    Metabolic and haemodynamic effects of oral glucose loading in young healthy men carrying the 825T-allele of the G protein β3 subunit

    BACKGROUND: A C825T polymorphism was recently identified in the gene encoding the β3 subunit of heterotrimeric G-proteins (GNB3). The T-allele is significantly associated with essential hypertension and obesity. In order to further explore a possible pathogenetic link between the T-allele and impaired glucose tolerance we studied metabolic and haemodynamic responses to oral glucose loading in young, healthy subjects with and without the 825T-allele. METHODS: Twelve subjects with and 10 without the 825T-allele were investigated at rest and following glucose ingestion (75 g). Blood glucose, serum insulin and haemodynamics were determined prior to and over 2 hours following glucose ingestion. We non-invasively measured stroke volume (SV, by impedance-cardiography), blood pressure (BP), heart rate (HR), and systolic-time-intervals. Cardiac output (CO) was calculated from HR and SV. Total peripheral resistance was calculated from CO and BP. Metabolic and haemodynamic changes were quantified by maximal responses and by calculation of areas under the concentration time profile (AUC). Significances of differences between subjects with and without the T-allele were determined by unpaired two-tailed t-tests. A p < 0.05 was considered statistically significant. RESULTS: Metabolic and haemodynamic parameters at baseline were very similar between both groups. The presence of the T-allele did not alter the response of any metabolic or haemodynamic parameter to glucose loading. CONCLUSIONS: In conclusion, this study does not support the hypothesis that the C825T polymorphism may serve as a genetic marker of early impaired glucose tolerance

    The First Post-Kepler Brightness Dips of KIC 8462852

    We present a photometric detection of the first brightness dips of the unique variable star KIC 8462852 since the end of the Kepler space mission in 2013 May. Our regular photometric surveillance started in October 2015, and a sequence of dipping began in 2017 May continuing on through the end of 2017, when the star was no longer visible from Earth. We distinguish four main 1-2.5% dips, named "Elsie," "Celeste," "Skara Brae," and "Angkor", which persist on timescales from several days to weeks. Our main results so far are: (i) there are no apparent changes of the stellar spectrum or polarization during the dips; (ii) the multiband photometry of the dips shows differential reddening favoring non-grey extinction. Therefore, our data are inconsistent with dip models that invoke optically thick material, but rather they are in-line with predictions for an occulter consisting primarily of ordinary dust, where much of the material must be optically thin with a size scale <<1um, and may also be consistent with models invoking variations intrinsic to the stellar photosphere. Notably, our data do not place constraints on the color of the longer-term "secular" dimming, which may be caused by independent processes, or probe different regimes of a single process

    Ethnic Context and Immigration Policy Preferences Among Latinos and Anglos

    Objective. We examine how the immigration policy preferences of Anglos and Latinos vary according to ethnic context. Specifically, we hypothesize that immigration policy attitudes are a product of both Latino immigrant and Latino native born group size. In contrast to previous work, which found that Latinos and Anglos react to contextual forces in an identical manner, we argue that Latino group size produces opposite reactions for Anglos and Latinos. Methods. These hypotheses are tested using an original state-wide survey of Anglos and Latinos in Texas conducted during 2006, which is supplemented with data from the 2000 Census. Results. Our findings show that residing in a heavily Latino area produces more liberal immigration attitudes among Latinos and more conservative attitudes among Anglos. However, this result is driven not by the size of the foreign-born Latino population, but by the size of the native-born Latino population. Conclusion. Anti-immigrant sentiments among Anglos appear to result, at least in part, from ethnic concerns

    Efficacy of manidipine/delapril versus losartan/hydrochlorothiazide fixed combinations in patients with hypertension and diabetes

    Background Hypertension markedly increases the already high risk for cardiovascular complications in patients with diabetes mellitus. Less than one in eight patients with hypertension and type 2 diabetes have adequately controlled blood pressure. As a result, antihypertensive combinations are now widely used in management of hypertension associated with diabetes.Methods This double-blind study investigated efficacy of a new fixed dose combination of a calcium antagonist, manidipine 10 mg, and an angiotensin-converting enzyme inhibitor, delapril 30 mg, compared with a combination of an angiotensin receptor blocker, losartan 50 mg, and a diuretic, hydrochlorothiazide 12.5 mg. Patients with hypertension ( blood pressure >= 130/ 80 mmHg) with controlled type 2 diabetes ( HbA1c <= 7.5%) were randomized to manidipine/delapril ( n=153) or losartan/hydrochlorothiazide ( n=161), administered once daily for 12 weeks. Patients underwent ambulatory blood pressure monitor evaluation at baseline and end of treatment.Results Mean decreases in 24- h systolic blood pressure were seen with both manidipine/ delapril ( -9.3mmHg) and losartan/hydrochlorothiazide ( -10.7mmHg) combinations. The mean ( 95% confidence interval) treatment difference was -1.4 ( -4.5/1.8) mmHg, demonstrating noninferiority of the manidipine/ delapril combination. Reduction in 24- h diastolic blood pressure ( -4.6 versus -4.5mmHg) and daytime ( systolic blood pressure -10.5 versus -11.1mmHg) and night-time ( systolic blood pressure -7.1 versus -9.3mmHg) blood pressure were also not significantly different between treatments.     Rapid searches for counterparts of GRB 930131

    A fading counterpart to a gamma-ray burst (GRB) would appear as a point source inside a GRB error region soon after the burst which dims on a timescale from minutes to days. The favorable circumstances of the burst GRB 930131 allowed for an international campaign to search for fading counterparts starting 6.8 hr after the burst. We report observations from many optical sites, two radio telescopes, and archival ROSAT data, including deep Schmidt exposures 35, 44, and 64 hr after the burst. No fading counterparts were detected with our observations

    IL-1β, IL-4 and IL-12 control the fate of group 2 innate lymphoid cells in human airway inflammation in the lungs

    Group 2 innate lymphoid cells (ILC2s) secrete type 2 cytokines, which protect against parasites but can also contribute to a variety of inflammatory airway diseases. We report here that interleukin 1β (IL-1β) directly activated human ILC2s and that IL-12 induced the conversion of these activated ILC2s into interferon-γ (IFN-γ)-producing ILC1s, which was reversed by IL-4. The plasticity of ILCs was manifested in diseased tissues of patients with severe chronic obstructive pulmonary disease (COPD) or chronic rhinosinusitis with nasal polyps (CRSwNP), which displayed IL-12 or IL-4 signatures and the accumulation of ILC1s or ILC2s, respectively. Eosinophils were a major cellular source of IL-4, which revealed cross-talk between IL-5-producing ILC2s and IL-4-producing eosinophils. We propose that IL-12 and IL-4 govern ILC2 functional identity and that their imbalance results in the perpetuation of type 1 or type 2 inflammatio
