    Between-Year Survival and Rank Transitions in Male Black-Capped Chickadees (\u3cem\u3ePoecile Atricapillus\u3c/em\u3e): A Multistate Modeling Approach

    In dominance-structured animal societies, variation in individual fitness is often related to social status. Like many passerine birds, Black-capped Chickadees (Poecile atricapillus) have a short average adult life-expectancy (Ø2 years); however, the maximum recorded life span is \u3e5× as long. Enhanced annual survival could contribute to greater lifetime reproductive success for male Black-capped Chickadees with high social rank. We used multistate capture–mark–recapture models to estimate annual survival of male Black-capped Chickadees in Ontario using resighting and recapture data collected from 1997 to 2002. Our goal was to evaluate support for an influence of rank on annual survival and estimate its effect size for a food-supplemented study site. We also statistically modeled the probability of between-year rank transitions. Model selection based on Akaike’s information criterion provided support for an effect of rank on survival. However, multimodel inference revealed that the size of the effect was rather small. Over the six study years, model-averaged estimates of the survival benefit of high versus low rank ranged from 5.0 to 7.3%. As expected, survival was strongly year-dependent, with model-averaged estimates of annual survival probability varying between 0.36 and 0.73. Age was an important predictor of the probability of rank transitions. Low-ranked second-year birds were less likely than older low-ranked birds to advance to high rank between years; likewise, high-ranked after-second-year birds were less likely to drop in rank. Other studies have found larger effects of rank on survival than we observed here. Future research should consider how interactions between social and environmental factors influence annual survival

    Role of School-Based Occupational Therapy Practitioners: Perspectives from Teachers and OTs

    Schools are the third largest employer of occupational therapists (OTs) in the United States (U.S. Bureau of Labor Statistics, 2021). Occupational Therapists have the distinctive ability to observe the child in their natural context of the school, a place where children participate in a variety of activities (Benson et al., 2015). The problem is that there continues to be discrepancies in the understanding and utilization of OT services in the school setting that affect the perceptions of both OTs and teachers regarding the OT role in school, and general service provision (Bolton & Plattner, 2020; Seruya & Garfinkel, 2020; Truong & Hodgetts, 2017). The purpose of this capstone project was to analyze perceptions of both OTs and teachers regarding the OT role in school, and service provision. An interview was conducted to gather these perceptions. Participants consisted of 20 OTs. It was concluded that the roles and responsibilities are understood by school-based Occupational Therapists, but there is a lack of knowledge by some teachers on the scope of school-based OT practice. There is a need for improved OT service delivery in the school system as well as a need for more school-based OT. A broadening of the realm of school-based OT practice to include mental health and social emotional functioning would also be of benefit to the practice. In addition, there is a need for improved education of school-based OT provided to OT students while in school.     Program to Increase Occupational Therapy Inclusion in Community Mental Health Practice

    Occupational therapists are qualified mental health providers, however, inadequate numbers of practitioners work in the community to treat individuals with mental health conditions. A program to advocate for the profession of occupational therapy in community-based mental health was created to increase capacity for occupational therapy practitioners in this setting. A two-hour virtual workshop was implemented to educate participants on occupational therapy\u27s role, the barriers that limit occupational therapy inclusion, and methods to increase occupational therapy inclusion. Results indicate the virtual workshop was effective at increasing an individuals knowledge on the roles, limitations, and opportunities for occupational therapy practitioners in community mental health settings. Future recommendations include providing additional workshops to improve advocacy efforts through increased attendance.https://soar.usa.edu/otdcapstones-spring2022/1024/thumbnail.jp

    Translational development of ABCB5+ dermal mesenchymal stem cells for therapeutic induction of angiogenesis in non-healing diabetic foot ulcers

    Background While rapid healing of diabetic foot ulcers (DFUs) is highly desirable to avoid infections, amputations and life-threatening complications, DFUs often respond poorly to standard treatment. GMP-manufactured skin-derived ABCB5+ mesenchymal stem cells (MSCs) might provide a new adjunctive DFU treatment, based on their remarkable skin wound homing and engraftment potential, their ability to adaptively respond to inflammatory signals, and their wound healing-promoting efficacy in mouse wound models and human chronic venous ulcers. Methods The angiogenic potential of ABCB5+ MSCs was characterized with respect to angiogenic factor expression at the mRNA and protein level, in vitro endothelial trans-differentiation and tube formation potential, and perfusion-restoring capacity in a mouse hindlimb ischemia model. Finally, the efficacy and safety of ABCB5+ MSCs for topical adjunctive treatment of chronic, standard therapy-refractory, neuropathic plantar DFUs were assessed in an open-label single-arm clinical trial. Results Hypoxic incubation of ABCB5+ MSCs led to posttranslational stabilization of the hypoxia-inducible transcription factor 1α (HIF-1α) and upregulation of HIF-1α mRNA levels. HIF-1α pathway activation was accompanied by upregulation of vascular endothelial growth factor (VEGF) transcription and increase in VEGF protein secretion. Upon culture in growth factor-supplemented medium, ABCB5+ MSCs expressed the endothelial-lineage marker CD31, and after seeding on gel matrix, ABCB5+ MSCs demonstrated formation of capillary-like structures comparable with human umbilical vein endothelial cells. Intramuscularly injected ABCB5+ MSCs to mice with surgically induced hindlimb ischemia accelerated perfusion recovery as measured by laser Doppler blood perfusion imaging and enhanced capillary proliferation and vascularization in the ischemic muscles. Adjunctive topical application of ABCB5+ MSCs onto therapy-refractory DFUs elicited median wound surface area reductions from baseline of 59 % (full analysis set, n = 23), 64 % (per-protocol set, n = 20) and 67 % (subgroup of responders, n = 17) at week 12, while no treatment-related adverse events were observed. Conclusions The present observations identify GMP-manufactured ABCB5+ dermal MSCs as a potential, safe candidate for adjunctive therapy of otherwise incurable DFUs and justify the conduct of a larger, randomized controlled trial to validate the clinical efficacy.     Effect of diabetes on caregiver burden in an observational study of individuals with Alzheimer’s disease

