    Fludarabine inhibits KV1.3 currents in human B lymphocytes

    Fludarabine (F-ara-A) is a purine analog commonly used in the treatment of indolent B cell malignancies that interferes with different aspects of DNA and RNA synthesis. KV1.3 K+ channels are membrane proteins involved in the maintenance of K+ homeostasis and the resting potential of the cell, thus controlling signaling events, proliferation and apoptosis in lymphocytes. Here we show that F-ara-A inhibits KV currents in human B lymphocytes. Our data indicate that KV1.3 is expressed in both BL2 and Dana B cell lines, although total KV1.3 levels were higher in BL2 than in Dana cells. However, KV currents in the plasma membrane were similar in both cell lines and were abrogated by the specific KV1.3 channel inhibitor PAP-1, indicating that KV1.3 accounts for most of the KV currents in these cell lines. F-ara-A, at a concentration (3.5 μM) similar to that achieved in the plasma of fludarabine phosphate-treated patients (3 μM), inhibited KV1.3 currents by 61 ± 6.3% and 52.3 ± 6.3% in BL2 and Dana B cells, respectively. The inhibitory effect of F-ara-A was concentration-dependent and showed an IC50 value of 0.36 ± 0.04 μM and a nH value of 1.07 ± 0.15 in BL2 cells and 0.34 ± 0.13 μM (IC50) and 0.77 ± 0.11 (nH) in Dana cells. F-ara-A inhibition of plasma membrane KV1.3 was observed irrespective of its cytotoxic effect on the cells, BL2 cells being sensitive and Dana cells resistant to F-ara-A cytotoxicity. Interestingly, PAP-1, at concentrations as high as 10 μM, did not affect the viability of BL2 and Dana cells, indicating that blockage of KV1.3 in these cells is not toxic. Finally, F-ara-A had no effect on ectopically expressed KV1.3 channels, suggesting an indirect mechanism of current inhibition. In summary, our results describe the inhibitory effect of F-ara-A on the activity of KV1.3 channel. Although KV1.3 inhibition is not sufficient to induce cell death, further research is needed to determine whether it might still contribute to F-ara-A cytotoxicity in sensitive cells or be accountable for some of the clinical side effects of the drug.This study was supported by MINECO (SAF2013-45800-R, SAF2016-75021-R, RD12/0042/0019, CB/11/00222) and ISCIII (PI12/01135 and PI16/00895).     Estabilidad de catalizadores Ni soportados en ZrO₂ durante la hidrogenación selectiva de CO₂ hacia metano

    En el presente trabajo se estudió el efecto del método de síntesis de soportes de ZrO₂, y el efecto de la carga metálica sobre la hidrogenación catalítica del CO₂ para la producción de metano. Los soportes utilizados fueron ZrO₂ comercial (ZrO₂-COM), y óxidos de zirconio sintetizados por los métodos de coprecipitación (ZrO₂-COP) y Sol-Gel (ZrO₂-SG). Para los catalizadores sintetizados se utilizaron cargas metálicas de níquel del 10 y 20% peso. La evaluación catalítica se llevó a cabo a presión atmosférica, en un intervalo de temperatura de 350-500°C, utilizando una relación molar estequiométrica de CO₂/H₂. Los resultados más sobresalientes se obtuvieron con los catalizadores 20%/NiZrO₂-COM y 20%/NiZrO₂-COP con conversiones de CO2 cercanas al 50% a temperaturas de 400°C. El catalizador 20%/NiZrO₂-COP presentó una buena estabilidad con una caída en la conversión de solo el 8% a un tiempo de corrida experimental de 200 horas.In this work the effect of synthesis method for ZrO₂ as support, and the effect of metal loading on the catalytic hydrogenation of CO₂ to methane production were studied. The supports were commercial ZrO₂ (ZrO₂-COM), and zirconia synthesized by the coprecipitation (ZrO₂-COP) and the Sol-Gel (ZrO₂-SG) methods. Nickel metal loading of 10 and 20 wt. % were impregnated on the synthetized catalyst. Catalytic evaluation test was performed at atmospheric pressure conditions, in a temperature range of 350-500°C, and using a stoichiometric CO₂/H₂ molar ratio. The most outstanding results were obtained with the catalysts 20%/NiZrO₂-COM and 20%/NiZrO₂-COP with CO2 conversions close to 50% at temperatures of 400°C. The catalyst 20%/NiZrO₂-COP showed a very good stability with a decrease in CO2 conversion of only 8% after 200 hours of experimental test

