    Ares I Aerodynamic Testing at the Boeing Polysonic Wind Tunnel

    Throughout three full design analysis cycles, the Ares I project within the Constellation program has consistently relied on the Boeing Polysonic Wind Tunnel (PSWT) for aerodynamic testing of the subsonic, transonic and supersonic portions of the atmospheric flight envelope (Mach=0.5 to 4.5). Each design cycle required the development of aerodynamic databases for the 6 degree-of-freedom (DOF) forces and moments, as well as distributed line-loads databases covering the full range of Mach number, total angle-of-attack, and aerodynamic roll angle. The high fidelity data collected in this facility has been consistent with the data collected in NASA Langley s Unitary Plan Wind Tunnel (UPWT) at the overlapping condition ofMach=1.6. Much insight into the aerodynamic behavior of the launch vehicle during all phases of flight was gained through wind tunnel testing. Important knowledge pertaining to slender launch vehicle aerodynamics in particular was accumulated. In conducting these wind tunnel tests and developing experimental aerodynamic databases, some challenges were encountered and are reported as lessons learned in this paper for the benefit of future crew launch vehicle aerodynamic developments

    N-Acetylmuramic Acid (MurNAc) Auxotrophy of the Oral Pathogen Tannerella forsythia: Characterization of a MurNAc Kinase and Analysis of Its Role in Cell Wall Metabolism

    Tannerella forsythia is an anaerobic, Gram-negative oral pathogen that thrives in multispecies gingival biofilms associated with periodontitis. The bacterium is auxotrophic for the commonly essential bacterial cell wall sugar N-acetylmuramic acid (MurNAc) and, thus, strictly depends on an exogenous supply of MurNAc for growth and maintenance of cell morphology. A MurNAc transporter (Tf_MurT; Tanf_08375) and an ortholog of the Escherichia coli etherase MurQ (Tf_MurQ; Tanf_08385) converting MurNAc-6-phosphate to GlcNAc-6-phosphate were recently described for T. forsythia. In between the respective genes on the T. forsythia genome, a putative kinase gene is located. In this study, the putative kinase (Tf_MurK; Tanf_08380) was produced as a recombinant protein and biochemically characterized. Kinetic studies revealed Tf_MurK to be a 6-kinase with stringent substrate specificity for MurNAc exhibiting a 6 × 104 - fold higher catalytic efficiency (kcat/Km) for MurNAc than for N-acetylglucosamine (GlcNAc) with kcat values of 10.5 s−1 and 0.1 s−1 and Km values of 200 µM and 116 mM, respectively. The enzyme kinetic data suggest that Tf_MurK is subject to substrate inhibition (Ki[S] = 4.2 mM). To assess the role of Tf_MurK in the cell wall metabolism of T. forsythia, a kinase deletion mutant (1Tf_murK::erm) was constructed. This mutant accumulated MurNAc intracellularly in the exponential phase, indicating the capability to take up MurNAc, but inability to catabolize MurNAc. In the stationary phase, the MurNAc level was reduced in the mutant, while the level of the peptidoglycan precursor UDP-MurNAc-pentapeptide was highly elevated. Further, according to scanning electron microscopy evidence, the 1Tf_murK::erm mutant was more tolerant toward low MurNAc concentration in the medium (below 0.5 µg/ml) before transition from healthy, rod-shaped to fusiform cells occurred, while the parent strain required > 1 µg/ml MurNAc for optimal growth. These data reveal that T. forsythia readily catabolizes exogenous MurNAc but simultaneously channels a proportion of the sugar into peptidoglycan biosynthesis. Deletion of Tf_murK blocks MurNAc catabolism and allows the direction of MurNAc solely to peptidoglycan biosynthesis, resulting in a growth advantage in MurNAc-depleted medium. This work increases our understanding of the T. forsythia cell wall metabolism and may pave new routes for lead finding in the treatment of periodontitis

    Space Launch System Liftoff and Transition Aerodynamic Characterization in the NASA Langley 14- by 22-Foot Subsonic Wind Tunnel

    A 1.75% scale force and moment model of the Space Launch System was tested in the NASA Langley Research Center 14- by 22-Foot Subsonic Wind Tunnel to quantify the aerodynamic forces that will be experienced by the launch vehicle during its liftoff and transition to ascent flight. The test consisted of two parts: the first was dedicated to measuring forces and moments for the entire range of angles of attack (0deg to 90deg) and roll angles (0 deg. to 360 deg.). The second was designed to measure the aerodynamic effects of the liftoff tower on the launch vehicle for ground winds from all azimuthal directions (0 deg. to 360 deg.), and vehicle liftoff height ratios from 0 to 0.94. This wind tunnel model also included a set of 154 surface static pressure ports. Details on the experimental setup, and results from both parts of testing are presented, along with a description of how the wind tunnel data was analyzed and post-processed in order to develop an aerodynamic database. Finally, lessons learned from experiencing significant dynamics in the mid-range angles of attack due to steady asymmetric vortex shedding are presented

    Empirical Evaluation of the Difficulty of Finding a Good Value of k for the Nearest Neighbor

    As an analysis of the classification accuracy bound for the Nearest Neighbor technique, in this work we have studied if it is possible to find a good value of the parmeter k for each example according to their attribute values. Or at least, if there is a pattern for the parameter k in the original search space. We have carried out different approaches based onthe Nearest Neighbor technique and calculated the prediction accuracy for a group of databases from the UCI repository. Based on the experimental results of our study, we can state that, in general, it is not possible to know a priori a specific value of k to correctly classify an unseen example

