    Modelling of natural convection flows with large temperature differences : a benchmark problem for low Mach number solvers. Part 1, Reference solutions

    There are very few reference solutions in the literature on non-Boussinesq natural convection flows. We propose here a test case problem which extends the well-known De Vahl Davis differentially heated square cavity problem to the case of large temperature differences for which the Boussinesq approximation is no longer valid. The paper is split in two parts: in this first part, we propose as yet unpublished reference solutions for cases characterized by a non-dimensional temperature difference of 0.6, (constant property and variable property cases) and (variable property case). These reference solutions were produced after a first international workshop organized by CEA and LIMSI in January 2000, in which the above authors volunteered to produce accurate numerical solutions from which the present reference solutions could be established

    SIgA, TGF-beta 1, IL-10, and TNF alpha in Colostrum Are Associated with Infant Group B Streptococcus Colonization

    Background: Group B Streptococcus (GBS) is a major cause of mortality and morbidity in infants and is associated with transmission from a colonized mother at birth and via infected breastmilk. Although maternal/infant colonization with GBS is common, the majority of infants exposed to GBS remain unaffected. The association between breastmilk immune factors and infant colonization and disease prevention has not been elucidated. Objectives: We have investigated the association between SIgA and cytokines in breastmilk and infant GBS colonization and clearance. Methods: Mother/infant GBS colonization was determined in a prospective cohort of 750 Gambian mother/infant pairs followed to day 89 of life. Anti-GBS secretory IgA bound to the surface of whole bacteria was assessed by flow cytometry and a panel of 12 cytokines quantified by mesoscale discovery in colostrum, breastmilk and serum. Results: Compared with infants receiving low anti-GBS SIgA in colostrum, infants receiving high anti-GBS SIgA were at decreased risk of GBS colonization for serotypes III and V. Infants colonized at day 6 were twice as likely to receive colostrum with high TGF-ÎČ1, TNFα, IL10, and IL-6 compared to uncolonized infants. Infants receiving high colostral TGF-ÎČ1, TNFα, and IL-6 had two-fold enhanced GBS clearance between birth and day 89. Conclusion: Our results suggest that the infant GBS colonization risk diminishes with increasing anti-GBS SIgA antibody in breastmilk and that key maternally derived cytokines might contribute to protection against infant colonization. These findings might be leveraged to develop interventions including maternal vaccination that may reduce infant GBS colonization

    Evaluation of harmful algal bloom outreach activities

Available from MDPI via the link in this record.With an apparent increase of harmful algal blooms (HABs) worldwide, healthcare providers, public health personnel and coastal managers are struggling to provide scientifically-based appropriately-targeted HAB outreach and education. Since 1998, the Florida Poison Information Center-Miami, with its 24 hour/365 day/year free Aquatic Toxins Hotline (1-888-232-8635) available in several languages, has received over 25,000 HAB-related calls. As part of HAB surveillance, all possible cases of HAB-related illness among callers are reported to the Florida Health Department. This pilot study evaluated an automated call processing menu system that allows callers to access bilingual HAB information, and to speak directly with a trained Poison Information Specialist. The majority (68%) of callers reported satisfaction with the information, and many provided specific suggestions for improvement. This pilot study, the first known evaluation of use and satisfaction with HAB educational outreach materials, demonstrated that the automated system provided useful HAB-related information for the majority of callers, and decreased the routine informational call workload for the Poison Information Specialists, allowing them to focus on callers needing immediate assistance and their healthcare providers. These results will lead to improvement of this valuable HAB outreach, education and surveillance tool.     Optical spectrum of proflavine and its ions

    Motivated by possible astrophysical and biological applications we calculate visible and near UV spectral lines of proflavine (C13H11N3, 3,6-diaminoacridine) in vacuum, as well as its anion, cation, and dication. The pseudopotential density functional and time-dependent density functional methods are used. We find a good agreement in spectral line positions calculated by two real-time propagation methods and the Lanczos chain method. Spectra of proflavine and its ions show characteristic UV lines which are good candidates for a detection of these molecules in interstellar space and various biological processes

    Contribution of Renal and Non‐Renal Clearance on Increased Total Clearance of Adalimumab in Glomerular Disease

