    Questionnaire on the Training Profile of a Learning Therapy Specialist: Creation and Validation of the Instrument.

    This article provides a description of the creation and validation process for a questionnaire designed to analyze the initial training received by students of degree programs in pre-primary and primary education, specializing in learning therapy. The instrument consists of 175 items in three sections: the first collects sociodemographic data; the second analyzes the initial training received; and the third and final section analyzes the perception of this initial training. The questionnaire was subjected to a double validation process: an evaluation by a panel of experts and a pilot group, these permitted the validation of the content and construct, as well as the corresponding reliability assessment. The results confirm the validity of the questionnaire with an excellent reliability index, allowing the e ective evaluation of the training profile of learning therapy specialists.post-print257 K

    Analysis of the Functioning of Families of Children with Autism Spectrum Disorder: A Psychometric Study of the Family APGAR Scale.

    Normalised family functioning is a predictor of individual well-being. The diagnosis of a family member with autism spectrum disorder (ASD) can alter the ordinary dynamics of family systems, having a variable impact on family functioning. This research employed a non-probability convenience sampling method to gather a sample of 327 families with and without children diagnosed with ASD. This study has dual objectives: to analyse the psychometric properties of the Spanish version of the Family APGAR Scale and to compare family functioning in families with and without a child with ASD. The results reveal several favourable psychometric properties in the application of the APGAR scale within families of children with ASD. The use of the Family APGAR Scale in the selected sample confirms that the functioning of families with children with ASD can be categorized as mildly dysfunctional, attributed to the inherent challenges in caring for and raising a child with ASD. The presence of ASD within family systems presents a challenge to typical family functioning, with significant differences observed between families with and without children with ASD. This underscores the necessity of implementing effective intervention programs based on empirical evidence to improve the quality of life for individuals with ASD and their families.post-print381 K

    Analysis of ASD Classrooms: Specialised Open Classrooms in the Community of Madrid.

    Autism spectrum disorder (ASD) classrooms are specialised spaces within mainstream schools that respond to the educational needs of students with autism spectrum disorder. This research aims to analyse ASD classrooms in the Community of Madrid, their features and internal functioning. The research follows a non-experimental, descriptive and comparative methodology, making a comparison between di erent classrooms and analysing their impact on the teaching-learning process of students with ASD. The variables refer to the physical environment, the characteristics of the children, training of professionals and collaboration with other organisations. The data was gathered using a questionnaire in digital format, collecting responses from 35 schools, using quantitative techniques to relate variables. The results show that most classrooms have a structured organisation that meet the needs of the students, who are generally diagnosed with moderate severity. Furthermore, the relationship between the teachers of ASD classrooms and the mainstream classrooms promotes inclusive educational practice. Finally, it was found that ASD classrooms o er many benefits, due to the personalised attention they provide and the specialised training of teachers.post-print4181 K

    La competencia digital en la atención del alumnado con necesidades educativas especiales. Una visión general del marco europeo para la competencia docente digital "DigCompEdu"

    We live in an age marked by technological advances. From an educational point of view, a model which requires methodologies adapted to digital media has consolidated, implying that the development of digital competence among the teachers has taken a special importance in order to adapt the learning and teaching process to each individual needs, favouring inclusion of students with special educational needs. This research aims to analyse the digital competency of teachers specialized in the attention to diversity. To this end, a non-experimental, descriptive investigation has been carried out, with a sample of 60 teachers currently working in both the Special and Ordinary Education models. The European questionnaire "DigCompEdu" has been used as analysis tool. Data obtained allows us to conclude that the level of digital competence of teachers who attend diversity in Special Education Schools is lower than the level shown by teachers of Ordinary Schools, showing statistically significative differences in three of the analysed areas. These areas are related to the use of digital competence, the adaptation of evaluation strategies and development of digital competency among the students.Vivimos en una época marcada por los avances tecnológicos. Desde el punto de vista educativo, se ha consolidado un modelo que requiere metodologías adaptadas a los medios digitales, lo que supone que el desarrollo de la competencia digital entre el profesorado sea un aspecto que ha cobrado especial importancia para adaptar la enseñanza a las necesidades individuales del alumnado, favoreciendo así la atención a las necesidades educativas especiales. Esta investigación tiene como objetivo analizar la competencia digital del profesorado especialista en atención a la diversidad. Para ello se ha llevado a cabo una investigación no experimental y descriptiva, con una muestra conformada por 60 docentes en activo del modelo educativo especial y del modelo ordinario. Como instrumento de análisis se ha utilizado el cuestionario europeo “DigCompEdu”. Los datos obtenidos permiten concluir que el nivel de competencia digital del profesorado que atiende a la diversidad del alumnado en los centros educativos especiales se encuentra en niveles inferiores respecto al manifestado por el profesorado de los centros ordinarios, observando diferencias estadísticamente significativas en tres de las áreas analizadas. Estas áreas están relacionadas con el uso de la competencia digital, la adaptación de estrategias de evaluación y el desarrollo de la competencia digital entre sus estudiantes

