    Out-of-Plane Transport of 1T-TaS2/Graphene-Based van der Waals Heterostructures

    Due to their anisotropy, layered materials are excellent candidates for studying the interplay between the in-plane and out-of-plane entanglement in strongly correlated systems. A relevant example is provided by 1T-TaS2, which exhibits a multifaceted electronic and magnetic scenario due to the existence of several charge density wave (CDW) configurations. It includes quantum hidden phases, superconductivity and exotic quantum spin liquid (QSL) states, which are highly dependent on the out-of-plane stacking of the CDW. In this system, the interlayer stacking of the CDW is crucial for interpreting the underlying electronic and magnetic phase diagram. Here, atomically thin-layers of 1T-TaS2 are integrated in vertical van der Waals heterostructures based on few-layers graphene contacts and their electrical transport properties are measured. Different activation energies in the conductance and a gap at the Fermi level are clearly observed. Our experimental findings are supported by fully self-consistent DFT+U calculations, which evidence the presence of an energy gap in the few-layer limit, not necessarily coming from the formation of out-of-plane spin-paired bilayers at low temperatures, as previously proposed for the bulk. These results highlight dimensionality as a key effect for understanding quantum materials as 1T-TaS2, enabling the possible experimental realization of low-dimensional QSLs

    Congreso online como herramienta docente para estudiantes de tercer ciclo en Electroquímica

    Los congresos científicos son una buena herramienta para que los estudiantes de tercer ciclo puedan ampliar sus conocimientos. Sin embargo, las intervenciones donde los estudiantes plantean dudas o preguntas son prácticamente nulas. Con el objetivo de fomentar la participación de los estudiantes en los congresos científicos, la nueva red continua con el trabajo anteriormente realizado, llevando a cabo la II edición del Congreso online de estudiantes dentro del programa interuniversitario “Electroquímica. Ciencia y Tecnología”. La red busca concienciar acerca de la importancia que para un investigador tiene un congreso científico y a su vez, incrementar y mejorar su participación. Para ello, se utiliza un formato más atractivo que en la edición anterior, que mejora el entorno de trabajo y favorece la interacción entre los estudiantes. Asimismo, se emplean estrategias de comunicación más desarrolladas para hacer crecer el número de participantes. Finalmente, se establecerán diferentes parámetros para evaluar la actividad durante el congreso y se entregarán premios para motivar la participación

    Digestibility of Duddingtonia flagrans chlamydospores in ruminants: in vitro and in vivo studies

    <p>Abstract</p> <p>Background</p> <p>The use of <it>Duddingtonia flagrans </it>as a tool for the biological control of gastrointestinal nematodes (GIN) is a promising alternative to anthelmintics. The chlamydospores of <it>D. flagrans </it>are orally dosed and their thick cell wall gives them the capacity to resist digestion and pass through the gastrointestinal tract (GIT). Chlamydospores reaching the faeces are able to germinate and trap nematode larvae. The efficacy of this control method is based on reducing the numbers of infective larvae leaving the faeces. Techniques have recently been developed for quantifying the numbers of chlamydospores in faeces. As the number of non-digested spores could be relevant in the design and optimization of dosing programmes for the control of GIN infective larvae, the aim of the present study was to estimate the loss of <it>D. flagrans </it>chlamydospores during their passage through the ruminant gastrointestinal tract using <it>in vitro </it>and <it>in vivo </it>techniques.</p> <p>Results</p> <p>After <it>in vitro </it>rumen digestion, chlamydospore recovery was not different from the quantity originally incubated (undigested spores) (P > 0.05). <it>In vitro </it>rumen+abomasum digestion caused nearly 36% loss of the chlamydospores originally incubated (P < 0.05). Germination of chlamydospores classified as viable was 24.3%. Chlamydospores classified as non-viable did not germinate. Rumen digestion resulted in more spore germination (R1 = 35.7% and R2 = 53.3%) compared to no digestion (time 0 h = 8.7%). Subsequent abomasal digestion reduced germination (R1+A = 25%) or stopped it (R2+A = 0%). <it>In vivo </it>apparent chlamydospore digestibility in sheep showed a loss of 89.7% of the chlamydospores (P < 0.05).</p> <p>Conclusions</p> <p>The loss of chlamydospores was evident under <it>in vitro </it>and <it>in vivo </it>conditions. Negligible amounts of spores were lost during the <it>in vitro </it>rumen digestion. However, <it>in vitro </it>rumen+abomasum digestion resulted in a chlamydospore loss of approximately 36%. <it>In vivo </it>passage through the sheep GIT resulted in a total loss of 89.7% of the orally administered spores.</p

    Aedes albopictus diversity and relationships in south-western Europe and Brazil by rDNA/mtDNA and phenotypic analyses: ITS-2, a useful marker for spread studies