    Space Launch System Ascent Static Aerodynamic Database Development

    This paper describes the wind tunnel testing work and data analysis required to characterize the static aerodynamic environment of NASA's Space Launch System (SLS) ascent portion of flight. Scaled models of the SLS have been tested in transonic and supersonic wind tunnels to gather the high fidelity data that is used to build aerodynamic databases. A detailed description of the wind tunnel test that was conducted to produce the latest version of the database is presented, and a representative set of aerodynamic data is shown. The wind tunnel data quality remains very high, however some concerns with wall interference effects through transonic Mach numbers are also discussed. Post-processing and analysis of the wind tunnel dataset are crucial for the development of a formal ascent aerodynamics database

    Starenje populacije je dominantno demografsko obilježje razvijenih zemalja. Stogodišnjaci su selekcionirana skupina i samo jedna od 7.000 do 10.000 osoba dosegne tu dob. Čimbenici dugovječnosti vjerojatno su brojni i uključuju gensko predodređenje (lokus na 4. kromosomu), zdrav okoliš i zdrave životne navike (prehrana s malo kalorija), redovita tjelesna i psihička aktivnost, kao i dostupnost te učinkovitost zdravstvene zaštite s primjenom geroprofi lakse. Stogodišnjaci se adaptiraju na novi život i na gubitak tjelesnih funkcija koji bivaju postupno sve izraženiji kako se dob povisuje. Granice ljudskog života produžuju se - do sada najstarija poznata osoba doživjela je 128 godina. Pojedina zemljopisna područja bilježe izrazito veći broj stogodišnjaka. Navedene su i neke dugovječne osobe s više od 100 godina u svijetu i na području Republike Hrvatske i nekih susjednih zemalja. Iako se uglavnom smatra da se granica trajanja života čovjeka ne može produžiti iznad 120 godina, za sada je ipak teško predvidjeti gdje su njezine granice.Aged population dominates in developed countries. Centenarians are a select group, and only one in 7,000 to 10,000 reach that age. Factors of longevity are numerous and include genetic predisposition (a locus on chromosome 4), environment, healthy lifestyle (hypocaloric diet, regular physical and mental exercise), accessible health services, and effi cient health protection at old age. Centenarians are well adapted to the new life and compensate for the loss of functions with age. The limits of human life are extended, so that nowadays the oldest person has reached the age of 128. Some geographic areas are characterised by higher numbers of centenarians. This article mentions a few individuals who outlived 100 years in the world, Croatia, and neighbouring countries. Although some argue that the limits of human life cannot be extended over the age of 120 years, for now we cannot predict the actual limits of human life

    Exploring the performance of resampling strategies for the class imbalance problem

    The present paper studies the influence of two distinct factors on the performance of some resampling strategies for handling imbalanced data sets. In particular, we focus on the nature of the classifier used, along with the ratio between minority and majority classes. Experiments using eight different classifiers show that the most significant differences are for data sets with low or moderate imbalance: over-sampling clearly appears as better than under-sampling for local classifiers, whereas some under-sampling strategies outperform over-sampling when employing classifiers with global learning

    OWA-FRPS: A Prototype Selection method based on Ordered Weighted Average Fuzzy Rough Set Theory

    The Nearest Neighbor (NN) algorithm is a well-known and effective classification algorithm. Prototype Selection (PS), which provides NN with a good training set to pick its neighbors from, is an important topic as NN is highly susceptible to noisy data. Accurate state-of-the-art PS methods are generally slow, which motivates us to propose a new PS method, called OWA-FRPS. Based on the Ordered Weighted Average (OWA) fuzzy rough set model, we express the quality of instances, and use a wrapper approach to decide which instances to select. An experimental evaluation shows that OWA-FRPS is significantly more accurate than state-of-the-art PS methods without requiring a high computational cost.Spanish Government TIN2011-2848

    Cisplatin and vinorelbine first-line chemotherapy in non-resectable malignant pleural mesothelioma

    The aim was to evaluate the activity of cisplatin and vinorelbine in previously untreated, inoperable patients having histologically verified malignant pleural mesothelioma (MPM), normal organ function, and performance status 0–2. Treatment was vinorelbine 25 mg m−2 i.v. weekly and cisplatin 100 mg m−2 i.v. every 4 weeks with hydration and standard prophylactic antiemetic treatment. Patients gave written informed consent. Characteristics of 54 consecutive patients were: males 85%, epithelial subtype 74%, IMIG stages III and IV 35 and 46%, performance status 0, 1, and 2, 26, 69, and 6%, and median age 63 years (31–78 years). CTC grade 3 or 4 toxicity occurred with respect to leukocytopenia (48% of patients, grade 4 in 13%), nausea (13%), neurotoxicity (11%), nephrotoxicity (4%), and other toxicities (9%). There were no toxic deaths. The median number of cycles was four. The fraction of patients alive at 1-, 2-, and 3-years were 61, 31, and 4%, respectively, and median survival and median time to progression were 16.8 months (0.5 to 46.4 +months) and 7.2 months (1.6 to 40.6 + months). There were two CRs and 14 PRs (response rate 29.6%). Cisplatin and intravenous vinorelbine is a highly active regimen in MPM with a response rate and survival comparable to the most active regimens so far reported
