    Background During the last years the quantification of immune response under immunological challenges, e.g. parasitation, has been a major focus of research. In this context, the expression of immune response genes in teleost fish has been surveyed for scientific and commercial purposes.     Constraining noncommutative field theories with holography

    An important window to quantum gravity phenomena in low energy noncommutative (NC) quantum field theories (QFTs) gets represented by a specific form of UV/IR mixing. Yet another important window to quantum gravity, a holography, manifests itself in effective QFTs as a distinct UV/IR connection. In matching these two principles, a useful relationship connecting the UV cutoff ΛUV\Lambda_{\rm UV}, the IR cutoff ΛIR\Lambda_{\rm IR} and the scale of noncommutativity ΛNC\Lambda_{\rm NC}, can be obtained. We show that an effective QFT endowed with both principles may not be capable to fit disparate experimental bounds simultaneously, like the muon g2g-2 and the masslessness of the photon. Also, the constraints from the muon g2g-2 preclude any possibility to observe the birefringence of the vacuum coming from objects at cosmological distances. On the other hand, in NC theories without the UV completion, where the perturbative aspect of the theory (obtained by truncating a power series in ΛNC2 \Lambda_{\rm NC}^{-2}) becomes important, a heuristic estimate of the region where the perturbative expansion is well-defined E/ΛNC1E/ \Lambda_{\rm NC} \lesssim 1, gets affected when holography is applied by providing the energy of the system EE a ΛNC\Lambda_{\rm NC}-dependent lower limit. This may affect models which try to infer the scale ΛNC\Lambda_{\rm NC} by using data from low-energy experiments.Comment: 4 pages, version to be published in JHE

    Development and preliminary testing of the new five-level version of EQ-5D (EQ-5D-5L)

    This article introduces the new 5-level EQ-5D (EQ-5D-5L) health status measure. EQ-5D currently measures health using three levels of severity in five dimensions. A EuroQol Group task force was established to find ways of improving the instrument's sensitivity and reducing ceiling effects by increasing the number of severity levels. The study was performed in the United Kingdom and Spain. Severity labels for 5 levels in each dimension were identified using response scaling. Focus groups were used to investigate the face and content validity of the new versions, including hypothetical health states generated from those versions. Selecting labels at approximately the 25th, 50th, and 75th centiles produced two alternative 5-level versions. Focus group work showed a slight preference for the wording 'slight-moderate-severe' problems, with anchors of 'no problems' and 'unable to do' in the EQ-5D functional dimensions. Similar wording was used in the Pain/Discomfort and Anxiety/Depression dimensions. Hypothetical health states were well understood though participants stressed the need for the internal coherence of health states. A 5-level version of the EQ-5D has been developed by the EuroQol Group. Further testing is required to determine whether the new version improves sensitivity and reduces ceiling effects

    Superantigens (SAgs) are potent exotoxins secreted by Staphylococcus aureus and Streptococcus pyogenes. They target a large fraction of T cell pools to set in motion a "cytokine storm" with severe and sometimes life-threatening consequences typically encountered in toxic shock syndrome (TSS). Given the rapidity with which TSS develops, designing timely and truly targeted therapies for this syndrome requires identification of key mediators of the cytokine storm's initial wave. Equally important, early host responses to SAgs can be accompanied or followed by a state of immunosuppression, which in turn jeopardizes the host's ability to combat and clear infections. Unlike in mouse models, the mechanisms underlying SAg-associated immunosuppression in humans are ill-defined. In this work, we have identified a population of innate-like T cells, called mucosa-associated invariant T (MAIT) cells, as the most powerful source of pro-inflammatory cytokines after exposure to SAgs. We have utilized primary human peripheral blood and hepatic mononuclear cells, mouse MAIT hybridoma lines, HLA-DR4-transgenic mice, MAIThighHLA-DR4+ bone marrow chimeras, and humanized NOD-scid IL-2Rγnull mice to demonstrate for the first time that: i) mouse and human MAIT cells are hyperresponsive to SAgs, typified by staphylococcal enterotoxin B (SEB); ii) the human MAIT cell response to SEB is rapid and far greater in magnitude than that launched by unfractionated conventional T, invariant natural killer T (iNKT) or γδ T cells, and is characterized by production of interferon (IFN)-γ, tumor necrosis factor (TNF)-α and interleukin (IL)-2, but not IL-17A; iii) high-affinity MHC class II interaction with SAgs, but not MHC-related protein 1 (MR1) participation, is required for MAIT cell activation; iv) MAIT cell responses to SEB can occur in a T cell receptor (TCR) Vβ-specific manner but are largely contributed by IL-12 and IL-18; v) as MAIT cells are primed by SAgs, they also begin to develop a molecular signature consistent with exhaustion and failure to participate in antimicrobial defense. Accordingly, they upregulate lymphocyte-activation gene 3 (LAG-3), T cell immunoglobulin and mucin-3 (TIM-3), and/or programmed cell death-1 (PD-1), and acquire an anergic phenotype that interferes with their cognate function against Klebsiella pneumoniae and Escherichia coli; vi) MAIT cell hyperactivation and anergy co-utilize a signaling pathway that is governed by p38 and MEK1/2. Collectively, our findings demonstrate a pathogenic, rather than protective, role for MAIT cells during infection. Furthermore, we propose a novel mechanism of SAg-associated immunosuppression in humans. MAIT cells may therefore provide an attractive therapeutic target for the management of both early and late phases of severe SAg-mediated illnesses

