    Industrial and environmental applications of white-rot fungi

    Abstract White-rot-fungi (WRF) are the only organisms able to degrade the whole wood components (i.e. lignin, cellulose and hemicellulose). This ability is due to the secretion of extracellular nonspecific ligninolytic enzymes during their secondary metabolism usually triggered by nutrient exhaustion. The non-specificity of these enzymes enables them to transform a great variety of recalcitrant and hazardous pollutants such as polycyclic aromatic hydrocarbons (PAHs), pesticides, fuels, alkanes, polychlorinated biphenyls (PCBs), explosives and synthetic dyes. In addition, their extracellular nature allows WRF to access non-polar and insoluble compounds. This makes WRF very appealing for their application to different industrial and biotechnological processes. Also, new potential commercial products and processes from the fungal treatment of lignocellulosic materials may arise. The implementation of such applications would contribute to the establishment of a more sustainable industry and the development of a circular economy

    Removal efficiency of nitrate, phosphate, fecal and total coliforms by horizontal subsurface flow-constructed wetland from domestic wastewater

    Background: Constructed wetlands are systems designed based on the utilization of natural processes, including vegetation, soil, and their associated microbial assemblage to assist in treating different types of wastewater. Methods: Two local Appalachian plants (Louis latifolia and Phragmites australis) were planted into smallscale constructed wetlands to treat domestic wastewater in the North of Iran. The influent wastewater and the effluent from each wetland were sampled daily for 120 days. Experiments were conducted based on the mean ± standard deviation (SD) by analysis of variance (ANOVA). Results: It was found that nitrate, phosphate, fecal and total coliforms were reduced by 84.4%, 94.4%, 96.3%, 93.9% for P. australis and 73.3%, 64.0%, 94.4%, 92.1% for L. latifolia, respectively. Conclusion: According to the results, by using the HF-CW technology with L. latifolia and P. australis plants, the treated wastewater fully meets the wastewater discharge parameters of WHO standards

    Enzymatic synthesis of Tinuvin

    Coupling of 3-(3-tert-butyl-4-hydroxyphenyl) propionic acid methylester to 1H-benzotriazole using a laccase from Trametes hirsuta was studied. The potentially resulting coupling product Tinuvin 1130 is an important UV-absorber used in polymer based materials. Oxidation of the phenol by the laccase led to homomolecular coupling reactions while the laccase did not attack 1H-benzotriazole. Due to the homomolecular reaction of the phenol in the presence of laccase coupling of phenol and 1H-benzotriazole was only observed when 1H-benzotriazole was applied in four-fold molar excess. The reaction was monitored by UV/vis spectroscopy, TLC and MS (ion trap) analysis. Coupling of 1H-benzotriazole took place in ortho position according to the postulated mechanism

    Immobilization of the white-rot fungus Anthracophyllum discolor to degrade the herbicide atrazine

    Herbicides cause environmental concerns because they are toxic and accumulate in the environment, food products and water supplies. There is a need to develop safe, efficient and economical methods to remove them from the environment, often by biodegradation. Atrazine is such herbicide. White-rot fungi have the ability to degrade herbicides of potential utility. This study formulated a novel pelletized support to immobilize the white-rot fungus Anthracophyllum discolor to improve its capability to degrade the atrazine using a biopurification system (BS). Different proportions of sawdust, starch, corn meal and flaxseed were used to generate three pelletized supports (F1, F2 and F3). In addition, immobilization with coated and uncoated pelletized supports (CPS and UPS, respectively) was assessed. UPS-F1 was determined as the most effective system as it provided high level of manganese peroxidase activity and fungal viability. The half-life (t1/2) of atrazine decreased from 14 to 6 days for the control and inoculated samples respectively. Inoculation with immobilized A. discolor produced an increase in the fungal taxa assessed by DGGE and on phenoloxidase activity determined.     Fermi/LAT discovery of gamma-ray emission from a relativistic jet in the narrow-line quasar PMN J0948+0022

    We report the discovery by the Large Area Telescope (LAT) onboard the Fermi Gamma-ray Space Telescope of high-energy gamma-ray emission from the peculiar quasar PMN J0948+0022 (z=0.5846). The optical spectrum of this object exhibits rather narrow Hbeta (FWHM(Hbeta) ~ 1500 km s^-1), weak forbidden lines and is therefore classified as a narrow-line type I quasar. This class of objects is thought to have relatively small black hole mass and to accrete at high Eddington ratio. The radio loudness and variability of the compact radio core indicates the presence of a relativistic jet. Quasi simultaneous radio-optical-X-ray and gamma-ray observations are presented. Both radio and gamma-ray emission (observed over 5-months) are strongly variable. The simultaneous optical and X-ray data from Swift show a blue continuum attributed to the accretion disk and a hard X-ray spectrum attributed to the jet. The resulting broad band spectral energy distribution (SED) and, in particular, the gamma-ray spectrum measured by Fermi are similar to those of more powerful FSRQ. A comparison of the radio and gamma-ray characteristics of PMN J0948+0022 with the other blazars detected by LAT shows that this source has a relatively low radio and gamma-ray power, with respect to other FSRQ. The physical parameters obtained from modelling the SED also fall at the low power end of the FSRQ parameter region discussed in Celotti & Ghisellini (2008). We suggest that the similarity of the SED of PMN J0948+0022 to that of more massive and more powerful quasars can be understood in a scenario in which the SED properties depend on the Eddington ratio rather than on the absolute power.Comment: 10 pages, 5 figures, accepted for publication on ApJ Main Journal.     Integrated use of residues from olive mill and winery for lipase production by solid state fermentation with Aspergillus sp

