    Vuodesta 2008 alkaen Euroopan unioni (EU) on joutunut pohtimaan ratkaisuja uudenlaisiin ongelmiin. 2010-lukua voikin pitää eräänlaisena kriisien vuosikymmenenä. Unionin on täytynyt miettiä, miten auttaa jäsenmaitansa ja niiden pankkeja velkaongelmissa sekä miten vastata nationalistisen ideologian nousuun, työttömyyteen, terrorismin luomaan epävakauteen ja turvapaikanhakijoiden määrän kasvuun. Vuosikymmeniä kestäneen laajentumisjakson jälkeen yhteistyön sujuminen on Euroopassa rakoillut kriisien takia. Esimerkiksi Unkari, Puola ja Kreikka ovat kyseenalaistaneet unionin legitimiteetin. Kesäkuussa 2016 unioni joutui täysin uudenlaiseen tilanteeseen, kun britit äänestivät EU-eron puolesta. Britanniaa vahvasti jakanut brexit-kampanjointi herätti myös keskustelun median objektiivisuudesta ja vaikutusvallasta. Äänestyksen yhteydessä osa äänestäjistä perusteli erohaluaan identiteetillä: he eivät tunteneet itseään eurooppalaiseksi vaan britiksi. Pro gradu -tutkielmassani tutkin diskurssianalyysin avulla ja vertailevalla otteella, miten EU:n tulevaisuus esitettiin kolmessa eurooppalaisessa lehdessä brexit-äänestyksen jälkeen. Tutkimukseni keskeisiä käsitteitä olivat identiteetti ja kuviteltu yhteisö. Diskurssianalyysin pohjana käytin Eero Vaaran diskurssijakoa (Vaara 2014). Tutkimukseni lähtökohta oli, että medialla on valtaa luoda yhteisöjä ja vaikuttaa siihen, millaisessa maailmassa lukijat kokevat elävänsä. Guardianin, Helsingin Sanomien ja Süddeutsche Zeitungin teksteistä piirtyi EU-myönteinen kuva, jossa brexit näytettiin Britannian kriisinä. Unioni ja sen legitimiteetti esitettiin pääosin vahvoina, eikä sen päätöksiä tai politiikkaa pidetty syynä Britannian eroon, vaikka EU:lta toivottiinkin aiempaa ihmislähtöisempää politiikkaa. Sekä Britanniaa johtavat konservatiivit että EU:n elinten ja jäsenvaltioiden poliittinen eliitti yrittivät eron jälkeen vakuutella mediassa, että Britannian lähtö saadaan hoidettua sulavasti. Äänestyksen jälkeen kansan toivottiin pysyvän tyynenä. Analyysini perusteella lehdet tukivat pääasiassa uusliberaalia, EU-myönteistä ideologiaa ja näkivät nationalismin sekä populismin koko Euroopan kannalta suurina uhkina

    Challenges of Managing a Network Business

    This study aims at identifying and analysing the key challenges in managing a network business. The starting point of the paper is the alleged need for new kinds of theoretical frameworks which take into account value creation situations businesses face today. Hence, this paper introduces the value co-production framework as an alternative perspective to the existing approaches built in the industrial era (i.e., value chain thinking). This framework elevates knowledge and relationships in the locus of business and argues for reconfiguration of roles, actions, and interactions among the networked actors. Based on our study on a network-like, two years old joint venture firm, we demonstrate how the value co-production framework functions in practice. Moreover, we suggest that the future management competence lies in company’s ability to manage interactions, not individuals or individual companies. Thus, studying different kinds of network approaches in relation to proposed framework would give a deeper understanding of the ways by which networks and relationships drive value co-production

    Multiple roles for UV RESISTANCE LOCUS8 in regulating gene expression and metabolite accumulation in arabidopsis under solar ultraviolet radiation

    Photomorphogenic responses triggered by low fluence rates of ultraviolet B radiation (UV-B; 280–315 nm) are mediated by the UV-B photoreceptor UV RESISTANCE LOCUS8 (UVR8). Beyond our understanding of the molecular mechanisms of UV-B perception by UVR8, there is still limited information on how the UVR8 pathway functions under natural sunlight. Here, wild-type Arabidopsis (Arabidopsis thaliana) and the uvr8-2 mutant were used in an experiment outdoors where UV-A (315–400 nm) and UV-B irradiances were attenuated using plastic films. Gene expression, PYRIDOXINE BIOSYNTHESIS1 (PDX1) accumulation, and leaf metabolite signatures were analyzed. The results show that UVR8 is required for transcript accumulation of genes involved in UV protection, oxidative stress, hormone signal transduction, and defense against herbivores under solar UV. Under natural UV-A irradiance, UVR8 is likely to interact with UV-A/blue light signaling pathways to moderate UV-B-driven transcript and PDX1 accumulation. UVR8 both positively and negatively affects UV-A-regulated gene expression and metabolite accumulation but is required for the UV-B induction of phenolics. Moreover, UVR8-dependent UV-B acclimation during the early stages of plant development may enhance normal growth under long-term exposure to solar UV

    Cystatin B deficiency results in sustained histone H3 tail cleavage in postnatal mouse brain mediated by increased chromatin-associated cathepsin L activity

