The curricular organization in the competency approach is currently a concern of the educational
community in Cape Verde. The perception that teachers and managers have of pedagogical and
epistemological practices based on this new approach, has directed its own effectiveness. The
implementation of teaching by competences in the teaching-learning process, as well as in the school
organizations leadership, has led to the disclosure of feelings and speculative positions, without
previous studies that redirect the curricular policies of professional technical training in an efficient and
effective manner. In this sense, the present communication reflects an underway study at the
Universidade Aberta, Portugal, which is part of the PhD in Education, focused on the way in which
educational agents understand the curricular management by competences in several secondary
technical schools in Cabo Verde. With this research, we intend to answer the following questions: 1)
To what extent and by what processes has the concept of competence changed the organization, the
teaching practices and leadership in the context of technical and vocational education in Cape Verde?;
2) What are the roles of leadership in promoting these changes?; 3) How has the implementation of
these changes been processed in teaching practice? In order to do so, we suggest a qualitative and
quantitative approach, collecting data through documentary analysis, semi-structured interviews and
questionnaire surveys.info:eu-repo/semantics/publishedVersio
School age Portuguese children who stutter : socio-cognitive performance and the impact of stuttering in their quality of life
outros intervenientes, quer do seio familiar, quer do seio escolar.
No primeiro,
avaliado através do questionário parental: Temperament in Middle Childhood
A avaliação do
for Children (MASC).

Stuttering is a multifaceted disorder resulting from the interaction between genetic,
neurophysiological, environmental, and other factors. The ways these factors interact
with each other is not yet fully understood, and is still subject of research. In recent
years, several authors have shown the impact of the disorder to reach far beyond the
surface components, with strong evidence about the psychosocial effects of stuttering
not only for the person who stutters but also for their family.
This research work seeks to clarify the interaction between factors that contribute to the
development of stuttering. In particular, it attempts to improve the assessment and
therapeutic programs for stuttering in Portugal. This work presents five chapters: the
first consists of a literature review based on the multidimensionality inherent to
stuttering. The following chapters consist of four studies that are detailed below.
The first study aimed to relate stuttering to temperament, executive functioning, and
anxiety, by comparing stuttering children and their nonstuttering peers. Participants
were 100 school-age children (mean age = 9.13 mos.; SD = 1.70 mos.), which included
50 children who stutter and 50 children who did not. In this study, participants were
divided into two subgroups, taking age into account: a younger group (7-9 years) and an
older group (10-12 years). Their temperament was evaluated through the parental
questionnaire, Temperament in Middle Childhood Questionnaire (TMCQ), translated
and adapted for this purpose. Executive functioning was assessed through the
Portuguese version of the neuropsychological instrument, Children Color Trails Test
(CCTT). Anxiety was assessed through the Portuguese version of the Multidimensional
Anxiety Scale for Children (MASC) self-assessment questionnaire.
The second study aimed to measure the impact of stuttering on the Portuguese
children’s lives. Participants were 50 children who stutter (Mean age = 9.10 mos.; SD =
1.7 mos.). For this study, the Overall Assessment of the Speaker's Experience of
Stuttering - School age (OASES-S) (OASES-S) was translated, adapted, and validated
for European Portuguese (OASES-S-PT). Good reliability and validity of the OASESS-
PT were verified for the assessment of impact in Portuguese school-age children.
The third and fourth studies were conducted in order to understand the parents' and
teachers' perspectives regarding the impact of stuttering on the lives of their children
and students, and to compare the results with the children's self-perception of impact.
ix
In the study related to the perception of parents, a subdivision of the group was
performed, taking into account the presence or absence of a family history of stuttering.
The participants were 50 parents divided into: i) group of parents with a family history
of stuttering (n= 30), and ii) group of parents with no history family of stuttering (n=
20). The study that aimed to investigate the teacher’s perceptions involved 27 teachers.
In both studies adaptations, of the OASES-S-PT instrument were made so that they
could be answered by parents (OASES-S-P) and teachers (OASES-S-T).
