    The Role of Trust in Relationship of Leadership Style and Organizational Effectiveness: Mediation Mechanism

    This study is aimed at explaining the effect of leadership style to organizational effectiveness through trust as mediators. The number of samples of this study was 267 university employee respondents at Universitas Negeri Semarang. The sampling technique used was purposive sampling. The methods of collecting data used were observation, interview, and questionnaire. Hypothesis test used was path coefficient value which was done through t-test using Smart PLS 3.0. Leadership style directly gave positive effect to organizational effectiveness. Furthermore, mediation mechanism had been proven that trust gave important role in determining the relationship between leadership style and organizational effectiveness. This study was expected to contribute to management practice in the field of university in which university need pay attention to effective leadership to encourage employees’ trust. As a result, trust can encourage organizational effectiveness. This study also gave contribution to the development of leadership style literature and organizational effectiveness in the field of non-profit organization. This study attempted to answer the need of previous study by adding mediation variable. It was expected that mediation could explain the mechanism of the relationship between leadership style and organizational effectiveness completely.     Improving Organizational Management Skills for Women of Association Members of Sido Rukun Guyub in Semarang City

    Tujuan dari program pengabdian ini selain untuk mendiseminasikan hasil penelitian penulis juga untuk memberikan pelatihan keterampilan manajemen pada pengusaha wanita. Metode pelaksanaan program dilakukan melalui pelatihan yang terstruktur, dimulai dari tahap pertama, brainstorming dengan pengusaha wanita yang tergabung dalam asosiasi. Tahapan kedua, mengadakan kegiatan pelatihan pada pengusaha wanita untuk mengenalkan konsep fungsi-fungsi manajemen. Dalam pelatihan ini terdapat kegiatan simulasi praktik membuat perencanaan manajerial. Tahap ketiga, evaluasi untuk merefleksikan sejauhmana pelatihan telah dipahami dengan baik. Dengan kegiatan pengabdian ini, diharapkan dapat menjadi sarana bagi pengusaha wanita dalam meningkatkan keterampilan manajerial pengelolaan usaha. Hasil yang didapat adalah pelaku UMKM asosiasi Guyub Sido Rukun sangat aktif dan mereka mampu melakukan pemetaan diri melalui pemahaman teori manajemen, menganalisis situasi yang dihadapi dan solusi yang hendak dilakukan ke depan.The purpose of this service program is to disseminate the results of the author's research as well as to provide management skills training to female entrepreneurs. The method of implementing the program is carried out through structured training, starting from the first stage, brainstorming with women entrepreneurs who are members of the association. The second stage, held training activities for women entrepreneurs to introduce the concept of management functions. In this training there are practical simulation activities to make managerial planning. The third stage is evaluation to reflect the extent to which training has been well understood. With this service activity, it is hoped that it can become a means for female entrepreneurs in improving business management managerial skills. The results obtained were the association of SMEs, Guyub Sido Rukun, who was very active and they were able to carry out self mapping through understanding management theory, analyzing the situation faced and the solutions to be carried forward

    Signal Transduction Pathways in the Pentameric Ligand-Gated Ion Channels

    The mechanisms of allosteric action within pentameric ligand-gated ion channels (pLGICs) remain to be determined. Using crystallography, site-directed mutagenesis, and two-electrode voltage clamp measurements, we identified two functionally relevant sites in the extracellular (EC) domain of the bacterial pLGIC from Gloeobacter violaceus (GLIC). One site is at the C-loop region, where the NQN mutation (D91N, E177Q, and D178N) eliminated inter-subunit salt bridges in the open-channel GLIC structure and thereby shifted the channel activation to a higher agonist concentration. The other site is below the C-loop, where binding of the anesthetic ketamine inhibited GLIC currents in a concentration dependent manner. To understand how a perturbation signal in the EC domain, either resulting from the NQN mutation or ketamine binding, is transduced to the channel gate, we have used the Perturbation-based Markovian Transmission (PMT) model to determine dynamic responses of the GLIC channel and signaling pathways upon initial perturbations in the EC domain of GLIC. Despite the existence of many possible routes for the initial perturbation signal to reach the channel gate, the PMT model in combination with Yen's algorithm revealed that perturbation signals with the highest probability flow travel either via the β1-β2 loop or through pre-TM1. The β1-β2 loop occurs in either intra- or inter-subunit pathways, while pre-TM1 occurs exclusively in inter-subunit pathways. Residues involved in both types of pathways are well supported by previous experimental data on nAChR. The direct coupling between pre-TM1 and TM2 of the adjacent subunit adds new insight into the allosteric signaling mechanism in pLGICs. © 2013 Mowrey et al