    Background: Aedes albopictus is a very invasive mosquito, which has recently colonized tropical and temperate regions worldwide. Of concern is its role in the spread of emerging or re-emerging mosquito-borne diseases. Ae. albopictus from south-western Europe and Brazil were studied to infer genetic and phenetic diversity at intra-individual, intra-population and inter-population levels, and to analyse its spread. Methods: Genotyping was made by rDNA 5.8S-ITS-2 and mtDNA cox1 sequencing to assess haplotype and nucleotide diversity, genetic distances and phylogenetic networks. Male and female phenotyping included combined landmark-and outlined-based geometric morphometrics of wing size and shape. Results: Specimens from seven populations from Spain, France and Brazil provided 12 cox1 and 162 5.8S-ITS-2 haplotypes, with great genetic variability difference between both markers (0.9% vs 31.2%). Five cox1 haplotypes were shared with other countries, mainly Italy, USA and China, but none was shared between Europe and Brazil. The 5.8S-ITS-2 showed 2–7 intra-individual (mean 4.7) and 16–34 intra-/inter-population haplotypes (24.7), including haplotypes shared between Spain, France and Brazil. A 4.3% of ITS-2 haplotypes were shared, mainly with Italy, USA and Thailand, evidencing worldwide spread and introductions from areas where recent outbreaks of Ae. albopictus-transmitted pathogens occurred. Wing size showed sex differences. Wing shape distinguished between Brazilian and European specimens. Both genetic and morphometric markers showed differences between insular Spain and continental Spain, France and Brazil. Conclusions: ITS-2 proves to be a useful marker to assess Ae. albopictus spread, providing pronouncedly more information than cox1, including intra-individual, intra-population and inter-population levels, furnishing a complete overview of the evolutionary exchanges followed by this mosquito. Wing morphometry proves to be a useful phenotyping marker, allowing to distinguish different populations at the level of both male and female specimens. Results indicate the need for periodic surveillance monitorings to verify that no Ae. albopictus with high virus transmission capacity is introduced into Europe.     Congreso online como herramienta docente para estudiantes de máster y doctorado

    Los congresos científicos son el segundo medio de difusión de resultados y conocimientos científicos y representan además, una gran oportunidad para contactar con otros colegas de profesión y establecer futuras colaboraciones. En el entorno de esta red se ha organizado la segunda edición de un congreso online para estudiantes del Programa de Máster y Doctorado en “Electroquímica. Ciencia y Tecnología”, con la firme intención de que sirva no sólo como medio de aprendizaje, sino también para que los estudiantes se habitúen al intercambio de dudas y preguntas sobre sus propios trabajos y los de otros investigadores y pasen a desempeñar un papel más activo en congresos científicos presenciales. Por otro lado, en esta segunda edición del congreso online, se han planteado objetivos más ambiciosos tanto en cuanto al número de participantes respecto a la edición anterior como en el grado de participación de los inscritos. En este sentido, las estrategias llevadas a cabo han sido satisfactorias y hemos logrado no sólo un importante aumento de participantes sino también un mayor grado de implicación de los mismos, lo que se refleja en el número de visitas a los pósteres como el número de preguntas y respuestas formuladas por los estudiantes

    Estrategias para la mejora de la cooperación y trabajo en equipo en un congreso

    La introducción de las tecnologías de la información y comunicación (TIC) en los campos de la docencia e investigación presenta numerosas ventajas en relación al fomento de la participación, el trabajo colaborativo y la cooperación de los estudiantes de grado, de máster o de doctorado. Este trabajo tiene como primer objetivo analizar el impacto de las plataformas online sobre el aprendizaje de los estudiantes y, en segundo lugar, desarrollar alternativas que fomenten la participación e interacción de los estudiantes utilizando estas plataformas educativas. Con estos objetivos, esta red educativa ha adquirido durante estos últimos años una notable experiencia en la organización de congresos online. Sin embargo, todas las potencialidades comunicativas que podría ofrecer un congreso online a sus participantes no han sido todavía exploradas: la incorporación de las herramientas utilizadas en las redes sociales para promover la participación de los estudiantes, o bien la posibilidad de que los estudiantes participen a través de otros formatos además del póster, entre otras, podrían ser altamente beneficiosas. En relación a este último aspecto, una posible alternativa sería la presentación de comunicaciones en formato de presentación flash, PowerPoint, o cualquier otro formato interactivo donde se muestren los aspectos claves de la investigación del participante

