    Impact of Caffeine on Ethanol-Induced Stimulation and Sensitization: Changes in ERK and DARPP-32 Phosphorylation in Nucleus Accumbens

    Background: Caffeine is frequently consumed with ethanol to reduce the impairing effects induced by ethanol, including psychomotor slowing or incoordination. Both drugs modulate dopamine (DA)-related markers in accumbens (Acb), and Acb DA is involved in voluntary locomotion and locomotor sensitization. The present study determined whether caffeine can affect locomotion induced by acute and repeated ethanol administration in adult male CD-1 mice. Methods: Acute administration of caffeine (7.5 to 30.0 mg/kg) was evaluated for its effects on acute ethanol-induced (1.5 to 3.5 g/kg) changes in open-field horizontal locomotion, supported rearing, and rearing not supported by the wall. DA receptor-dependent phosphorylation markers were assessed: extracellular signal-regulated kinase (pERK), and dopamine-and cAMP-regulated phosphoprotein Mr32kDa phosphorylated at threonine 75 site (pDARPP-32-Thr75) in Acb core and shell. Acutely administered caffeine was also evaluated in ethanol-sensitized (1.5 g/kg) mice. Results: Acute ethanol decreased both types of rearing. Caffeine increased supported rearing but did not block ethanol -induced decreases in rearing. Both substances increased horizontal locomotion in a biphasic manner, and caffeine potentiated ethanol-induced locomotion. Although ethanol administered repeatedly induced sensitization of locomotion and unsupported rearing, acute administration of caffeine to ethanol-sensitized mice in an ethanol-free state resulted in blunted stimulant effects compared with those seen in ethanol-naĂŻve mice. Ethanol increased pERK immunoreactivity in both subregions of the Acb, but coadministration with caffeine blunted this increase. There were no effects on pDARPP-32(Thr75) immunoreactivity. Conclusions: The present results demonstrated that, after the first administration, caffeine potentiated the stimulating actions of ethanol, but did not counteract its suppressant or ataxic effects. Moreover, our results show that caffeine has less activating effects in ethanol-sensitized animals

    Synthesis and homopolymerization studies of vinylimidazolium salts

    The preparations and characterizations of several monomeric vinylimidazolium salts are presented from the quaternizations of 1-vinylimidazole and 2-methyl-1-vinylimidazole with n-alkyl iodides and with dimethyl sulphate. Although vinylimidazolium salts have been reported in the patent literature, many of these salts were not isolated and characterized prior to their polymerization. From the reactions of 1-vinylimidazole with n-alkyl iodides, a homologous series of 3-n-alkyl-1-vinylimidazolium iodides were prepared in which the longer chain derivatives appeared to form micelles in aqueous solution. Dicationic crosslinking agents were also prepared through similar quaternization reactions. All the cationic vinyl monomers were homopolymerized in aqueous solution by free radical initiation.     Tunanetra adalah tidak berfungsinya indra penglihatan yaitu mata karena kecelakaan atau kecacatan. Menyebabkan tunanetra sulit memahami aturan minum obat setiap harinya ini memicu pengaruh pemahaman dan kepatuhan minum obat pasien penyandang tunanetra. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan etiket Braille terhadap tingkat kepahaman dan kepatuhan aturan minum obat. Jenis penelitian termasuk metode Pra-eskperimen dengan rancangan perbandingan kelompok statis (Static Group Comprarison),dengan pengambilan sampel Consecutive sampling, instrument penelitian dengan kuesioner kepada 30 responden. Hasil penelitian menunjukkan tingkat pemahaman pada kelompok kontrol 61,67%, sementara kelompok perlakuan sebesar 100%. Untuk kepatuhan kelompok kontrol sebesar 3,65%, dan kelompok perlakuan sebesar 7,92%. Hasil uji Mann Whitey didapat nilai p signifikansi 0,000 < 0,05 terhadap pengaruh penggunaan etiket Braille terhadap pemahaman dan kepatuhan minum obat.     Bupropion Increases Selection of High Effort Activity in Rats Tested on a Progressive Ratio/Chow Feeding Choice Procedure: Implications for Treatment of Effort-Related Motivational Symptoms