    Two phase olive mill waste (TPOMW) is presently the major waste produced by the olive mill industry. This waste has potential to be used as substrate for solid state fermentation (SSF) despite of its high concentration of phenolic compounds and low nitrogen content. In this work, it is demonstrated that mixtures of TPOMW with winery wastes support the production of lipase by Aspergillus spp. By agar plate screening, Aspergillus niger MUM 03.58, Aspergillus ibericus MUM 03.49 and Aspergillus uvarum MUM 08.01 were chosen for lipase production by SSF. Plackett-Burman experimental design was employed to evaluate the effect of substrate composition and time on lipase production. The highest amounts of lipase were produced by A. ibericus on a mixture of TPOMW, urea and exhausted grape mark (EGM). Urea was found to be the most influent factor for the lipase production. Further optimization of lipase production by A. ibericus using a full factorial design (32) conducted to optimal conditions of substrate composition (0.073 g urea/g and 25% of EGM) achieving 18.67 U/g of lipolytic activity.Jose Manuel Salgado is grateful for Postdoctoral fellowship (EX-2010-0402) of Education Ministry of Spanish Government.     Bioinformatic Analysis Reveals High Diversity of Bacterial Genes for Laccase-Like Enzymes

    Fungal laccases have been used in various fields ranging from processes in wood and paper industries to environmental applications. Although a few bacterial laccases have been characterized in recent years, prokaryotes have largely been neglected as a source of novel enzymes, in part due to the lack of knowledge about the diversity and distribution of laccases within Bacteria. In this work genes for laccase-like enzymes were searched for in over 2,200 complete and draft bacterial genomes and four metagenomic datasets, using the custom profile Hidden Markov Models for two- and three- domain laccases. More than 1,200 putative genes for laccase-like enzymes were retrieved from chromosomes and plasmids of diverse bacteria. In 76% of the genes, signal peptides were predicted, indicating that these bacterial laccases may be exported from the cytoplasm, which contrasts with the current belief. Moreover, several examples of putatively horizontally transferred bacterial laccase genes were described. Many metagenomic sequences encoding fragments of laccase-like enzymes could not be phylogenetically assigned, indicating considerable novelty. Laccase-like genes were also found in anaerobic bacteria, autotrophs and alkaliphiles, thus opening new hypotheses regarding their ecological functions. Bacteria identified as carrying laccase genes represent potential sources for future biotechnological applications

    Characterization of Tajogaite volcanic plumes detected over the Iberian Peninsula from a set of satellite and ground-based remote sensing instrumentation

    Three volcanic plumes were detected during the Tajogaite volcano eruptive activity (Canary Islands, Spain, September–December 2021) over the Iberian Peninsula. The spatiotemporal evolution of these events is characterised by combining passive satellite remote sensing and ground-based lidar and sun-photometer systems. The inversion algorithm GRASP is used with a suite of ground-based remote sensing instruments such as lidar/ ceilometer and sun-photometer from eight sites at different locations throughout the Iberian Peninsula. Satellite observations showed that the volcanic ash plumes remained nearby the Canary Islands covering a mean area of 120 ± 202 km2 during the whole period of eruptive activity and that sulphur dioxide plumes reached the Iberian Peninsula

    Lenalidomide and dexamethasone with or without clarithromycin in patients with multiple myeloma ineligible for autologous transplant: a randomized trial

    Although case-control analyses have suggested an additive value with the association of clarithromycin to continuous lenalidomide and dexamethasone (Rd), there are not phase III trials confirming these results. In this phase III trial, 286 patients with MM ineligible for ASCT received Rd with or without clarithromycin until disease progression or unacceptable toxicity. The primary endpoint was progression-free survival (PFS). With a median follow-up of 19 months (range, 0-54), no significant differences in the median PFS were observed between the two arms (C-Rd 23 months, Rd 29 months; HR 0.783, p = 0.14), despite a higher rate of complete response (CR) or better in the C-Rd group (22.6% vs 14.4%, p = 0.048). The most common G3-4 adverse events were neutropenia [12% vs 19%] and infections [30% vs 25%], similar between the two arms; however, the percentage of toxic deaths was higher in the C-Rd group (36/50 [72%] vs 22/40 [55%], p = 0.09). The addition of clarithromycin to Rd in untreated transplant ineligible MM patients does not improve PFS despite increasing the ?CR rate due to the higher number of toxic deaths in the C-Rd arm. Side effects related to overexposure to steroids due to its delayed clearance induced by clarithromycin in this elderly population could explain these results.
