    Gene expression profile of bladder tissue of patients with ulcerative interstitial cystitis

    BACKGROUND: Interstitial cystitis (IC), a chronic bladder disease with an increasing incidence, is diagnosed using subjective symptoms in combination with cystoscopic and histological evidence. By cystoscopic examination, IC can be classified into an ulcerative and a non-ulcerative subtype. To better understand this debilitating disease on a molecular level, a comparative gene expression profile of bladder biopsies from patients with ulcerative IC and control patients has been performed. RESULTS: Gene expression profiles from bladder biopsies of five patients with ulcerative IC and six control patients were generated using Affymetrix GeneChip expression arrays (Affymetrix--GeneChip Human Genome U133 Plus 2.0). More than 31,000 of > 54,000 tested probe sets were present (detection p-value < 0.05). The difference between the two groups was significant for over 3,500 signals (t-test p-value < 0.01), and approximately 2,000 of the signals (corresponding to approximately 1,000 genes) showed an IC-to-healthy expression ratio greater than two. The IC pattern had similarities to patterns from immune system, lymphatic, and autoimmune diseases. The dominant biological processes were the immune and inflammatory responses. Many of the up-regulated genes were expressed in leukocytes, suggesting that leukocyte invasion into the bladder wall is a dominant feature of ulcerative IC. Histopathological data supported these findings. CONCLUSION: GeneChip expression arrays present a global picture of ulcerative IC and provide us with a series of marker genes characteristic for this subtype of the disease. Evaluation of biopsies from other bladder patients with similar symptoms (e.g. patients with non-ulcerative IC) will further indicate whether the data presented here will be valuable for the diagnosis of IC

    Oscillating Between Hope and Despair:Understanding Migrants’ Reflections on Ambivalence in ‘Transit’

    This paper investigates the under-explored question of how migrants in so-called ‘transit countries’ make sense of migration aspirations. Drawing from recent scholarship on migration-related ambivalence, we focus on how people reflect on the past and present of their migration aspirations, employing a migrant-centered approach. Based on semi-structured interviews with refugees in Libya and Bosnia and Herzegovina as well as complementary expert interviews, we find that ambivalence, which (re-)shapes migration aspirations, is a necessary reaction to the structures of uncertainty installed in current European externalization measures beyond EU borders. By adopting a migrant-centered approach and taking into account the multidimensional and processual nature of ambivalence, our research contributes to a better understanding of migrants as self-critical and reflective actors facing the challenges of making decisions in situations of uncertainty; thus, ambivalence is produced in a dialectic interplay between migrants’ agency and the opportunities/limitations of changing structures that surround them which, in turn, informs the interplay between forward and backward migration aspirations

    Exploring the mechanical character of molybdenum grain boundaries via nanoindentation and three-point-bending

    The interactions of interfaces with dislocations have been extensively studied in the past. Still, there is a lack of high throughput methods, which can potentially be used for systematic studies to cover a wide range of grain boundary types. Nanoindentation offers the opportunity to combine a high spatial resolution with high effectiveness, thus enabling to obtain comprehensive mechanical data in the vicinity of grain boundaries. The present study on coarse-grained molybdenum will show results of mechanical property mapping near grain boundaries. Here, for the first time also the indenter tip rotation angle with respect to the loading axis as well as the grain orientation are considered. Results will show that neglecting these parameters can bias interpretations of the interface/dislocation interactions, as the localized deformation paths around the indentation are thereby significantly changed. Systematic experiments on commercially pure, recrystallized molybdenum have been performed to investigate the dependence of the hardness increase near grain boundaries with respect to the boundary misorientation angle. As a complementary method, three-point-bending is applied on mm-sized specimens until individual grain boundaries delaminate, which in turn will be identified and cross-checked with findings of the nanoindentation tests. Doping molybdenum with elements like carbon and/or boron is known to suppress intercrystalline failure. For this reason, the presented grain boundary characterization methods will be applied to extract mechanical changes caused by these doping elements

    Is there an association between social determinants and care dependency risk? A multi‐state model analysis of a longitudinal study