    The contribution of renal and non‐renal clearance toward targeted concentrations and/or effects of therapeutic proteins in nephrotic patients are unknown. This study dissected the contribution of clearance pathways to adalimumab elimination in patients with focal segmental glomerulosclerosis (FSGS). Urine was collected from seven patients treated with adalimumab. Renal clearance (Cl R ) was measured and non‐renal clearance (Cl NR ) was calculated as the difference between total clearance and Cl R . Differences in cumulative amount in urine, Cl R , and Cl NR between study weeks 1 and 16 and relationships between proteinuria (protein:creatinine ratio (Up/c)), and Cl R and Cl NR were evaluated. Up to 13% of the adalimumab dose was lost in urine. Cl NR contributed more than Cl R to enhanced total clearance. There was a nonlinear relationship between Up/c and Cl R (R 2 0.7059); an increase in Cl R beginning at Up/c of 12 mg/mg [slope 1.755, (C.I. −7.825 to 11.34)]. There was a linear relationship between Up/c and Cl NR (R 2 0.5039); for every one unit increase in Up/c, Cl NR would increase by 3.5 mL/hr ( P  = 0.01). Both Cl R and Cl NR contribute to enhanced total clearance of adalimumab in glomerular disease secondary to FSGS.     Theoretical evaluation of PAH dications properties

    Aims. We present a systematic theoretical study on 40 polycyclic aromatic hydrocarbons dications (PAHs++) containing up to 66 carbon atoms. Methods. We performed our calculations using well established quantum-chemical techniques in the framework of the density functional theory (DFT) to obtain the electronic ground-state properties, and of the time-dependent DFT (TD-DFT) to evaluate the excited\textendash state properties. Results. For each PAH++ considered we computed the absolute visible-UV photo-absorption cross-section up to about 30 eV. We also evaluated their vibrational properties and compared them to those of the corresponding neutral and singly-ionised species. We estimated the adiabatic and vertical second ionisation energy DeltaI through total energy differences. Conclusions. The DeltaI values obtained fall in the energy range 8-13 eV, confirming that PAHs could reach the doubly-ionised state in HI regions. The total integrated IR absorption cross-sections show a marked increase upon ionization, being on the average about two and five times larger for PAHs++ than for PAHs+ and PAHs, respectively. The visible-UV photo-absorption cross-sections for the 0, +1 and +2 charge-states show comparable features but PAHs++ are found to absorb slightly less than their parent neutrals and singly ionized species between ~7 and ~12 eV. Combining these pieces of information we found that PAHs++ should actually be more stable against photodissociation than PAHs and PAHs+,_if_ dissociation tresholds are nearly unchanged by ionization.Comment: 13 pages, 7 figures, accepted for pubblication in A&A (25/09/06

    Tidal propagation in strongly convergent channels

    Simple first‐ and second‐order analytic solutions, which diverge markedly from classical views of cooscillating tides, are derived for tidal propagation in strongly convergent channels. Theoretical predictions compare well with observations from typical examples of shallow, “funnel‐shaped” tidal estuaries. A scaling of the governing equations appropriate to these channels indicates that at first order, gradients in cross‐sectional area dominate velocity gradients in the continuity equation and the friction term dominates acceleration in the momentum equation. Finite amplitude effects, velocity gradients due to wave propagation, and local acceleration enter the equations at second order. Applying this scaling, the first‐order governing equation becomes a first‐order wave equation, which is inconsistent with the presence of a reflected wave. The solution is of constant amplitude and has a phase speed near the frictionless wave speed, like a classical progressive wave, yet velocity leads elevation by 90°, like a classical standing wave. The second‐order solution at the dominant frequency is also a unidirectional wave; however, its amplitude is exponentially modulated. If inertia is finite and convergence is strong, amplitude increases along channel, whereas if inertia is weak and convergence is limited, amplitude decays. Compact solutions for second‐order tidal harmonics quantify the partially canceling effects of (1) time variations in channel depth, which slow the propagation of low water, and (2) time variations in channel width, which slow the propagation of high water. Finally, it is suggested that phase speed, along‐channel amplitude growth, and tidal harmonics in strongly convergent channels are all linked by morphodynamic feedback

