Modelling tools to predict potential distribution of forest species : using Pico Island and the Azores as study case
Tese de Doutoramento, Biologia, 16 de novembro de 2018, Universidade dos Açores.Os modelos de distribuição de espécies (SDMs) têm sido aplicados em diferentes áreas da ecologia, nomeadamente para modelar a distribuição potencial de espécies invasoras, para avaliar espécies prioritárias no âmbito da conservação e para apoiar o planeamento florestal. Um SDM é uma descrição matemática da distribuição de uma espécie no espaço ambiental, a qual pode ser utilizada para prever a distribuição da espécie no espaço geográfico. O avanço ao nível da capacidade computacional disponibilizou uma diversidade de métodos estatísticos, que anteriormente não era possível utilizar. Esta diversidade de métodos reflete-se num número crescente de publicações direcionadas ao estudo e aplicação dos SDMs e também numa variedade crescente de métodos de modelação. Nos Açores, a abundância crescente de dados corológicos, a diversidade geomorfológica do arquipélago e os diferentes padrões espaciais que é possível encontrar em diferentes ilhas e em diferentes espécies, contribuem para que o arquipélago seja um bom modelo para a comparação de diferentes abordagens de modelação, bem como para testar possíveis constrangimentos inerentes ao processo de modelação. As perguntas de investigação a que pretendemos responder nesta tese foram as seguintes: (i) As abordagens de modelação, baseadas em diferentes fundamentos teóricos, originam resultados semelhantes, ao nível da distribuição potencial das espécies florestais estudadas? (ii) Existe alguma diferença relevante, entre o cálculo de Modelos Lineares Generalizados (GLMs) usando métodos de máxima verossimilhança ou métodos bayesianos? (iii) Existe alguma vantagem, no uso de um campo aleatório relativo à estrutura espacial dos dados, em comparação com os modelos que incluem apenas os efeitos fixos das variáveis ambientais? (iv) As diferentes abordagens de modelação originam resultados consistentes, em particular quando o número de variáveis ambientais utilizadas na modelação é reduzido? (v) As diferentes técnicas de modelação são afetadas de um modo relevante pela dimensão da amostra, pelo tipo de distribuição da espécie e pelas alterações no uso do solo? Para responder a estas questões, foram desenvolvidos três exercícios de modelação: (i) Uma comparação da Análise Fatorial do Nicho Ecológico (ENFA) e da modelação baseada na Máxima Entropia (MaxEnt), utilizando dados relativos à presença de três espécies (Pittosporum undulatum, Acacia melanoxylon e Morella faya) em três ilhas (Pico, Terceira e São Miguel), e incluindo o efeito da redução da dimensão da amostra; (ii) A comparação de modelos com efeitos fixos ou mistos, utilizando a plataforma R para o cálculo de GLMs e da aproximação de Laplace (INLA), permitindo o cálculo da estrutura espacial dos dados (função de covariância de Matérn), baseada em dados de duas ilhas (Pico e São Miguel) para duas espécies (P. undulatum e M. faya), e incluindo o efeito da redução da dimensão da amostra; e (iii) A comparação de GLMs e de uma seleção de algoritmos de autoaprendizagem (Machine Learning), usados para modelar as possíveis alterações nas áreas de distribuição de P. undulatum, A. melanoxylon e M. faya nas três ilhas, resultantes das alterações climáticas previstas para 2100. Em relação ao primeiro exercício, ambas as abordagens originaram cenários semelhantes, particularmente quando a quantidade de informação explicada pela ENFA era elevada; os resultados da modelação foram afetados pela redução do tamanho da amostra; os modelos com melhor capacidade de previsão incluíam um conjunto variado de variáveis ambientais (topográficas, climáticas e de uso do solo); e os modelos eram afetados pela transferência para um novo habitat (i.e. ilha). Os resultados do segundo exercício de modelação indicaram que os GLMs, calculados através de métodos de máxima verossimilhança ou métodos bayesianos originaram resultados similares, mesmo nos casos em que a dimensão da amostra era reduzida; e que a adição de um campo aleatório aumentou o ajustamento dos modelos, particularmente para a árvore menos abundante, M. faya, embora a estrutura do campo aleatório fosse claramente afetada pela dimensão da amostra. O terceiro exercício de modelação revelou que existem várias limitações quando se modela o efeito das alterações climáticas na distribuição das