    Despite a growing body of knowledge about the morbidities and functional impairment that frequently lead to care dependency, the role of social determinants is not yet well understood. The purpose of this study was to examine the effect of social determinants on care dependency onset and progression. We used data from the Berlin Initiative Study, a prospective, population-based cohort study including 2,069 older participants living in Berlin. Care dependency was defined as requiring substantial assistance in at least two activities of daily living for 90 min daily (level 1) or 3+ hours daily (level 2). Multi-state time to event regression modeling was used to estimate the effects of social determinants (partnership status, education, income, and sex), morbidities, and health behaviors, characteristics, and conditions. During the study period, 556 participants (27.5%) changed their status of care dependency. Participants without a partner at baseline were at a higher risk to become care-dependent than participants with a partner (hazard ratio [HR], 95% confidence interval [CI]: 1.24 (1.02-1.51)). After adjustment for other social determinants, morbidities and health behaviors, characteristics, and conditions the risk decreased to a HR of 1.19 (95% CI: 0.79-1.79). Results indicate that older people without a partner may tend to be at higher risk of care dependency onset but not at higher risk of care dependency progression. Clinicians should inquire about and consider patients' partnership status as they evaluate care needs

    Investigating the zoonotic origin of the West African Ebola epidemic

    The severe Ebola virus disease epidemic occurring in West Africa stems from a single zoonotic transmission event to a 2‐year‐old boy in Meliandou, Guinea. We investigated the zoonotic origins of the epidemic using wildlife surveys, interviews, and molecular analyses of bat and environmental samples. We found no evidence for a concurrent outbreak in larger wildlife. Exposure to fruit bats is common in the region, but the index case may have been infected by playing in a hollow tree housing a colony of insectivorous free‐tailed bats (Mops condylurus). Bats in this family have previously been discussed as potential sources for Ebola virus outbreaks, and experimental data have shown that this species can survive experimental infection. These analyses expand the range of possible Ebola virus sources to include insectivorous bats and reiterate the importance of broader sampling efforts for understanding Ebola virus ecology

    Feebly-Interacting Particles:FIPs 2020 Workshop Report

    With the establishment and maturation of the experimental programs searching for new physics with sizeable couplings at the LHC, there is an increasing interest in the broader particle and astrophysics community for exploring the physics of light and feebly-interacting particles as a paradigm complementary to a New Physics sector at the TeV scale and beyond. FIPs 2020 has been the first workshop fully dedicated to the physics of feebly-interacting particles and was held virtually from 31 August to 4 September 2020. The workshop has gathered together experts from collider, beam dump, fixed target experiments, as well as from astrophysics, axions/ALPs searches, current/future neutrino experiments, and dark matter direct detection communities to discuss progress in experimental searches and underlying theory models for FIPs physics, and to enhance the cross-fertilisation across different fields. FIPs 2020 has been complemented by the topical workshop "Physics Beyond Colliders meets theory", held at CERN from 7 June to 9 June 2020. This document presents the summary of the talks presented at the workshops and the outcome of the subsequent discussions held immediately after. It aims to provide a clear picture of this blooming field and proposes a few recommendations for the next round of experimental results.Comment: 240 pages, 71 figure

    The Role of Occupants in Buildings’ Energy Performance Gap: Myth or Reality?

    Buildings’ expected (projected, simulated) energy use frequently does not match actual observations. This is commonly referred to as the energy performance gap. As such, many factors can contribute to the disagreement between expectations and observations. These include, for instance, uncertainty about buildings’ geometry, construction, systems, and weather conditions. However, the role of occupants in the energy performance gap has recently attracted much attention. It has even been suggested that occupants are the main cause of the energy performance gap. This, in turn, has led to suggestions that better models of occupant behavior can reduce the energy performance gap. The present effort aims at the review and evaluation of the evidence for such claims. To this end, a systematic literature search was conducted and relevant publications were identified and reviewed in detail. The review entailed the categorization of the studies according to the scope and strength of the evidence for occupants’ role in the energy performance gap. Moreover, deployed calculation and monitoring methods, normalization procedures, and reported causes and magnitudes of the energy performance gap were documented and evaluated. The results suggest that the role of occupants as significant or exclusive contributors to the energy performance gap is not sufficiently substantiated by evidence.</jats:p
