Deconstructing Diagnosis: Four Commentaries on a Diagnostic Tool to Assess Individuals for Autism Spectrum Disorders

research and clinical practice to assess and evaluate autism
symptoms for both children and adults. These tools typically
involve observing the child or adult under assessment, and
rating their behaviour for signs or so-called symptoms of
autism.
In order to examine how autism diagnosis is constructed, how
diagnostic tools are positioned, and how their trainings are
delivered, we paid for four places on a training course for a
diagnostic tool. We asked the attendees (the first four authors)
to each produce a critical commentary about their impressions
of the training and the diagnostic tool itself. Their commentaries
are published here in full. They have various disciplinary
backgrounds: one is a social scientist, one an ethicist, one a
psychiatrist, and one a developmental psychologist.
The commentaries are followed by a concluding section that
summarises the themes, commonalities, and differences
between their accounts of the training course. Authors differed
as to whether the diagnostic tool is a useful and necessary
endeavour. Nevertheless, all critiqued of the tool’s lack of
transparency, recognizing context, emotion, and differences in
interpretation and power imbalances as playing an unidentified
role in the assessment process. Based on this project, we
recommend that training for raters for such tools should be
accessible to a wider group of people, and incorporate more
explicit recognition of its own limitations and commercialisation.Wellcome Trus
A comprehensive evaluation of the activity and selectivity profile of ligands for RGD-binding integrins
Integrins, a diverse class of heterodimeric cell surface receptors, are key regulators of cell structure and behaviour, affecting cell morphology, proliferation, survival and differentiation. Consequently, mutations in specific integrins, or their deregulated expression, are associated with a variety of diseases. In the last decades, many integrin-specific ligands have been developed and used for modulation of integrin function in medical as well as biophysical studies. The IC50-values reported for these ligands strongly vary and are measured using different cell-based and cell-free systems. A systematic comparison of these values is of high importance for selecting the optimal ligands for given applications. The Hidden Curriculum of Veterinary Education: Mediators and Moderators of Its Effects
The “hidden curriculum” has long been supposed to have an effect on students' learning during their clinical education, and in particular in shaping their ideas of what it means to be a professional. Despite this, there has been little evidence linking specific changes in professional attitudes to the individual components of the hidden curriculum. This study aimed to recognize those components that led to a change in students' professional attitudes at a UK veterinary school, as well as to identify the attitudes most affected. Observations were made of 11 student groups across five clinical rotations, followed by semi-structured interviews with 23 students at the end of their rotation experience. Data were combined and analyzed thematically, taking both an inductive and deductive approach. Views about the importance of technical competence and communication skills were promoted as a result of students' interaction with the hidden curriculum, and tensions were revealed in relation to their attitudes toward compassion and empathy, autonomy and responsibility, and lifestyle ethic. The assessment processes of rotations and the clinical service organization served to communicate the messages of the hidden curriculum, bringing about changes in student professional attitudes, while student-selected role models and the student rotation groups moderated the effects of these influences
European Sea Bass (Dicentrarchus labrax) immune status and disease resistance are impaired by arginine dietary supplementation
Infectious diseases and fish feeds management are probably the major expenses in the aquaculture business. Hence, it is a priority to define sustainable strategies which simultaneously avoid therapeutic procedures and reinforce fish immunity. Currently, one preferred approach is the use of immunostimulants which can be supplemented to the fish diets. Arginine is a versatile amino acid with important mechanisms closely related to the immune response. Aiming at finding out how arginine affects the innate immune status or improve disease resistance of European seabass (Dicentrarchus labrax) against vibriosis, fish were fed two arginine-supplemented diets (1% and 2% arginine supplementation). A third diet meeting arginine requirement level for seabass served as control diet. Following 15 or 29 days of feeding, fish were sampled for blood, spleen and gut to assess cell-mediated immune parameters and immune-related gene expression. At the same time, fish from each dietary group were challenged against Vibrio anguillarum and survival was monitored. Cell-mediated immune parameters such as the extracellular superoxide and nitric oxide decreased in fish fed arginine-supplemented diets. Interleukins and immune-cell marker transcripts were down-regulated by the highest supplementation level. Disease resistance data were in accordance with a generally depressed immune status, with increased susceptibility to vibriosis in fish fed arginine supplemented diets. Altogether, these results suggest a general inhibitory effect of arginine on the immune defences and disease resistance of European seabass. A mass accuracy sensitive probability based scoring algorithm for database searching of tandem mass spectrometry data
