    PENGUKURAN TINGKAT PENGETAHUAN DAN KEPEDULIAN TERHADAP PENERAPAN GREEN COMPUTING (STUDI KASUS: FAKULTAS TEKNIK UNIVERSITAS PASUNDAN)

    Penerapan green computing bertujuan untuk mencegah pemborosan sumber daya alam dan energi, agar pengguna teknologi paham mengenai penggunaan teknologi yang efisen dan ramah lingkungan. Seperti halnya di FT-UNPAS pengetahuan mengenai green computing sepertinya belum sepenuhnya diketahui oleh para penggunanya. Karena pentingnya pengetahuan mengenai green computing ini maka penulis merasa perlu dilakukan penelitian mengenai tingkatan seberapa jauh pengguna teknologi mengetahui mengenai green computing dan juga kepedulian pengguna teknologi tersebut terhadap penerapan green computing. Pengguna teknologi di FT-UNPAS diantaranya adalah Mahasiswa, Dosen dan Karyawan. Tugas akhir ini bertujuan untuk mengetahui sejauh mana tingkat pengetahuan dan kepedulian pengguna teknologi komputer di FT-UNPAS terhadap penerapan green computing dalam penggunaan teknologi komputer. Pengukuran menggunakan kuesioner dengan item instrumen yang dibuat berdasarkan teori Taksonomi Bloom. Dalam tugas akhir ini juga, membandingkan tingkat pengetahuan dan kesadaran karakteristik dari masing-masing responden (mahasiswa, dosen dan karyawan) dan mengetahui hubungan antara tingkat pengetahuan dan kepedulian penerapan green computing. Hasil akhir dari tugas akhir ini adalah dapat mengetahui tingkat pengetahuan dan kepedulian pengguna teknologi komputer di FT-UNPAS dan seberapa besar pengaruh tingkat pengetahuan terhadap kepedulian penerapan green computing.     Partial agonism improves the anti-hyperglycaemic efficacy of an oxyntomodulin-derived GLP-1R/GCGR co-agonist

    OBJECTIVE: Glucagon-like peptide-1 and glucagon receptor (GLP-1R/GCGR) co-agonism can maximise weight loss and improve glycaemic control in type 2 diabetes and obesity. In this study we investigated the cellular and metabolic effects of modulating the balance between G protein and β-arrestin-2 recruitment at GLP-1R and GCGR using oxyntomodulin (OXM)-derived co-agonists. This strategy has been previously shown to improve the duration of action of GLP-1R mono-agonists by reducing target desensitisation and downregulation. METHODS: Dipeptidyl dipeptidase-4 (DPP-4)-resistant OXM analogues were generated and assessed for a variety of cellular readouts. Molecular dynamic simulations were used to gain insights into the molecular interactions involved. In vivo studies were performed in mice to identify effects on glucose homeostasis and weight loss. RESULTS: Ligand-specific reductions in β-arrestin-2 recruitment were associated with slower GLP-1R internalisation and prolonged glucose-lowering action in vivo. The putative benefits of GCGR agonism were retained, with equivalent weight loss compared to the GLP-1R mono-agonist liraglutide in spite of a lesser degree of food intake suppression. The compounds tested showed only a minor degree of biased agonism between G protein and β-arrestin-2 recruitment at both receptors and were best classified as partial agonists for the two pathways measured. CONCLUSIONS: Diminishing β-arrestin-2 recruitment may be an effective way to increase the therapeutic efficacy of GLP-1R/GCGR co-agonists. These benefits can be achieved by partial rather than biased agonism

    Compound-based Chinese medicine formula: From discovery to compatibility mechanism.

    ETHNOPHARMACOLOGICAL RELEVANCE: Chinese medicine formula (CMF) has a long history of clinical use in the treatment of various diseases under the guidance of traditional Chinese medicine (TCM) theory. The application of CMF can be divided into three levels, crude extracts, homologous compounds mixture, and specific compounds. However, the modern scientific connotation of the CMF theory has not been clarified. AIM OF THE REVIEW: To critically evaluate the research strategy for the investigation of compound-based CMF (CCMF). MATERIALS AND METHODS: The related information was collected from the scientific databases, including CNKI, Elsevier, ScienceDirect, PubMed, SpringerLink, Web of Science, and Wiley Online. RESULTS: The research design including discovery, screening, optimization, pharmacodynamics models, and target research techniques including the targets for compatibility compounds were evaluated. Essentially it has been evaluated that the in vitro multicellular three-dimensional culture or organoid model has been proposed for the optimization model for compatibility research of CCMF. Based on these, the traditional compatibility theory of CMF, such as Monarch-Minister-Assistant-Guide (Jun-Chen-Zuo-Shi in Chinese), can probably be elucidated by the CCMF research. CONCLUSIONS: CCMF has the clear advantage of providing the exact composition and controllable quality of modern medicines, in addition to having the characteristics of multi-ingredients and multi-targets synergistic effects of TCM. However, CCMF is still associated with challenges which need to be addressed for its future use

