In experiments with rats, P. nigriventer venom (PNV) temporarily disrupts the properties of the BBB by affecting both the transcellular and the paracellular route. However, it is unclear how cells and/or proteins participate in the transient opening of the BBB. The present study demonstrates that PNV is a substrate for the multidrug resistance protein-1 (MRP1) in cultured astrocyte and endothelial cells (HUVEC) and increases mrp1 and cx43 and down-regulates glut1 mRNA transcripts in cultured astrocytes. The inhibition of nNOS by 7-nitroindazole suggests that NO derived from nNOS mediates some of these effects by either accentuating or opposing the effects of PNV. In vivo, MRP1, GLUT1 and Cx43 protein expression is increased differentially in the hippocampus and cerebellum, indicating region-related modulation of effects. PNV contains a plethora of Ca(2+), K(+) and Na(+) channel-acting neurotoxins that interfere with glutamate handling. It is suggested that the findings of the present study are the result of a complex interaction of signaling pathways, one of which is the NO, which regulates BBB-associated proteins in response to PNV interference on ions physiology. The present study provides additional insight into PNV-induced BBB dysfunction and shows that a protective mechanism is activated against the venom.     Opening a window on the past: paleo-horizons across Pleistocene and Holocene. Madonna della Grotta, Praia a Mare, Southern Italy

    The goal of this study was to identify and characterize three paleo-horizons along a stratigraphic succession embracing sediments from Paleolithic to historic that has been preserved inside a grotto close to the Tyrrhenian Sea in southern Italy. The studied paleo-horizons date back to the transition period between Pleistocene and Holocene, when swift alternations of cold and warm phases profoundly altered the range dynamics of many organisms, including salmonids like Salmo trutta and, consequently, the behavior of the humans that occupied the grotto. Morphological observations and preliminary analysis were run to assess if the paleo-horizons preserved information of the human modified behavior. Results confirmed that one of the three paleo-horizons (Bud horizon, dating back to 10,300 cal YBP) had a strongly affected anthropic origin as it contains the rests of terrestrial (mammals, birds, and snails) and sea food (Salmo trutta and Patella spp.), and showed a hard compaction acquired via hydroconsolidation

    Three novel series of 1,2-benzisothiazole derivatives have been developed as inhibitors of carbonic anhydrase isoform IX. Compounds 5c and 5j, tested in vitro on the human colon cell line HT-29, blocked the growth of cells cultured under chemically induced hypoxic conditions, displaying a specific activity against cancer cells characterized by CAIX up-regulation. Moreover, a synergistic activity of 5c with SN-38 (the active metabolite of irinotecan) and 5-fluorouracil on cell proliferation under hypoxic conditions was demonstrated

    GEN-O-MA project: an Italian network studying clinical course and pathogenic pathways of moyamoya disease—study protocol and preliminary results

    Background: GENetics of mOyaMoyA (GEN-O-MA) project is a multicenter observational study implemented in Italy aimed at creating a network of centers involved in moyamoya angiopathy (MA) care and research and at collecting a large series and bio-repository of MA patients, finally aimed at describing the disease phenotype and clinical course as well as at identifying biological or cellular markers for disease progression. The present paper resumes the most important study methodological issues and preliminary results. Methods: Nineteen centers are participating to the study. Patients with both bilateral and unilateral radiologically defined MA are included in the study. For each patient, detailed demographic and clinical as well as neuroimaging data are being collected. When available, biological samples (blood, DNA, CSF, middle cerebral artery samples) are being also collected for biological and cellular studies. Results: Ninety-eight patients (age of onset mean ± SD 35.5 ± 19.6 years; 68.4% females) have been collected so far. 65.3% of patients presented ischemic (50%) and haemorrhagic (15.3%) stroke. A higher female predominance concomitantly with a similar age of onset and clinical features to what was reported in previous studies on Western patients has been confirmed. Conclusion: An accurate and detailed clinical and neuroimaging classification represents the best strategy to provide the characterization of the disease phenotype and clinical course. The collection of a large number of biological samples will permit the identification of biological markers and genetic factors associated with the disease susceptibility in Italy

    The recent advances in the trifluoromethyl aldimine synthesis and reactivity were here reported. The relevance in the construction of various nitrogenated molecules enriched with a -CF3 substituent has prompted much research in this field, especially due to the increase of their use in the pharmaceutical field. Unlike their unfluorinated analogues, trifluoromethyl aldimines are good electrophiles owing to the electron-withdrawing effect of the -CF3 group and therefore have been largely studied in addition reactions, emerging as powerful building blocks to construct important molecules in many fields of application