espécies, uma vez que os melhores modelos incluíram variáveis topográficas, demonstrando que a modelação baseada somente no clima poderá não ser fiável; verificou-se igualmente que o ajuste dos modelos variava de forma relevante entre as diferentes abordagens de modelação, e que o algoritmo Random Forest apresentou, em geral, os melhores resultados. De uma forma geral, os resultados desta investigação poderão ser aplicados como forma de apoio à gestão da floresta açoriana. Poderão ser replicados em outros sistemas insulares e noutras regiões florestais, não somente em projetos direcionados para a ecologia das espécies florestais, mas também em questões de investigação relacionadas com a previsão do sucesso e expansão das plantas invasoras, a deteção de áreas adequadas para projetos de restauro, a modelação baseada em dados de deteção remota e a modelação do efeito potencial das alterações climáticas.ABSTRACT: Species distribution models (SDMs) have been used in different areas within ecology, namely to model the potential spread of invasive species, to evaluate and manage priority species for conservation and to support forest management. An SDM is a mathematical description of the species distribution in the environmental space that can be used to predict the distribution of the species in the geographic space. The advances in computational capabilities have provided increasingly greater and more intensive statistical algorithms than was previously possible, as reflected by the increasing number of publications addressing SDMs and also the growing variety of modelling approaches. In the Azores, the growing abundance of the species distribution data, the diversity on island size and morphology, and the different spatial patterns that are possible among islands and species, make the archipelago a good model for the comparison of different modelling approaches and to test possible modelling constraints. Overall, the results of this research can be expanded to support Azorean forestry management, and could be replicated in other island systems and forest regions, not only in projects addressing the ecology of particular forest species, but also when handling research questions related with the prediction of plant invader success and expansion, the detection of areas potentially suited for restoration projects, modelling based on remote sense data, and modelling of the potential effect of climate change
O papel da lipocalin-2 na patofisiologia da esclerose múltipla: um olhar para além do cérebro
Dissertação de mestrado em Ciências da SaúdeMultiple sclerosis (MS) is an immune-mediated demyelinating disease of the central nervous system
(CNS), characterized by the presence of demyelination plaques, inflammation and gliosis that
consequently lead to axonal damage. The sequence of events that leads to demyelination remains
unclear and the pathophysiological mechanisms are diverse. Also, although this is a disease of the
CNS, there is no doubt that, in terms of peripheral organs, the thymus, as the organ of T cell
differentiation and maturation, plays an important role in the pathophysiology of the disease.
Recently, the levels of lipocalin 2 (LCN2), an acute phase protein that is part of the defense system
against bacteria, by binding to iron-loaded siderophores, were found to be increased in cerebrospinal
fluid (CSF) and serum of MS patients, when compared to control subjects. Similarly, using the MS
animal model of experimental autoimmune encephalomyelitis (EAE), LCN2 was detected in brain
parenchyma astrocytes, in regions typically affected in MS patients. This expression by astrocytes,
together with an increased LCN2 level in the CSF, occurs during the active phases of the disease, which
could point towards a role for LCN2 secreted by astrocytes in the mediation of inflammatory responses
in the EAE model. Altogether, these findings support LCN2 as a valuable molecule for the
diagnostic/monitoring of MS and suggest its potential involvement as a disease modulator. Of
relevance, the exact role of LCN2 in the pathophysiology of the disease remains largely unknown and
contradictory data exists on its potential protective or deleterious effect. Therefore, we sought to
investigate the role of LCN2 in the onset and progression of the disease. Herein, we tackled the
disease, by evaluating the role of LCN2, not only in the perspective of the CNS, but also on the
perspective of peripheral organs such as the thymus.
