    Revisiting the footprints of climate change in Arctic marine food webs: An assessment of knowledge gained since 2010

    In 2011, a first comprehensive assessment of the footprints of climate change on Arctic marine ecosystems (such as altered distribution ranges, abundances, growth and body conditions, behaviours and phenologies, as well as community and regime shifts) was published. Here, we re-assess the climate-driven impacts reported since then, to elucidate to which extent and how observed ecological footprints have changed in the following decade (2011 to 2021). In total, 98 footprints have been described and analysed. Most of those impacts reported in the 2011 assessment are reconfirmed and can, hence, be assumed as continuing trends. In addition, novel footprints (behavioural changes, diet changes, altered competition and pathogen load) are described. As in 2011, most reported footprints are related to changes in distribution ranges, abundances, biomass and production. Range shifts have mostly been observed for fish species, while behavioural changes have mainly been reported for mammals. Primary production has been observed to further increase in Arctic seas. The footprints on pelagic herbivores, particularly the key species Calanus spp., are less clear. In comparison to 2011, more complex, cascading effects of climate change, such as increased bowhead whale body conditions due to increased primary production, have been reported. The observed footprints, and the trends that they indicate, strongly suggest that due to further northward range shifts of sub-Arctic and boreal species Arctic seas are likely to experience increasing species richness in the future. However, a tipping point may be reached, characterized by subsequent biodiversity decline, when Arctic-endemic species will go extinct as ocean warming and/or acidification will exceed their physiological adaptation capacity.Overall, the future Arctic Ocean will very likely experience increasing numbers and intensities of climate-change footprints

    Factor structure and psychometric properties of the trier inventory for chronic stress (TICS) in a representative german sample

    <p>Abstract</p> <p>Background</p> <p>Chronic stress results from an imbalance of personal traits, resources and the demands placed upon an individual by social and occupational situations. This chronic stress can be measured using the Trier Inventory for Chronic Stress (TICS). Aims of the present study are to test the factorial structure of the TICS, report its psychometric properties, and evaluate the influence of gender and age on chronic stress.</p> <p>Methods</p> <p>The TICS was answered by <it>N </it>= 2,339 healthy participants aged 14 to 99. The sample was selected by random-route sampling. Exploratory factor analyses with Oblimin-rotated Principal Axis extraction were calculated. Confirmatory factor analyses applying Robust Maximum Likelihood estimations (MLM) tested model fit and configural invariance as well as the measurement invariance for gender and age. Reliability estimations and effect sizes are reported.</p> <p>Results</p> <p>In the exploratory factor analyses, both a two-factor and a nine-factor model emerged. Confirmatory factor analyses resulted in acceptable model fit (RMSEA), with model comparison fit statistics corroborating the superiority of the nine-factor model. Most factors were moderately to highly intercorrelated. Reliabilities were good to very good. Measurement invariance tests gave evidence for differential effects of gender and age on the factor structure. Furthermore, women and younger individuals, especially those aged 35 to 44, tended to report more chronic stress than men and older individuals.</p> <p>Conclusions</p> <p>The proposed nine-factor structure could be factorially validated, results in good scale reliability, and heuristically can be grouped by two higher-order factors: "High Demands" and "Lack of Satisfaction". Age and gender represent differentiable and meaningful contributors to the perception of chronic stress.</p

    Are perinatal measures associated with a dolescent mental health? A retrospective e xploration with original data from psychiatric c ohorts