The results indicated that children who stutter may experience lower ability to orient
attention and may have greater emotional reactivity compared with their nonstuttering
peers. Portuguese children who stutter experience a moderate impact of stuttering in
their lives. In general, the results of parents and teachers were in agreement with the
perceptions of the children; however, parents with a family history of stuttering reported
significantly higher negative impact. This finding supports the idea that refinements in
clinical and educational practices are needed; findings also highlight the importance of
teamwork with parents and teachers
The impact of endocrine disruptors on spermatogonia survival: the case of methoxychlor
Endocrine disruptors (EDCs) are xenobiotics that have the ability to interfere with hormone synthesis, secretion and metabolism, thus affecting the reproductive system. Today we are ubiquitously exposed to these compounds through various pathways, such as ingestion, inhalation and dermal absorption. Methoxychlor (MXC) is an organochloride pesticide with moderate persistence in the environment capable of adversely affecting spermatogenesis, deregulating the development of germ cells, and interfering with sperm function. Among germ cells, type B spermatogonia are the first differentiated germline cell population and the initiator of the spermatogenic process. However, little is known about the role of MXC in modulating the survival pathways of type B spermatogonia. In the present dissertation, the impact of MXC on survival/apoptosis, and on the antioxidant defences of spermatogonia, was studied. For this purpose, a type B spermatogonia cell line, the GC-1spg, was cultured in the presence (5, 10, 25, 50 and 100 µM) and without MXC for 48 hours. The effect of MXC on GC-1spg viability was analysed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium (MTT) bromide assays. Moreover, protein expression of key regulators of apoptosis, estrogen receptors alpha and beta (ERa and ERß, respectively) and androgen receptor (AR) was analysed by Western Blot and the activity of antioxidant enzymes and caspase-3 by colorimetric assays. The obtained results demonstrated that MXC decreases the viability of GC-1spg cells in a concentration-dependent manner, increasing the activity of the antioxidant enzymes superoxide dismutase and glutathione peroxidase, and caspase-3. Despite the increase in the Bax (pro-apoptotic protein)/Bcl-2 (anti-apoptotic protein) ratio, there was a decrease in caspase-8 and p53 expression in GC-1spg exposed to MXC. In addition, MXC treatment decreased the expression of both estrogen receptors (ERa and ERß) and AR in GC-1spg. The present study demonstrates the modulation of GC1-spg apoptosis by MXC, being the first to demonstrate the expression of ERß in GC-1spg and to show the impact of MXC on the expression of these sex steroid receptors. By investigating the mechanisms and pathways by which EDCs exert harmful effects on male reproductive health, the present study widens the horizons for the identification of targets for developing new strategies for preserving male fertility and eventual treatments.
Morphological and molecular adaptation of Aspergillus niger to simulated spaceflight and Mars-like conditions
The International Space Station (ISS) is an indoor-closed environment in Low Earth Orbit (LEO). Outside of
the ISS, radiation is the most challenging factor outside. In turn, inside the ISS, spaceflight microgravity is
the one factor that cannot be evaded. Aspergillus niger and Penicillium rubens are two common isolates
of the ISS microbiota. As filamentous fungi, they form highly resistant airborne spores that can easily
spread and colonize the spacecraft habitat. Fungi surface-associated growth (or biofilm formation), can
biodegrade surfaces and clog life-support systems, and their spores can potentially infect the humans on
board. In contrast, on Earth filamentous fungi play an important role in biotechnology, producing a widerange of compounds of interest, from food to antibiotics. Because of this, envisioned long-term spaceflight
missions going far beyond low Earth orbit, to the Moon or Mars, will require an intensification of the fungal
research, not only in relation to astronaut health and spacecraft safety, but also establishing opportunities
for fungal-based biotechnology in space. Thus, this thesis aims to answer three main questions: i) can A.
niger spores resist space radiation, and if yes, could they endure interplanetary space travel? ii) if brought
to the surface of Mars, could A. niger spores survive the martian environment? and iii) how does simulated
microgravity affect A. niger colony growth and biofilm formation? In total, four strains of A. niger were
analyzed in this thesis: the industrial and highly pigmented wild-type strain (N402), a strain defective in
pigmentation (ΔfwnA), a strain defective in DNA repair (ΔkusA), and a strain defective in polar growth
(ΔracA). To assess the level of resistance and survival limits of fungal spores in a long-term interplanetary
mission scenario, A. niger spores were exposed to high radiation doses of X-rays and cosmic radiation
(helium- and iron-ions) and of UV-C radiation. Results show that wild-type spores of A. niger were able to
withstand high doses of the all tested types of space radiation. This suggests that A. niger spores might
endure space travel, when considering the radiation factor alone. To evaluate the survival of A. niger to
Mars surface conditions, dried spores were launched in a stratospheric balloon mission called MARSBOx.
