Ancient Amazonian populations left lasting impacts on forest structure
Amazonia contains a vast expanse of contiguous tropical forest and is influential in global carbon and hydrological cycles. Whether ancient Amazonia was highly disturbed or modestly impacted, and how ancient disturbances have shaped current forest ecosystem processes, is still under debate. Amazonian Dark Earths (ADEs), which are anthropic soil types with enriched nutrient levels, are one of the primary lines of evidence for ancient human presence and landscape modifications in settings that mostly lack stone structures and which are today covered by vegetation. We assessed the potential of using moderate spatial resolution optical satellite imagery to predict ADEs across the Amazon Basin. Maximum entropy modeling was used to develop a predictive model using locations of ADEs across the basin and satelliteâderived remotely sensed indices. Amazonian Dark Earth sites were predicted to be primarily along the main rivers and in eastern Amazonia. Amazonian Dark Earth sites, when compared with randomly selected forested sites located within 50 km of ADE sites, were less green canopies (lower normalized difference vegetation index) and had lower canopy water content. This difference was accentuated in two drought years, 2005 and 2010. This is contrary to our expectation that ADE sites would have nutrientârich soils that support trees with greener canopies and forests on ADE soils being more resilient to drought. Biomass and tree height were lower on ADE sites in comparison with randomly selected adjacent sites. Our results suggested that ADEârelated ancient human impact on the forest is measurable across the entirety of the 6 million km2 of Amazon Basin using remotely sensed data

Germline deletion and a somatic mutation of the PRKAR1A gene in a Carney complex-related pituitary adenoma
Objective: The objective was to assess involvement of loss of the PRKAR1A gene encoding a type 1α regulatory subunit of cAMP-dependent protein kinase A located on 17q24 in a Carney complex (CNC)-related pituitary adenoma.
Design: We investigated aberrations of the PRKAR1A gene in a CNC patient with a GH-producing pituitary adenoma, whose family has 3 other members with probable CNC.
Methods: A gene mutation was identified by a standard DNA sequencing method based on PCR. DNA copy number was measured to evaluate allelic loss on 17q24 by quantitative PCR. The breakpoints of deletion were determined by cloning a
rearranged region in the deleted allele.
Results: A PRKAR1A mutation of c.751_758del8 (p.S251LfsX16) was found in genomic DNA obtained from a pituitary adenoma, but not leukocytes from the patient.
Reduced DNA copy number at loci including the PRKAR1A gene on 17q24 was detected in both the tumor and leukocytes, suggesting a deletion at the loci at the germline level. The deletion size was determined to be approximately 0.5 Mb and this large deletion was also found in other 2 family members.
Conclusion: This is the first case showing a CNC-related pituitary adenoma with the combination of somatic mutation and a large inherited deletion of the PRKAR1A gene.
Biallelic inactivation of PRKAR1A may be necessary for the development of CNC-related pituitary adenoma
Medulloblastoma and ependymoma cells display levels of 5-carboxylcytosine and elevated TET1 expression
Background
Alteration of DNA methylation (5-methylcytosine, 5mC) patterns represents one of the causes of tumorigenesis and cancer progression. Tet proteins can oxidize 5mC to 5-hydroxymethylcytosine (5hmC), 5-formylcytosine and 5-carboxylcytosine (5caC). Although the roles of these oxidised forms of 5mC (oxi-mCs) in cancer pathogenesis are still largely unknown, there are indications that they may be involved in the mechanisms of malignant transformation. Thus, reduction of 5hmC content represents an epigenetic hallmark of human tumours and, according to our recent report; 5caC is enriched in a proportion of breast cancers and gliomas. Nevertheless, the distribution of oxi-mCs in paediatric brain tumours has not been assessed.
