    Super-resolution Imaging of the T cell Central Supramolecular Signaling Cluster Using Stimulated Emission Depletion Microscopy

    Supramolecular signaling assemblies are of interest for their unique signaling properties. A µm scale signaling assembly, the central supramolecular signaling cluster (cSMAC), forms at the center interface of T cells activated by antigen presenting cells (APC). The adaptor protein linker for activation of T cells (LAT) is a key cSMAC component. The cSMAC has widely been studied using total internal reflection fluorescence microscopy of CD4(+) T cells activated by planar APC substitutes. Here we provide a protocol to image the cSMAC in its cellular context at the interface between a T cell and an APC. Super resolution stimulated emission depletion microscopy (STED) was utilized to determine the localization of LAT, that of its active, phosphorylated form and its entire pool. Agonist peptide-loaded APCs were incubated with TCR transgenic CD4(+) T cells for 4.5 min before fixation and antibody staining. Fixed cell couples were imaged using a 100x 1.4 NA objective on a Leica SP8 AOBS confocal laser scanning microscope. LAT clustered in multiple supramolecular complexes and their number and size distributions were determined. Using this protocol, cSMAC properties in its cellular context at the interface between a T cell and an APC could be quantified

    The author's procedures and methods were described for compiling General Volume Tables for teak, from which Local Tables can be derived. Only easily and rather accurately measurable characteristics should be used: tree height, length of trunk free of branches and timber-yielding length of trunk and trunk girth at 1.30 metre. Girthes at 1.50 metre and 1.70 metre were also necessary, to avoid abnormal trunk bases. The Table should also contain estimated volumes of whole tree and its parts with bark, and diameter, girths and volumes of the tree without bark at different heights.But more data were needed to construct the Table, primarily the mathematical expression of the trunk form. Two true form quotients are required to represent the trunk form of teak with satisfactory accuracy in a simple equation. Estimation of the form class of an individual tree was only possible by direct measurements, so average form classes were used. These classes for the various trees and stands of one single kind of wood usually varied so little that for any one Table practically only one form class was required. The slight variation in form class of teak can easily be incorporated in the Volume Tables.</p

    Ground and excited state communication within a ruthenium containing benzimidazole metallopolymer

    Emission spectroscopy and electrochemistry has been used to probe the electronic communication between adjacent metal centres and the conjugated backbone within a family of imidazole based metallopolymer, [Ru(bpy)2(PPyBBIM)n]2+, in the ground and excited states, bpy is 2,2’-bipyridyl, PPyBBIM is poly[2-(2-pyridyl)-bibenzimidazole] and n = 3, 10 or 20. Electronic communication in the excited state is not efficient and upon optical excitation dual emission is observed, i.e., both the polymer backbone and the metal centres emit. Coupling the ruthenium moiety to the imidazole backbone results in a red shift of approximately 50 nm in the emission spectrum. Luminescent lifetimes of up to 120 ns were also recorded. Cyclic voltammetry was also utilized to illustrate the distance dependence of the electron hopping rates between adjacent metal centres with ground state communication reduced by up to an order of magnitude compared to previously reported results when the metal to backbone ratio was not altered. DCT and De values of up to 3.96 x 10-10 and 5.32 x 10-10 cm2S-1 were observed with corresponding conductivity values of up to 2.34 x 10-8 Scm-1

    Computational spatiotemporal analysis identifies WAVE2 and cofilin as joint regulators of costimulation-mediated T cell actin dynamics

    Fluorescence microscopy is one of the most important tools in cell biology research and it provides spatial and temporal information to investigate regulatory systems inside cells. This technique can generate data in the form of signal intensities at thousands of positions resolved inside individual live cells; however, given extensive cell-to-cell variation, methods do not currently exist to assemble these data into three- or four-dimensional maps of protein concentration that can be compared across different cells and conditions. Here, we have developed one such method and applied it to investigate actin dynamics in T cell activation. Antigen recognition in T cells by the T cell receptor (TCR) is amplified by engagement of the costimulatory receptor CD28 and we have determined how CD28 modulates actin dynamics. We imaged actin and eight core actin regulators under conditions where CD28 in the context of a strong TCR signal was engaged or blocked to yield over a thousand movies. Our computational analysis identified diminished recruitment of the activator of actin nucleation WAVE2 and the actin severing protein cofilin to F-actin as the dominant difference upon costimulation blockade. Reconstitution of WAVE2 and cofilin activity restored the defect in actin signaling dynamics upon costimulation blockade. Thus we have developed and validated an approach to quantify protein distributions in time and space for analysis of complex regulatory systems

    Water-soluble gold nanoparticles : from catalytic selective nitroarene reduction in water to refractive index sensing

    Water-soluble gold nanoparticles (Au NPs) stabilized by a nitrogen-rich PEG-tagged substrate have been prepared by reduction of HAuClâ‚„ with NaBHâ‚„ in water at room temperature. The morphology and size of the nanoparticles can be controlled by simply varying the gold/stabilizer ratio. The nanoparticles have been fully characterized by TEM, HRTEM, ED, EDS, UV-vis, p-XRD and elemental analysis. The material is efficient as a recyclable catalyst for the selective reduction of nitroarenes with NaBHâ‚„ to yield the corresponding anilines in water at room temperature. Furthermore, the potential ability of the Au NPs as a refractive index sensor due to their localized surface plasmon resonance (LSPR) effect has also been assessed

    PKCθ links proximal T cell and Notch signaling through localized regulation of the actin cytoskeleton

    Notch is a critical regulator of T cell differentiation and is activated through proteolytic cleavage in response to ligand engagement. Using murine myelin-reactive CD4 T cells, we demonstrate that proximal T cell signaling modulates Notch activation by a spatiotemporally constrained mechanism. The protein kinase PKCθ is a critical mediator of signaling by the T cell antigen receptor and the principal costimulatory receptor CD28. PKCθ selectively inactivates the negative regulator of F-actin generation, Coronin 1A, at the center of the T cell interface with the antigen presenting cell (APC). This allows for effective generation of the large actin-based lamellum required for recruitment of the Notch-processing membrane metalloproteinase ADAM10. Such enhancement of Notch activation is critical for efficient T cell proliferation and Th17 differentiation. We reveal a novel mechanism that, through modulation of the cytoskeleton, controls Notch activation at the T cell:APC interface thereby linking T cell receptor and Notch signaling pathways.
