    Propagation of Rayleighs waves in cracked media

    Este trabajo está enfocado a la obtención de resultados numéricos que permitan la detección y caracterización de grietas sub-superficiales en sólidos mediante la incidencia de ondas elásticas de Rayleigh. Los resultados se obtienen a partir de ecuaciones integrales de frontera, que pertenecen al campo de la elastodinámica. Una vez que se aplican las condiciones de frontera se obtiene un sistema de ecuaciones integrales del tipo Fredholm de segunda especie y orden cero, el cual es resuelto mediante eliminación gaussiana. El método que se emplea para la discretización de dichas ecuaciones es conocido como «método indirecto de elementos frontera», el cual puede ser visto como una derivación del teorema clásico de Somigliana. A partir de los análisis realizados en el dominio de la frecuencia emergen picos de resonancia que permiten inferir la presencia de grietas mediante los cocientes espectrales. Se analizaron varios modelos de medios agrietados donde se pretende evidenciar la gran utilidad que presenta el uso de los cocientes espectrales para identificar grietas. Se estudiaron los efectos de la orientación y la localización de las grietas. Los resultados obtenidos presentan buena concordancia con los publicados previamente.This work is focused on the finding of numerical results for detection and characterization of sub-surface cracks in solids under the incidence of Rayleighs elastic waves. The results are obtained from boundary integral equations, which belong to the field of dynamics of elasticity. Once applied the boundary conditions, a system of Fredholms integral equations of second kind and zero order is obtained, which is solved using Gaussian elimination. The method that is used for the solution of such integral equations is known as the Indirect Boundary Element Method, which can be seen as a derivation of the Somiglianas classic theorem. On the basis of the analysis made in the frequency domain, resonance peaks emerge and allow us to infer the presence of cracks through the spectral ratios. Several models of cracked media were analyzed, where analyses reveal the great utility that displays the use of spectral ratios to identify cracks.     El libro “Los espacios universitarios como objeto de estudio. Análisis geográfico, ambiental y ecológico del Cerro de Coatepec, Universidad Autónoma del Estado de México”, es resultado de la participación de un grupo multidisciplinario integrado por investigadores, profesores y estudiantes de la Universidad Autónoma del Estado de México, así como del grupo de asesores y consultores del Colegio de Ciencias Geográficas del Estado de México, A. C. (COCIGEM), interesados en la problemática ambiental y ecológica que ocurre en la Ciudad Universitaria ubicada geográficamente en el Cerro de Coatepec, Toluca Estado de México, en el cual se encuentran establecidas algunas edificaciones de dependencias de educación superior, centros e institutos de investigación, difusión cultural, dependencias administrativas, bibliotecas, museos e instalaciones deportivas.Los ejes rectores de análisis contenidos en este volumen son siete: a) la caracterización geográfica, ecológica, ambiental, sociocultural e infraestructural del espacio que comprende el Cerro de Coatepec; b) el análisis del Cerro de Coatepec como un ecosistema urbano en el contexto de la Zona Metropolitana del Valle de Toluca, Estado de México; c) la multifuncionalidad de las áreas verdes, las áreas arboladas y los espacios naturales del ecosistema; d) los impactos ambientales que han ocasionado y que ocasionan las etapas de preparación del sitio, la construcción de edificaciones y la ejecución de actividades docentes, de investigación, difusión, deportivas y socioculturales; e) los riesgos que ocurren y pueden ocurrir en las áreas verdes, las áreas arboladas, los ambientes naturales y la infraestructura de las edificaciones; f) la responsabilidad ambiental e importancia de la participación de los actores universitaritos en el cuidado del entorno universitario; y g) propuesta de acciones generales para el mejoramiento de las condiciones del paisaje y manejo del entorno universitario.     Transcriptomic changes linked to age-dependent neuromelanin accumulation in a new Parkinson's disease mouse model

    Resumen del trabajo presentado en el 50th Annual Meeting Society for Neuroscience, celebrado de forma virtual del 8 al 11 de noviembre de 2021In Parkinson's disease (PD) there is a preferential degeneration of neuromelanin (NM)-containing neurons, especially neurons from the Substantia Nigra (SN) but also from the Ventral Tegmental Area (VTA) and Locus Coeruleus (LC). We generated a new NM-producing mouse model, based on the tissue-specific constitutive expression of human tyrosinase (hTyr) under the tyrosine hydroxylase (TH) promoter (tgNM), that mimics the distribution and age-dependent accumulation of NM in the human brain (i.e. catecholaminergic groups A1-A14). TgNM mice exhibited major PD features, including both motor and non-motor behavioral alterations, inclusion body formation, neuronal degeneration in lower brainstem areas (LC) together with neuronal dysfunction in higher brainstem areas (SN and VTA). In order to understand the mechanisms by which NM accumulation in specific brain areas ultimately interferes with the normal functioning of cells, we characterized genome-wide transcriptomic changes linked to the intracellular presence and progressive accumulation of NM in two NM-accumulating neuronal subpopulations (SN and VTA) that are known to be differentially susceptible to PD pathology. We selectively isolated single dopaminergic NM-containing neurons by laser capture microdissection from male and female wild-type and tgNM animals at 3 months, 12 months and 20 months of age (n=4-6 mice per group). We performed differential expression analysis, resulting in statistically significant differentially expressed genes at all ages (p-value<0.5). Gene-set enrichment analysis (GSEA) with Reactome Pathway Database led to the identification of altered biological pathways in tgNM related to neuroinflammation, vesicle-mediated transport and lipid metabolism, transcription and translation, mitochondrial function and cell cycle (senescence) (False Discovery Rate<0.05). Targeted-based validation of candidate RNA species was performed in microdissected samples by quantitative real-time PCR and candidate biological pathways were validated at the protein level by western blot in dissected ventral midbrain tissues from biological replicates. The transcriptomic profiles identified in this project contribute to our understanding of selective vulnerability in PD and brain aging, and points to key biological pathways and molecular targets in prodromal and early PD

