    Unlike other tumor necrosis factor family members, the cytotoxic ligand tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)/Apo-2L contains an unpaired cysteine residue (Cys(230)) in its receptor-binding domain. Here we show that the biological activity of both soluble recombinant TRAIL and cell-associated, full-length TRAIL is critically dependent on the presence of Cys(230). Mutation of Cys(230) to alanine or serine strongly affected its ability to kill target cells. Binding to its receptors was decreased by at least 200-fold, and the stability of its trimeric structure was reduced. In recombinant TRAIL, Cys(230) was found engaged either in interchain disulfide bridge formation, resulting in poorly active TRAIL, or in the chelation of one zinc atom per TRAIL trimer in the active, pro-apoptotic form of TRAIL

    Ground state non-universality in the random field Ising model

    Two attractive and often used ideas, namely universality and the concept of a zero temperature fixed point, are violated in the infinite-range random-field Ising model. In the ground state we show that the exponents can depend continuously on the disorder and so are non-universal. However, we also show that at finite temperature the thermal order parameter exponent one half is restored so that temperature is a relevant variable.     Differential Effects of the Mitochondria-Active Tetrapeptide SS-31 (D-Arg-dimethylTyr-Lys-Phe-NH<sub>2</sub>) and Its Peptidase-Targeted Prodrugs in Experimental Acute Kidney Injury.

    The mitochondria-active tetrapeptide SS-31 can control oxidative tissue damage in kidney diseases. To investigate other potential beneficial nephroprotective effects of SS-31, in vivo murine models of acute tubular injury and glomerular damage were developed. Reduction of acute kidney injury was demonstrated in mice treated with SS-31. The expression of mRNAs involved in acute inflammatory and oxidative stress responses in the diseased kidneys confirmed that SS-31 could regulate these pathways in our in vivo models. Furthermore, ex vivo histoenzymography of mouse kidneys showed that aminopeptidase A (APA), the enzyme involved in the processing of angiotensin (Ang) II to Ang III, was induced in the diseased kidneys, and its activity was inhibited by SS-31. As the renin-angiotensin system (RAS) is a main regulator of kidney functions, the modulation of Ang receptors (ATR) and APA by SS-31 was further investigated using mRNAs extracted from diseased kidneys. Following acute tubular and/or glomerular damage, the expression of the AT &lt;sub&gt;1&lt;/sub&gt; R mRNA was upregulated, which could be selectively downregulated upon SS-31 administration to the animals. At the same time, SS-31 was able to increase the expression of the AT &lt;sub&gt;2&lt;/sub&gt; R, which may contribute to limit renal damage. Consequently, SS-31-based prodrugs were developed as substrates and/or inhibitors for APA and were screened using cells expressing high levels of APA, showing its selective regulation by α-Glu-SS-31. Thus, a link between SS-31 and the RAS opens new therapeutic implications for SS-31 in kidney diseases

    Large time existence for 3D water-waves and asymptotics

    We rigorously justify in 3D the main asymptotic models used in coastal oceanography, including: shallow-water equations, Boussinesq systems, Kadomtsev-Petviashvili (KP) approximation, Green-Naghdi equations, Serre approximation and full-dispersion model. We first introduce a ``variable'' nondimensionalized version of the water-waves equations which vary from shallow to deep water, and which involves four dimensionless parameters. Using a nonlocal energy adapted to the equations, we can prove a well-posedness theorem, uniformly with respect to all the parameters. Its validity ranges therefore from shallow to deep-water, from small to large surface and bottom variations, and from fully to weakly transverse waves. The physical regimes corresponding to the aforementioned models can therefore be studied as particular cases; it turns out that the existence time and the energy bounds given by the theorem are always those needed to justify the asymptotic models.     Mass-spectrometric identification of primary biological particle markers and application to pristine submicron aerosol measurements in Amazonia

    The detection of primary biological material in submicron aerosol by means of thermal desorption/electron impact ionization aerosol mass spectrometry was investigated. Mass spectra of amino acids, carbohydrates, small peptides, and proteins, all of which are key building blocks of biological particles, were recorded in laboratory experiments. Several characteristic marker fragments were identified. The intensity of the marker signals relative to the total organic mass spectrum allows for an estimation of the content of primary biological material in ambient organic aerosol. The developed method was applied to mass spectra recorded during AMAZE-08, a field campaign conducted in the pristine rainforest of the central Amazon Basin, Brazil, during the wet season of February and March 2008. The low abundance of identified marker fragments places upper limits of 7.5% for amino acids and 5.6% for carbohydrates on the contribution of primary biological aerosol particles (PBAP) to the submicron organic aerosol mass concentration during this time period. Upper limits for the absolute submicron concentrations for both compound classes range from 0.01 to 0.1 μg m−3. Carbohydrates and proteins (composed of amino acids) make up for about two thirds of the dry mass of a biological cell. Thus, our findings suggest an upper limit for the PBAP mass fraction of about 20% to the submicron organic aerosol measured in Amazonia during AMAZE-08

    In the regime of weakly transverse long waves, given long-wave initial data, we prove that the nondimensionalized water wave system in an infinite strip under influence of gravity and surface tension on the upper free interface has a unique solution on [0,{T}/\eps] for some \eps independent of constant T.T.     Effective damping in the Raman cooling of trapped ions

    We present a method of treating the interaction of a single three-level ion with two laser beams. The idea is to apply a unitary transformation such that the exact transformed Hamiltonian has one of the three levels decoupled for all values of the detunings. When one takes into account damping, the evolution of the system is governed by a master equation usually obtained via adiabatic approximation under the assumption of far-detuned lasers.
