Acid stress response in campylobacter jejuni

and the expressive increasing of incidence of Campylobacter jejuni in recent years as
etiologic agent of human campylobacteriosis, this work aimed to evaluate the ability of C.
jejuni to survive under acid stress. Strains of different origins (reference, turkey isolates) and
tolerance to particular stresses (acid and temperature) were tested for survival in liquid
defined media with pH values of 5.0 and 7.0 adjusted with 1M HCl. Experimental conditions
were performed at 2 different temperatures: 4 ºC and 37 ºC. C. jejuni cells were found to be
very sensitive to acid stress at 37 ºC, whereas they were more resistant at 4 ºC. A strain effect
was observed at 37 ºC. Temperature of 37 ºC combined with acid stress allowed a rapid
decrease in the C. jejuni population, whereas low temperature (4ºC) considerably decreased
the effect of acid stress. The survival curves were either described by the Weibull or
traditional first-order model and goodness of fit of these models was investigated. Regression
coefficients (R2), root mean square error (RMSE) and correlation plots suggestted that
Weibull model produced a better fit to the data than the traditional model.
Fluorescence Ratio Imaging Microscopy (FRIM) was used to determine intracellular pH (pHi)
as an indicator of the physiological state of C. jejuni cells at the single cell level after
treatment with hydrochloride acid (1M HCl, pH 4.0) in liquid defined medium. For all the
tested strains pHi of healthy cells was found to be above 7.0. After exposure to HCl in liquid
medium an immediate decline in pHi to 5.5 (detection limit) was observed in the majority of
cells (75%) of one strain within 15 minutes. The FRIM results revealed that the
subpopulations with pHi>5.5 increased for this strain, especially subpopulations with
5.5<pHi<6.0 and 6.0<pHi<6.5 following 200 min. of exposure to HCl. This indicates that
some strains of C. jejuni may employ certain recovery strategies to extrude protons and to
increase pHi.RESUMO -
Considerando que a tolerância ao ácido é um importante factor de virulência de agentes
patogénicos veiculados por alimentos, condicionando a sobrevivência dos microrganismos, e
o expressivo aumento, nos últimos anos, da incidência de Campylobacter jejuni como agente
etiológico de campilobacteriose humana, este trabalho teve como objectivo avaliar a
capacidade do patogénico C. jejuni sobreviver em condições de stresse ácido. Estirpes de
diferentes origens (referência, isolados de peru) sujeitas a diferentes factores de stresse
(acidez e temperatura) foram testadas avaliando-se a sua sobrevivência em meio líquido (pH
5.0 e 7.0). As condições experimentais foram efectuadas a duas temperaturas diferentes: 4 ºC
e 37 ºC. Células de C. jejuni mostraram-se muitos sensíveis a stresse ácido a 37 ºC, enquanto
a 4 ºC foram mais resistentes. O efeito estirpe foi evidenciado a 37 ºC. A temperatura 37ºC
em combinação com um pH 5.0 (stresse ácido) causou rápido decréscimo da população de C.
jejuni, enquanto que a 4ºC o efeito do stresse ácido diminuiu consideravelmente. As curvas de
sobrevivência foram descritas pelos modelos Weibull ou pelo modelo clássico de inactivação
de primeira ordem, tendo-se concluído através dos coeficientes de regressão (R2), raízes
quadradas do erro médio (RMSE) e gráficos de correlação que o modelo Weibull se adequa
melhor aos dados apresentados que o modelo tradicionalmente utilizado.
O racio da emissão de fluorescência medida pela técnica de microscopia óptica invertida foi
utilizado para determinar o pH intracelular (pHi) como indicador do estado fisiológico das
células de C. jejuni, a um nível celular único, após aplicação de um tratamento com ácido
clorídrico (1M HCl, pH 4.0) em meio de cultura líquido. Para todas as estirpes testadas, o pHi
de células saudáveis encontrava-se acima de 7.0. Após exposição a HCl em meio líquido, foi
detectado para uma das estirpes, um decréscimo imediato do pHi para 5.5 (limite de detecção)
na maioria das células observadas (75%) ao fim de 15 minutos. Os resultados da medição de
fluorescência revelaram um aumento de subpopulações com pHi>5.5 para essa estirpe,
nomeadamente subpopulações com 5.5<pHi<6.0 e 6.0<pHi<6.5 em 200 min. de exposição a
HCl. Estes resultados indicam que algumas estirpes de C. jejuni poderão utilizar determinadas
estratégias de recuperação para exteriorizar protões e elevar o pHi
A methodology to produce geographical information for land planning using very-high resolution images
Actualmente, os municípios são obrigados a produzir, no âmbito da elaboração dos instrumentos de gestão territorial, cartografia homologada pela autoridade nacional. O Plano Director Municipal (PDM) tem um período de vigência de 10 anos. Porém, no que diz respeito à cartografia para estes planos, principalmente em municípios onde a pressão urbanística é elevada, esta periodicidade não é compatível com a dinâmica de alteração de uso do solo. Emerge assim, a necessidade de um processo de produção mais eficaz, que permita a obtenção de uma nova cartografia de base e temática mais frequentemente. Em Portugal recorre-se à fotografia aérea como informação de base para a produção de cartografia de grande escala. Por um lado, embora este suporte de informação resulte em mapas bastante rigorosos e detalhados, a sua produção têm custos muito elevados e consomem muito tempo.
