    Peroxiredoxin 6 is required for blood vessel integrity in wounded skin

    Peroxiredoxin 6 (Prdx6) is a cytoprotective enzyme with largely unknown in vivo functions. Here, we use Prdx6 knockout mice to determine its role in UV protection and wound healing. UV-mediated keratinocyte apoptosis is enhanced in Prdx6-deficient mice. Upon skin injury, we observe a severe hemorrhage in the granulation tissue of knockout animals, which correlates with the extent of oxidative stress. At the ultrastructural level endothelial cells appear highly damaged, and their rate of apoptosis is enhanced. Knock-down of Prdx6 in cultured endothelial cells also increases their susceptibility to oxidative stress, thus confirming the sensitivity of this cell type to loss of Prdx6. Wound healing studies in bone marrow chimeric mice demonstrate that Prdx6-deficient inflammatory and endothelial cells contribute to the hemorrhage phenotype. These results provide insight into the cross-talk between hematopoietic and resident cells at the wound site and the role of reactive oxygen species in this interplay

    Science in the Swiss public: the state of science communication and public engagement with science in Switzerland

    Science communication and public engagement with science have repeatedly been called for in recent years, particularly during the COVID-19 pandemic. Therefore, die Swiss Academies of the Arts and Sciences have set up an expert group to assess the state of science communication in Switzerland, and to provide recommendations for how to improve it. The expert group report is based on a comprehensive review of the available interdisciplinary scholarship analyzing science communication and public engagement with science in Switzerland. Selectively, it also incorporates original data, international findings, and secondary analyses where little or no published scholarly work was available. The report covers a wide range of facets of science communication and public engagement in Switzerland, from public attitudes towards science over individuals and organizations engaging in science communication and engagement formats to news and social media representations of science. On this basis, it formulates 20 recommendations for improving science communication in Switzerland

    Bei Projekten zur Digitalisierung in Geisteswissenschaften ist heute die Realisierung grĂ¶ĂŸerer, ĂŒberregionaler und ĂŒber das World Wide Web abfragbarer Lösungen erforderlich. Die BeitrĂ€ge dieses Bandes wurden auf der Tagung des Staatsarchivs Hamburg und des Zentrums "Geisteswissenschaften in der digitalen Welt" an der UniversitĂ€t Hamburg am 10. und 11. April 2006 gehalten. Sie leisten einen interdisziplinĂ€ren Beitrag zur erforderlichen Standardisierung dieser Angebote, die erst den dringend notwendigen Austausch erleichtern und die gemeinsame Nutzung strukturierter Daten ermöglichen kann.Today, digitization projects in the Humanities require the implementation of larger, supraregional solutions. The contributions in this volume were presented at the conference of the Hamburg State Archives and the Center for the Humanities in the Digital World at the University of Hamburg on April 10 and 11, 2006. They make an interdisciplinary contribution to the required standardisation of corresponding services which can only facilitate the urgently needed exchange of information and make it possible to share structured data

    Die vorliegende Publikation versteht sich als Ideengeber fĂŒr die universitĂ€re Lehre in der Lehrerbildung in Gestalt eines sogenannten Transferpakets. Sie berichtet ĂŒber Ergebnisse des Teilprojekts ‚Spotlights-Lehre‘ des Projekts ‚Schnittstellen gestalten – Lehrerbildung entlang des Leitbildes des "Reflective Practitioner" an der UniversitĂ€t Bremen' im BMBF-Programm ‚QualitĂ€tsoffensive Lehrerbildung‘. Inhaltlich geht es um innovative Lehrprojekte zur Verzahnung und Vernetzung von Fachwissenschaft und Fachdidaktik, die das Ziel verfolgen, Fragmentierungserfahrungen von Lehramtsstudierenden in den FĂ€chern Mathematik, Englisch, Romanistik, Geschichte und Inklusive Didaktik zu reduzieren. Zentrales Anliegen dieses Transferpakets ist es, Transferstrategien bereitzustellen, an Beispielen zu illustrieren und in Lehrbeschreibungen einzubetten. Umfassend wird gezeigt, wie die Designprozesse zur Lehre in den beiden zentralen Modellprojekten der FĂ€cher Englisch und Mathematik gestaltet sowie die Transfer- und Vernetzungsstrategien gewonnen wurden

    Elimusertib has anti-tumor activity in preclinical patient-derived pediatric solid tumor models

    The small molecule inhibitor of ataxia telangiectasia and Rad3-related protein (ATR), elimusertib, is currently being tested clinically in various cancer entities in adults and children. Its preclinical anti-tumor activity in pediatric malignancies, however, is largely unknown. We here assessed the preclinical activity of elimusertib in 38 cell lines and 32 patient-derived xenograft (PDX) models derived from common pediatric solid tumor entities. Detailed in vitro and in vivo molecular characterization of the treated models enabled the evaluation of response biomarkers. Pronounced objective response rates were observed for elimusertib monotherapy in PDX, when treated with a regimen currently used in clinical trials. Strikingly, elimusertib showed stronger anti-tumor effects than some standard of care chemotherapies, particularly in alveolar rhabdomysarcoma PDX. Thus, elimusertib has strong preclinical anti-tumor activity in pediatric solid tumor models, which may translate to clinically meaningful responses in patients

    The First European Interdisciplinary Ewing Sarcoma Research Summit

    The European Network for Cancer Research in Children and Adolescents (ENCCA) provides an interaction platform for stakeholders in research and care of children with cancer. Among ENCCA objectives is the establishment of biology-based prioritization mechanisms for the selection of innovative targets, drugs, and prognostic markers for validation in clinical trials. Specifically for sarcomas, there is a burning need for novel treatment options, since current chemotherapeutic treatment protocols have met their limits. This is most obvious for metastatic Ewing sarcoma (ES), where long term survival rates are still below 20%. Despite significant progress in our understanding of ES biology, clinical translation of promising laboratory results has not yet taken place due to fragmentation of research and lack of an institutionalized discussion forum. To fill this gap, ENCCA assembled 30 European expert scientists and five North American opinion leaders in December 2011 to exchange thoughts and discuss the state of the art in ES research and latest results from the bench, and to propose biological studies and novel promising therapeutics for the upcoming European EWING2008 and EWING2012 clinical trials
