Characterization of Zika virus endocytic pathways in human glioblastoma cells
Zika virus (ZIKV) infections can cause microcephaly and neurological disorders. However, the early infection events of ZIKV in neural cells remain to be characterized. Here, by using a combination of pharmacological and molecular approaches and the human glioblastoma cell T98G as a model, we first observed that ZIKV infection was inhibited by chloroquine and NH4Cl, indicating a requirement of low intracellular pH. We further showed that dynamin is required as the ZIKV entry was affected by the specific inhibitor dynasore, small interfering RNA (siRNA) knockdown of dynamin, or by expressing the dominant-negative K44A mutant. Moreover, the ZIKV entry was significantly inhibited by chlorpromazine, pitstop2, or siRNA knockdown of clathrin heavy chain, indicating an involvement of clathrin-mediated endocytosis. In addition, genistein treatment, siRNA knockdown of caveolin-1, or overexpression of a dominant-negative caveolin mutant impacted the ZIKV entry, with ZIKV particles being observed to colocalize with caveolin-1, implying that caveola endocytosis can also be involved. Furthermore, we found that the endocytosis of ZIKV is dependent on membrane cholesterol, microtubules, and actin cytoskeleton. Importantly, ZIKV infection was inhibited by silencing of Rab5 and Rab7, while confocal microscopy showed that ZIKV particles localized in Rab5- and Rab7-postive endosomes. These results indicated that, after internalization, ZIKV likely moves to Rab5-positive early endosome and Rab7-positive late endosomes before delivering its RNA into the cytoplasm. The role of TG2 in regulating S100A4-mediated mammary tumour cell migration
The importance of S100A4, a Ca2+-binding protein, in mediating tumour cell migration, both intracellularly and extracellularly, is well documented. Tissue transglutaminase (TG2) a Ca2+-dependent protein crosslinking enzyme, has also been shown to enhance cell migration. Here by using the well characterised non-metastatic rat mammary R37 cells (transfected with empty vector) and highly metastatic KP1 cells (R37 cells transfected with S100A4), we demonstrate that inhibition of TG2 either by TG2 inhibitors or transfection of cells with TG2 shRNA block S100A4-accelerated cell migration in the KP1cells and in R37 cells treated with exogenous S100A4. Cell migration was also blocked by the treatment with the non-cell permeabilizing TG2 inhibitor R294, in the human breast cancer cell line MDA-MB-231 (Clone 16, which has a high level of TG2 expression). Inhibition was paralleled by a decrease in S100A4 polymer formation. co-immunoprecipitation and Far Western blotting assays and cross-linking assays showed not only the direct interaction between TG2 and S100A4, but also confirmed S100A4 as a substrate for TG2. Using specific functional blocking antibodies, a targeting peptide and a recombinant protein as a competitive treatment, we revealed the involvement of syndecan-4 and a5Ă1 integrin co-signalling pathways linked by activation of PKCa in this TG2 and S100A4-mediated cell migration. We propose a mechanism for TG2-regulated S100A4-related mediated cell migration, which is dependent on TG2 crosslinking
Causality and defect formation in the dynamics of an engineered quantum phase transition in a coupled binary Bose-Einstein condensate
Continuous phase transitions occur in a wide range of physical systems, and
provide a context for the study of non-equilibrium dynamics and the formation
of topological defects. The Kibble-Zurek (KZ) mechanism predicts the scaling of
the resulting density of defects as a function of the quench rate through a
critical point, and this can provide an estimate of the critical exponents of a
phase transition. In this work we extend our previous study of the
miscible-immiscible phase transition of a binary Bose-Einstein condensate (BEC)
composed of two hyperfine states in which the spin dynamics are confined to one
dimension [J. Sabbatini et al., Phys. Rev. Lett. 107, 230402 (2011)]. The
transition is engineered by controlling a Hamiltonian quench of the coupling
amplitude of the two hyperfine states, and results in the formation of a random
pattern of spatial domains. Using the numerical truncated Wigner phase space
method, we show that in a ring BEC the number of domains formed in the phase
transitions scales as predicted by the KZ theory. We also consider the same
experiment performed with a harmonically trapped BEC, and investigate how the
density inhomogeneity modifies the dynamics of the phase transition and the KZ
scaling law for the number of domains. We then make use of the symmetry between
inhomogeneous phase transitions in anisotropic systems, and an inhomogeneous
quench in a homogeneous system, to engineer coupling quenches that allow us to
quantify several aspects of inhomogeneous phase transitions. In particular, we
quantify the effect of causality in the propagation of the phase transition
front on the resulting formation of domain walls, and find indications that the
density of defects is determined during the impulse to adiabatic transition