First we intended to perform a characterization of the thymus regarding thymocyte populations and
histological morphology, in wild-type (WT) animals induced with EAE, in the onset and chronic phases of
disease. Next, to further understand the role of LCN2 in MS pathology, we induced EAE both in LCN2-
null mice and in WT littermates. Non-induced EAE animals were used as controls.
The thymus of EAE animals was atrophied, as assessed by its weight, normalized for total body weight,
and by the number of total cells. Also, we found a decrease in total cell number of all thymocyte
populations, during the onset and chronic phases of EAE. In relative terms, the percentage of double
positive cells was decreased, and the percentages of the cluster of differentiation (CD)4 and CD8 single
positive cells were increased, during the onset phase. At the chronic phase, the proportions between
the different populations were restored.
LCN2-null mice induced with EAE did not present major alterations in terms of the clinical score, when
compared with WT littermates also induced with EAE. Likewise, their thymic alterations were similar to
the ones observed in WT EAE animals. Of relevance, as for the inflammatory profile in the cerebellum,
LCN2-null mice presented less inflammation, as assessed by decreased expression levels of proinflammatory
cytokines interferon (Ifn)-gamma, interleukin (Il)12a and Il17a. Also of interest, the
cerebellum of LCN2-null mice presented a decrease in the percentage of lesioned areas. Finally, EAE
animals, from both genotypes, presented an increase in the area positive for glial fibrillary acidic protein
(GFAP), in the white matter of the cerebellum, in both the onset and chronic phases of disease. On the
contrary, the expression levels of Gfap in the cerebellum were only increased at the onset phase of
disease.A Esclerose Múltipla (EM) é uma doença autoimune desmielinizante do sistema nervosa central (SNC),
caracterizada pela presença de placas de desmielinização, inflamação e gliose, que tem como
consequência dano axonal. A sequência de eventos que induzem desmielinização permanecem
desconhecidos, e os mecanismos patofisiológicos são diversos. Embora esta seja uma doença do SNC,
não há dúvidas que, em termos de órgãos periféricos, o timo, sendo o órgão de maturação e
diferenciação das células T, desempenha um papel importante na patofisiologia da doença.
Recentemente, os níveis de lipocalin-2 (LCN2), uma proteína de fase aguda que participa no sistema
de defesa contra infeções bacterianas, através da ligação a sideróforos, foram encontrados como
estando elevados no líquido cefalorraquidiano (LCR) e no soro de doentes com EM, comparativamente
aos controlos. Da mesma maneira, usando o modelo animal de EM de encefalomielite autoimune
experimental (EAE), a LCN2 foi detetada em astrócitos do parênquima, em regiões tipicamente
afetadas em doentes com EM. Esta expressão pelos astrócitos, associada a um aumento de LCN2 no
LCR, ocorreu durante as fases ativas da doença, o que aponta para um papel da LCN2 secretada pelos
astrócitos na mediação da response inflamatória no modelo de EAE. No seu conjunto, estas evidências
suportam o papel da LCN2 como uma molécula importante no diagnóstico e/ou monitorização da EM,
e sugere o seu possível envolvimento como moduladora da doença. É relevante dizer que o papel exato
da LCN2 na patofisiologia da doença permanece desconhecido, e existem dados contraditórios no que
diz respeito ao seu potencial efeito protetor ou deletério. Por isso, nós procurámos perceber o papel da
LCN2 no onset e na progressão da doença. Assim, nós investigámos o papel da LCN2 na doença, não
só na perspetiva do SNC, mas também dos órgãos periféricos, nomeadamente do timo.
Primeiro pretendemos caracterizar o timo em relação às populações de timócitos e morfologia
histológica, em animais wild-type (WT) induzidos com EAE, no onset e na fase crónica da doença. De
seguida, para melhor entender o papel da LCN2 na patologia da EM, induzimos EAE em animais LCN2-
null e em WT da mesma ninhada. Para além disso, usámos animais não induzidos como controlos.