Throughout the mission, A. niger spores were exposed to desiccation, simulated martian atmosphere and
pressure, as well as to full UV-VIS radiation. Results revealed that the highly pigmented wild-type spores
would survive in a Mars-like middle stratosphere environment with radiation exposure, even as a spore
monolayer (106 spores/ml), i.e. with no self-shielding. Spore survival to space radiation and martian
conditions suggest that current planetary protection guidelines should be revisited integrating the high
resistance of fungal spores. Furthermore, A. niger colony growth and biofilm formation under simulated
microgravity was investigated. Scanning Electron Microscopy (SEM) pictures reveal never-before seen
ultrastructure of A. niger colonies and biofilms (i.e. vegetative mycelium embedded in extracellular
matrix). Results reveal changes in biofilm thickness, spore production and dry biomass, suggesting an
increased potential for A. niger to colonize spaceflight habitats. Lastly, P. rubens was proven as a model
organism for a spaceflight biofilm experiment aboard the International Space Station. Overall, this thesis
highlights the extraordinary resistance of fungal spores to extraterrestrial conditions and reveals their
ability to cope with spaceflight microgravity. This advocates for future research that will enable better
monitoring and controlling of fungal contaminations in space habitats, and that will help establish
filamentous fungi as valuable companions of human space exploration.2021-10-2
The world is aging and therefore it is essential to understand how the
development of this process in organisms is, especially in human
beings. Its knowing that this phenomenon does not occur in the same
way in all individuals, and that there are some with a chronological
age of 80 years and yet, they present a condition of a younger
subject. Thus, it is essential to join efforts to understand this event.
Currently, there are several models for the study of those process,
from animal models, to computational or cellular models, with the
latter having more advantages. Cell models, namely cell models of
fibroblasts, are thus increasingly used for metabolomics research in
the aging research since through these techniques we are able to
indirectly study the pathways of this process, since this gives
valuable insights into the composition of metabolites present in a
sample. An approach that combines several of these tools, such as
FTIR, MS or NMR, will be an asset and will allow for the study of
aging at a molecular level and possible reveal new discoveries.O mundo está a envelhecer e, torna-se fundamental perceber

High-glucose feeding of gilthead seabream (Sparus aurata) larvae: effects on molecular and metabolic pathways
Nutritional programming has begun to arouse interest as a novel tool to alter specific metabolic pathways or functions in farmed animals. The aim of the present study was to explore the potential of early glucose stimuli to induce changes in nutrient metabolism of gilthead seabream. Nutritional conditioning was performed by delivering glucose-rich feed at three distinct recurrent periods of larval feeding regime: during first-feeding with rotifers (3 days after hatching, DAH) and mid-feeding with Artemia metanauplii (20DAH) and the beginning of inert diet feeding (30DAH), called the Recurrent treatment (REC). As opposed, the control treatment (CTRL) did not experience any glucose stimuli. At post-larval stage (from 50 to 60DAH), both treatments were challenged with a high-carbohydrate diet (50%). The immediate response to the early stimuli was assessed through gene expression of metabolic markers and by nutrient metabolism using [C-14] tracers. Each dietary stimulus induced metabolic changes on REC larvae, shown by altered expression of some genes, including those involved in glycolysis, and by a different pattern of glucose utilization. However, none of the molecular adaptations (except G6PDH gene) were persistent in the viscera and muscle of challenged post-larvae from REC group. In contrast, the glucose metabolism of challenged REC post-larvae revealed a shift towards a higher catabolism and lower glucose retention in tissues, compared to the CTRL group, suggesting an improvement of glucose oxidation pathways. In addition, the REC group showed a higher bio-conversion of glucose into lipids, indicating enhanced hepatic lipogenesis. The early stimuli did not affect the relative retention or use of amino acids or the growth and survival of challenged fish, up to 60DAH. In summary, although not substantiated at a molecular level, our data reveal that a recurrent high-glucose stimulus during larval stages affects the short-term modulation of pathways for glucose utilization in gilthead seabream.