<p>Abstract</p> <p>Background</p> <p>Liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) has become one of the most used tools in mass spectrometry based proteomics. Various algorithms have since been developed to automate the process for modern high-throughput LC-MS/MS experiments.</p> <p>Results</p> <p>A probability based statistical scoring model for assessing peptide and protein matches in tandem MS database search was derived. The statistical scores in the model represent the probability that a peptide match is a random occurrence based on the number or the total abundance of matched product ions in the experimental spectrum. The model also calculates probability based scores to assess protein matches. Thus the protein scores in the model reflect the significance of protein matches and can be used to differentiate true from random protein matches.</p> <p>Conclusion</p> <p>The model is sensitive to high mass accuracy and implicitly takes mass accuracy into account during scoring. High mass accuracy will not only reduce false positives, but also improves the scores of true positive matches. The algorithm is incorporated in an automated database search program MassMatrix.</p
Analogue peptides for the immunotherapy of human acute myeloid leukemia

However, responses are often short-lived and rarely induce notable memory responses. The reason is that self-antigens have already been presented to the immune system as the tumor develops, leading to tolerance or some degree of host tumor cell destruction. To try to break tolerance against self-antigens, one of the methods employed has been to modify peptides at the anchor residues to enhance their ability to bind major histocompatibility complex molecules, extending their exposure to the T-cell receptor. These modified or analogue peptides have been investigated as stimulators of the immune system in patients with different cancers with variable but sometimes notable success. In this review we describe the background and recent developments in the use of analogue peptides for the immunotherapy of acute myeloid leukemia describing knowledge useful for the application of analogue peptide treatments for other malignancies
Searches for the Shell Swept up by the Stellar Wind from Cyg OB2
We investigated the kinematics of ionized gas in an extended (20 degrees by
15 degrees) region containing the X-ray Superbubble in Cygnus with the aim of
finding the shell swept up by a strong wind from Cyg OB2. H-alpha observations
were carried out with high angular and spectral resolutions using a Fabry-Perot
interferometer attached to the 125-cm telescope at the Crimean Observatory of
the Sternberg Astronomical Institute. We detected high-velocity gas motions,
which could result from the expansion of the hypothetical shell at a velocity
of 25-50 km/s. Given the number of OB stars increased by Knoedlseder (2000) by
an order of magnitude, Cyg OB2 is shown to possess a wind that is strong enough
[Lw ~= (1-2)x10^39 erg/s] to produce a shell comparable in size to the X-ray
Superbubble and to a giant system of optical filaments. Based on our
measurements and on X-ray and infrared observations, we discuss possible
observational manifestations of the shell swept up by the wind.Comment: 14 pages, Astronomy Letter
Revival of the magnetar PSR J1622-4950: observations with MeerKAT, Parkes, XMM-Newton, Swift, Chandra, and NuSTAR
New radio (MeerKAT and Parkes) and X-ray (XMM-Newton, Swift, Chandra, and
NuSTAR) observations of PSR J1622-4950 indicate that the magnetar, in a
quiescent state since at least early 2015, reactivated between 2017 March 19
and April 5. The radio flux density, while variable, is approximately 100x
larger than during its dormant state. The X-ray flux one month after
reactivation was at least 800x larger than during quiescence, and has been
decaying exponentially on a 111+/-19 day timescale. This high-flux state,
together with a radio-derived rotational ephemeris, enabled for the first time
the detection of X-ray pulsations for this magnetar. At 5%, the 0.3-6 keV
pulsed fraction is comparable to the smallest observed for magnetars. The
overall pulsar geometry inferred from polarized radio emission appears to be
broadly consistent with that determined 6-8 years earlier. However, rotating
vector model fits suggest that we are now seeing radio emission from a
different location in the magnetosphere than previously. This indicates a novel
way in which radio emission from magnetars can differ from that of ordinary
pulsars. The torque on the neutron star is varying rapidly and unsteadily, as
is common for magnetars following outburst, having changed by a factor of 7
