    Feeding behaviour of Lumbriculus variegatus as an ecological indicator of in situ sediment contamination

    Previous studies have demonstrated that the feeding behaviour of Lumbriculus variegatus may be significantly inhibited during exposure to toxic substances. The potential use of an in situ sediment bioassay, using L.variegatus post-exposure feeding inhibition as an endpoint, was investigated. The bioassay consisted of exposing animals in the field for a six-day exposure period and feeding rates were measured immediately afterwards over a twenty-four hour post-exposure period. The bioassay methodology developed in the laboratory produced a consistent baseline response that was reliable and repeatable. Endpoint sensitivity was demonstrated under laboratory conditions, where bioassay organisms exhibited delayed recovery from feeding inhibition after previous exposure to sediment-associated contaminants. The apparent insensitivity of the bioassay to sediment-associated metals means that the technique should only be used as part of a suite of bioassays that employ representative deposit feeders. The ecological relevance of the bioassay endpoint was also demonstrated by comparing short-term measures of post-exposure feeding inhibition with the longer-term effects of a toxicant on L.variegatus populations. The bioassay methodology was successfully adapted for in situ use. Post-exposure feeding inhibition was detected at contaminated field sites. However, the consistent baseline response produced in the laboratory could not be replicated during deployments of the bioassay at upstream (“clean”) field sites. Increased environmental “noise” may have been a result of a number of confounding factors that could limit the sensitivity of the bioassay endpoint if not adequately controlled. Despite the above concerns, the in situ bioassay is suggested to represent a useful tool, which uses a more realistic field exposure scenario to investigate the effects of sediment-associated toxicants with an important functional component of aquatic ecosystems

    Our panel is Local Exchange Service in the Next Century--What Still must be Done to Bring Us to Where We Want to Be? We have four excellent panelists to address these questions. I expect the panelists to agree, in part, where we want to be, and they won\u27t spend a whole lot of time about increased service, new technology, lower prices, and all those things. I am sure we\u27ll hear some assurances along those lines, at least from three of the panelists. Most of the time will be spent on what still must be done to get us there. I hope we will have not just what we, the Commission, has to do to get us there, but also perhaps what they may have to do to get us there. Also, perhaps, some references to some new technologies

    Infrared Diode Laser Spectrometer for the Study of Jet Cooled Gases

    We have constructed a tunable, high resolution, infrared diode laser spectrometer and combined it with a pulsed supersonic jet expansion. A modified version of the Perry multipass cell has been incorporated into the spectrometer to increase the effective absorption path length. Performance capabilities of the spectrometer are evaluated by examining ro-vibrational spectra for the carbon monoxide molecule in the five micron region of the infrared. From these measurements, an instrumental absorption sensitivity is determined. Finally, since one of our immediate goals is the infrared study of jet cooled transition metal carbonyls, we present high resolution data obtained by entraining the vapor above a solid metal carbonyl and injecting it into the pulsed jet expansion

    Polymorphic organization in a planktonic graptoloid (Hemichordata: Pterobranchia) colony of Late Ordovician age

    Graptolites are common fossils in Early Palaeozoic strata, but little is known of their soft-part anatomy. However, we report a long-overlooked specimen of Dicranograptus aff. ramosus from Late Ordovician strata of southern Scotland that preserves a strongly polymorphic, recalcitrant, organic-walled network hitherto unseen in graptoloid graptolites. This network displays three morphologies: proximally, a strap-like pattern, likely of flattened tubes; these transform distally into isolated, hourglass-shaped structures; then, yet more distally, revert to a (simpler) strap-like pattern. The network most likely represents a stolon-like system, hitherto unknown in graptoloids, that connected individual zooids. Its alternative interpretation, as colonial xenobionts that infested a graptoloid colony and mimicked its architecture, is considered less likely on taphonomic and palaeobiological grounds. Such polymorphism is not known in non-graptolite pterobranchs, which are less diverse and morphologically more conservative: a division of labour between graptoloid zooids for such functions as feeding, breeding and rhabdosome construction may have been the key to their remarkable evolutionary success

    Polymerase manager protein UmuD directly regulates Escherichia coli DNA polymerase III α binding to ssDNA

    Replication by Escherichia coli DNA polymerase III is disrupted on encountering DNA damage. Consequently, specialized Y-family DNA polymerases are used to bypass DNA damage. The protein UmuD is extensively involved in modulating cellular responses to DNA damage and may play a role in DNA polymerase exchange for damage tolerance. In the absence of DNA, UmuD interacts with the α subunit of DNA polymerase III at two distinct binding sites, one of which is adjacent to the single-stranded DNA-binding site of α. Here, we use single molecule DNA stretching experiments to demonstrate that UmuD specifically inhibits binding of α to ssDNA. We predict using molecular modeling that UmuD residues D91 and G92 are involved in this interaction and demonstrate that mutation of these residues disrupts the interaction. Our results suggest that competition between UmuD and ssDNA for α binding is a new mechanism for polymerase exchange

    Quantifying forensic investigations involving bloodstain pattern analysis within the UK

    Research informed by practice is more likely to have a greater impact on society. However, forensic cases are confidential and thus, real-life data regarding the details of violent crime is usually withheld from the public and academia. Through a partnership between the authors institution and a UK police service, casefiles from 78 criminal investigations from 2012 to 2020 involving Bloodstain Pattern Analysis (BPA) were examined and quantified. The most common methods of assault and weapons used were identified as well as the frequency of different bloodstain pattern classifications. The results of this study will help inform researchers and supply forensic training providers with data derived from forensic practice. Despite a significant body of literature exploring impact patterns and software for calculating the Area of Origin (AO), impact pattern was classified at only 22% of scenes, with sharp-force trauma being the most prevalent form of assault. This paper recommends a review of the BPA terminology, to include additional commonly encountered patterns that are not defined by the current standard

    An early Cambrian greenhouse climate

    The oceans of the early Cambrian (~541 to 509 million years ago) were the setting for a marked diversification of animal life. However, sea temperatures — a key component of the early Cambrian marine environment — remain unconstrained, in part because of a substantial time gap in the stable oxygen isotope (delta 18 O) record before the evolution of euconodonts. We show that previously overlooked sources of fossil biogenic phosphate have the potential to fill this gap. Pristine phosphatic microfossils from the Comley Limestones, UK, yield a robust delta 18 O signature, suggesting sea surface temperatures of 20° to 25°C at high southern paleolatitudes (~65°S to 70°S) between ~514 and 509 million years ago. These sea temperatures are consistent with the distribution of coeval evaporite and calcrete deposits, peak continental weathering rates, and also our climate model simulations for this interval. Our results support an early Cambrian greenhouse climate comparable to those of the late Mesozoic and early Cenozoic, offering a framework for exploring the interplay between biotic and environmental controls on Cambrian animal diversification