    Background The burden on caregivers of patients with Alzheimer’s disease (AD) is associated with the patient’s functional status and may also be influenced by chronic comorbid medical conditions, such as diabetes. This post-hoc exploratory analysis assessed whether comorbid diabetes in patients with AD affects caregiver burden, and whether caregivers with diabetes experience greater burden than caregivers without diabetes. Caregiver and patient healthcare resource use (HCRU) were also assessed. Methods Baseline data from the GERAS observational study of patients with AD and their caregivers (both n = 1495) in France, Germany and the UK were analyzed. Caregiver burden was assessed using the Zarit Burden Interview (ZBI). Caregiver time on activities of daily living (ADL: basic ADL; instrumental ADL, iADL) and supervision (hours/month), and caregiver and patient HCRU (outpatient visits, emergency room visits, nights hospitalized) were assessed using the Resource Utilization in Dementia instrument for the month before the baseline visit. Regression analyses were adjusted for relevant covariates. Time on supervision and basic ADL was analyzed using zero-inflated negative binomial regression. Results Caregivers of patients with diabetes (n = 188) were younger and more likely to be female (both p < 0.05), compared with caregivers of patients without diabetes (n = 1307). Analyses showed caregivers of patients with diabetes spent significantly more time on iADL (+16 %; p = 0.03; increases were also observed for basic ADL and total caregiver time but did not reach statistical significance) and had a trend towards increased ZBI score. Patients with diabetes had a 63 % increase in the odds of requiring supervision versus those without diabetes (p = 0.01). Caregiver and patient HCRU did not differ according to patient diabetes. Caregivers with diabetes (n = 127) did not differ from those without diabetes (n = 1367) regarding burden/time, but caregivers with diabetes had a 91 % increase in the odds of having outpatient visits (p = 0.01). Conclusions This cross-sectional analysis found caregiver time on iADL and supervision was higher for caregivers of patients with AD and diabetes versus without diabetes, while HCRU was unaffected by patient diabetes. Longitudinal analyses assessing change in caregiver burden over time by patient diabetes status may help clarify the cumulative impact of diabetes and AD dementia on caregiver burden

    Neuronal Profilin Isoforms Are Addressed by Different Signalling Pathways

    Profilins are prominent regulators of actin dynamics. While most mammalian cells express only one profilin, two isoforms, PFN1 and PFN2a are present in the CNS. To challenge the hypothesis that the expression of two profilin isoforms is linked to the complex shape of neurons and to the activity-dependent structural plasticity, we analysed how PFN1 and PFN2a respond to changes of neuronal activity. Simultaneous labelling of rodent embryonic neurons with isoform-specific monoclonal antibodies revealed both isoforms in the same synapse. Immunoelectron microscopy on brain sections demonstrated both profilins in synapses of the mature rodent cortex, hippocampus and cerebellum. Both isoforms were significantly more abundant in postsynaptic than in presynaptic structures. Immunofluorescence showed PFN2a associated with gephyrin clusters of the postsynaptic active zone in inhibitory synapses of embryonic neurons. When cultures were stimulated in order to change their activity level, active synapses that were identified by the uptake of synaptotagmin antibodies, displayed significantly higher amounts of both isoforms than non-stimulated controls. Specific inhibition of NMDA receptors by the antagonist APV in cultured rat hippocampal neurons resulted in a decrease of PFN2a but left PFN1 unaffected. Stimulation by the brain derived neurotrophic factor (BDNF), on the other hand, led to a significant increase in both synaptic PFN1 and PFN2a. Analogous results were obtained for neuronal nuclei: both isoforms were localized in the same nucleus, and their levels rose significantly in response to KCl stimulation, whereas BDNF caused here a higher increase in PFN1 than in PFN2a. Our results strongly support the notion of an isoform specific role for profilins as regulators of actin dynamics in different signalling pathways, in excitatory as well as in inhibitory synapses. Furthermore, they suggest a functional role for both profilins in neuronal nuclei

    Reconstructing the Deep Population History of Central and South America

    We report genome-wide ancient DNA from 49 individuals forming four parallel time transects in Belize, Brazil, the Central Andes, and the Southern Cone, each dating to at least 9,000 years ago. The common ancestral population radiated rapidly from just one of the two early branches that contributed to Native Americans today. We document two previously unappreciated streams of gene flow between North and South America. One affected the Central Andes by 4,200 years ago, while the other explains an affinity between the oldest North American genome associated with the Clovis culture and the oldest Central and South Americans from Chile, Brazil, and Belize. However, this was not the primary source for later South Americans, as the other ancient individuals derive from lineages without specific affinity to the Clovis-associated genome, suggesting a population replacement that began at least 9,000 years ago and was followed by substantial population continuity in multiple regions