As imagens de satélite de muito alta-resolução espacial podem constituir uma alternativa, mas sem substituir as fotografias aéreas na produção de cartografia temática, a grande escala.
O tema da tese trata assim da satisfação das necessidades municipais em informação geográfica actualizada. Para melhor conhecer o valor e utilidade desta informação, realizou-se um inquérito aos municípios Portugueses. Este passo foi essencial para avaliar a pertinência e a utilidade da introdução de imagens de satélite de muito alta-resolução espacial na cadeia de procedimentos de actualização de alguns temas, quer na cartografia de base quer na cartografia temática.
A abordagem proposta para solução do problema identificado baseia-se no uso de imagens de satélite e outros dados digitais em ambiente de Sistemas de Informação Geográfica.
A experimentação teve como objectivo a extracção automática de elementos de interesse municipal a partir de imagens de muito alta-resolução espacial (fotografias aéreas ortorectificadas, imagem QuickBird, e imagem IKONOS), bem como de dados altimétricos (dados LiDAR).
Avaliaram-se as potencialidades da informação geográfica extraídas das imagens para fins cartográficos e analíticos. Desenvolveram-se quatro casos de estudo que reflectem diferentes usos para os dados geográficos a nível municipal, e que traduzem aplicações com exigências diferentes. No primeiro caso de estudo, propõe-se uma metodologia para actualização periódica de cartografia a grande escala, que faz uso de fotografias aéreas

ortorectificadas na área da Alta de Lisboa. Esta é uma aplicação quantitativa onde as qualidades posicionais e geométricas dos elementos extraídos são mais exigentes. No segundo caso de estudo, criou-se um sistema de alarme para áreas potencialmente alteradas, com recurso a uma imagem QuickBird e dados LiDAR, no Bairro da Madre de Deus, com objectivo de auxiliar a actualização de cartografia de grande escala. No terceiro caso de estudo avaliou-se o potencial solar de topos de edifícios nas Avenidas Novas, com recurso a dados LiDAR. No quarto caso de estudo, propõe-se uma série de indicadores municipais de monitorização territorial, obtidos pelo processamento de uma imagem IKONOS que cobre toda a área do concelho de Lisboa. Esta é uma aplicação com fins analíticos onde a qualidade temática da extracção é mais relevante.Currently, the Portuguese municipalities are required to produce homologated cartography, under the Territorial Management Instruments framework. The Municipal Master Plan (PDM) has to be revised every 10 years, as well as the topographic and thematic maps that describe the municipal territory. However, this period is inadequate for representing counties where urban pressure is high, and where the changes in the land use are very dynamic. Consequently, emerges the need for a more efficient mapping process, allowing obtaining recent geographic information more often. Several countries, including Portugal, continue to use aerial photography for large-scale mapping. Although this data enables highly accurate maps, its acquisition and visual interpretation are very costly and time consuming.
Very-High Resolution (VHR) satellite imagery can be an alternative data source, without replacing the aerial images, for producing large-scale thematic cartography.
The focus of the thesis is the demand for updated geographic information in the land planning process. To better understand the value and usefulness of this information, a survey of all Portuguese municipalities was carried out. This step was essential for assessing the relevance and usefulness of the introduction of VHR satellite imagery in the chain of procedures for updating land information.
The proposed methodology is based on the use of VHR satellite imagery, and other digital data, in a Geographic Information Systems (GIS) environment.
Different algorithms for feature extraction that take into account the variation in texture, color and shape of objects in the image, were tested. The trials aimed for automatic extraction of features of municipal interest, based on aerial and satellite high-resolution (orthophotos, QuickBird and IKONOS imagery) as well as elevation data (altimetric information and LiDAR data).