      In general, when the river flow increases, a scour phenomenon will occur, in which the substructure of the bridge, be it abutment and / or piers, will be affected, resulting in a partial or total collapse of the structure. All of the above is the key to achieving our research objectives, which is to carry out an evaluation and protection design against the hydraulic scour process of the Unocolla bridge in the city of Juliaca, Puno. The population will be the Unocolla bridge and the sample will be a substructure of the bridge. The method used is applied and its design is pure experimental, using quantitative methods, in which the research problem is described as realistic results. To determine the scour effects, topographic, hydrological and sedimentological information was collected to later carry out hydraulic modeling in the Hec-Ras program. Scour depths were determined in reference to pillars, determining in pillar 1 scour depth 1.2 m, in pillar 2 scour depth 2.46 m, in pillar 3 scour depth 1.2 m, in pillar 4 scour depth 1.2 m, for which the castling process is proposed as an alternative solution.El objetivo de la investigación ha sido determinar la relación que ha existido entre la tributación y el gasto público en el Perú en el periodo 2000-2018. Con ese propósito, recolectamos los datos sobre los indicadores de las variables del portal de las instituciones públicas, particularmente de la Superintendencia Nacional de Administración Tributaria y Aduanas (Sunat) y del Ministerio de Economía y Finanzas (MEF). A partir de dichos datos, elaboramos una base de datos utilizando el SPSS Versión 25, que nos permitió describir la evolución de las variables (la estadística descriptiva) y realizar las pruebas de hipótesis correspondientes (la estadística inferencial). La principal conclusión a que hemos arribado, con la investigación, ha sido que existe una relación significativa entre la tributación y el gasto público en el Perú en el periodo 2000-2018

    Non-typeable Haemophilus influenzae and Streptococcus pneumoniae as primary causes of acute otitis media in colombian children: a prospective study

    <p>Abstract</p> <p>Background</p> <p>Acute otitis media (AOM) is one of the most frequently encountered bacterial infections in children aged < 5 years; <it>Streptococcus pneumoniae </it>(<it>S. pneumoniae</it>) and non-typeable <it>Haemophilus influenzae </it>(NTHi) are historically identified as primary AOM causes. Nevertheless, recent data on bacterial pathogens causing AOM in Latin America are limited. This prospective study aimed to identify and characterize bacterial etiology and serotypes of AOM cases including antimicrobial susceptibility in < 5 year old Colombian children.</p> <p>Methods</p> <p>From February 2008 to January 2009, children ≥3 months and < 5 years of age presenting with AOM and for whom a middle ear fluid (MEF) sample was available were enrolled in two medical centers in Cali, Colombia. MEF samples were collected either by tympanocentesis procedure or spontaneous otorrhea swab sampling. Bacteria were identified using standard laboratory methods, and antimicrobial resistance testing was performed based on the 2009 Clinical and Laboratory Standards Institute (CLSI) criteria. Most of the cases included in the study were sporadic in nature.</p> <p>Results</p> <p>Of the 106 enrolled children, 99 were included in the analysis. Bacteria were cultured from 62/99 (63%) of samples with <it>S. pneumoniae, H. influenzae, or S. pyogenes</it>. The most commonly isolated bacteria were <it>H. influenzae </it>in 31/99 (31%) and <it>S. pneumoniae </it>in 30/99 (30%) of samples. The majority of <it>H. influenzae </it>episodes were NTHi (27/31; 87%). 19F was the most frequently isolated pneumococcal serotype (10/30; 33%). Of the 30 <it>S. pneumoniae </it>positive samples, 8/30 (27%) were resistant to tetracycline, 5/30 (17%) to erythromycin and 8/30 (27%) had intermediate resistance to penicillin. All <it>H. influenzae </it>isolates tested were negative to beta-lactamase.</p> <p>Conclusions</p> <p>NTHi and <it>S. pneumoniae </it>are the leading causes of AOM in Colombian children. A pneumococcal conjugate vaccine that prevents both pathogens could be useful in maximizing protection against AOM.</p

    QTL analysis and genomic selection using RADseq derived markers in Sitka spruce: the potential utility of within family data

    Sitka spruce (Picea sitchensis (Bong.) Carr) is the most common commercial plantation species in Britain and a breeding programme based on traditional lines has been in operation since the early 1960s. Rotation lengths of 40-years have led breeders to adopt a process of indirect selection at younger ages based on traits well correlated with final selection, but still the generation interval is unlikely to reduce much below twenty years. Recent successful developments with genomic selection in animal breeding have led tree breeders to consider the application of this technology. In this study a RAD sequence assay was developed as a means of investigating the potential of molecular breeding in a non-model species. DNA was extracted from nearly 500 clonally replicated trees growing in a single full-sibling family at one site in Britain. The technique proved successful in identifying 132 QTLs for 5-year bud-burst and 2 QTLs for 6-year height. In addition, the accuracy of predicting phenotypes by genomic selection was strikingly high at 0.62 and 0.59 respectively. Sensitivity analysis with 200 offspring found only a slight fall in correlation values (0.54 and 0.38) although when the training population reduced to 50 offspring predictive values fell further (0.33 and 0.25). This proved an encouraging first investigation into the potential use of genomic selection in the breeding of Sitka spruce. The authors investigate how problems associated with effective population size and linkage disequilibrium can be avoided and suggest a practical way of incorporating genomic selection into a dynamic breeding programme
