    Structural optimization and biological evaluation of 2-substituted 5-hydroxyindole-3-carboxylates as potent inhibitors of human 5-lipoxygenase.

    Pharmacological suppression of leukotriene biosynthesis by inhibitors of 5-lipoxygenase (5-LO) is a strategy to intervene with inflammatory and allergic disorders. We recently presented 2-amino-5-hydroxy-1H-indoles as efficient 5-LO inhibitors in cell-based and cell-free assays. Structural optimization led to novel benzo[g]indole-3-carboxylates exemplified by ethyl 2-(3-chlorobenzyl)-5- hydroxy-1H-benzo[g]indole-3-carboxylate (compound 11a), which inhibits 5-LO activity in human neutrophils and recombinant human 5-LO with IC50 values of 0.23 and 0.086 ÎŒM, respectively. Notably, 11a efficiently blocks 5-LO product formation in human whole blood assays (IC50 = 0.83-1.6 ÎŒM) and significantly prevented leukotriene B4 production in pleural exudates of carrageenan-treated rats, associated with reduced severity of pleurisy. Together, on the basis of their high potency against 5-LO and the marked efficacy in biological systems, these novel and straightforward benzo[g]indole-3-carboxylates may have potential as anti-inflammatory therapeutics

    Reachability in Parametric Interval Markov Chains using Constraints

    Parametric Interval Markov Chains (pIMCs) are a specification formalism that extend Markov Chains (MCs) and Interval Markov Chains (IMCs) by taking into account imprecision in the transition probability values: transitions in pIMCs are labeled with parametric intervals of probabilities. In this work, we study the difference between pIMCs and other Markov Chain abstractions models and investigate the two usual semantics for IMCs: once-and-for-all and at-every-step. In particular, we prove that both semantics agree on the maximal/minimal reachability probabilities of a given IMC. We then investigate solutions to several parameter synthesis problems in the context of pIMCs -- consistency, qualitative reachability and quantitative reachability -- that rely on constraint encodings. Finally, we propose a prototype implementation of our constraint encodings with promising results

    Selective inhibition of pro-inflammatory prostaglandin (PG)E2 formation via microsomal PGE2 synthase-1 (mPGES-1) might be superior over inhibition of all cyclooxygenase (COX)-derived products by non-steroidal anti-inflammatory drugs (NSAIDs) and coxibs. We recently showed that benzo[g]indol-3-carboxylates potently suppress leukotriene biosynthesis by inhibiting 5-lipoxygenase. Here, we describe the discovery of benzo[g]indol-3-carboxylates as a novel class of potent mPGES-1 inhibitors (IC50 ≄ 0.1 ÎŒM). Ethyl 2-(3-chlorobenzyl)-5-hydroxy-1H-benzo[g]indole-3-carboxylate (compound 7a) inhibits human mPGES-1 in a cell-free assay (IC50 = 0.6 ÎŒM) as well as in intact A549 cells (IC50 = 2 ÎŒM), and suppressed PGE2 pleural levels in rat carrageenan-induced pleurisy. Inhibition of cellular COX-1/2 activity was significantly less pronounced. Compound 7a significantly reduced inflammatory reactions in the carrageenan-induced mouse paw edema and rat pleurisy. Together, based on the select and potent inhibition of mPGES-1 and 5-lipoxygenase, benzo[g]indol-3-carboxylates possess potential as novel anti-inflammatory drugs with a valuable pharmacological profile

    Performance of organically grown tomato cultivars under greenhouse conditions

Dentre as hortaliças cultivadas em sistema orgĂąnico, o tomate constitui grande desafio para os produtores devido Ă  inexistĂȘncia de recomendaçÔes de manejo cultural e de cultivares desenvolvidas especĂ­ficas para esse sistema de cultivo. Diante disso, o presente trabalho teve como objetivo avaliar o desempenho de cultivares de tomate sob sistema orgĂąnico, em ambiente protegido. O experimento foi conduzido de maio a outubro de 2004 em campo, no municĂ­pio de Araraquara-SP. O delineamento experimental adotado foi blocos ao acaso, com quatro repetiçÔes e oito tratamentos (cultivares Avalon, Colibri, HTX-5415, HTX-8027, Sahel, San Marzano, San Vito e Jane). As plantas foram conduzidas em fileiras duplas, com duas hastes por planta no espaçamento de 0,8 m entre linhas e 0,6 m entre plantas (cerca de 20.000 plantas ha-1), sem poda apical. O hĂ­brido Sahel destacou-se com o melhor desempenho para rendimento comercial. A traça-do-tomateiro se revelou como fator limitante Ă  produção de tomate em sistema orgĂąnico, sendo responsĂĄvel, em mĂ©dia, por 17% de danos nos frutos colhidos das oito cultivares que foram avaliadas.Organic agriculture in Brazil has been increasing about 30% per year over the last few years, since consumers are seeking for healthier foods, i.e. nutritious and free of pesticide residues. Among organically grown vegetable crops, tomato is an attractive economic opportunity for growers. However, the lack of information about management practices and adapted cultivars to organic production systems under protected cultivation are pointed out as important constraints that prevents this activity to expand. This work aimed at evaluating the performance of indeterminate tomato cultivars in organic management systems in unheated plastic greenhouse. In both experiments tomato plants were staked. Plant spacing was 1.5 m between rows and 0.35 m between plants. They were planted in double line and spaced 0.8 m between lines and 0.6 m between plants (about 20,000 plants ha-1). The experiment was set up in a randomized complete block design, with four replications and eight treatments (cultivars: Avalon, Colibri, HTX-5415, HTX-8027, Sahel, San Marzano, San Vito, and Jane). Sahel hybrid exhibited an outstanding performance for marketable yield under organic cultivation. Tomato pinworm (Tuta absoluta) was a serious limiting pest for organically grown tomatoes, responding for 17% of damage in the fruits harvested from the evaluated cultivars