    The JAK3Q988P mutation reveals oncogenic potential and resistance to ruxolitinib

    T-cell acute lymphoblastic leukemia (T-ALL) arises from the malignant transformation of T-cell progenitors at various differentiation stages. Given that patients who relapse have a dismal prognosis, there is an urgent need to identify the molecular alterations that are present in such patients and promote leukemogenesis to implement personalized therapies with higher efficacy and fewer adverse effects. In the present manuscript, we identified the JAK3Q988P mutation in a T-ALL patient who did not achieve a durable response after the conventional treatment and whose tumor cells at relapse presented constitutive activation of the JAK/STAT pathway. Although JAK3Q988P has been previously identified in T-ALL patients from different studies, the functional consequences exerted by this mutation remain unexplored. Through the combination of different hematopoietic cellular models, we functionally characterize JAK3Q988P as an oncogenic mutation that contributes to leukemogenesis. Notably, JAK3Q988P not only promotes constitutive activation of the JAK/STAT pathway in the absence of cytokines and growth factors, as is the case for other JAK3 mutations that have been functionally characterized as oncogenic, but also functions independently of JAK1 and IL2RG, resulting in high oncogenic potential as well as resistance to ruxolitinib.     SPR Biosensing MUA/Poly-L-lysine platform for the detection of 2,4-Dinitrophenol as small molecule model system

    Surface Plasmon Resonance assays are being developed as alternative biodetection methods for a great number of pesticides and toxins.These substances typically have low molecular weight, making it necessary to perform competitive inhibition immunoassays. In most of the cases, the strategy is to immobilize a protein derivative of the analyte, which usually involves the appearance of nonspecific protein binding which limits the detection range of the assay. In this work we present results of a poly-L-lysine (Au-MUA-PLL) based sensor platform for quantitative determination of 2,4-dinitrophenol as model system for small molecular weight substances detection. The prepared sensor chip was characterized by means of Atomic Force Microscopy, Surface Plasmon Resonance, and Surface Enhanced Raman Spectroscopy. Experiments verified the absence of nonspecific protein adsorption to Au-MUA-PLL surfaces and the improvement of the competitive inhibition assays performance in comparison with single and mixed thiol self-assembled monolayers. The possibility of directly immobilizing 2,4-dinitrophenol to the poly-L-lysine containing platforms leads to an improvement in the detection of the soluble analyte by the competitive inhibition assay avoiding undesirable nonspecific protein adsorption.Therefore, Au-MUA-PLL surfaces constitute a suitable alternative for quantitative detection of small molecules when nonspecific adsorption cannot be avoided.Fil: Daza Millone, Maria Antonieta.     A Gesture Recognition System for Detecting Behavioral Patterns of ADHD

    We present an application of gesture recognition using an extension of dynamic time warping (DTW) to recognize behavioral patterns of attention deficit hyperactivity disorder (ADHD). We propose an extension of DTW using one-class classifiers in order to be able to encode the variability of a gesture category, and thus, perform an alignment between a gesture sample and a gesture class. We model the set of gesture samples of a certain gesture category using either Gaussian mixture models or an approximation of convex hulls. Thus, we add a theoretical contribution to classical warping path in DTW by including local modeling of intraclass gesture variability. This methodology is applied in a clinical context, detecting a group of ADHD behavioral patterns defined by experts in psychology/psychiatry, to provide support to clinicians in the diagnose procedure. The proposed methodology is tested on a novel multimodal dataset (RGB plus depth) of ADHD children recordings with behavioral patterns. We obtain satisfying results when compared to standard state-of-the-art approaches in the DTW context

    SOCS3 deregulation contributes to aberrant activation of the JAK/STAT pathway in precursor T-cell neoplasms

    Despite the Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway being frequently altered in T-ALL/LBL, no specific therapy has been approved for T-ALL/LBL patients with constitutive signalling by JAK/STAT, so there is an urgent need to identify pathway members that may be potential therapeutic targets. In the present study, we searched for JAK/STAT pathway members potentially modulated through aberrant methylation and identified SOCS3 hypermethylation as a recurrent event in T-ALL/LBL. Additionally, we explored the implications of SOCS3 deregulation in T-ALL/LBL and demonstrated that SOCS3 counteracts the constitutive activation of the JAK/STAT pathway through different molecular mechanisms.     Cellular and humoral functional responses after BNT162b2 mRNA vaccination differ longitudinally between naive and subjects recovered from COVID-19

    We have analyzed BNT162b2 vaccine-induced immune responses in naive subjects and individuals recovered from coronavirus disease 2019 (COVID-19), both soon after (14 days) and later after (almost 8 months) vaccination. Plasma spike (S)-specific immunoglobulins peak after one vaccine shot in individuals recovered from COVID-19, while a second dose is needed in naive subjects, although the latter group shows reduced levels all along the analyzed period. Despite how the neutralization capacity against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) mirrors this behavior early after vaccination, both groups show comparable neutralizing antibodies and S-specific B cell levels late post-vaccination. When studying cellular responses, naive individuals exhibit higher SARS-CoV-2-specific cytokine production, CD4+ T cell activation, and proliferation than do individuals recovered from COVID-19, with patent inverse correlations between humoral and cellular variables early post-vaccination. However, almost 8 months post-vaccination, SARS-CoV-2-specific responses are comparable between both groups.