Muscarinic Agonist-Mediated Heterologous Desensitization in Isolated Ileum Requires Activation of Both Muscarinic M2 and M3 Receptors
We investigated the subtypes of the muscarinic receptor mediating short-term heterologous desensitization in the isolated ileum. Treatment of the ileum from C57BL/6 mice with acetylcholine (30 ΌM) for 20 min caused a subsequent decrease in contractile sensitivity to both prostaglandin F2α (PGF2α) and the muscarinic agonist, oxotremorine-M. This subsensitivity was characterized by 7- and 3-fold increases in the EC50 values of the agonists, respectively, with no significant effect on the maximal response. The subsensitivity to PGF2α was prevented in both M2 and M3 muscarinic receptor knockout mice. Similarly, the subsensitivity to oxotremorine-M was prevented in M2 knockout mice. Acetylcholine-mediated desensitization of histamine-induced contractions in the guinea pig ileum was inhibited by both M2- and M3-selective muscarinic antagonists with high potency, although careful analysis of the data suggested behavior more consistent with an M2 antagonistic profile. Modeling studies showed that the competitive antagonism of response contingent upon activation of two receptor subtypes should exhibit a pharmacological profile similar to that of the least sensitive signaling pathway. Our results demonstrate that muscarinic agonist-mediated short-term heterologous desensitization of intestinal smooth muscle is contingent upon activation of both M2 and M3 muscarinic receptors and that activation of either receptor by itself is insufficient to cause desensitization

Clinical effectiveness of a manual based coping strategy programme (START, STrAtegies for RelaTives) in promoting the mental health of carers of family members with dementia: pragmatic randomised controlled trial
Objective
To assess whether a manual based coping strategy compared with treatment as usual reduces depression and anxiety symptoms in carers of family members with dementia.
Design
Randomised, parallel group, superiority trial.
Setting
Three mental health community services and one neurological outpatient dementia service in London and Essex, UK.
Participants
260 carers of family members with dementia.
Intervention
A manual based coping intervention comprising eight sessions and delivered by supervised psychology graduates to carers of family members with dementia. The programme consisted of psychoeducation about dementia, carersâ stress, and where to get emotional support; understanding behaviours of the family member being cared for, and behavioural management techniques; changing unhelpful thoughts; promoting acceptance; assertive communication; relaxation; planning for the future; increasing pleasant activities; and maintaining skills learnt. Carers practised these techniques at home, using the manual and relaxation CDs.
Main outcome measures
Affective symptoms (hospital anxiety and depression total score) at four and eight months. Secondary outcomes were depression and anxiety caseness on the hospital anxiety and depression scale; quality of life of both the carer (health status questionnaire, mental health) and the recipient of care (quality of life-Alzheimerâs disease); and potentially abusive behaviour by the carer towards the recipient of care (modified conflict tactics scale).
Results
260 carers were recruited; 173 were randomised to the intervention and 87 to treatment as usual. Mean total scores on the hospital anxiety and depression scale were lower in the intervention group than in the treatment as usual group over the eight month evaluation period: adjusted difference in means â1.80 points (95% confidence interval â3.29 to â0.31; P=0.02) and absolute difference in means â2.0 points. Carers in the intervention group were less likely to have case level depression (odds ratio 0.24, 95% confidence interval 0.07 to 0.76) and there was a non-significant trend towards reduced case level anxiety (0.30, 0.08 to 1.05). Carersâ quality of life was higher in the intervention group (difference in means 4.09, 95% confidence interval 0.34 to 7.83) but not for the recipient of care (difference in means 0.59, â0.72 to 1.89). Carers in the intervention group reported less abusive behaviour towards the recipient of care compared with those in the treatment as usual group (odds ratio 0.47, 95% confidence interval 0.18 to 1.23), although this was not significant.
Conclusions
A manual based coping strategy was effective in reducing affective symptoms and case level depression in carers of family members with dementia. The carersâ quality of life also improved.

