Molecular cloning of ion channels in Felis catus that are related to periodic paralyses in man: a contribution to the understanding of the genetic susceptibility to feline neck ventroflexion and paralysis

Neck ventroflexion in cats has different causes; however, the most common is the hypokalemia associated with flaccid paralysis secondary to chronic renal failure. in humans, the most common causes of acute flaccid paralysis are hypokalemia precipitated by thyrotoxicosis and familial forms linked to mutations in sodium, potassium, and calcium channel genes. Here, we describe the sequencing and analysis of skeletal muscle ion channels in Felis catus that could be related to periodic paralyses in humans, contributing to the understanding of the genetic susceptibility to feline neck ventroflexion and paralysis. We studied genomic DNA from eleven cats, including five animals that were hyperthyroid with hypokalemia, although only one presented with muscle weakness, and six healthy control domestic cats. We identified the ion channel ortholog genes KCNJ2, KCNJ12, KCNJ14, CACNA1S and SCN4A in the Felis catus genome, together with several polymorphic variants. Upon comparative alignment with other genomes, we found that Felis catus provides evidence for a high genomic conservation of ion channel sequences. Although we hypothesized that neck ventroflexion in cats could be associated with a thyrotoxic or familial periodic paralysis channel mutation, we did not identify any previously detected human channel mutation in the hyperthyroid cat presenting hypokalemia. However, based on the small number of affected cats in this study, we cannot yet rule out this molecular mechanism. Notwithstanding, hyperthyroidism should still be considered as a differential diagnosis in hypokalemic feline paralysis.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Universidade Federal de São Paulo, Escola Paulista Med, Dept Med, Div Endocrinol,Lab Mol & Translat Endocrinol, BR-04039032 São Paulo, BrazilUniversidade Federal de São Paulo, Escola Paulista Med, Dept Med, Div Endocrinol,Lab Mol & Translat Endocrinol, BR-04039032 São Paulo, BrazilFAPESP: 2011/20747-8FAPESP: 2012/02529Web of Scienc

Probing the role of grain boundaries in single Cu nanoparticle oxidation by in situ plasmonic scattering

Grain boundaries determine physical properties of bulk materials including ductility, diffusivity, and electrical conductivity. However, the role of grain boundaries in nanostructures and nanoparticles is much less understood, despite the wide application of nanoparticles in nanophotonics, nanoelectronics, and heterogeneous catalysis. Here, we investigate the role of high-angle grain boundaries in the oxidation of Cu nanoparticles, using a combination of in situ single particle plasmonic nanoimaging and postmortem transmission electron microscopy image analysis, together with ab initio and classical electromagnetic calculations. We find an initial growth of a 5-nm-thick Cu2O shell on all nanoparticles, irrespective of different grain morphologies. This insensitivity of the Cu2O shell on the grain morphology is rationalized by extraction of Cu atoms from the metal lattice being the rate limiting step, as proposed by density functional theory calculations. Furthermore, we find that the change in optical scattering intensity measured from the individual particles can be deconvoluted into one contribution from the oxide layer growth and one contribution that is directly proportional to the grain boundary density. The latter contribution signals accumulation of Cu vacancies at the grain boundaries, which, as corroborated by calculations of the optical scattering, leads to increased absorption losses and thus a decrease of the scattering, thereby manifesting the role of grain boundaries as vacancy sinks and nuclei for Kirkendall void formation at a later stage of the oxidation process

Through the diffusion techniques in culture media with paper disc and stainless steel cylinders the antimicrobial activity in 123 pharmaceutical preparations was evaluated: nystatin (43 samples of vaginal cream), erythromycin (14 samples of tablets and 9 of oral suspension), gentamicin (33 samples of injectable liquid) and neomycin (24 samples of cream of topic use), against the microorganisms: Saccharomyces cerevisiae ATCC 2601 to nystatin; Staphylococcus epidermidis12228 to gentamicin and neomycin, and Micrococcus luteusATCC 9341 to erythromycin. The preparation of the test organisms, standard solution, test solutions and culture media in both methods followed the recomendations of Brazilian Pharmacopoeia IV (1988) and United States Pharmacopoeia 24 (2000). The results was statically analysed by tests of Fisher (p=0,05) and Student (p=0,05). Among the 43 samples of vaginal cream nystatin analysed, checked out by the Student's test, differences of antimicrobial activity between methods of disc and cylinder were observed. However, the Fisher's test does not presented difference. The samples of erythromycin, gentamicin and neomycin did not show any difference when submitted to Fisher and Student tests in conclusion, to the erythromycin, gentamicin, neomycin and nystatin, the analysis of antimicrobial activity by the technique of paper discs can substitute the technique of the cylinders of stainless steel.

Latin America does not research on the problems of digital divide related to gender. This was one of the findings in the research made at Universidade Estadual Paulista, which included the most cited literature in English, Spanish and Portuguese in the domain of the digital divide. Women's lack of access to digital information technology or their inability to benefit from it, due to their gender, define a type of exclusion that is being studied in developed countries, but Latin America is neglected. To contribute to the development of gender research in librarian and information sciences, this study compiled the most relevant works about gender digital exclusion, identifying the main problems they refer to, articulating them in a conceptual map that allows to perceive the challenges and opportunities of research that they imply to the region. The target is to break academic silence in librarian and information sciences, and spread out what we know about gender digital exclusion, articulating it comprehensively to boost new projects, valuing female exclusion in science and technology, and specifically in information and communication technology, as a central problem of the research agenda of librarian and information sciences, of all Social Sciences, and of Latin American education and research institutions