To evaluate the potential of geographic information extracted from VHR images, two areas of application were identified: mapping and analytical purposes. Four case studies that reflect different uses of geographic data at the municipal level, with different accuracy requirements, were considered. The first case study presents a methodology for periodic updating of large-scale maps based on orthophotos, in the area of Alta de Lisboa. This is a situation where the positional and geometric accuracy of the extracted information are more demanding, since technical mapping standards must be complied. In the second case study, an alarm system that indicates the location of potential changes in building areas, using a QuickBird image and LiDAR data, was developed for the area of Bairro da Madre de Deus. The goal of the system is to assist the updating of large scale mapping, providing a layer that can be used by the municipal technicians as the basis for manual editing. In the third case study, the analysis of the most suitable roof-tops for installing solar systems, using LiDAR data, was performed in the area of Avenidas Novas. A set of urban environment indicators obtained from VHR imagery is presented. The concept is demonstrated for the entire city of Lisbon, through IKONOS imagery processing. Communicating (in) wine tourism: what are the paths for harmonising the sector and the Translation Process?
Wine tourism is an emerging area of specialisation to which several areas of knowledge (marketing, economics, anthropology, viticulture, etc.) converge. Portugal’s wine culture has a long tradition and is internationally recognised, placing it at the forefront of economic, professional, and academic initiatives in this sector. Communication between specialists and between specialists and national and international wine tourists requires an international terminology that is, simultaneously, mindful of tradition and that favours inclusive, efficient, and competitive trade exchanges. 226
Our research aims to contribute to the terminological harmonisation of Portuguese wine tourism, even though no ISO/IPQ standards have been issued in this emerging transdisciplinary area. In this article, two comparable academic sub-corpora (10 theses) on wine tourism will be analysed. Their comparison was carried out with the Sketch Engine programme, which allows, in addition to corpus management, to extract terms, identify keywords and represent their conceptual organisation. Our methodological approach included the analysis of the results based on the 50 most relevant terms in each of the corpus. Ten case studies taken from the corpora emphasise the diversity of terminogenic patterns in each language, the influence of cultural factors in the specialised wine tourism terminology of both languages, and, lastly, the influence of the English language on Portuguese wine tourism terminology. Evaluating the Impact of the Urban Requalification Project of Praça Duque De Saldanha and Avenida Da República in Lisbon

SFRH/BPD/76893/2011In dense urban areas, reducing traffic and increasing green areas is foreseen as a way of promoting urban comfort. Using the urban requalification project for Lisbon’s Avenidas Novas neighbourhood as a case study, the effects of vegetation on microclimate and urban comfort are evaluated. In this context, the ENVI-met software is used to model the present and the future urban scenario. The simulation results indicate: i) increased urban comfort in the morning and in the afternoon resulting from the decrease in temperature due to the presence of new green areas foreseen by the urban project; ii) a reduction of up to 3 degrees in the morning (9h) and up to 3 degrees in the afternoon (15h); iii) the model implemented in ENVI-met suits the urban and environmental characteristics of Lisbon and it is, therefore, desirable that future urban rehabilitation projects should consider ex ante simulation of this typepublishersversionpublishe
High-frequency audiometry hearing on monitoring of individuals exposed to occupational noise: a systematic review
The literature reports on high-frequency audiometry as one of the exams used on hearing monitoring of individuals exposed to high sound pressure in their work environment, due to the method́s greater sensitivity in early identification of hearing loss caused by noise. The frequencies that compose the exam are generally between 9 KHz and 20KHz, depending on the equipment. This study aims to perform a retrospective and secondary systematic revision of publications on high-frequency audiometry on hearing monitoring of individuals exposed to occupational noise. This systematic revision followed the methodology proposed in the Cochrane Handbook, focusing on the question: “Is High-frequency Audiometry more sensitive than Conventional Audiometry in the screening of early hearing loss individuals exposed to occupational noise?” The search was based on PubMed data, Base, Web of Science (Capes), Biblioteca Virtual em Saúde (BVS), and in the references cited in identified and selected articles. The search resulted in 6059 articles in total. Of these, only six studies were compatible with the criteria proposed in this study. The performed meta-analysis does not definitively answer the study's proposed question. It indicates that the 16 KHz high frequency audiometry (HFA) frequency is sensitive in early identification of hearing loss in the control group (medium difference (MD = 8.33)), as well as the 4 KHz frequency (CA), this one being a little less expressive (MD = 5.72). Thus, others studies are necessary to confirm the HFA importance for the early screening of hearing loss on individuals exposed to noise at the workplace.20328128