    Background: Depression and related disorders are characterized by deficits in behavioral activation, exertion of effort, and other psychomotor/motivational dysfunctions. Depressed patients show alterations in effort-related decision making and a bias towards selection of low effort activities. It has been suggested that animal tests of effort-related decision making could be useful as models of motivational dysfunctions seen in psychopathology. Methods: Because clinical studies have suggested that inhibition of catecholamine uptake may be a useful strategy for treatment of effort-related motivational symptoms, the present research assessed the ability of bupropion to increase work output in rats responding on a test of effort-related decision-making (ie, a progressive ratio/chow feeding choice task). With this task, rats can choose between working for a preferred food (high-carbohydrate pellets) by lever pressing on a progressive ratio schedule vs obtaining a less preferred laboratory chow that is freely available in the chamber. Results: Bupropion (10.0–40.0 mg/kg intraperitoneal) significantly increased all measures of progressive ratio lever pressing, but decreased chow intake. These effects were greatest in animals with low baseline levels of work output on the progressive ratio schedule. Because accumbens dopamine is implicated in effort-related processes, the effects of bupropion on markers of accumbens dopamine transmission were examined. Bupropion elevated extracellular dopamine levels in accumbens core as measured by microdialysis and increased phosphorylated dopamine and cyclic-AMP related phosphoprotein 32 kDaltons (pDARPP-32) immunoreactivity in a manner consistent with D1 and D2 receptor stimulation. Conclusion: The ability of bupropion to increase exertion of effort in instrumental behavior may have implications for the pathophysiology and treatment of effort-related motivational symptoms in humans

    Flunitrazepam Excretion Patterns using the Abuscreen OnTrak and OnLine Immunoassays: Comparison with GC-MS

    A study was conducted to compare the performance of the OnLine and OnTrak immunoassays for benzodiazepines with gas chromatographic-mass spectrometric (GC-MS) analysis in detecting flunitrazepam (FNP) and its metabolites in human urine. Urine was collected over a 72-h period from six individuals (four male and two female) who had taken a single oral dose of either 1 or 4 mg of FNP. The OnTrak assay was run at a 100-ng/mL cutoff of nordiazepam (NDP), and the OnLine assay was run with a standard curve from zero to 200 ng/mL of NDP with and without β-glucuronidase treatment. Each sample was analyzed by GC-MS using FNP, 7-amino-FNP, 3-hydroxy-FNP, desmethyl-FNP, 7-amino-3-hydroxy-FNP, and desmethyl-3-hydroxy-FNP as standards with β-glucuronidase treatment. The specimens from the 1-mg dose did not yield a positive result by immunoassay over the 72-h collection period. Specimens from the 4-mg dose did yield positive results in both immunoassays. The time of the first positive result ranged from 4 to 12 h, and the time to the last positive result ranged from 18 to 60 h. Treatment of the samples with β-glucuronidase increased the OnLine values between 20 and 60%, but it did not appreciably increase the detection time. GC-MS analysis showed no detectable levels of FNP, 3-hydroxy-FNP, desmethyl-FNP, 7-amino-3-hydroxy-FNP, and desmethyl-3-hydroxy-FNP. However, all samples collected past time zero showed detectable levels of 7-amino-FNP (> 2 ng/mL) with peak concentrations at 12-36 h. The peak levels of 7-amino-FNP by GC-MS paralleled the peak levels of the immunoassay response. The amount of 7-amino-FNP metabolite quantitated by GC-MS, however, accounted for only 15-20% of the total immunoassay crossreactive FNP metabolite

    Effort-Related Motivational Effects of the VMAT-2 Inhibitor Tetrabenazine: Implications for Animal Models of the Motivational Symptoms of Depression

    Motivated behaviors are often characterized by a high degree of behavioral activation, and work output and organisms frequently make effort-related decisions based upon cost/benefit analyses. Moreover, people with major depression and other disorders often show effort-related motivational symptoms such as anergia, psychomotor retardation, and fatigue. It has been suggested that tasks measuring effort-related choice behavior could be used as animal models of the motivational symptoms of depression, and the present studies characterized the effort-related effects of the vesicular monoamine transport (VMAT) inhibitor tetrabenazine. Tetrabenazine produces depressive symptoms in humans and, because of its selective inhibition of VMAT-2, it preferentially depletes dopamine (DA). Rats were assessed using a concurrent fixed-ratio 5/chow feeding choice task that is known to be sensitive to dopaminergic manipulations. Tetrabenazine shifted response choice in rats, producing a dose-related decrease in lever pressing and a concomitant increase in chow intake. However, it did not alter food intake or preference in parallel free-feeding choice studies. The effects of tetrabenazine on effort-related choice were reversed by the adenosine A2A antagonist MSX-3 and the antidepressant bupropion. A behaviorally active dose of tetrabenazine decreased extracellular DA in nucleus accumbens and increased expression of DARPP-32 in accumbens medium spiny neurons in a pattern indicative of reduced transmission at both D1 and D2 DA receptors. These experiments demonstrate that tetrabenazine, which is used in animal models to produce depression-like effects, can alter effort-related choice behavior. These studies have implications for the development of animal models of the motivational symptoms of depression and related disorders

