    Penilaian media bercetak untuk pendidikan pemakanan kanak-kanak berlebihan berat badan dan obes

    Media pendidikan bercetak merupakan saluran komunikasi yang utama dalam pendidikan kesihatan awam. Namun, kajian tempatan yang menilai penerimaan media pendidikan bercetak masih terhad di Malaysia. Maka, kajian ini dilakukan untuk menilai media pendidikan bercetak sedia ada yang mensasarkan obesiti di kalangan kanak-kanak untuk kegunaan dalam program pendidikan atau promosi pemakanan. Kajian ini melibatkan dua peringkat. Peringkat pertama merupakan tinjauan keperluan iaitu tinjauan terhadap media pendidikan bercetak sedia ada yang mensasarkan kanakkanak yang mengalami masalah berlebihan berat badan dan obes dan kajian rintis terhadap borang soal selidik yang dibentuk. Manakala peringkat kedua adalah penilaian penerimaan subjek kajian terhadap media pendidikan bercetak dan perubahan tahap pengetahuan mereka selepas membaca media pendidikan bercetak yang diberikan. Skala Likert digunakan dalam penilaian ini iaitu daripada 1 (sangat tidak memuaskan) hingga 5 (sangat memuaskan). Jumlah subjek dalam kajian ini adalah seramai 67 orang yang terdiri daripada golongan profesional dalam bidang pendidikan dan kesihatan (n = 7), subjek dewasa (n = 30) dan subjek kanak-kanak (n = 30). Di dalam kajian ini, subjek dewasa diberi satu buku kecil yang mengandungi maklumat berkenaan pencegahan dan pengurusan masalah obesiti di kalangan kanak-kanak seperti komplikasi masalah obesiti dan penyediaan makanan. Subjek kanak-kanak pula menerima satu buku kecil yang mengandungi maklumat berkenaan piramid makanan, kepentingan sarapan, aktiviti fi zikal dan sebagainya di samping satu poster yang mengandungi maklumat berkenaan pemilihan makanan sihat. Manakala golongan profesional menerima semua media pendidikan bercetak ini. Kajian ini mendapati penerimaan subjek terhadap isi kandungan, gambar, jadual dan rajah, warna, susun atur gambar, susun atur kandungan, bahasa dan saiz huruf bagi ketiga-tiga media pendidikan bercetak adalah pelbagai, iaitu daripada sangat tidak memuaskan hingga sangat memuaskan. Min skor penilaian keseluruhan terhadap buku kecil yang mensasarkan populasi dewasa adalah 3.9 ± 0.5 manakala min skor penilaian keseluruhan terhadap buku kecil yang mensasarkan populasi kanak-kanak adalah 4.3 ± 0.4 dan poster adalah 3.9 ± 0.9. Skor pengetahuan pemakanan golongan profesional meningkat dari 88.6 ± 11.2% kepada 95.5 ± 3.2% selepas membaca media tersebut. Skor pengetahuan pemakanan subjek dewasa juga meningkat secara signifi kan (p < 0.001) daripada 67.7 ± 13.6% kepada 84.8 ± 15.9%; bagi subjek kanak-kanak, skor pengetahuan pemakanan juga meningkat secara signifi kan (p < 0.001) daripada 65.3 ± 14.4% kepada 84.0 ± 11.4%. Kesimpulannya, media pendidikan bercetak sedia ada mendapat penerimaan yang baik dan dapat meningkatkan tahap pengetahuan kumpulan sasaran. Oleh itu, media pendidikan bercetak tersebut sesuai digunakan dalam mana-mana program pendidikan pemakanan yang berkaitan dengan obesiti kanak-kanak

    Growth performance of brown-golden marine microalga, Isochrysis sp., cultivated in alternative algal culture media

    The present study aimed to evaluate the suitability of introduced algal culture media as an alternative to the general enriched seawater media in the laboratory cultivation of Isochrysis sp., a marine microalga commonly cultivated for aquaculture purposes. Isochrysis sp. was established into culture in three replications using three experimental algal culture media (Walne’s medium as a control, China-contributed culture medium (CCM) and CCM supplemented with vitamins (CCM + Vit)). The experiment was performed with a continuous illumination for a period of seven days at 25±1°C. The effect of introduced algal culture media (CCM and CCM + Vit) on the growth performance of Isochrysis sp. was highlighted. CCM demonstrated promising results for the cultivation of Isochrysis sp. A maximum cell density of 9.16×106 ± 5.40×105 cells mL-1, which corresponded to an instantaneous growth rate (r) of 0.21 cell day-1 and a doubling time (T2) of 3.29 days, was observed in Isochrysis sp. cultivated in CCM. In view of its potential and reduced preparation labour, CCM may be recommended as an alternative to the general enriched seawater media in the cultivation of Isochrysis sp. in laboratories with basic facility as well as small- and medium-scale aquaculture hatcheries

    Multivariate biophysical markers predictive of mesenchymal stromal cell multipotency