    The transport mechanism of the mitochondrial ADP/ATP carrier

    The mitochondrial ADP/ATP carrier imports ADP from the cytosol and exports ATP from the mitochondrial matrix, which are key transport steps for oxidative phosphorylation in eukaryotic organisms. The transport protein belongs to the mitochondrial carrier family, a large transporter family in the inner membrane of mitochondria. It is one of the best studied members of the family and serves as a paradigm for the molecular mechanism of mitochondrial carriers. Structurally, the carrier consists of three homologous domains, each composed of two transmembrane α-helices linked with a loop and short α-helix on the matrix side. The transporter cycles between a cytoplasmic and matrix state in which a central substrate binding site is alternately accessible to these compartments for binding of ADP or ATP. On both the cytoplasmic and matrix side of the carrier are networks consisting of three salt bridges each. In the cytoplasmic state, the matrix salt bridge network is formed and the cytoplasmic network is disrupted, opening the central substrate binding site to the intermembrane space and cytosol, whereas the converse occurs in the matrix state. In the transport cycle, tighter substrate binding in the intermediate states allows the interconversion of conformations by lowering the energy barrier for disruption and formation of these networks, opening and closing the carrier to either side of the membrane in an alternating way. Conversion between cytoplasmic and matrix states might require the simultaneous rotation of three domains around a central translocation pathway, constituting a unique mechanism among transport proteins.     Oligomeric Status and Nucleotide Binding Properties of the Plastid ATP/ADP Transporter 1: Toward a Molecular Understanding of the Transport Mechanism

    Background: Chloroplast ATP/ADP transporters are essential to energy homeostasis in plant cells. However, their molecular mechanism remains poorly understood, primarily due to the difficulty of producing and purifying functional recombinant forms of these transporters. Methodology/Principal Findings: In this work, we describe an expression and purification protocol providing good yields and efficient solubilization of NTT1 protein from Arabidopsis thaliana. By biochemical and biophysical analyses, we identified the best detergent for solubilization and purification of functional proteins, LAPAO. Purified NTT1 was found to accumulate as two independent pools of well folded, stable monomers and dimers. ATP and ADP binding properties were determined, and Pi, a co-substrate of ADP, was confirmed to be essential for nucleotide steady-state transport. Nucleotide binding studies and analysis of NTT1 mutants lead us to suggest the existence of two distinct and probably inter-dependent binding sites. Finally, fusion and deletion experiments demonstrated that the C-terminus of NTT1 is not essential for multimerization, but probably plays a regulatory role, controlling the nucleotide exchange rate.     Factors associated with teenage marital pregnancy among Bangladeshi women

    <p>Abstract</p> <p>Background</p> <p>Teenage pregnancy is a public health concern both in developed and developing world. In Bangladesh, most of the first pregnancies occur immediately after marriage, especially among teenagers. Although women aged 15-29 years are the most fertility contributing women in Bangladesh, studies are not yet conducted on teenage pregnancy within this group of women. In the current study, an attempt had been made to identify the factors affecting teenage marital pregnancy in women aged 15-29 years.</p> <p>Methods</p> <p>A cross sectional survey was carried out in 389 women, selected with a convenience sampling technique. Participants were selected on the basis of two criteria, such as married women and age within 15-29 years. We excluded women aged more than 29 years as we attempted to conduct study within high fertility contributing women and with the assumption that they may provide data subjected to relatively high level of recall bias as marital pregnancy may be a longer past event to them. In the analysis, we applied bi-variate and multi-variate logistic regression technique to find out odds ratio of teenage marital pregnancy.</p> <p>Results</p> <p>Result revealed that 72.5% of the participants experienced first marital pregnancy during their teenage, with a mean age of 17.88 years (SD = 2.813). Multivariate logistic regression analysis revealed that participants aged 20-24 years had higher likelihood (OR 1.971, 95% CI 1.132 to 3.434), whereas participants aged 25-29 years had lower likelihood (OR 0.054, 95% CI 0.016 to 0.190) of experiencing teenage marital pregnancy compared to participants aged 15-19 years. In addition, participants desired for >2 children had significant higher odds (OR 3.573, 95% CI 1.910 to 6.684) and participants born in urban area had significant lower odds (OR 0.458, 95% CI 0.228 to 0.919) for teenage marital pregnancy.</p> <p>Conclusions</p> <p>Based on the findings, we conclude that in order to reduce teenage marital pregnancy, consideration should be given on women's desired number of children and birth place so that women's desired number of children is limited to within two children, and that rural women get increased working and other related opportunities that may contribute in delaying teenage pregnancy.</p

    A propofol binding site on mammalian GABAA receptors identified by photolabeling

    Propofol is the most important intravenous general anesthetic in current clinical use. It acts by potentiating GABA(A) receptors, but where it binds to this receptor is not known and has been a matter of some controversy. We have synthesized a novel propofol analogue photolabeling reagent that has a biological activity very similar to that of propofol. We confirmed that this reagent labeled known propofol binding sites in human serum albumin which have been identified using X-ray crystallography. Using a combination of the protiated label and a deuterated version, and mammalian receptors labeled in intact membranes, we have identified a novel binding site for propofol in GABA(A) receptors consisting of both β(3) homopentamers and α(1)β(3) heteropentamers. The binding site is located within the β subunit, at the interface between the transmembrane domains and the extracellular domain, and lies close to known determinants of anesthetic sensitivity in transmembrane segments TM1 and TM2