    Background Perinatal markers of prenatal development are associated with offspring psychiatric symptoms. However, there is little research investigating the specificity of perinatal markers for the development of specific disorders. This study aimed to explore if perinatal markers are specifically associated with adolescent substance use disorder (SUDs). Methods Adolescent participants from two study centers, one for SUD patients (n = 196) and one for general psychopathology (n = 307), were recruited for participation. Since the SUD participants presented with a number of comorbid disorders, we performed a 1-on-1 matching procedure, based on age, gender, and specific pattern of comorbid disorders. This procedure resulted in n = 51 participants from each group. From all participants and their mothers we recorded perinatal markers (mode of birth, weeks of completed pregnancy, birth weight, Apgar score after 5 min) as well as intelligence quotient (IQ). The SUD sample additionally filled out the Youth Safe Report (YSR) as well as the PQ-16 and the DUDIT. We aimed to distinguish the two groups (SUD sample vs. general psychiatric sample) based on the perinatal variables via a logistic regression analysis. Additionally, linear regressions were performed for the total group and the subgroups to assess the relationship between perinatal variables and IQ, YSR, DUDIT and PQ-16. Results The perinatal variables were not able to predict group membership (X2 [4] = 4.77, p = .312, Cox & Snell RÂČ = 0.053). Odds ratios indicated a small increase in probability to belonging to the general psychiatric sample instead of the SUD sample if birth was completed via C-section. After Bonferroni-correction, the linear regression models showed no relation between perinatal markers and IQ (p = .60, RÂČ = 0.068), YSR (p = .09, RÂČ = 0.121), DUDIT (p = .65, RÂČ = 0.020), and PQ-16 (p = .73, RÂČ =0.021)

    Re-expression of CD14 in response to a combined IL-10/TLR stimulus defines monocyte-derived cells with an immunoregulatory phenotype

    Interleukin 10 is a central regulator of the antigen-presenting function of myeloid cells. It exerts immunomodulatory effects in vivo and induces a regulatory phenotype in monocyte-derived cells in vitro. We analyzed phenotype and function of monocytic cells in vitro in relation to the cytokine milieu and the timing of TLR-based activation. In GM-CSF/IL-4 cultured human monocytic cells, we identified two, mutually exclusive cell populations arising from undifferentiated cells: CD83+^+ fully activated dendritic cells and CD14+^+ macrophage like cells. Re-expression of CD14 occurs primarily after a sequential trigger with a TLR signal following IL-10 preincubation. This cell population with re-expressed CD14 greatly differs in phenotype and function from the CD83+ cells. Detailed analysis of individual subpopulations reveals that exogenous IL-10 is critical for inducing the shift toward the CD14+^+ population, but does not affect individual changes in marker expression or cell function in most cases. Thus, plasticity of CD14 expression, defining a subset of immunoregulatory cells, is highly relevant for the composition of cellular products (such as DC vaccines) as it affects the function of the total product

    Crowdsourcing Scholarly Discourse Annotations

    The number of scholarly publications grows steadily every year and it becomes harder to find, assess and compare scholarly knowledge effectively. Scholarly knowledge graphs have the potential to address these challenges. However, creating such graphs remains a complex task. We propose a method to crowdsource structured scholarly knowledge from paper authors with a web-based user interface supported by artificial intelligence. The interface enables authors to select key sentences for annotation. It integrates multiple machine learning algorithms to assist authors during the annotation, including class recommendation and key sentence highlighting. We envision that the interface is integrated in paper submission processes for which we define three main task requirements: The task has to be . We evaluated the interface with a user study in which participants were assigned the task to annotate one of their own articles. With the resulting data, we determined whether the participants were successfully able to perform the task. Furthermore, we evaluated the interface's usability and the participant's attitude towards the interface with a survey. The results suggest that sentence annotation is a feasible task for researchers and that they do not object to annotate their articles during the submission process

    Susceptibility to interference between Pavlovian and instrumental control predisposes risky alcohol use developmental trajectory from ages 18 to 24

