    The expression of VDACs and Bcl2 family genes in pituitary adenomas : clinical correlations and postsurgical outcomes

    Introduction: Pituitary adenomas (PAs) are benign tumors with high prevalence and, occasionally, aggressive course. The tumorigenesis of these lesions is not completely understood at the molecular level. BAK1 and BAX proteins play fundamental roles in apoptosis and seem to interact with VDAC proteins, whose expressions have been markedly altered in cancer, impacting their prognosis. Objective: to evaluate the gene expression of VDAC1, VDAC2, BAK1 and BAX and their association with clinical and imaging characteristics in PA. Methods: Clinical-epidemiological data were collected from 117 tumor samples from patients affected by PA. Invasiveness was assessed by the Knosp scale. Gene expression was examined by real-time PCR. Relative expression analysis was performed by 2^(-DDCt) method. Results: The sample was mainly composed of women (69/117 - 57.2%). Tumor subtypes observed were Non-Functioning (NF) (73/117 - 62.4%), Acromegaly (24/117 - 20.5%) and Cushing's Disease (CD) (20/117 - 17.1%). Compared to normal tissue, there was a significant reduction in VDAC1 expression in the Acromegaly (p=0.029) and NF (p=0.002) groups. BAX expression was lower in all groups (p <0.001; p=0.007; P =0.005). No difference was found in VDAC2 and BAK1 expression, compared to normal pituitary. Overexpression of VDAC2 occurred in PAs with post-surgical regrowth (p=0.042). A strongly negative correlation was observed in BAX and BAK1 expression in CD. Conclusion: The results indicated that downregulations of VDAC1 and BAX may be related to resistance to apoptosis. In contrast, overexpression of VDAC2 in regrowing PAs suggests an antiapoptotic role for this gene. In summary, the genes evaluated might be involved in the biopathology of PAs

    The rapid atmospheric monitoring system of the Pierre Auger Observatory

    The Pierre Auger Observatory is a facility built to detect air showers produced by cosmic rays above 10(17) eV. During clear nights with a low illuminated moon fraction, the UV fluorescence light produced by air showers is recorded by optical telescopes at the Observatory. To correct the observations for variations in atmospheric conditions, atmospheric monitoring is performed at regular intervals ranging from several minutes (for cloud identification) to several hours (for aerosol conditions) to several days (for vertical profiles of temperature, pressure, and humidity). In 2009, the monitoring program was upgraded to allow for additional targeted measurements of atmospheric conditions shortly after the detection of air showers of special interest, e. g., showers produced by very high-energy cosmic rays or showers with atypical longitudinal profiles. The former events are of particular importance for the determination of the energy scale of the Observatory, and the latter are characteristic of unusual air shower physics or exotic primary particle types. The purpose of targeted (or 'rapid') monitoring is to improve the resolution of the atmospheric measurements for such events. In this paper, we report on the implementation of the rapid monitoring program and its current status. The rapid monitoring data have been analyzed and applied to the reconstruction of air showers of high interest, and indicate that the air fluorescence measurements affected by clouds and aerosols are effectively corrected using measurements from the regular atmospheric monitoring program. We find that the rapid monitoring program has potential for supporting dedicated physics analyses beyond the standard event reconstruction

    Wound-healing evaluation of ointment from Stryphnodendron adstringens (barbatimão) in rat skin

    This study evaluated the cicatrizant effect of an ointment containing 1% of the ethyl-acetate fraction extracted from the stem bark of "barbatimão" (Stryphnodendron adstringens), in wounds made in the skin of rats, after 4, 7 and 10 days of treatment. Control wounds were treated with a base ointment without extract. The proliferation of keratinocytes in the area of reepithelialization was evaluated by counting the number of epithelial cells that were blocked in metaphase by vincristine sulfate. The length of the reepithelialized margin and the contraction of the wound were measured. Topical application of the "barbatimão" ointment stimulated proliferation of the keratinocytes, but had no effect on the length of the epithelium or on the contraction of the wounds.Neste estudo, avaliou-se a atividade cicatrizante de uma pomada contendo uma fração acetato de etila 1% obtida de cascas de "barbatimão" (Stryphnodendron adstringens) em feridas excisionais na pele de ratos após 4, 7 e 10 dias de tratamento. Feridas controle foram tratadas com pomada base, sem extrato. A proliferação dos queratinócitos na área reepitelizada foi avaliada através da contagem do número de queratinócitos bloqueados em metáfase, pelo sulfato de vincristina. O comprimento da margem reepitelizada e a contração das feridas foram mensurados. As feridas tratadas com barbatimão apresentaram um maior número de mitoses do que aquelas tratadas com a pomada base, em todos os tempos avaliados. A aplicação tópica da pomada de "barbatimão" estimulou a proliferação epitelial contudo não teve efeito sobre a migração dos queratinócitos ou sobre a contração das feridas

    Anatomia do cone medular aplicada à via epidural de administração de fármacos em macacos-prego ( Sapajus libidinosus )

    Resumo: Com este estudo objetivou-se descrever a topografia do cone medular do macaco-prego (Sapajus libidinosus) a fim de fornecer suporte para que a realização de procedimentos anestésicos, bem como exames de mielografia e coleta de líquor, dentre outros procedimentos que utilizam a via epidural. Para tanto foram dissecados oito animais, sendo seis machos e duas fêmeas, de diferentes faixas etárias. Rebateu-se a pele para retirada da musculatura da região dorsal, exposição de toda a coluna vertebral e identificação das vértebras lombares e sacrais. Para estabelecer o final da medula espinhal e medir o comprimento do cone medular, foi aberto todo o canal vertebral lombossacro, seccionando-se lateralmente os arcos vertebrais. Em seguida a duramáter foi seccionada para visualização do cone medular e observação da relação topográfica deste com as vértebras. Todos os animais apresentaram cinco vértebras lombares e três vértebras sacrais. As vértebras se apresentaram, de forma geral, muito próximas e com os processos espinhosos bem desenvolvidos e direcionados em sentido cranial. O cone medular dos macacos-prego situou-se entre as vértebras L2 e L5, com a base localizando-se com maior frequência na altura da vértebra L3, enquanto o ápice em L4. O comprimento corporal (espaço interarcual occiptoatlântico até o espaço interarcual sacrocaudal) variou de 22,9 a 31,8cm, com média de 27,44 ±3,1cm enquanto que comprimento do cone medular variou de 1,70 a 3,51cm, com média de 2,47 ±0,57cm. Não houve correlação entre o tamanho do corpo e o comprimento do cone medular (r = 0,212). Conclui-se que apesar das variações do comprimento e posicionamento do cone medular, o seu ápice não ultrapassa a articulação lombossacral, tornando seguro o acesso ao espaço epidural por esta via