    Observation of the thermal Casimir force

    Quantum theory predicts the existence of the Casimir force between macroscopic bodies, due to the zero-point energy of electromagnetic field modes around them. This quantum fluctuation-induced force has been experimentally observed for metallic and semiconducting bodies, although the measurements to date have been unable to clearly settle the question of the correct low-frequency form of the dielectric constant dispersion (the Drude model or the plasma model) to be used for calculating the Casimir forces. At finite temperature a thermal Casimir force, due to thermal, rather than quantum, fluctuations of the electromagnetic field, has been theoretically predicted long ago. Here we report the experimental observation of the thermal Casimir force between two gold plates. We measured the attractive force between a flat and a spherical plate for separations between 0.7 ÎŒ\mum and 7 ÎŒ\mum. An electrostatic force caused by potential patches on the plates' surfaces is included in the analysis. The experimental results are in excellent agreement (reduced χ2\chi^2 of 1.04) with the Casimir force calculated using the Drude model, including the T=300 K thermal force, which dominates over the quantum fluctuation-induced force at separations greater than 3 ÎŒ\mum. The plasma model result is excluded in the measured separation range.Comment: 6 page

    Alterations in vascular function in primary aldosteronism - a cardiovascular magnetic resonance imaging study

    Introduction: Excess aldosterone is associated with increased cardiovascular risk. Aldosterone has a permissive effect on vascular fibrosis. Cardiovascular magnetic resonance imaging (CMR) allows study of vascular function by measuring aortic distensibility. We compared aortic distensibility in primary aldosteronism (PA), essential hypertension (EH) and normal controls and explored the relationship between aortic distensibility and pulse wave velocity (PWV).<p></p> Methods: We studied PA (n=14) and EH (n=33) subjects and age-matched healthy controls (n=17) with CMR, including measurement of aortic distensibility, and measured PWV using applanation tonometry. At recruitment, PA and EH patients had similar blood pressure and left ventricular mass.<p></p> Results: Subjects with PA had significantly lower aortic distensibilty and higher PWV compared to EH and healthy controls. These changes were independent of other factors associated with reduced aortic distensibility, including aging. There was a significant relationship between increasing aortic stiffness and age in keeping with physical and vascular aging. As expected, aortic distensibility and PWV were closely correlated.<p></p> Conclusion: These results demonstrate that PA patients display increased arterial stiffness compared to EH, independent of vascular aging. The implication is that aldosterone invokes functional impairment of arterial function. The long-term implications of arterial stiffening in aldosterone excess require further study.<p></p&gt

    Short- and long-lasting tinnitus relief induced by transcranial direct current stimulation

    A significant proportion of the population suffers from tinnitus, a bothersome auditory phantom perception that can severely alter the quality of life. Numerous experimental studies suggests that a maladaptive plasticity of the auditory and limbic cortical areas may underlie tinnitus. Accordingly, repetitive transcranial magnetic stimulation (rTMS) has been repeatedly used with success to reduce tinnitus intensity. The potential of transcranial direct current stimulation (tDCS), another promising method of noninvasive brain stimulation, to relieve tinnitus has not been explored systematically. In a double-blind, placebo-controlled and balanced order design, 20 patients suffering from chronic untreatable tinnitus were submitted to 20 minutes of 1 mA anodal, cathodal and sham tDCS targeting the left temporoparietal area. The primary outcome measure was a change in tinnitus intensity or discomfort assessed with a Visual Analogic Scale (VAS) change-scale immediately after tDCS and 1 hour later. Compared to sham tDCS, anodal tDCS significantly reduced tinnitus intensity immediately after stimulation; whereas cathodal tDCS failed to do so. The variances of the tinnitus intensity and discomfort VAS change-scales increased dramatically after anodal and cathodal tDCS, whereas they remained virtually unchanged after sham tDCS. Moreover, several patients unexpectedly reported longer-lasting effects (at least several days) such as tinnitus improvement, worsening, or changes in tinnitus features, more frequently after real than sham tDCS. Anodal tDCS is a promising therapeutic tool for modulating tinnitus perception. Moreover, both anodal and cathodal tDCS seem able to alter tinnitus perception and could, thus, be used to trigger plastic changes

    Molecular and clinical characterization of albinism in a large cohort of Italian patients.