Nós observámos que o timo dos animais induzidos com EAE estava atrofiado, com base no seu peso,
após normalização para o peso total do animal, e no número total de células. Para além disso,
encontrámos uma diminuição no número total de células de todas as principais populações de
timócitos, durante o onset e fase crónica da doença. No que diz respeito à percentagem de cada uma
das populações de timócitos, durante o onset da doença, a percentagem de células duplas positivas
encontrava-se diminuída, enquanto as percentagens das populações CD4+CD8- e CD4-CD8+ se
encontrava aumentada. Na fase crónica da doença, as proporções entre as diferentes populações
foram reestabelecidas.
Os animais LCN2-null induzidos com EAE não apresentaram grandes alterações em termos de score
clínico, quando comparados com os animais WT da mesma ninhada também induzidos com EAE. Para
além disso, as alterações observadas no timo foram semelhantes às encontradas nos animais WT EAE.
De relevância, no que diz respeito ao perfil inflamatório no cerebelo, os animais LCN2-null
apresentaram menos inflamação, o que é suportado por níveis diminuídos dos níveis de expressão das
citoquinas pró-inflamatórias interferão-gama, e interleucinas 12 e 17. É importante também referir que
os cerebelos de animais LCN2-null apresentaram uma diminuição na percentagem de áreas com
lesões. Os animais EAE, de ambos os genótipos, apresentaram um aumento na área positiva para
GFAP, na substância branca do cerebelo, no onset e na fase crónica da doença. Pelo contrário, os
níveis de expressão de Gfap no cerebelo só foram encontrados elevados no onset da doença
Desenvolvimento de novos materiais híbridos capazes de incorporar compostos com atividade farmacológica
Despite its intrinsic ability to regenerate after an injury, bone tissue can be challenged by a huge variety of pathological conditions that lead to massive bone loss, necessitating the application of a bone implant. Recent advances in the development of biomaterials, specifically in the area of hybrid materials, have provided better alternatives to commonly used implants, offering wide medical applications in the restoration of damaged bone. In this work we specifically focus on a hybrid material composed of polydimethylsiloxane and tetraethyl orthosilicate This system, obtained through the sol-gel method, allows the incorporation of the organic component in an inorganic network, with interactions that lead to the obtention of hybrid materials with morphology and multifunctional properties suitable for bone applications.
Resveratrol (RES) is a bioactive molecule with many beneficial properties for human health, specifically, in the metabolism involved in bone regeneration, making it an excellent candidate for application in therapies associated with bone diseases. However, its physicochemical characteristics, specifically the low solubility and bioavailability, create limitations to its application. To combat these problems, drug delivery systems capable of increasing absorption and overcoming the limitations of RES have been developed.
In this work, a new hybrid material combined with RES was developed and characterized, in order to evaluate the system's ability to incorporate and release this compound. RES was loaded with high efficiency (loading efficiency > 75%) into hybrid materials using the rotary evaporator technique. The incorporation promoted its amorphization and increased solubility and in vitro release when compared to free RES (non-encapsulated). All the release profiles obtained showed a pH dependence and were clearly faster at pH 5.2 than at pH 7.4. The release of RES from the loaded material was gradual over time, had an initial time lag of approximately 4 hours, and was well described by the Weibull mathematical model. In vitro cytotoxicity studies were performed on human osteosarcoma cells (MG-63) and these showed a decrease in cell viability with increasing concentration of RES-loaded samples (for concentrations > 100μg mL⁻¹). Hybrid materials without the addition of RES did not show cytotoxicity regardless of the concentration studied.Apesar da sua capacidade intrínseca de regeneração após uma lesão, o tecido ósseo pode ser desafiado por uma enorme variedade de condições patológicas que levam a grandes perdas e impedem a sua regeneração, sendo necessária a aplicação de um substituto (implante) ósseo. Os recentes avanços no desenvolvimento de biomateriais, especificamente na área dos materiais híbridos, têm providenciado melhores alternativas aos implantes utilizados normalmente, oferendo amplas aplicações médicas na restauração do osso danificado. Neste trabalho focamo-nos especificamente num material híbrido composto por polidimetilsiloxano e tetraetilortosilicato. Este sistema, obtido através do método de sol-gel, permite a incorporação da componente orgânica numa rede inorgânica, com interações que levam à obtenção de materiais híbridos com morfologia e propriedades multifuncionais adequadas para aplicação óssea.
