    Glacial geomorphology of the northern Kivalliq region, Nunavut, Canada, with an emphasis on meltwater drainage systems

    This paper presents a glacial geomorphological map of glacial lineations, ribbed terrain, moraines, meltwater channels (subglacial and ice-marginal/proglacial), eskers, glaciofluvial deposits, ice-contact outwash fans and deltas and abandoned shorelines on the bed of the former Laurentide Ice Sheet in northern Canada. Mapping was compiled from satellite imagery and digital elevation data and landforms were digitised directly into a Geographical Information System. The map reveals a complex glacial history characterised by multiple ice-flow events, including fast-flowing ice-streams. Moraines record a series of pauses or re-advances during overall SE retreat towards the Keewatin Ice Divide. The distribution of subglacial meltwater landforms indicates that several distinctive scales and modes of drainage system operated beneath the retreating ice sheet. This includes a large (>100 km) integrated network of meltwater channels, eskers, ice-contact outwash fans and deltas and glaciofluvial deposits that originates at the northern edge of Aberdeen Lake. The map comprises zone 66 of the Canadian National Topographic System, which encompasses an area of 160,000 km2. It is presented at a scale of 1:500,000 and is designed to be printed at A0 size

    LecturePg. 107-116Improving centrifugal compressor seal performance and reliability can be accomplished through development of new technology and/or by improvement of existing technology. An improved dry gas lubricating seal of single rotor design has been designed for high pressure gas compressors. A more durable seal of simpler design has been created through implementation of ductile materials and reduction of components. Currently, the tandem dry gas seal configuration is the accepted standard for high pressure natural gas pipeline compressor applications. These applications still suffer problems with process and lube oil contamination, brittle fracture of carbide materials, and explosive decompression of 0-rings. Real costs associated with these problems are considerable. Development and potential benefits of the single rotor design seal, along with field application, in a high pressure natural gas centrifugal compressor is discussed

    Molecular probe technology detects bacteria without culture

    <p>Abstract</p> <p>Background</p> <p>Our ultimate goal is to detect the entire human microbiome, in health and in disease, in a single reaction tube, and employing only commercially available reagents. To that end, we adapted molecular inversion probes to detect bacteria using solely a massively multiplex molecular technology. This molecular probe technology does not require growth of the bacteria in culture. Rather, the molecular probe technology requires only a sequence of forty sequential bases unique to the genome of the bacterium of interest. In this communication, we report the first results of employing our molecular probes to detect bacteria in clinical samples.</p> <p>Results</p> <p>While the assay on Affymetrix GenFlex Tag16K arrays allows the multiplexing of the detection of the bacteria in each clinical sample, one Affymetrix GenFlex Tag16K array must be used for each clinical sample. To multiplex the clinical samples, we introduce a second, independent assay for the molecular probes employing Sequencing by Oligonucleotide Ligation and Detection. By adding one unique oligonucleotide barcode for each clinical sample, we combine the samples after processing, but before sequencing, and sequence them together.</p> <p>Conclusions</p> <p>Overall, we have employed 192 molecular probes representing 40 bacteria to detect the bacteria in twenty-one vaginal swabs as assessed by the Affymetrix GenFlex Tag16K assay and fourteen of those by the Sequencing by Oligonucleotide Ligation and Detection assay. The correlations among the assays were excellent.</p

    A study of physical activity comparing people with Charcot Marie Tooth disease to normal control subjects

