Langeres Erhitzen .des Benzoyl- bzw. p-Nitrobenzoylesters des monocyclischen Sesquiterpens Elemol (III) auf 190-2100 liefert ein Gemisch von Kohlenwasserstoffen C15H 24 (VII), welches bei der katalytischen Hydrierung nach Aufnahme von 2,1 Mol Wasserstoff in den bekannten, bicyclischen Kohlenwasserstoff Eudesman (VIII) iibergeht . Obwohl dieser Befund eine direkte experimentelle Ve1!lmiipfung der Sesquiterpenalkohole Eudesmol (IV) und Elemol (III) darstellt, kann er fiir die Bestimmung der absoluten Konfig uration des Elemols vorlii.ufig nicht h erangezogen werden
Loss of XIAP facilitates switch to TNFα-induced necroptosis in mouse neutrophils.
Neutrophils are essential players in the first-line defense against invading bacteria and fungi. Besides its antiapoptotic role, the inhibitor of apoptosis protein (IAP) family member X-linked IAP (XIAP) has been shown to regulate innate immune signaling. Whereas the role of XIAP in innate signaling pathways is derived mostly from work in macrophages and dendritic cells, it is not known if and how XIAP contributes to these pathways in neutrophils. Here we show that in response to bacterial lipopolysaccharides (LPS), mouse neutrophils secreted considerable amounts of tumor necrosis factor-α (TNFα) and interleukin-1β (IL-1β) and, in accordance with earlier reports, XIAP prevented LPS-induced hypersecretion of IL-1β also in neutrophils. Interestingly, and in contrast to macrophages or dendritic cells, Xiap-deficient neutrophils were insensitive to LPS-induced cell death. However, combined loss of function of XIAP and cIAP1/-2 resulted in rapid neutrophil cell death in response to LPS. This cell death occurred by classical apoptosis initiated by a TNFα- and RIPK1-dependent, but RIPK3- and MLKL-independent, pathway. Inhibition of caspases under the same experimental conditions caused a shift to RIPK3-dependent cell death. Accordingly, we demonstrate that treatment of neutrophils with high concentrations of TNFα induced apoptotic cell death, which was fully blockable by pancaspase inhibition in wild-type neutrophils. However, in the absence of XIAP, caspase inhibition resulted in a shift from apoptosis to RIPK3- and MLKL-dependent necroptosis. Loss of XIAP further sensitized granulocyte-macrophage colony-stimulating factor (GM-CSF)-primed neutrophils to TNFα-induced killing. These data suggest that XIAP antagonizes the switch from TNFα-induced apoptosis to necroptosis in mouse neutrophils. Moreover, our data may implicate an important role of neutrophils in the development of hyperinflammation and disease progression of patients diagnosed with X-linked lymphoproliferative syndrome type 2, which are deficient in XIAP
A Cell Motility Screen Reveals Role for MARCKS-Related Protein in Adherens Junction Formation and Tumorigenesis
Invasion through the extracellular matrix (ECM) is important for wound healing, immunological responses and metastasis. We established an invasion-based cell motility screen using Boyden chambers overlaid with Matrigel to select for pro-invasive genes. By this method we identified antisense to MARCKS related protein (MRP), whose family member MARCKS is a target of miR-21, a microRNA involved in tumor growth, invasion and metastasis in multiple human cancers. We confirmed that targeted knockdown of MRP, in both EpRas mammary epithelial cells and PC3 prostate cancer cells, promoted in vitro cell migration that was blocked by trifluoperazine. Additionally, we observed increased immunofluoresence of E-cadherin, β-catenin and APC at sites of cell-cell contact in EpRas cells with MRP knockdown suggesting formation of adherens junctions. By wound healing assay we observed that reduced MRP supported collective cell migration, a type of cell movement where adherens junctions are maintained. However, destabilized adherens junctions, like those seen in EpRas cells, are frequently important for oncogenic signaling. Consequently, knockdown of MRP in EpRas caused loss of tumorigenesis in vivo, and reduced Wnt3a induced TCF reporter signaling in vitro. Together our data suggest that reducing MRP expression promotes formation of adherens junctions in EpRas cells, allowing collective cell migration, but interferes with oncogenic β-catenin signaling and tumorigenesis
Isotopic Composition of Light Nuclei in Cosmic Rays: Results from AMS-01
The variety of isotopes in cosmic rays allows us to study different aspects
of the processes that cosmic rays undergo between the time they are produced
and the time of their arrival in the heliosphere. In this paper we present
measurements of the isotopic ratios 2H/4He, 3He/4He, 6Li/7Li, 7Be/(9Be+10Be)
and 10B/11B in the range 0.2-1.4 GeV of kinetic energy per nucleon. The
measurements are based on the data collected by the Alpha Magnetic
Spectrometer, AMS-01, during the STS-91 flight in 1998 June.Comment: To appear in ApJ. 12 pages, 11 figures, 6 table
Protons in near earth orbit
The proton spectrum in the kinetic energy range 0.1 to 200 GeV was measured
by the Alpha Magnetic Spectrometer (AMS) during space shuttle flight STS-91 at
an altitude of 380 km. Above the geomagnetic cutoff the observed spectrum is
parameterized by a power law. Below the geomagnetic cutoff a substantial second
spectrum was observed concentrated at equatorial latitudes with a flux ~ 70
m^-2 sec^-1 sr^-1. Most of these second spectrum protons follow a complicated
trajectory and originate from a restricted geographic region.Comment: 19 pages, Latex, 7 .eps figure
Podoplanin Associates with CD44 to Promote Directional Cell Migration
Podoplanin, a cancer-associated glycoprotein, interacts with CD44. Both glycoproteins are coordinately upregulated during tumor progression. Podoplanin–CD44 interaction in the cell membrane occurs mainly in migrating cells, and it seems to be required for podoplanin-mediated cell migration and directionality
A Study of Cosmic Ray Secondaries Induced by the Mir Space Station Using AMS-01
The Alpha Magnetic Spectrometer (AMS-02) is a high energy particle physics
experiment that will study cosmic rays in the to range and will be installed on the International Space Station
(ISS) for at least 3 years. A first version of AMS-02, AMS-01, flew aboard the
space shuttle \emph{Discovery} from June 2 to June 12, 1998, and collected
cosmic ray triggers. Part of the \emph{Mir} space station was within the
AMS-01 field of view during the four day \emph{Mir} docking phase of this
flight. We have reconstructed an image of this part of the \emph{Mir} space
station using secondary and emissions from primary cosmic rays
interacting with \emph{Mir}. This is the first time this reconstruction was
performed in AMS-01, and it is important for understanding potential

The High Radiosensitizing Efficiency of a Trace of Gadolinium-Based Nanoparticles in Tumors
International audienceWe recently developed the synthesis of ultrasmall gadolinium-based nanoparticles (GBN), (hydrodynamic diameter <5 nm) characterized by a safe behavior after intravenous injection (renal clearance, preferential accumulation in tumors). Owing to the presence of gadolinium ions, GBN can be used as contrast agents for magnetic resonance imaging (MRI) and as radiosensitizers. The attempt to determine the most opportune delay between the intravenous injection of GBN and the irradiation showed that a very low content of radiosensitizing nanoparticles in the tumor area is sufficient (0.1 μg/g of particles, i.e. 15 ppb of gadolinium) for an important increase of the therapeutic effect of irradiation. Such a promising and unexpected result is assigned to a suited distribution of GBN within the tumor, as revealed by the X-ray fluorescence (XRF) maps