O resveratrol (RES) é uma molécula bioativa com múltiplas propriedades benéficas para a saúde humana, em específico, no metabolismo envolvido na regeneração óssea, tornando-o um excelente candidato para aplicação em terapias associadas a doenças ósseas. No entanto, as suas caraterísticas físico-químicas, especificamente a baixa solubilidade e biodisponibilidade, criam limitações à sua aplicação. Para combater estes problemas têm sido desenvolvidos sistemas de entrega de fármacos capazes de aumentar a absorção e ultrapassar as suas limitações.
Neste trabalho foi desenvolvido e caraterizado um novo material híbrido combinado com RES, com o objetivo de avaliar a capacidade do sistema para incorporar e libertar este composto. O RES foi carregado com elevada eficácia (eficácia de carregamento > 75%) nos materiais híbridos através da técnica do evaporador rotativo. A incorporação promoveu a sua amorfização e aumentou a solubilidade e libertação in vitro quando comparado com o RES livre (não encapsulado). Todos os perfis de libertação obtidos apresentaram uma dependência do pH e mostraram ser claramente mais rápidos em pH 5,2 do que em pH 7,4. A libertação de RES do material carregado foi gradual ao longo do tempo, apresentou um atraso inicial de aproximadamente 4 horas e foi bem descrita pelo modelo matemático de Weibull. Foram realizados estudos de citotoxicidade in vitro em células humanas de osteossarcoma (MG-63) e, estes mostraram uma diminuição da viabilidade celular com o aumento da concentração das amostras carregadas com RES (para concentrações > 100μg mL⁻¹). Novel Excipient Candidates for RNA Stabilization
Until recently, the study of therapies using the RNA molecule as a therapeutic
agent was considerably low when compared to therapies using DNA and proteins. For a
long time, the RNA molecule was considered as a mere intermediary in the production
of proteins. More recently, the study of the RNA molecule has led to a better
understanding of its function, thus bringing a new perspective to potential therapeutic
applications of this molecule. The use of therapeutic RNAs namely, mRNAs and
microRNAs has been increasing, especially due to the recent use of mRNA in vaccination
against COVID-19. One of the limitations in the use of mRNA vaccines, which has seen a
lot of discussion, is the temperature at which they have to be stored, in order to maintain
vaccine stability and effectiveness. This reveals the importance and urgency in
identifying compounds capable of stabilizing the RNA molecule in order to facilitate, the
storage and handling, while maintaining its biological activity. Taking into account the
growing research in the therapeutic RNAs field, identifying stabilizers might have an
impact when working with other RNA molecules with different applications.
In the present work, Circular Dichroism (CD) assays were performed to assess
the stability of the RNA structure, after incubation with different categories of excipients,
namely amino acids (arginine and glycine), ionic liquids (N-acetyl amino acid N-alkyl
cholinium-based ILs), sugars (glucose, sucrose and trehalose) and Deep Eutectic
Solvents (DES) (glucose-based DES and trehalose-based DES). These studies also
involved the assessment of stability over time (0, 7, 15, 30 and 60 days) at different
storage temperatures (4 ° C and room temperature). In general, the results obtained
demonstrate the potential of amino acids to stabilize RNA, for 60 days of storage at both
temperatures. Moreover, the samples stabilized by the new excipient candidates also
demonstrated great potential, taking into account that the samples stabilized by
trehalose-based DES not only maintained their stability after 60 days at both
temperatures, but also increased the melting temperature by around 18 °C. Data
gathered by molecular docking simulations allowed for a better understanding of the
interactions that happen between DES and RNA and which of the compounds might have
a higher contribution for the RNA stabilization. Results using amino acid-based ionic
liquids as stabilizers were also very encouraging, as most of the samples tested
maintained stability over the testing period at both temperatures.
Considering the results obtained, these confirm the stabilizing capacity of the
classes already used as excipients and the potential of new classes as RNA stabilizers.