    Detection of ‘long-haired’ Saprolegnia (S. parasitica) isolates using monoclonal antibodies

    P. 726-733The ability of five monoclonal antibodies (Mabs) raised against a pathogenic Saprolegnia parasitica isolate from brown trout to detect and differentiate between isolates with bundles of long hairs (S. parasitica) and other Saprolegnia species was determined by means of an indirect immunofluorescence assay. Four of the Mabs used recognized some of the long-haired S. parasitica isolates but also cross-reacted with other Saprolegnia species without bundles of hairs and with Achlya sp. The other Mab (named 18A6) was able to differentiate between the asexual and most of the sexual isolates in the group of long-haired S. parasitica isolates, but did not recognize Achlya sp. or the Saprolegnia species without bundles of hairs, with the exception of S. hypogyna. These results indicate that isolates with bundles of long hairs are closely related with other members of genus Saprolegnia and share several antigens. However, Mab 18A6 seems to recognize an epitope that is expressed mainly in the asexual isolates in the long-haired S. parasitica isolates.S

    Dendritic cell deficiencies persist seven months after SARS-CoV-2 infection

    Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV)-2 infection induces an exacerbated inflammation driven by innate immunity components. Dendritic cells (DCs) play a key role in the defense against viral infections, for instance plasmacytoid DCs (pDCs), have the capacity to produce vast amounts of interferon-alpha (IFN-α). In COVID-19 there is a deficit in DC numbers and IFN-α production, which has been associated with disease severity. In this work, we described that in addition to the DC deficiency, several DC activation and homing markers were altered in acute COVID-19 patients, which were associated with multiple inflammatory markers. Remarkably, previously hospitalized and nonhospitalized patients remained with decreased numbers of CD1c+ myeloid DCs and pDCs seven months after SARS-CoV-2 infection. Moreover, the expression of DC markers such as CD86 and CD4 were only restored in previously nonhospitalized patients, while no restoration of integrin β7 and indoleamine 2,3-dyoxigenase (IDO) levels were observed. These findings contribute to a better understanding of the immunological sequelae of COVID-19

    Hydrothermal alteration of andesitic lava domes can lead to explosive volcanic behaviour

    Dome-forming volcanoes are among the most hazardous volcanoes on Earth. Magmatic outgassing can be hindered if the permeability of a lava dome is reduced, promoting pore pressure augmentation and explosive behaviour. Laboratory data show that acid-sulphate alteration, common to volcanoes worldwide, can reduce the permeability on the sample lengthscale by up to four orders of magnitude and is the result of pore- and microfracture-filling mineral precipitation. Calculations using these data demonstrate that intense alteration can reduce the equivalent permeability of a dome by two orders of magnitude, which we show using numerical modelling to be sufficient to increase pore pressure. The fragmentation criterion shows that the predicted pore pressure increase is capable of fragmenting the majority of dome-forming materials, thus promoting explosive volcanism. It is crucial that hydrothermal alteration, which develops over months to years, is monitored at dome-forming volcanoes and is incorporated into real-time hazard assessments

    SARS-CoV-2 viral load in nasopharyngeal swabs is not an independent predictor of unfavorable outcome

    The aim was to assess the ability of nasopharyngeal SARS-CoV-2 viral load at first patient’s hospital evaluation to predict unfavorable outcomes. We conducted a prospective cohort study including 321 adult patients with confirmed COVID-19 through RT-PCR in nasopharyngeal swabs. Quantitative Synthetic SARS-CoV-2 RNA cycle threshold values were used to calculate the viral load in log10 copies/mL. Disease severity at the end of follow up was categorized into mild, moderate, and severe. Primary endpoint was a composite of intensive care unit (ICU) admission and/or death (n = 85, 26.4%). Univariable and multivariable logistic regression analyses were performed. Nasopharyngeal SARS-CoV-2 viral load over the second quartile (≥ 7.35 log10 copies/mL, p = 0.003) and second tertile (≥ 8.27 log10 copies/mL, p = 0.01) were associated to unfavorable outcome in the unadjusted logistic regression analysis. However, in the final multivariable analysis, viral load was not independently associated with an unfavorable outcome. Five predictors were independently associated with increased odds of ICU admission and/or death: age ≥ 70 years, SpO2, neutrophils > 7.5 × 103/µL, lactate dehydrogenase ≥ 300 U/L, and C-reactive protein ≥ 100 mg/L. In summary, nasopharyngeal SARS-CoV-2 viral load on admission is generally high in patients with COVID-19, regardless of illness severity, but it cannot be used as an independent predictor of unfavorable clinical outcome