    The capacity to produce therapeutically relevant quantities of multipotent mesenchymal stromal cells (MSCs) via in vitro culture is a common prerequisite for stem cell-based therapies. Although culture expanded MSCs are widely studied and considered for therapeutic applications, it has remained challenging to identify a unique set of characteristics that enables robust identification and isolation of the multipotent stem cells. New means to describe and separate this rare cell type and its downstream progenitor cells within heterogeneous cell populations will contribute significantly to basic biological understanding and can potentially improve efficacy of stem and progenitor cell-based therapies. Here, we use multivariate biophysical analysis of culture-expanded, bone marrow-derived MSCs, correlating these quantitative measures with biomolecular markers and in vitro and in vivo functionality. We find that, although no single biophysical property robustly predicts stem cell multipotency, there exists a unique and minimal set of three biophysical markers that together are predictive of multipotent subpopulations, in vitro and in vivo. Subpopulations of culture-expanded stromal cells from both adult and fetal bone marrow that exhibit sufficiently small cell diameter, low cell stiffness, and high nuclear membrane fluctuations are highly clonogenic and also exhibit gene, protein, and functional signatures of multipotency. Further, we show that high-throughput inertial microfluidics enables efficient sorting of committed osteoprogenitor cells, as distinct from these mesenchymal stem cells, in adult bone marrow.     Length-weight relationship and relative condition factor of pearl oyster, Pinctada fucata martensii, cultured in the Tieshangang Bay of the Beibu Gulf, Guangxi Province, China

    Tieshangang Bay in the Beibu Gulf, Guangxi of China, is a strategic location for pearl farming. Although water pollution has been reported in this bay but the general health of the pearl oyster, Pinctada fucata martensii,farmed there has never been assessed. The present study examined the condition of P. fucata martensii farmed in the Tieshangang Bay by analyzing its length-weight relationship (LWR) and relative condition factor (RCF). A total of 111 specimens were sampled for measuring their shell height and total weight for determining the LWR and RCF. The coefficient of correlation of the LWR was high (R2= 0.93), significant at 0.01 level. Negative allometric growth (b= 2.7048) was observed. However, P.fucata martensiiachieved the expected growth in terms of weight, as determined through the RCF (mean 1.13). Negative allometric growth is commonly reported on the wild Pinctadaspp. collected from different regions. Apparently, the water pollution in the Tieshangang Bay did not compromise the general health of the pearl oyster cultured there. Nevertheless, further study on the farm’s surrounding water quality and plankton availability is necessary to investigate the interaction between the growth of the oyster and its culture environment. In conclusion, the P. fucata martensiifarmed in the Tieshangang Bay was considered healthy and the bay is still suitable for pearl oyster farming

    Manipulating the monolayer : responsive and reversible control of colloidal inorganic nanoparticle properties

    Funding: EPSRC EP/K016342/1; Leverhulme Trust: RPG-2015-042For a wide range of nanomaterials, surface-bound molecules play a central role in defining properties, and are key to integration with other components – be they molecules, surfaces, or other nanoparticles. Predictable and general methods for manipulating the surface monolayer are therefore crucial to exploiting this new region of chemical space. This review highlights limitations of the few established methods for controlling nanoparticle-bound molecular functionality, then focuses on emerging new strategies. In particular, approaches that can achieve stimuli-responsive and reversible modification of surface-bound molecules in colloidal solution are examined, with an emphasis on using these methods to control nanoparticle properties such as solvent compatibility, catalytic activity and cytotoxicity. Finally, the outstanding challenges and future potential for precisely controlled nanoparticle bound monolayers are discussed.Publisher PDFPeer reviewe

    4-phenylbutyric Acid Regulates Collagen Synthesis and Secretion Induced by High Concentrations of Glucose in Human Gingival Fibroblasts

    High glucose leads to physio/pathological alterations in diabetes patients. We investigated collagen production in human gingival cells that were cultured in high concentrations of glucose. Collagen synthesis and secretion were increased when the cells were exposed to high concentrations of glucose. We examined endoplasmic reticulum (ER) stress response because glucose metabolism is related to ER functional status. An ER stress response including the expression of glucose regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), inositol requiring enzyme alpha (IRE-1α) and phosphoreukaryotic initiation factor alpha (p-eIF-2α) was activated in the presence of high glucose. Activating transcription factor 4 (ATF-4), a downstream protein of p-eIF-2α as well as a transcription factor for collagen, was also phosphorylated and translocalized into the nucleus. The chemical chaperone 4-PBA inhibited the ER stress response and ATF-4 phosphorylation as well as nuclear translocation. Our results suggest that high concentrations of glucose-induced collagen are linked to ER stress and the associated phosphorylation and nuclear translocation of ATF-4

    Characterization of wash-off from urban impervious surfaces and SuDS design criteria for source control under semi-arid conditions

    [EN] Knowledge about pollutant wash-off from urban impervious surfaces is a key feature for developing effective management strategies. Accordingly, further information is required about urban areas under semi-arid climate conditions at the sub-catchment scale. This is important for designing source control systems for pollution. In this study, a characterization of pollutant wash-off has been performed over sixteen months, at the sub-catchment scale for urban roads as impervious surfaces. The study was conducted in Valencia, Spain, a city with a Mediterranean climate. The results show high event mean concentrations for suspended solids (98 mg/l), organic matter (142 mgCOD/l, 25 mg BOD5/l), nutrients (3.7mgTN/l, 0.4 mg TP/l), and metals (0.23, 0.32, 0.62 and 0.17 mg/l for Cu, Ni, Pb, and Zn, respectively). The results of the runoff characterization highlight the need to control this pollution at its source, separately from wastewater because of their different characteristics. The wash-off, defined in terms of mobilized mass (g/m(2)) fits well with both process-based and statistical models, with the runoff volume and rainfall depth being the main explanatory variables. Based on these results and using information collected from hydrographs and pollutographs, an approach for sizing sustainable urban drainage systems (SuDS), focusing on water quality and quantity variables, has been proposed. By setting a concentration-based target (TSS discharged to receiving waters < 35 mg/l), the results indicate that for a SuDS type detention basin (DB), an off-line configuration performs better than an on-line configuration. The resulting design criterion, expressed as SuDS volume per unit catchment area, assuming a DB type SuDS, varies between 7 and 10 l/m(2).This research was funded through the SUPRIS-SUReS projects (Ref.