    Pavlovian cues can influence ongoing instrumental behaviour via Pavlovian-to-instrumental transfer (PIT) processes. While appetitive Pavlovian cues tend to promote instrumental approach, they are detrimental when avoidance behaviour is required, and vice versa for aversive cues. We recently reported that susceptibility to interference between Pavlovian and instrumental control assessed via a PIT task was associated with risky alcohol use at age 18. We now investigated whether such susceptibility also predicts drinking trajectories until age 24, based on AUDIT (Alcohol Use Disorders Identification Test) consumption and binge drinking (gramme alcohol/drinking occasion) scores. The interference PIT effect, assessed at ages 18 and 21 during fMRI, was characterized by increased error rates (ER) and enhanced neural responses in the ventral striatum (VS), the lateral and dorsomedial prefrontal cortices (dmPFC) during conflict, that is, when an instrumental approach was required in the presence of an aversive Pavlovian cue or vice versa. We found that a stronger VS response during conflict at age 18 was associated with a higher starting point of both drinking trajectories but predicted a decrease in binge drinking. At age 21, high ER and enhanced neural responses in the dmPFC were associated with increasing AUDIT-C scores over the next 3 years until age 24. Overall, susceptibility to interference between Pavlovian and instrumental control might be viewed as a predisposing mechanism towards hazardous alcohol use during young adulthood, and the identified high-risk group may profit from targeted interventions

    FĂŒr den Forschungsstandort Kiel wird an der Christian Albrechts-UniversitĂ€t zu Kiel und am GEOMAR Helmholtz-Zentrum fĂŒr Ozeanforschung Kiel ein einheitliches Modell zum Umgang mit Forschungsdaten entwickelt. Das Ziel ist, dass alle Disziplinen am Forschungsstandort mit ihren jeweils unterschiedlichen Anforderungen einen Baukasten nutzen können, der den Instituten, Arbeitsgruppen und Projekten Bausteine und Elemente zur Erstellung einheitlicher Regelungen zum Umgang mit Forschungsdaten zur VerfĂŒgung stellt. Die Basis ist der seit 2010 am GEOMAR zusammen mit dem Exzellenzcluster Future Ocean entwickelte und bereitgestellte Data-Policy-Baukasten fĂŒr die Kiel Marine Science. Um die Erstellung von spezifischen Standards zum Umgang mit Forschungsdaten durch die Forschenden selbst zu ermöglichen, soll der Baukasten aus den Disziplinen heraus dynamisch um notwendige, fachspezifische Komponenten erweitert werden können. Eine Versionierung der Baukastenelemente und der Regelwerke soll dabei fĂŒr eine nachhaltige Nutzungbarkeit und Nachvollziehbarkeit der erhobenen Forschungsdaten sorgen und gleichzeitig der Dynamik der sich rasch entwickelnden Forschungsbereiche genĂŒge tun. Perspektivisch soll so ein Rahmenwerk fĂŒr Datenmanagement als – ggf. auch automatisierter – Service entstehen. Ein zentrales Gremium von Forschungsdatenmanagern aus den Disziplinen soll die Pflege des Baukastens und der daraus entwickelten Standards im Hinblick auf ihre QualitĂ€t begleiten

    Carriage of linezolid-resistant enterococci (LRE) among humans and animals in Nigeria: coexistence of the cfr, optrA, and poxtA genes in Enterococcus faecium of animal origin

    Objectives: In contrast to increasing reports of the emergence of linezolid-resistant enterococci (LRE) emanating from many countries in Europe, Asia, and North America, data on its status and dissemination from the African continent remain scarce, with the information available limited to countries in North Africa. This study investigated the carriage of LRE and the genetic mechanism of resistance among Enterococcus faecium and Enterococcus faecalis strains recovered from humans and animals in Makurdi, Nigeria. Methods: We conducted a cross-sectional study between June 2020 and July 2021 during which 630 nonduplicate human and animal faecal samples were collected and processed for the recovery of LRE. The genetic mechanisms for resistance were investigated using polymerase chain reaction (PCR) and Sanger sequencing. Results: Linezolid-resistant enterococci were recovered from 5.87% (37/630; 95% CI: 4.17–8.00) of the samples, with the prevalence in animals and humans being 6.22% [(28/450); 95% CI: 4.17–8.87] and 5.00% [(9/180); 95% CI: 2.31–9.28], respectively. All isolates remained susceptible to vancomycin. No known point mutation mediating linezolid resistance was detected in the 23S rRNA and ribosomal protein genes; however, acquisition of one or more potentially transferable genes (cfr, optrA, and poxtA) was observed in 26 of the 37 LRE isolates. Co-existence of all three transferable genes in a single isolate was found in four E. faecium strains of animal origin. Conclusion: This study provides baseline evidence for the emergence and active circulation of LRE driven majorly by the acquisition of the optrA gene in Nigeria. To the best of our knowledge, our study is the first to report a co-carriage of all three transferable linezolid resistance determinants in E. faecium. Active LRE surveillance is urgently required to understand the extent of LRE spread across sub-Saharan Africa and to develop tailored mitigation strategies