    O impacto da psoríase na autoimagem e sua influência no trabalho

    Psoriasis is a skin disease that affects the body in a systemic way of affected by it, may also affect the joints. Its impact goes beyond the skin, causing psychosocial damage and occupational arising from prejudice and social stigma. This study aimed to evaluate the impact of psoriasis on self-image and their influence on the work of the individuals affected. It is a qualitative study of the case study type with 7 patients in the Reference Center, Support and Treatment of Carriers of Paraiba State of psoriasis, installed at the Dermatology Clinic of the University Hospital Lauro Wanderley - HULW/UFPB - João Pessoa / PB. Data were collected through semi-structured interview, categorized and treated according to the Content Analysis. The results showed interference in the self-image of participants, mainly caused by situations involving prejudice, but there were few significant influences to the work of the respondents, only difficulties of functional performance in cases of arthropathic psoriasis. They also suggest that it is important to comprehensive care by multidisciplinary team and social support for individuals with psoriasis for good promotion of quality of life and are necessary awareness actions to society in order to reduce situations of prejudice and stigma.A psoríase é uma doença dermatológica que afeta de maneira sistêmica o organismo dos acometidos por ela, podendo afetar também as articulações. Seu impacto vai além da pele, ocasionando prejuízos psicossociais e ocupacionais, decorrentes do preconceito e estigma social. O presente trabalho objetivou avaliar o impacto da psoríase na autoimagem e sua influência no trabalho dos indivíduos acometidos. É um estudo qualitativo do tipo estudo de caso com 7 pacientes do Centro de Referência, Apoio e Tratamento aos Portadores da Psoríase do Estado da Paraíba, instalado no ambulatório de dermatologia do Hospital Universitário Lauro Wanderley – HULW/UFPB – João Pessoa/PB. Os dados foram coletados por meio de entrevista semiestruturada, categorizados e tratados de acordo com a Análise de Conteúdo. Os resultados indicaram interferência na autoimagem dos participantes, causadas principalmente por situações que envolvem preconceito, porém houveram poucas influências significativas em relação ao trabalho dos entrevistados, apenas dificuldades de desempenho funcional nos casos de psoríase artropática. Também sugerem que é importante a assistência integral por equipe multiprofissional e o apoio social para os indivíduos com psoríase para a boa promoção da qualidade de vida, bem como são necessárias ações de conscientização para a sociedade, a fim de reduzir situações de preconceito e estigma

    No association between a candidate TCF7L2 variant and risk of breast or ovarian cancer

    <p>Abstract</p> <p>Background</p> <p>TCF7L2 is a transcription factor involved in Wnt/β-catenin signaling which has a variant known to be associated with risk of Type 2 diabetes and, in some studies, with risk of certain cancers, including familial breast cancer. No studies of ovarian cancer have been reported to date.</p> <p>Methods</p> <p>Two clinic-based case-control studies at the Mayo Clinic were assessed including 798 breast cancer cases, 843 breast cancer controls, 391 ovarian cancer cases, and 458 ovarian cancer controls. Genotyping at <it>TCF7L2 </it>rs12255372 used a 5' endonuclease assay, and statistical analysis used logistic regression among participants as a whole and among <it>a priori</it>-defined subsets.</p> <p>Results</p> <p>No associations with risk of breast or ovarian cancer were observed (ordinal model, p = 0.62 and p = 0.75, respectively). In addition, no associations were observed among sub-groups defined by age, BMI, family history, stage, grade, histology, or tumor behavior.</p> <p>Conclusion</p> <p>Although the biology of the Wnt/β-catenin signaling pathway and prior association between rs12255372 and numerous phenotypes warranted examination of this <it>TCF7L2 </it>SNP, no compelling evidence for association with breast or ovarian cancer was observed.</p

    Multiple primary tumours in women following breast cancer, 1973–2000

    We investigated the predictors of the risk of developing a second primary cancer after breast cancer, this occurring in about 12% of affected women. The analysis included 335 191 females, registered in the National Cancer Institute's Surveillance Epidemiology and End Results (SEER) database, who had been diagnosed with breast cancer. Observed numbers of subsequent cancers in the SEER database with a first breast cancer diagnosed from 1973 to 2000 were compared with the expected numbers based on age-adjusted incidence rates to calculate standardised incidence ratios. Kaplan–Meier curves were conducted to determine the median time until the second primary cancer diagnosis. Average number of years until diagnosis varied by site and by age as well as median years until second cancer diagnosis. Most cancer risks decreased with age, but there was an increase in aging-related cancers such as lung cancer. The median years of follow-up were well beyond the 5-year mark. Breast cancer survivors should be advised of their increased risk for developing certain cancers in their lifetime