    Cystatin B (CSTB) is a cysteine cathepsin inhibitor whose biallelic loss-of-function mutations in human result in defects in brain development and in neurodegeneration. The physiological function of CSTB is largely unknown, and the mechanisms underlying the human brain diseases remain poorly understood. We previously showed that CSTB modulates the proteolysis of the N-terminal tail of histone H3 (H3cs1) during in vitro neurogenesis. Here we investigated the significance of this mechanism in postnatal mouse brain. Spatiotemporal analysis of H3cs1 intensity showed that while H3cs1 in wild-type (wt) mice was found at varying levels during the first postnatal month, it was virtually absent in adult brain. We further showed that the high level of H3cs1 coincides with chromatin association of de novo synthesized cathepsin L suggesting a role for nuclear cathepsin L in brain development and maturation. On the contrary, the brains of Cstb(-/-) mice showed sustained H3cs1 proteolysis to adulthood with increased chromatin-associated cathepsin L activity, implying that CSTB regulates chromatin-associated cathepsin L activity in the postnatal mouse brain. As H3 tail proteolysis has been linked to cellular senescence in vitro, we explored the presence of several cellular senescence markers in the maturing Cstb(-/-) cerebellum, where we see increased levels of H3cs1. While several markers showed alterations in Cstb(-/-) mice, the results remained inconclusive regarding the association of deficient CSTB function with H3cs1-induced senescence. Together, we identify a molecular role for CSTB in brain with implications for brain development and disease.Peer reviewe

    DigiWeb - a workflow environment for quality assurance of transcription in digitization of natural history collections

    Data produced by digitization increases the scientific use of natural history collections. However, in mass digitization, attention must be paid to the flawless management of the workflows, and high quantities of end results should not be compromised by a low standard of quality. A web-based environment DigiWeb was created for controlling the workflow of transcribing data from images of natural history specimens. Using DigiWeb, it was possible to manage the workflow of transcription and data proofing, include all participants to the workflow, allow collaboration and training, and also to provide useful processing features. The data emerging from this process passes quality control standards which are supported by DigiWeb and based on the strict requirements of the ISO 2859 standard

    Comparison of a high and a low intensity smoking cessation intervention in a dentistry setting in Sweden – a randomized trial

    <p>Abstract</p> <p>Background</p> <p>Tobacco is still the number one life style risk factor for ill health and premature death and also one of the major contributors to oral problems and diseases. Dentistry may be a potential setting for several aspects of clinical public health interventions and there is a growing interest in several countries to develop tobacco cessation support in dentistry setting. The aim of the present study was to assess the relative effectiveness of a high intensity intervention compared with a low intensity intervention for smoking cessation support in a dental clinic setting.</p> <p>Methods</p> <p>300 smokers attending dental or general health care were randomly assigned to two arms and referred to the local dental clinic for smoking cessation support. One arm received support with low intensity treatment (LIT), whereas the other group was assigned to high intensity treatment (HIT) support. The main outcome measures included self-reported point prevalence and continuous abstinence (≥ 183 days) at the 12-month follow-up.</p> <p>Results</p> <p>Follow-up questionnaires were returned from 86% of the participants. People in the HIT-arm were twice as likely to report continuous abstinence compared with the LIT-arm (18% vs. 9%, p = 0.02). There was a difference (not significant) between the arms in point prevalence abstinence in favour of the HIT-protocol (23% vs. 16%). However, point prevalence cessation rates in the LIT-arm reporting additional support were relatively high (23%) compared with available data assessing abstinence in smokers trying to quit without professional support.</p> <p>Conclusion</p> <p>Screening for willingness to quit smoking within the health care system and offering smoking cessation support within dentistry may be an effective model for smoking cessation support in Sweden. The LIT approach is less expensive and time consuming and may be appropriate as a first treatment option, but should be integrated with other forms of available support in the community. The more extensive and expensive HIT-protocol should be offered to those who are unable to quit with the LIT approach in combination with other support.</p> <p>Trial Registration</p> <p>Trial registration number: NCT00670514</p

    Eating disorders - knowledge, attitudes, management and clinical experience of Norwegian dentists

    Background The purpose of this study was to investigate knowledge, attitudes and clinical experience with regard to patients with eating disorders (ED) among Norwegian dentists. Methods In 2010, a questionnaire was sent to all dentists in Norway (N = 4282) comprising 33 questions related to demographics of the participating dentists, their knowledge of ED (general and oral health aspects), clinical experience, attitudes and perceived management preferences. Results The participation rate was 40 % (47 % women and 53 % men). Their knowledge about ED was often retrieved from common media sources and the greater part of the participants reported they had seen very few patients with ED during their professional career. Female dentists reported superior knowledge about ED compared to males, but the former experienced greater difficulties to inform about the condition. Referrals of the patient to other health facilities were significantly more common among female compared to male dentists. The majority of dentists (76 %) reported a need of more education related to ED management. Conclusions The Norwegian dentists in this study reported limited clinical experience and insufficient knowledge regarding ED. There is therefore a need to increase both undergraduate and continuing education in this field, which can improve preventive and management measures that a dentist can provide for ED patients.publishedVersio