    Oncogenic Role of Secreted Engrailed Homeobox 2 (EN2) in Prostate Cancer

    Engrailed variant-2 (EN2) has been suggested as a potential diagnostic biomarker; however, its presence and functional role in prostate cancer (PCa) cells is still controversial or unknown. Here, we analyzed 1) the expression/secretion profile of EN2 in five independent samples cohorts from PCa patients and controls (prostate tissues and/or urine) to determine its utility as a PCa biomarker; and 2) the functional role of EN2 in normal (RWPE1) and tumor (LNCaP/22Rv1/PC3) prostate cells to explore its potential value as therapeutic target. EN2 was overexpressed in our two cohorts of PCa tissues compared to control and in tumor cell lines compared with normal-like prostate cells. This profile was corroborated in silico in three independent data sets [The Cancer Genome Atlas(TCGA)/Memorial Sloan Kettering Cancer Center (MSKCC)/Grasso]. Consistently, urine EN2 levels were elevated and enabled discrimination between PCa and control patients. EN2 treatment increased cell proliferation in LNCaP/22Rv1/PC3 cells, migration in RWPE1/PC3 cells, and PSA secretion in LNCaP cells. These effects were associated, at least in the androgen-sensitive LNCaP cells, with increased AKT and androgen-receptor phosphorylation levels and with modulation of key cancer-associated genes. Consistently, EN2 treatment also regulated androgen-receptor activity (full-length and splicing variants) in androgen-sensitive 22Rv1 cells. Altogether, this study demonstrates the potential utility of EN2 as a non-invasive diagnostic biomarker for PCa and provides novel and valuable information to further investigate its putative utility to develop new therapeutic tools in PCa

    Activation of Type 1 Cannabinoid Receptor (CB1R) promotes neurogenesis in murine subventricular zone cell cultures

    The endocannabinoid system has been implicated in the modulation of adult neurogenesis. Here, we describe the effect of type 1 cannabinoid receptor (CB1R) activation on self-renewal, proliferation and neuronal differentiation in mouse neonatal subventricular zone (SVZ) stem/progenitor cell cultures. Expression of CB1R was detected in SVZ-derived immature cells (Nestin-positive), neurons and astrocytes. Stimulation of the CB1R by (R)-(+)-Methanandamide (R-m-AEA) increased self-renewal of SVZ cells, as assessed by counting the number of secondary neurospheres and the number of Sox2+/+ cell pairs, an effect blocked by Notch pathway inhibition. Moreover, R-m-AEA treatment for 48 h, increased proliferation as assessed by BrdU incorporation assay, an effect mediated by activation of MAPK-ERK and AKT pathways. Surprisingly, stimulation of CB1R by R-m-AEA also promoted neuronal differentiation (without affecting glial differentiation), at 7 days, as shown by counting the number of NeuN-positive neurons in the cultures. Moreover, by monitoring intracellular calcium concentrations ([Ca2+](i)) in single cells following KCl and histamine stimuli, a method that allows the functional evaluation of neuronal differentiation, we observed an increase in neuronal-like cells. This proneurogenic effect was blocked when SVZ cells were co-incubated with R-m-AEA and the CB1R antagonist AM 251, for 7 days, thus indicating that this effect involves CB1R activation. In accordance with an effect on neuronal differentiation and maturation, R-m-AEA also increased neurite growth, as evaluated by quantifying and measuring the number of MAP2-positive processes.     Genetic landscape of 6089 inherited retinal dystrophies affected cases in Spain and their therapeutic and extended epidemiological implications

    ESRETNET Study Group, The ERDC Study Group, The Associated Clinical Study Group.Inherited retinal diseases (IRDs), defined by dysfunction or progressive loss of photoreceptors, are disorders characterized by elevated heterogeneity, both at the clinical and genetic levels. Our main goal was to address the genetic landscape of IRD in the largest cohort of Spanish patients reported to date. A retrospective hospital-based cross-sectional study was carried out on 6089 IRD affected individuals (from 4403 unrelated families), referred for genetic testing from all the Spanish autonomous communities. Clinical, demographic and familiar data were collected from each patient, including family pedigree, age of appearance of visual symptoms, presence of any systemic findings and geographical origin. Genetic studies were performed to the 3951 families with available DNA using different molecular techniques. Overall, 53.2% (2100/3951) of the studied families were genetically characterized, and 1549 different likely causative variants in 142 genes were identified. The most common phenotype encountered is retinitis pigmentosa (RP) (55.6% of families, 2447/4403). The most recurrently mutated genes were PRPH2, ABCA4 and RS1 in autosomal dominant (AD), autosomal recessive (AR) and X-linked (XL) NON-RP cases, respectively; RHO, USH2A and RPGR in AD, AR and XL for non-syndromic RP; and USH2A and MYO7A in syndromic IRD. Pathogenic variants c.3386G > T (p.Arg1129Leu) in ABCA4 and c.2276G > T (p.Cys759Phe) in USH2A were the most frequent variants identified. Our study provides the general landscape for IRD in Spain, reporting the largest cohort ever presented.