Thus opening the door to new possibilities for formulating therapeutic products.Até recentemente, o estudo de terapias usando a molécula de RNA como agente
terapêutico era consideravelmente reduzido, quando comparado com terapias usando
DNA e proteínas. A molécula de RNA era considerada como um mero intermediário na
produção de proteínas. Mais recentemente, o estudo da molécula de RNA levou à
identificação de outras funções, trazendo uma nova perspetiva para a potencial aplicação
terapêutica desta molécula. A utilização de RNAs terapêuticos nomeadamente, os
mRNAs e microRNAs tem vindo a aumentar, especialmente devido ao uso recente do
mRNA na vacinação contra a COVID-19. Uma das principais limitações na distribuição
global e utilização das vacinas de mRNA, que muito tem sido discutida, é a temperatura
de armazenamento necessária à manutenção de estabilidade e eficácia da vacina. Isto
revela a importância e urgência na identificação de compostos capazes de estabilizar a
molécula de RNA de modo a facilitar, o seu armazenamento e distribuição, garantindo a
sua atividade biológica. Considerando a crescente investigação na área dos RNAs
terapêuticos, a identificação de estabilizadores pode ter um impacto transversal e
alargado a outras moléculas de RNA, com diferentes aplicações.
No presente trabalho foram realizados ensaios de Dicroísmo Circular (CD) para
avaliar a estabilidade da estrutura do RNA, após a incubação com diferentes categorias
de excipientes, nomeadamente aminoácidos (arginina e glicina), líquidos iónicos (Nacetyl amino acid N-alkyl cholinium-based), açúcares (glucose, sacarose e trealose) e
Deep Eutectic Solvents (DES) (glucose-based DES e trehalose-based DES). Os estudos
de estabilidade envolveram também a avaliação da estabilidade de um RNA modelo ao
longo do tempo (0, 7, 15, 30 e 60 dias) e também quando armazenado a diferentes
temperaturas (4°C e temperatura ambiente). De uma maneira geral os resultados obtidos
demonstraram a possibilidade de estabilizar o RNA na presença de aminoácidos, durante
os 60 dias de armazenamento, quer a 4°C quer à temperatura ambiente. As novas classes
de estabilizadores também demonstraram grande potencial, tendo em conta que as
amostras estabilizadas por DES composto por trehalose e colina mantiveram a sua
estabilidade após os 60 dias de armazenamento, a ambas as temperaturas, com um
aumento da temperatura de melting de cerca de 18 °C. Os resultados obtidos por docking
molecular permitiram um melhor entendimento das interações estabelecidas entre os
DES e a molécula de RNA, permitindo estimar qual dos componentes do DES é que
apresenta uma maior contribuição para a estabilização global da molécula de RNA. Outra
das novas classes estudadas foram os líquidos iónicos com aminoácidos acetilados que
também demonstraram resultados encorajadores. Neste caso, grande parte das amostras
de RNA manteve a estabilidade ao longo do tempo, para ambas as temperaturas, demonstrando assim uma melhoria relativamente à amostra de RNA sem
estabilizadores.
Considerando os resultados obtidos, foi possível confirmar a possibilidade de
estabilizar RNA usando alguns excipientes já estabelecidos e verificar o potencial de
novas classes na indução da estabilidade desta molécula por um tempo médio de
armazenamento à temperatura ambiente ou em refrigeração a 4 °C. Com este trabalho,
criam-se novas perspetivas e novas possibilidades de formulação de produtos
terapêuticos de base génica
Two generations sharing Adult Training (EFA) Courses - the impact of EFA certification on Lusophone immigrants and their descendants
This paper is part of my on ongoing PhD research entitled “Lives recounted – the impact of the EFA experience on the life trajectories of Lusophone labour migrants and their descendants”.