    This research is to describe a process that occurs in structuring the regional government. In the environment of the City Government of Palangka Raya which currently applies government regulation Number 18/2016 concerning regional devices as well as supporting factors and inhibitors. Indicators in organizational arrangement have two roles, efficiency and effectiveness. In this study the authors used qualitative research methods with emphasis to the Regional Secretariat of the City of Palangka Raya where the existence of the Organization Section was discussed. The technique of collecting data uses observation, interviews, and documentation. Based on the results of the study (1) Structuring Regional organizations based on government regulation Number18/2016 is still not going well, this is analyzed from indicators of the regional organizational arrangement process, namely Environment, Strategy and Technology as government implementation so that public services can be implemented regions, which are determined by the decision of the Mayor of Palangka Raya in order to realize a regional apparatus institution in accordance with government regulation Number 18/2016. (2) Supporting factors: Facilities and Infrastructure and inhibiting factors: HR and number of employees (3) Suggestion: must always evaluating and reviewing its institutions, not increasing the number of civil servants who are not functional and contract workers, and to better utilize the potential of the organizational section as apparatus resources need to be placed.     Dalam pengelolaan sumber daya manusia yang baik harus diperhatikan agar menunjang strategi bisnis yang akan diterapkan karena jika praktik pengelolaan tersebut mendukung strategi bisnis organisasi akan memberikan kontribusi dalam menciptakan keunggulan kompetitif yang merupakan kunci kesuksesan organisasi dalam lingkungan bisnisnya. Oleh karena itu, agar perusahaan memiliki sumber daya manusia yang terampil, memiliki pengetahuan yang luas, perilaku dan sikap yang baik, perusahaan harus memiliki program-program yang mampu mengembangkan dan meningkatkan potensi yang dimiliki oleh setiap karyawan di perusahaan agar prestasi kerja dalam perusahaan meningkat diantaranya melalui desain organisasi dan budaya organisasi. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh desain organisasi dan budaya organisasi terhadap motivasi kerja karyawan pada Maternal Disaster Bandung. Metode yang digunakan oleh penulis adalah metode penelitian deskriptif dan verifikatif. Teknik pengambilan sampel yang digunakan oleh penulis dalam penelitian ini adalah dengan menggunakan teknik simple Probability Sampling. Sampel yang digunakan sebanyak 38 responden. Pengolahan data dilakukan dengan analisis koefisien korelasi, koefisien determinasi, dan pengujian hipotesis. Berdasarkan hasil penelitian dapat disimpulkan bahwa secara bersama (simultan) Desain organisasi dan Budaya organisasi berpengaruh terhadap Motivasi kerja karyawan. Desain organisasi berpengaruh terhadap Motivasi kerja karyawan. Begitu juga Budaya organisasi berpengaruh terhadap Motivasi kerja karyawan

    Construction and Functional Analysis of the Recombinant Bacteriocins Weissellicin-MBF from Weissella confusa MBF8-1

    Background: Bacteriocins (Bac1, Bac2, and Bac3) from Weissella confusa MBF8-1, weissellicin- MBF, have been reported as potential alternative substances as well as complements to the existing antibiotics against many antimicrobial-resistant pathogens. Previously, the genes encoded in the large plasmid, pWcMBF8-1, and the spermicidal activity of their synthetic peptides, originally discovered Indonesia, have been studied. Three synthetic bacteriocins peptides of this weissellicin-MBF have been reported for their potential activities, i.e. antibacterial and spermicidal. Objective: The aim of this study was to construct the recombinant Bacteriocin (r-Bac) genes, as well as to investigate the gene expressions and their functional analysis. Methods: Here, the recombinant Bacteriocin (r-Bac) genes were constructed and the recombinant peptides (r-Bac1, r-Bac2, and r-Bac3) in B. subtilis DB403 cells were produced on a large scale. After purification, using the His-tag affinity column, their potential bioactivities were measured as well as their antibacterial minimum inhibitory concentrations against Leuconostoc mesenteroides and Micrococcus luteus, were determined. Results: Pure His-tag-recombinant Bac1, Bac2, and Bac3 were obtained and they could inhibit the growth of L. mesenteroides and M. luteus. Conclusion: The recombinant bacteriocin could be obtained although with weak activity in inhibiting gram-positive bacterial growth