    PURPOSE: The purpose of this study was to identify the molecular basis of albinism in a large cohort of Italian patients showing typical ocular landmarks of the disease and to provide a full characterization of the clinical ophthalmic manifestations. METHODS: DNA samples from 45 patients with ocular manifestations of albinism were analyzed by direct sequencing analysis of five genes responsible for albinism: TYR, P, TYRP1, SLC45A2 (MATP), and OA1. All patients studied showed a variable degree of skin and hair hypopigmentation. Eighteen patients with distinct mutations in each gene associated with OCA were evaluated by detailed ophthalmic analysis, optical coherence tomography (OCT), and fundus autofluorescence. RESULTS: Disease-causing mutations were identified in more than 95% of analyzed patients with OCA (28/45 [62.2%] cases with two or more mutations; 15/45 [33.3%] cases with one mutation). Thirty-five different mutant alleles were identified of which 15 were novel. Mutations in TYR were the most frequent (73.3%), whereas mutations in P occurred more rarely (13.3%) than previously reported. Novel mutations were also identified in rare loci such as TYRP1 and MATP. Mutations in the OA1 gene were not detected. Clinical assessment revealed that patients with iris and macular pigmentation had significantly higher visual acuity than did severe hypopigmented phenotypes. CONCLUSIONS: TYR gene mutations represent a relevant cause of oculocutaneous albinism in Italy, whereas mutations in P present a lower frequency than that found in other populations.     ERP evidence suggests executive dysfunction in ecstasy polydrug users

    Background: Deficits in executive functions such as access to semantic/long-term memory have been shown in ecstasy users in previous research. Equally, there have been many reports of equivocal findings in this area. The current study sought to further investigate behavioural and electro-physiological measures of this executive function in ecstasy users. Method: Twenty ecstasy–polydrug users, 20 non-ecstasy–polydrug users and 20 drug-naïve controls were recruited. Participants completed background questionnaires about their drug use, sleep quality, fluid intelligence and mood state. Each individual also completed a semantic retrieval task whilst 64 channel Electroencephalography (EEG) measures were recorded. Results: Analysis of Variance (ANOVA) revealed no between-group differences in behavioural performance on the task. Mixed ANOVA on event-related potential (ERP) components P2, N2 and P3 revealed significant between-group differences in the N2 component. Subsequent exploratory univariate ANOVAs on the N2 component revealed marginally significant between-group differences, generally showing greater negativity at occipito-parietal electrodes in ecstasy users compared to drug-naïve controls. Despite absence of behavioural differences, differences in N2 magnitude are evidence of abnormal executive functioning in ecstasy–polydrug users

    A case of polymicrogyria in macaque monkey: impact on anatomy and function of the motor system

    Background: Polymicrogyria is a malformation of the cerebral cortex often resulting in epilepsy or mental retardation. It remains unclear whether this pathology affects the structure and function of the corticospinal (CS) system. The anatomy and histology of the brain of one macaque monkey exhibiting a spontaneous polymicrogyria (PMG monkey) were examined and compared to the brain of normal monkeys. The CS tract was labelled by injecting a neuronal tracer (BDA) unilaterally in a region where low intensity electrical microstimulation elicited contralateral hand movements (presumably the primary motor cortex in the PMG monkey). Results: The examination of the brain showed a large number of microgyri at macro- and microscopic levels, covering mainly the frontoparietal regions. The layered cortical organization was locally disrupted and the number of SMI-32 stained pyramidal neurons in the cortical layer III of the presumed motor cortex was reduced. We compared the distribution of labelled CS axons in the PMG monkey at spinal cervical level C5. The cumulated length of CS axon arbors in the spinal grey matter was not significantly different in the PMG monkey. In the red nucleus, numerous neurons presented large vesicles. We also assessed its motor performances by comparing its capacity to execute a complex reach and grasp behavioral task. The PMG monkey exhibited an increase of reaction time without any modification of other motor parameters, an observation in line with a normal CS tract organisation. Conclusion: In spite of substantial cortical malformations in the frontal and parietal lobes, the PMG monkey exhibits surprisingly normal structure and function of the corticospinal system