    Diagnostic Accuracy of the Drug Use Disorder Identification Test and Its Short Form, the DUDIT-C, in German Adolescent Psychiatric Patients

    Background A common screening instrument for substance use disorders (SUDs) is the Drug Use Disorders Identification Test (DUDIT) which includes a short form regarding only drug consumption (DUDIT-C). We aim to assess if a German version of the DUDIT, adapted for adolescents, is a suitable screening instrument in a sample of adolescent psychiatric patients. Methods N = 124 (54 female) German adolescent (M = 15.6 + 1.5 years) psychiatric patients completed the DUDIT and received a diagnostic interview (MINI-KID) assessing DSM-5 SUD criteria. A confirmatory factor analysis (CFA), receiver operating characteristic (ROC) curves, the area under the curve (AUC), and Youden’s Index were calculated. Results A two-factor model of the DUDIT shows the best model fit (CFI = 0.995, SRMR = 0.055, RMSEA = 0.059, WRMR = 0.603). The DUDIT as well as the DUDIT-C show high diagnostic accuracy, with AUC = 0.95 and AUC = 0.88, respectively. For the DUDIT a cut-off value of 8.5 was optimal (sensitivity = 0.93, specificity = 0.91, J = 0.84), while for the DUDIT-C the optimal cut-off value was at 1.5 (sensitivity = 0.86, specificity = 0.84, J = 0.70). Conclusion This is the first psychometric evaluation of the DUDIT in German, adolescent psychiatric outpatients, using the DSM-5 diagnostic criteria. The DUDIT as well as the DUDIT-C are well suited for use in this population. Since in our sample only few patients presented with a mild or moderate SUD, our results need to be replicated in a sample of adolescents with mild SUD

    The Association of Non–Drug-Related Pavlovian-to-Instrumental Transfer Effect in Nucleus Accumbens With Relapse in Alcohol Dependence: A Replication

    BACKGROUND: The Pavlovian-to-instrumental transfer (PIT) paradigm measures the effects of Pavlovian conditioned cues on instrumental behavior in the laboratory. A previous study conducted by our research group observed activity in the left nucleus accumbens (NAcc) elicited by a non–drug-related PIT task across patients with alcohol dependence (AD) and healthy control subjects, and the left NAcc PIT effect differentiated patients who subsequently relapsed from those who remained abstinent. In this study, we aimed to examine whether such effects were present in a larger sample collected at a later date. METHODS: A total of 129 recently detoxified patients with AD (21 females) and 74 healthy, age- and gender-matched control subjects (12 females) performing a PIT task during functional magnetic resonance imaging were examined. After task assessments, patients were followed for 6 months. Forty-seven patients relapsed and 37 remained abstinent. RESULTS: We found a significant behavioral non–drug-related PIT effect and PIT-related activity in the NAcc across all participants. Moreover, subsequent relapsers showed stronger behavioral and left NAcc PIT effects than abstainers. These findings are consistent with our previous findings. CONCLUSIONS: Behavioral non–drug-related PIT and neural PIT correlates are associated with prospective relapse risk in AD. This study replicated previous findings and provides evidence for the clinical relevance of PIT mechanisms to treatment outcome in AD. The observed difference between prospective relapsers and abstainers in the NAcc PIT effect in our study is small overall. Future studies are needed to further elucidate the mechanisms and the possible modulators of neural PIT in relapse in AD