My proposal in this paper is to explain the path that led me to introduce the issue of intergenerational relationships into my PhD research. Initially, the goal was to understand the impact of EFA certification2 on Lusophone immigrants’ lives. According to the official statistics, it is immigrants who rely most on EFA training courses. However, the first biographical interviews that I carried out showed that the category ‘foreign’ includes not only immigrants but also the descendants of immigrants who, for family or legal reasons, do not have Portuguese nationality. Many immigrant descendants, in fact, have difficulty in acquiring citizenship, which affects various dimensions of their lives, including access to and continuation in the educational system and labour market. In the official statistics these immigrants and their descendants appear in the category ‘foreigners’, i.e. despite being born in Portugal, a significant number of the descendants of immigrants have never been able to acquire Portuguese nationality.
The situation found in the fieldwork has thus led to the resizing of the sample, which now considers not only Lusophone immigrants but also the descendants of Lusophone immigrants as a key object of analysis. I aim to understand the different impacts of EFA certification on their lives, treating the generational differences as a crucial point of analysis. Despite these two generations sharing the certification processes, I start from the assumption that the discourses on the impact of the EFA experience in their lives can be differentiated.
In this paper I intend to show that the official statistics in the ‘foreign’ category embrace different groups and different generations, which appear interchangeably in a single category. On the basis of interviews conducted to date, I argue that being a Lusophone immigrants and the descendants of Lusophone immigrants form two distinct categories – for we are referring here to different generations that, despite the firmly established relationships between them, ultimately (self)evaluate the EFA experience differently

Dissertation to obtain the degree of Master in Chemical and Biochemical EngineeringThe work present in this thesis was conducted in Portucel Soporcel mill, in the industrial complex of Setúbal, and had as main objective the modelling of the treatment process of the effluents from this industry, using for this purpose the software GPS-X. This program has a clear-cut graphical interface and uses a specialized translator that converts the graphical process into material balance equations, based on dynamic models. These models allow, besides the kinetic descripton of the treatment process carried out at the WWTP, to simulate new scenarios towards the study of critical parameters for the process as well as optimization and control of the WWTP.
The effluent that arrives to Portucel’s WWTP, from the pulp and paper mills of the complex, is particularly rich on fibers (solids), lignin, chlorinated and sulphur compounds, resin acids, phenols and starch. It has a brown colour due to the presence of lignin and has a high oxygen chemical demand (about 1,095 g O2/m3). The WWTP uses the activated sludge process with extended aeration. This method allows an efficient removal of organics at the same time as it minimizes the sludge production.
For the modelling of the process it was necessary to collect historical data related to the WWTP’s performance over the last 3 years. This data was used as input values for the influent characterisation and as output values to achieve the treated effluent characterisation. Since the first simulation did not lead to the desired output results, it was necessary to proceed to the model calibration, by means of a more detailed study concerning the nutrient and organic fractions of the influent.
Once the model was calibrated, a study of the urea flowrate was conducted. The urea is added to the influent, before the beginning of the biological oxidation, as a way to satisfy the nitrogen requirements along the treatment process. However, this flowrate was never submitted to a study that evaluated, in a higher detail, the effective requirements of this nutrient. Thus, some simulations were done using the software, by decreasing successively the value of the urea flowrate and the results obtained were analyzed. Furthermore, these simulations were validated in the WWTP itself, at Portucel, through the decrease of the urea flowrate to half the normal value. Both the simulations and Portucel’s results showed that, actually, the addition of urea is not necessary because it does not affect the treatment process in a significant way, namely in terms of the removal of chemical oxygen demand. The simulations have also showed that the concentration of nitrogen in the final effluent diminishes significantly with the reduction of the urea flowrate, which could be advantageous in an environmental point of view
O papel da ubiquitinação na degradação da conexina em hiperglicemia

Uma das formas de regular a comunicação intercelular é através da degradação das Cx. A ubiquitinação de Cx43 é um sinal bem conhecido para a internalização e degradação das GJ. Apesar de estar bem descrito que em condições de hiperglicemia (HG) ocorre um aumento da degradação da Cx43, o mecanismo endocítico associado é ainda desconhecido. Assim, o principal objetivo deste trabalho é identificar os mecanismos e vias de sinalização envolvidos na degradação da Cx43 em células expostas a altas concentrações de glucose (HG). Os resultados obtidos neste estudo mostraram que em HG ocorre um aumento da ubiquitinação da Cx43, mediada pela proteína ligase de ubiquitina Nedd4, levando a uma diminuição nos seus níveis na membrana plasmática (MP) e a um concomitante aumento na sua degradaçãoCommunication between adjacent cells occurs through gap junctions (GJ) and these are formed of channels composed of proteins called connexins (Cx). The obtained results showed that in HG occurs an increase in Cx43 ubiquitination, mediated by the ubiquitin protein ligase Nedd4, resulting in a decrease in its levels on the plasma membrane, with the concomitant increase in its degradation
Designing a framework for mobile learning adoption and sustainable development
La digitalització del món és imparable, les funcionalitats que ofereix mobile learning proporcionen possibilitats
il·limitades per millorar l'aprenentatge, i la literatura existent sobre mobile learning ha evidenciat resultats positius. Tot i
això, hi ha una bretxa entre la disponibilitat de tecnologia i l'ús en educació, fins i tot quan succeeix, sovint no optimitza
els resultats. Existeixen pocs models per a l'adopció de mobile learning. En aquest context, l'objectiu principal
d'aquesta investigació és millorar l'adopció i l'ús sostenible de mobile learning a través de la creació d'un marc teòric.