Findings
Here we analyse the global levels and spatial distribution of 5hmC and 5caC in 4 brain tumour cell lines derived from paediatric sonic hedgehog (SHH) pathway activated medulloblastomas (Daoy and UW228-3) and ependymomas (BXD-1425EPN and DKFZEP1NS). We show that, unlike HeLa cells, the paediatric tumour cell lines possess both 5hmC and 5caC at immunochemically detectable levels, and demonstrate that both modifications display high degrees of spatial overlap in the nuclei of medulloblastomas and ependymomas. Moreover, although 5hmC levels are comparable in the 4 brain tumour cell lines, 5caC staining intensities differ dramatically between them with highest levels of this mark in a subpopulation of DKFZ-EP1NS cells. Remarkably, the 5caC enrichment does not correlate with 5hmC levels and is not associated with alterations in Thymine DNA Glycosylase (TDG) expression in SHH medulloblastoma and ependymoma cell lines, but corresponds to elevated levels of TET1 transcript in UW228-3 and DKFZ-EP1NS cells.
Conclusions
We demonstrate that both 5caC enrichment and elevated TET1 expression are observed in SHH medulloblastomas and ependymomas. Our results suggest that increased Tet-dependent 5mC oxidation may represent one of the epigenetic signatures of cancers with neural stem cell origin and, thus, may contribute to development of novel approaches for diagnosis and therapy of the brain tumours

The study of unorthodox taxes will be preceded by a more general discussion of how law is made under unorthodoxy, and what are the special features of unorthodox legal policy. Unorthodoxy challenges equality before the law and is critical towards mass democracies. It also raises doubts on the operability of the rule of law, relying on personal skills, or loyalty, rather than on impersonal mechanisms arising from checks and balances as developed by the division of political power. Contextualizing legal norms: a multi-dimensional view of the 2014 legal capital reform in China
This paper intends to shed light on the contentious theme of the reception of legal transplantation in the host environment, by examining the 2014 legislative reform of legal capital in China, which at least on paper imitates the enabling settings of US Revised Model Business Corporation Act (RMBCA). The paper looks at the interconnections between national-specific contextual elements, the resultant complexities, and the spillover effects of transplanted configurations in the unique Chinese socio-cultural setting, implicating the discrepancy between the âlaw in practiceâ and the borrowed words âon the booksâ, and suggesting the importance of gaining a holistic understanding of âlawâ involving the legal traditions in both the donor country and the recipient nation
Legal linked data ecosystems and the rule of law
This chapter introduces the notions of meta-rule of law and socio-legal ecosystems to both foster and regulate linked democracy. It explores the way of stimulating innovative regulations and building a regulatory quadrant for the rule of law. The chapter summarises briefly (i) the notions of responsive, better and smart regulation; (ii) requirements for legal interchange languages (legal interoperability); (iii) and cognitive ecology approaches. It shows how the protections of the substantive rule of law can be embedded into the semantic languages of the web of data and reflects on the conditions that make possible their enactment and implementation as a socio-legal ecosystem. The chapter suggests in the end a reusable multi-levelled meta-model and four notions of legal validity: positive, composite, formal, and ecological
Resources, Capabilities, and Routines in Public Organizations
States, state agencies, multilateral agencies, and other non-market actors are relatively under-studied in strategic management and organization science. While important contributions to the study of public actors have been made within the agency-theoretic and transaction-cost traditions, there is little research in political economy that builds on resource-based, dynamic capabilities, and behavioral approaches to the firm. Yet public organizations can be characterized as stocks of human and non-human resources, including routines and capabilities; they can possess excess capacity in these resources; and they may grow and diversify in predictable patterns according to behavioral and Penrosean logic. This paper shows how resource-based, dynamic capabilities, and behavioral approaches to understanding public agencies and organizations shed light on their nature and governance
Empirically-grounded development of legal ontologies: a socio-legal perspective
This paper shows the multiple relationships between empirical data and semantic content in the legal field. One of the well-known problems of ontology construction is the "knowledge acquisition bottleneck problem" pointed out by Edward Feigenbaum and others, many years ago. In the next generation of Semantic Web developments this problem has not been completely solved. It is our con-tention that an accurate description of the legal environment, and well-grounded previous sociological studies may help to face it in a more satisfactory way. This means adopting a user-centered approach for legal ontologies, in what we will call an "iterative and integrated pragmatic circle" involving legal theorists, socio-legal researchers, professional people (lawyers, magistrates, prosecutorsâŠ) and com-puter scientists.
