    Multi-County Approach to Master Gardener Program in Rural Areas Yields Results

    Training and developing Master Gardeners (MG) on a multi-county basis benefited Extension personnel and clientele in an Appalachian region in Ohio. Rural clientele increasingly expect credible, research-based information on horticultural issues. Agents within an Extension district in eastern Ohio organized the first rural-based MG program in the state and developed a curriculum specific for the region. Cooperative development of MG allowed a number of counties to benefit from trained volunteers responding to horticultural questions. Since 1994, 10,000 hours of volunteer time have been documented with over 20,000 individual contacts made. The first Master Gardener of the Year in Ohio was a member of the first rural MG group

    The Piscataqua River/Great Bay estuary is a shared coastal embayment that forms the southernmost boundary between the states of Maine and New Hampshire. This rich coastal bay provides critical ecological, economic, and social benefits to the southern Maine and coastal New Hampshire region. The Great Bay estuary is such an important coastal resource that it is officially recognized as a coastal area of national significance by both the federal National Estuary Program and the federal National Estuarine Research Reserve program. The Piscataqua River/Great Bay estuary is fed by many rivers in New Hampshire, and by the Salmon Falls River, Great Works River, and Spruce Creek water- sheds in Maine. Collectively, the land area that contributes water flow to this treasured bi-state estuarine system is referred to as the “Piscataqua Region.” Within Maine, this region includes portions or all of ten Maine communities: Acton, Berwick, Eliot, Kittery, Lebanon, North Berwick, Sanford, South Berwick, Wells, and York

    Microstructures to control elasticity in 3D printing

    We propose a method for fabricating deformable objects with spatially varying elasticity using 3D printing. Using a single, relatively stiff printer material, our method designs an assembly of small-scale microstructures that have the effect of a softer material at the object scale, with properties depending on the microstructure used in each part of the object. We build on work in the area of metamaterials, using numerical optimization to design tiled microstructures with desired properties, but with the key difference that our method designs families of related structures that can be interpolated to smoothly vary the material properties over a wide range. To create an object with spatially varying elastic properties, we tile the object's interior with microstructures drawn from these families, generating a different microstructure for each cell using an efficient algorithm to select compatible structures for neighboring cells. We show results computed for both 2D and 3D objects, validating several 2D and 3D printed structures using standard material tests as well as demonstrating various example applications

    The staphylococcal QacR multidrug regulator binds a correctly spaced operator as a pair of dimers

    Expression of the Staphylococcus aureus plasmid-encoded QacA multidrug transporter is regulated by the divergently encoded QacR repressor protein. To circumvent the formation of disulfide-bonded degradation products, site-directed mutagenesis to replace the two cysteine residues in wild-type QacR was undertaken. Analysis of a resultant cysteineless QacR derivative indicated that it retained full DNA-binding activities in vivo and in vitro and continued to be fully proficient for the mediation of induction of qacA expression in response to a range of structurally dissimilar multidrug transporter substrates. The cysteineless QacR protein was used in cross-linking and dynamic light-scattering experiments to show that its native form was a dimer, whereas gel filtration indicated that four QacR molecules bound per DNA operator site. The addition of inducing compounds led to the dissociation of the four operator-bound QacR molecules from the DNA as dimers. Binding of QacR dimers to DNA was found to be dependent on the correct spacing of the operator half-sites. A revised model proposed for the regulation of qacA expression by QacR features the unusual characteristic of one dimer of the regulatory protein binding to each operator half-site by a process that does not appear to require the prior self-assembly of QacR into tetramers

Copyright @ 2013 Elsevier B.V.Numerous research studies have explored the effect of hypermedia on learners' performance using Web Based Instruction (WBI). A learner's performance is determined by their varying skills and abilities as well as various differences such as gender, cognitive style and prior knowledge. In this paper, we investigate how differences between individuals influenced learner's performance using a hypermedia system to accommodate an individual's preferences. The effect of learning performance is investigated to explore relationships between measurement attributes including gain scores (post-test minus pre-test), number of pages visited in a WBI program, and time spent on such pages. A data mining approach was used to analyze the results by comparing two clustering algorithms (K-Means and Hierarchical) with two different numbers of clusters. Individual differences had a significant impact on learner behavior in our WBI program. Additionally, we found that the relationship between attributes that measure performance played an influential role in exploring performance level; the relationship between such attributes induced rules in measuring level of a learners' performance

    Elimination of Porcine Epidemic Diarrhea Virus in an Animal Feed Manufacturing Facility

PLoS ONE 12(1): e0169612. doi:10.1371/journal.pone.0169612Porcine Epidemic Diarrhea Virus (PEDV) was the first virus of wide scale concern to be linked to possible transmission by livestock feed or ingredients. Measures to exclude pathogens, prevent cross-contamination, and actively reduce the pathogenic load of feed and ingredients are being developed. However, research thus far has focused on the role of chemicals or thermal treatment to reduce the RNA in the actual feedstuffs, and has not addressed potential residual contamination within the manufacturing facility that may lead to continuous contamination of finished feeds. The purpose of this experiment was to evaluate the use of a standardized protocol to sanitize an animal feed manufacturing facility contaminated with PEDV. Environmental swabs were collected throughout the facility during the manufacturing of a swine diet inoculated with PEDV. To monitor facility contamination of the virus, swabs were collected at: 1) baseline prior to inoculation, 2) after production of the inoculated feed, 3) after application of a quaternary ammonium-glutaraldehyde blend cleaner, 4) after application of a sodium hypochlorite sanitizing solution, and 5) after facility heat-up to 60°C for 48 hours. Decontamination step, surface, type, zone and their interactions were all found to impact the quantity of detectable PEDV RNA (P < 0.05). As expected, all samples collected from equipment surfaces contained PEDV RNA after production of the contaminated feed. Additionally, the majority of samples collected from non-direct feed contact surfaces were also positive for PEDV RNA after the production of the contaminated feed, emphasizing the potential role dust plays in cross-contamination of pathogen throughout a manufacturing facility. Application of the cleaner, sanitizer, and heat were effective at reducing PEDV genomic material (P < 0.05), but did not completely eliminate it