Design-based research (DBR) és la metodologia que guia aquesta investigació. S'ha desenvolupat en quatre cicles de
disseny amb la utilització de múltiples metodologies d'investigació per desenvolupar, provar i refinar el marc teòric per
a l'adopció de mobile learning. El primer cicle de disseny identifica els problemes i les preguntes de recerca. Durant el
segon cicle de disseny s'identifiquen els principis de disseny i en base a aquests principis, es desenvolupa el primer
prototip. El tercer cicle de disseny inclou tres iteracions amb l'objectiu de testejar i refinar el disseny inicial. Una
primera iteració detalla els factors que afecten l'adopció de mobile learning a Catalunya i es basa en un judici
d'experts. Una segona iteració valida i ordena els factors identificats, un qüestionari recull les percepcions de 147
docents. Finalment, una tercera iteració va identificar els principals indicadors per avaluar i ajustar l'adopció de mobile

to improve learning, and the existing literature on mobile learning has proven positive results. However, there is a gap
between the availability of technology and its use in education, even when it happens, it often does not optimize the
results. There are few models for the adoption of mobile learning. In this context, the main objective of this research is
to improve the adoption and sustainable use of mobile learning by developing a framework. Design-based research
(DBR) is the methodology that guides this research. It has been developed in four design cycles to develop, test, and
refine the theoretical framework for the adoption of mobile learning. The first design cycle identifies problems and
research questions. During the second design cycle, the design principles are identified and based on these principles,
the first prototype is developed. The third design cycle includes three iterations with the aim of testing and refining the
initial design. A first iteration details the factors that affect the adoption of mobile learning in Catalonia and is based on
an expert judgment. A second iteration validates and orders the identified factors, a questionnaire collects the
perceptions of 147 teachers. Finally, a third iteration identifies the main indicators to evaluate and adjust the adoption
of mobile learning and proposes a scorecard. Based on the three design cycles, the fourth cycle proposes design
principles and presents a refined version of the first prototype of the framework

Quentin Tarantino’s 1994 masterpiece transformed cinematic storytelling by deviating from traditional narrative structures. Tarantino’s non-linear approach mirrors the deliberate fragmentation seen in postmodern literature, eliciting diverse audience interpretations. Taborda-Hernández’s audiovisual essay delves into Tarantino’s departure from linear storytelling, introducing an experimental linear perspective that prompts inquiries into the story’s efficacy in both linear and non-linear frameworks. This exploration highlights the complex interplay between narrative, characters, and the film’s non-linear structure. Thematic depth within non-linear storytelling enriches the viewing experience prompting reflection on the complex interweaving of human existence and morality, echoing Pulp Fiction’s multi-layered structure. This article contends that the film not only promoted but successfully achieved a continuous dialogue between the audience and the filmmaker, catalyzed by the film’s use of a non-linear structure and its consequential effects
