell glycosylation can be used to rationally design feeding strategies that ensure optimal and consistent rTP glycosylation

El ensayo propone un marco de análisis de las nuevas formas de operación del poder como dispositivos de producción afectiva y su operación a través del paradigma del pancoreográfico, o conjunto de dispositivos de distribución global de coreografías estándar. Plantea una redefinición de la corporalidad como campo de fuerzas relacionales en movimiento y la posibilidad de desarrollar resistencias a los mecanismos de estandarización de cuerpos del afectocapital, a través de nuevas tecnologías del movimiento a través del paradigma del cuerpo pos-anatómico.This essay proposes a frame of analysis of the new forms of power operating, as the devices of affective production trhough the paradygm of panchoreographics, or the sum of all the devices of global distribution of standard choreographies. El modo de vida actual fomenta la instantaneidad y la prisa. En el periodismo, ligado por definición a la noticia y la actualidad, este apresuramiento se hace especialmente palmario, en detrimento, demasiadas veces, del análisis y la reflexión. El periodismo cultural, por la naturaleza de su propio objeto (la cultura, el conocimiento, las artes), aparece como un reducto de libertad de pensamiento y expresión en que la servidumbre a la inmediatez es menor, pero con demasiada frecuencia la industria cultural pesa más que la cultura a secas, y el criterio de oportunidad comercial se impone. El presente artículo analiza esta situación y aboga por valores como la reflexión, la elaboración y la vocación minoritarias como antídoto contra la mercantilización y banalización de la prensa cultural
Undoing Anatomy: Resisting Global Choreographies in the Capitalism of Affects
Underlying the imaginaries staged and projected in texts, theater pieces, video, cinema and publicity there are certain anatomic constructions of the senses that are the conditions of possibility for those imaginaries. Such sensory anatomies underlie implicit power mechanisms in late capitalist societies, which operate at the level of the production of affect and desire of consumers as well as of the concealment of global violence through which the economic system operates. I will propose a framework of analysis of such power operations through the analysis of movement, and more precisely of what I will call the panchoreographic: a set of technological devices of global distribution that disseminate discrete, standard choreographies in bodies, thus contributing to the production and dissemination of standard affects. Finally, I will show some examples of the work developed by REVERSO in recent years that aims at the radical redefinition of the sensory anatomies that underlie media culture and information society, aiming at the production of a post-anatomical relational body
Efficient processing of an antigenic sequence for presentation by MHC class I molecules depends on its neighboring residues in the protein
Processing of endogenously synthesized proteins generates short peptides that are presented by MHC class I molecules to CD8 T lymphocytes. Here it is documented that not only the sequence of the presented peptide but also the residues by which it is flanked in the protein determine the efficiency of processing and presentation. This became evident when a viral sequence of proven antigenicity was inserted at different positions into an unrelated carrier protein. Not different peptides, but different amounts of the antigenic insert itself were retrieved by isolation of naturally processed peptides from cells expressing the different chimeric proteins. Low yield of antigenic peptide from an unfavorable integration site could be overcome by flanking the insert with oligo-alanine to space it from disruptive neighboring sequences. Notably, the degree of protection against lethal virus disease related directly to the amount of naturally processed antigenic peptide
Analysis of the Machining Process of Titanium Ti6Al-4V Parts Manufactured by Wire Arc Additive Manufacturing (WAAM)
In the current days, the new range of machine tools allows the production of titanium
alloy parts for the aeronautical sector through additive technologies. The quality of the materials
produced is being studied extensively by the research community. This new manufacturing
paradigm also opens important challenges such as the definition and analysis of the optimal
strategies for finishing-oriented machining in this type of part. Researchers in both materials and
manufacturing processes are making numerous advances in this field. This article discusses the
analysis of the production and subsequent machining in the quality of TI6Al4V produced by Wire
Arc Additive Manufacturing (WAAM), more specifically Plasma Arc Welding (PAW). The