    Myocardial dysfunction in the periinfarct and remote regions following anterior infarction in rats quantified by 2D radial strain echocardiography: An observational cohort study

    <p>Abstract</p> <p>Background</p> <p>Heart failure from adverse ventricular remodeling follows myocardial infarction, but the contribution of periinfarct and remote myocardium to the development of cardiomyopathy remains poorly defined. 2D strain echocardiography (2DSE) is a novel and sensitive tool to measure regional myocardial mechanics. The aim is to quantify radial strain in infarcted (I), periinfarct (PI) and remote (R) myocardial regions acutely and chronically following anterior infarction in rats.</p> <p>Methods</p> <p>The left anterior coronary artery of male Sprague-Dawley rats (270–370 g) were occluded for 20–30 minutes and 2DSE was performed in the acute setting (n = 10; baseline and 60 minutes post-reperfusion) and in the chronic setting (n = 14; baseline, 1, 3 and 6 weeks). Using software, radial strain was measured in the mid-ventricle in short axis view. The ventricle was divided into 3 regions: I (anteroseptum, anterior and anterolateral), PI – (inferoseptum and inferolateral) and R – (inferior). Infarct size was measured using triphenyl tetrazolium chloride in the acute group.</p> <p>Results</p> <p>Following infarct, adverse remodeling occurred with progressive increase in left ventricular size, mass and reduced fractional shortening within 6 weeks. Radial strain decreased not only in the infarct but also in the periinfarct and remote regions acutely and chronically (I, PI, R, change vs. baseline, 60 minutes -32.7 ± 8.7, -17.4 ± 9.4, -13.5 ± 11.6%; 6 weeks -24.4 ± 8.2, -17.7 ± 8.3, -15.2 ± 8.4% respectively, all p < 0.05). Reduced radial strain in periinfarct and remote regions occurred despite minimal or absent necrosis (area of necrosis I, PI, R: 48.8 ± 23, 5.1 ± 6.6, 0 ± 0%, p < 0.001 vs. I).</p> <p>Conclusion</p> <p>Following left anterior coronary occlusion, radial strain decreased at 60 minutes and up to 6 weeks in the periinfarct and remote regions, similar to the reduction in the infarct region. This demonstrates early and chronic myopathic process in periinfarct and remote regions following myocardial infarction that may be an under recognized but important contributor to adverse left ventricular remodeling and progression to ischemic cardiomyopathy.</p

    High content live cell imaging for the discovery of new antimalarial marine natural products

    <p>Abstract</p> <p>Background</p> <p>The human malaria parasite remains a burden in developing nations. It is responsible for up to one million deaths a year, a number that could rise due to increasing multi-drug resistance to all antimalarial drugs currently available. Therefore, there is an urgent need for the discovery of new drug therapies. Recently, our laboratory developed a simple one-step fluorescence-based live cell-imaging assay to integrate the complex biology of the human malaria parasite into drug discovery. Here we used our newly developed live cell-imaging platform to discover novel marine natural products and their cellular phenotypic effects against the most lethal malaria parasite, <it>Plasmodium falciparum</it>.</p> <p>Methods</p> <p>A high content live cell imaging platform was used to screen marine extracts effects on malaria. Parasites were grown <it>in vitro </it>in the presence of extracts, stained with RNA sensitive dye, and imaged at timed intervals with the BD Pathway HT automated confocal microscope.</p> <p>Results</p> <p>Image analysis validated our new methodology at a larger scale level and revealed potential antimalarial activity of selected extracts with a minimal cytotoxic effect on host red blood cells. To further validate our assay, we investigated parasite's phenotypes when incubated with the purified bioactive natural product bromophycolide A. We show that bromophycolide A has a strong and specific morphological effect on parasites, similar to the ones observed from the initial extracts.</p> <p>Conclusion</p> <p>Collectively, our results show that high-content live cell-imaging (HCLCI) can be used to screen chemical libraries and identify parasite specific inhibitors with limited host cytotoxic effects. All together we provide new leads for the discovery of novel antimalarials.</p