    PURPOSE: Charcot Marie Tooth disease (CMT) describes a group of hereditary neuropathies that present with distal weakness, wasting and sensory loss. Small studies indicate that people with CMT have reduced daily activity levels. This raises concerns as physical inactivity increases the risk of a range of co- morbidities, an important consideration in the long-term management of this disease. This study aimed to compare physical activity, patterns of sedentary behavior and overall energy expenditure of people with CMT and healthy matched controls. METHODS: We compared 20 people with CMT and 20 matched controls in a comparison of physical activity measurement over seven days, using an activity monitor. Patterns of sedentary behavior were explored through a power law analysis. RESULTS: Results showed a decrease in daily steps taken in the CMT group, but somewhat paradoxically, they demonstrate shorter bouts of sedentary activity and more frequent transitions from sedentary to active behaviors. No differences were seen in energy expenditure or time spent in sedentary, moderate or vigorous activity. CONCLUSION: The discrepancy between energy expenditure and number of steps could be due to higher energy requirements for walking, but also may be due to an over-estimation of energy expenditure by the activity monitor in the presence of muscle wasting. Alternatively, this finding may indicate that people with CMT engage more in activities or movement not related to walking. Implications for Rehabilitation Charcot-Marie-Tooth disease: • People with Charcot-Marie-Tooth disease did not show a difference in energy expenditure over seven days compared to healthy controls, but this may be due to higher energy costs of walking, and/or an over estimation of energy expenditure by the activity monitor in a population where there is muscle wasting. This needs to be considered when interpreting activity monitor data in people with neuromuscular diseases. • Compared to healthy controls, people with Charcot-Marie-Tooth disease had a lower step count over seven days, but exhibited more frequent transitions from sedentary to active behaviors • High Body Mass Index and increased time spent sedentary were related factors that have implications for general health status. • Understanding the profile of physical activity and behavior can allow targeting of rehabilitation interventions to address mobility and fitness

    International audienceThe evaluation of demography for proto- and prehistoric times remains complex due to the rareness of human remains. Here we propose an original approach based on the combination of geophysical and organic geochemical techniques applied to the Late Bronze Age sedimentary infill of Lake le Bourget. Deep sediments were mapped and their volume estimated by sub-bottom seismic profiling calibrated (accoustically and chronologically) on piston cores. Multibeam bathymetry combined to a subaquatic archaeological survey allowed to precisely identify the extension of Late Bronze Age palafittic stations in shallow waters and to determine the volume of the associated organic rich deposits

    Spatial variation in exhumation rates across Ladakh and the Karakoram: New apatite fission track data from the Eastern Karakoram, NW India

    Characterization of low-temperature cooling histories and associated exhumation rates is critical for deciphering the recent evolution of orogenic regions. However, these may vary significantly over relatively short distances within orogens. It is pertinent therefore to constrain cooling histories and hence exhumation rates across major tectonic boundaries. We report the first apatite fission track ages from the Karakoram Fault Zone in the Eastern Karakoram range, which forms part of the western margin of the Tibetan Plateau. Ten samples, from elevations of 3477–4875m, have apatite fission track dates from 3.3 ± 0.3 Ma to 7.4± 1.1Ma. The ages correspond to modeled average erosional exhumation rates of 0.67+ 0.27-0.18mm/yr across the Eastern Karakoram. The results are consistent with a trend northward from the Indus suture zone, across the Ladakh terrane and into the Karakoram, in which tectonic uplift associated with crustal thickening increases toward the north, raising elevation and promoting glaciation and generation of extreme relief. As a result, erosion and exhumation rates increase south to north. Present-day precipitation on the other hand varies little within the study area and on a larger scale decreases southwest to northeast across this portion of the orogen. The Eastern Karakoram results highlight the diverse patterns of exhumation driven by regional variations in tectonic response to collision along the western margin of Tibet

    Albiglutide, a Long Lasting Glucagon-Like Peptide-1 Analog, Protects the Rat Heart against Ischemia/Reperfusion Injury: Evidence for Improving Cardiac Metabolic Efficiency