    Theory of quantum radiation observed as sonoluminescence

    Sonoluminescence is explained in terms of quantum radiation by moving interfaces between media of different polarizability. In a stationary dielectric the zero-point fluctuations of the electromagnetic field excite virtual two-photon states which become real under perturbation due to motion of the dielectric. The sonoluminescent bubble is modelled as an optically empty cavity in a homogeneous dielectric. The problem of the photon emission by a cavity of time-dependent radius is handled in a Hamiltonian formalism which is dealt with perturbatively up to first order in the velocity of the bubble surface over the speed of light. A parameter-dependence of the zero-order Hamiltonian in addition to the first-order perturbation calls for a new perturbative method combining standard perturbation theory with an adiabatic approximation. In this way the transition amplitude from the vacuum into a two-photon state is obtained, and expressions for the single-photon spectrum and the total energy radiated during one flash are given both in full and in the short-wavelengths approximation when the bubble is larger than the wavelengths of the emitted light. It is shown analytically that the spectral density has the same frequency-dependence as black-body radiation; this is purely an effect of correlated quantum fluctuations at zero temperature. The present theory clarifies a number of hitherto unsolved problems and suggests explanations for several more.     Dynamical Casimir effect without boundary conditions

    The moving-mirror problem is microscopically formulated without invoking the external boundary conditions. The moving mirrors are described by the quantized matter field interacting with the photon field, forming dynamical cavity polaritons: photons in the cavity are dressed by electrons in the moving mirrors.     Genetic inhibition of neurotransmission reveals role of glutamatergic input to dopamine neurons in high-effort behavior

    Midbrain dopamine neurons are crucial for many behavioral and cognitive functions. As the major excitatory input, glutamatergic afferents are important for control of the activity and plasticity of dopamine neurons. However, the role of glutamatergic input as a whole onto dopamine neurons remains unclear. Here we developed a mouse line in which glutamatergic inputs onto dopamine neurons are specifically impaired, and utilized this genetic model to directly test the role of glutamatergic inputs in dopamine-related functions. We found that while motor coordination and reward learning were largely unchanged, these animals showed prominent deficits in effort-related behavioral tasks. These results provide genetic evidence that glutamatergic transmission onto dopaminergic neurons underlies incentive motivation, a willingness to exert high levels of effort to obtain reinforcers, and have important implications for understanding the normal function of the midbrain dopamine system.Fil: Hutchison, M.     Propiedades microbiolĂłgicas en un suelo de la Patagonia Argentina bajo la influencia de diferentes especies forestales

    191-197In forest systems, leaf litter is usually the main source of nutrients for the vegetation and microorganisms. Leaf litter composition may vary according to the biomass and quality of the materials produced by different species. Tree roots affect the development and activity of microflora. The aim of this study was to evaluate the influence of two implanted tree species, with different composition of plant residues, on the soil characteristics. The soil is classified as an Andisol and is located in the Trevelín Forest Station, Chubut, Argentina, Lat. 43° S, Long 71°31' West. The samples were extracted from the surface soil of two forest plots with a dominant species each: Radiata Pine (Pinus radiata D. Don.) and European Oak (Quercus robur). The following analytical determinations were carried out on soil samples: organic carbon, microbial respiration, deshydrogenase activity, microbial count (bacterial communities, amilolytic, actinomyces and fungi) as well as the functional diversity of associated microbial communities. The Shannon-Weaver index (H) was calculated. The soil under Oak trees showed the highest values of microbial respiration, organic carbon and amilolytic and bacterial community counts. No significant differences were found in deshydrogenase activity, actinomyces and fungi counts between species. The principal component analysis showed significant variations in the physiology of microbial communities associated with these two tree species. The microorganism H diversity index was significantly greater for Pine. Plants residues of broadleaf species such as Oak have lower contents of recalcitrant substances than conifer species, and this fact, associated with the higher development of total and amylolytic bacteria, could result in an increase in the decomposition of plant residues in the soil and in higher organic carbon and microbial respiration values