Herschel observations of embedded protostellar clusters in the Rosette Molecular Cloud
The Herschel OB young stellar objects survey (HOBYS) has observed the Rosette
molecular cloud, providing an unprecedented view of its star formation
activity. These new far-infrared data reveal a population of compact young
stellar objects whose physical properties we aim to characterise. We compiled a
sample of protostars and their spectral energy distributions that covers the
near-infrared to submillimetre wavelength range. These were used to constrain
key properties in the protostellar evolution, bolometric luminosity, and
envelope mass and to build an evolutionary diagram. Several clusters are
distinguished including the cloud centre, the embedded clusters in the vicinity
of luminous infrared sources, and the interaction region. The analysed
protostellar population in Rosette ranges from 0.1 to about 15 Msun with
luminosities between 1 and 150 Lsun, which extends the evolutionary diagram
from low-mass protostars into the high-mass regime. Some sources lack
counterparts at near- to mid-infrared wavelengths, indicating extreme youth.
The central cluster and the Phelps & Lada 7 cluster appear less evolved than
the remainder of the analysed protostellar population. For the central cluster,
we find indications that about 25% of the protostars classified as Class I from
near- to mid-infrared data are actually candidate Class 0 objects. As a
showcase for protostellar evolution, we analysed four protostars of low- to
intermediate-mass in a single dense core, and they represent different
evolutionary stages from Class 0 to Class I. Their mid- to far-infrared
spectral slopes flatten towards the Class I stage, and the 160 to 70um flux
ratio is greatest for the presumed Class 0 source. This shows that the Herschel
observations characterise the earliest stages of protostellar evolution in

Identification and characterization of an irreversible inhibitor of CDK2
Irreversible inhibitors that modify cysteine or lysine
residues within a protein kinase ATP binding site offer, through their distinctive mode of action, an alternative to ATP-competitive agents. 4-((6-(Cyclohexylmethoxy)-
9H-purin-2-yl)amino)benzenesulfonamide (NU6102) is a potent and selective ATP-competitive inhibitor of CDK2 in which the sulfonamide moiety is positioned close to a pair of lysine residues. Guided by the CDK2/NU6102 structure, we designed 6-(cyclohexylmethoxy)-N-(4-(vinylsulfonyl)phenyl)-9H-purin-2-amine (NU6300), which binds covalently to CDK2 as shown by a co-complex crystal
structure. Acute incubation with NU6300 produced a durable inhibition of Rb phosphorylation in SKUT-1B cells, consistent with it acting as an irreversible CDK2 inhibitor. NU6300 is the first covalent CDK2 inhibitor to be described, and illustrates the potential of vinyl sulfones for the design of more potent and selective compounds
Liquid racism and the Danish Prophet Muhammad cartoons

Copyright @ 2010 The Author.This article examines reactions to the October 2005 publication of the Prophet Muhammad cartoons in the Danish newspaper Jyllands-Posten. It does so by using the concept of âliquid racismâ. While the controversy arose because it is considered blasphemous by many Muslims to create images of the Prophet Muhammad, the article argues that the meaning of the cartoons is multidimensional, that their analysis is significantly more complex than most commentators acknowledge, and that this complexity can best be addressed via the concept of liquid racism. The article examines the liquidity of the cartoons in relation to four readings. These see the cartoons as: (1) a criticism of Islamic fundamentalism; (2) blasphemous images; (3) Islamophobic and racist; and (4) satire and a defence of freedom of speech. Finally, the relationship between postmodernity and the rise of fundamentalism is discussed because the cartoons, reactions to them, and Islamic fundamentalism, all contain an important postmodern dimension.ESR