O direito à informação não é uma regalia, mas sim a expressão da liberdade e participação do cidadão. A informação transparente dos gastos públicos transformou-se em lei no ano de 2011 e a partir desta data, todo cidadão com acesso à internet pode consultar os gastos administrativos do país. O acesso à informação faz o cidadão ser mais participativo politicamente. Este trabalho consiste em analisar todos os dados no período de fevereiro de 2014 a junho de 2018 via portal de transparência da cidade de Florianópolis, do balancete dos vereadores do município, implementando uma ferramenta que os torna graficamente visíveis e permite, em sequência, uma análise não fragmentada do orçamento governamental. O desenvolvimento da ferramenta utilizou conceitos de banco de dados e programação PHP. Inicialmente é feita uma pesquisa bibliográfica, depois o levantamento dos dados do portal da transparência de Florianópolis, a conversão dos dados de PDF para CSV, o desenvolvimento em PHP para interpretar os dados em CSV e popular o banco, a leitura do banco de dados para apresentação gráfica e por final elabora-se um questionário para verificar a utilidade do sistema de visualização
A Novel Rework Costing Methodology Applied To a Bus Manufacturing Company
This paper focuses on the quality costs analysis in the automotive industry, specifically a bus manufacturing company. The main goal was to improve the quality costs indicator, by providing means to evaluate the failure cost within the productive process. Kaizen-lean principles were used as methodology. A quality cost analysis was made based on literature review and in an ISO standard in quality costs. Process analysis was made based on the PDCA (Plan, Do, Check, Act) cycle. As the main achievements of this article, the quality costs were identified as well as its sources. Then, by collecting a very significant amount of data, from various departments, regarding each of the identified costs, it was possible to measure these costs and create a database to scorecard them – including rework costs. Due to the lack of rework costs information, a great part of this analysis focused on data collection and estimating/calculating these associated costs. Thus, it was possible to develop associated KPI’s (Key Performance Indicators) to support decision making and a graphic indicator to show the overall balance between quality costs and non-quality costs. In this process, some difficulties were found as well as some opportunities for improvement. The critical ones were implemented during this project, while the others are scheduled for future work. In terms of a more technological approach to these studies, some autonomous systems can exist, although it comes at a great cost. However, the 4.0 Industry concept is evolving and fits perfectly in the quality costs monitorization context: an integrated information system with a global data network allows for a quick check-up of all the quality costs, by category, while freeing employees from doing this work manually.info:eu-repo/semantics/publishedVersio
Construcción del formulario de evaluación de la competencia del autocuidado en la persona con ostomía de eliminación intestinal
Background: An intestinal stoma implies multiple and significant changes that tend to be overcome through the development of
ostomy self-care skills. The identification of the patient’s specific needs during this adaptation process will help nurses’ decisionmaking
and contribute to a rigorous nursing diagnosis process, thus improving the quality and continuity of care delivery to this
population group.
Objectives: To develop an instrument for assessing the self-care competence of the person with an intestinal stoma.
Methodology: Methodological study consisting of the development of a form. Content validity was ensured by expert review
using a focus group. The form was applied to a convenience sample of 180 participants.
Results: A form was developed with 45 measurement indicators divided into 6 domains. It showed very good internal consistency
(overall Cronbach’s alpha > .90) and solid content validity.
Conclusion: The form proved to be a reliable and easy to use method to assess the development of self-care competence in
people with an intestinal stoma.Enquadramento: A criação de uma ostomia intestinal impõe
à pessoa múltiplas e significativas mudanças que tendem a
ser superadas através do desenvolvimento da competência de
autocuidado ao estoma. Conhecer as necessidades específicas da
pessoa a vivenciar este processo de adaptação auxiliará na tomada
de decisão dos enfermeiros, imprimindo maior rigor ao processo
de diagnóstico de enfermagem, com repercussões na melhoria e
na continuidade da assistência a este grupo populacional.
Objetivos: Construir um instrumento de avaliação da competência
de autocuidado na pessoa com ostomia intestinal.
Metodologia: Estudo metodológico que consistiu na construção
de um formulário. A validade de conteúdo foi garantida pela
avaliação de peritos através de um focus group. O formulário foi
aplicado a uma amostra de conveniência de 180 participantes.
Resultados: Formulário com 45 indicadores de medida,
agrupados em 6 domínios, apresentando muito boa consistência
interna (alfa de Cronbach global > 0,90) e reconhecida validade
de conteúdo.
Conclusão: O formulário construído mostrou ser fidedigno e de
fácil aplicação na avaliação do desenvolvimento da competência
de autocuidado à pessoa com ostomia intestinal.Marco contextual: La creación de una ostomía intestinal

tienden a superarse a través del desarrollo de la competencia del

en el proceso del diagnóstico de enfermería, lo que repercutirá
en la mejora y la continuidad de la asistencia a este tipo de
pacientes.