    BACKGROUND: The cardioprotective effects of glucagon-like peptide-1 (GLP-1) and analogs have been previously reported. We tested the hypothesis that albiglutide, a novel long half-life analog of GLP-1, may protect the heart against I/R injury by increasing carbohydrate utilization and improving cardiac energetic efficiency. METHODS/PRINCIPAL FINDINGS: Sprague-Dawley rats were treated with albiglutide and subjected to 30 min myocardial ischemia followed by 24 h reperfusion. Left ventricle infarct size, hemodynamics, function and energetics were determined. In addition, cardiac glucose disposal, carbohydrate metabolism and metabolic gene expression were assessed. Albiglutide significantly reduced infarct size and concomitantly improved post-ischemic hemodynamics, cardiac function and energetic parameters. Albiglutide markedly increased both in vivo and ex vivo cardiac glucose uptake while reducing lactate efflux. Analysis of metabolic substrate utilization directly in the heart showed that albiglutide increased the relative carbohydrate versus fat oxidation which in part was due to an increase in both glucose and lactate oxidation. Metabolic gene expression analysis indicated upregulation of key glucose metabolism genes in the non-ischemic myocardium by albiglutide. CONCLUSION/SIGNIFICANCE: Albiglutide reduced myocardial infarct size and improved cardiac function and energetics following myocardial I/R injury. The observed benefits were associated with enhanced myocardial glucose uptake and a shift toward a more energetically favorable substrate metabolism by increasing both glucose and lactate oxidation. These findings suggest that albiglutide may have direct therapeutic potential for improving cardiac energetics and function

    De novo CCND2 mutations leading to stabilization of cyclin D2 cause megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome

    Activating mutations in genes encoding phosphatidylinositol 3-kinase (PI3K)-AKT pathway components cause megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome (MPPH, OMIM 603387). Here we report that individuals with MPPH lacking upstream PI3K-AKT pathway mutations carry de novo mutations in CCND2 (encoding cyclin D2) that are clustered around a residue that can be phosphorylated by glycogen synthase kinase 3β (GSK-3β). Mutant CCND2 was resistant to proteasomal degradation in vitro compared to wild-type CCND2. The PI3K-AKT pathway modulates GSK-3β activity, and cells from individuals with PIK3CA, PIK3R2 or AKT3 mutations showed similar CCND2 accumulation. CCND2 was expressed at higher levels in brains of mouse embryos expressing activated AKT3. In utero electroporation of mutant CCND2 into embryonic mouse brains produced more proliferating transfected progenitors and a smaller fraction of progenitors exiting the cell cycle compared to cells electroporated with wild-type CCND2. These observations suggest that cyclin D2 stabilization, caused by CCND2 mutation or PI3K-AKT activation, is a unifying mechanism in PI3K-AKT–related megalencephaly syndromes

    Gene Annotation and Drug Target Discovery in Candida albicans with a Tagged Transposon Mutant Collection

    Candida albicans is the most common human fungal pathogen, causing infections that can be lethal in immunocompromised patients. Although Saccharomyces cerevisiae has been used as a model for C. albicans, it lacks C. albicans' diverse morphogenic forms and is primarily non-pathogenic. Comprehensive genetic analyses that have been instrumental for determining gene function in S. cerevisiae are hampered in C. albicans, due in part to limited resources to systematically assay phenotypes of loss-of-function alleles. Here, we constructed and screened a library of 3633 tagged heterozygous transposon disruption mutants, using them in a competitive growth assay to examine nutrient- and drug-dependent haploinsufficiency. We identified 269 genes that were haploinsufficient in four growth conditions, the majority of which were condition-specific. These screens identified two new genes necessary for filamentous growth as well as ten genes that function in essential processes. We also screened 57 chemically diverse compounds that more potently inhibited growth of C. albicans versus S. cerevisiae. For four of these compounds, we examined the genetic basis of this differential inhibition. Notably, Sec7p was identified as the target of brefeldin A in C. albicans screens, while S. cerevisiae screens with this compound failed to identify this target. We also uncovered a new C. albicans-specific target, Tfp1p, for the synthetic compound 0136-0228. These results highlight the value of haploinsufficiency screens directly in this pathogen for gene annotation and drug target identification
