    Contribution of lateral gene transfers to the genome composition and parasitic ability of root-knot nematodes.

    International audienceLateral gene transfers (LGT), species to species transmission of genes by means other than direct inheritance from a common ancestor, have played significant role in shaping prokaryotic genomes and are involved in gain or transfer of important biological processes. Whether LGT significantly contributed to the composition of an animal genome is currently unclear. In nematodes, multiple LGT are suspected to have favored emergence of plant-parasitism. With the availability of whole genome sequences it is now possible to assess whether LGT have significantly contributed to the composition of an animal genome and to establish a comprehensive list of these events. We generated clusters of homologous genes and automated phylogenetic inference, to detect LGT in the genomes of root-knot nematodes and found that up to 3.34% of the genes originate from LGT of non-metazoan origin. After their acquisition, the majority of genes underwent series of duplications. Compared to the rest of the genes in these species, several predicted functional categories showed a skewed distribution in the set of genes acquired via LGT. Interestingly, functions related to metabolism, degradation or modification of carbohydrates or proteins were substantially more frequent. This suggests that genes involved in these processes, related to a parasitic lifestyle, have been more frequently fixed in these parasites after their acquisition. Genes from soil bacteria, including plant-pathogens were the most frequent closest relatives, suggesting donors were preferentially bacteria from the rhizosphere. Several of these bacterial genes are plasmid-borne, pointing to a possible role of these mobile genetic elements in the transfer mechanism. Our analysis provides the first comprehensive description of the ensemble of genes of non-metazoan origin in an animal genome. Besides being involved in important processes regarding plant-parasitism, genes acquired via LGT now constitute a substantial proportion of protein-coding genes in these nematode genomes

    Genome Expression Dynamics Reveal the Parasitism Regulatory Landscape of the Root-Knot Nematode Meloidogyne incognita and a Promoter Motif Associated with Effector Genes.

    Root-knot nematodes (genus Meloidogyne) are the major contributor to crop losses caused by nematodes. These nematodes secrete effector proteins into the plant, derived from two sets of pharyngeal gland cells, to manipulate host physiology and immunity. Successful completion of the life cycle, involving successive molts from egg to adult, covers morphologically and functionally distinct stages and will require precise control of gene expression, including effector genes. The details of how root-knot nematodes regulate transcription remain sparse. Here, we report a life stage-specific transcriptome of Meloidogyne incognita. Combined with an available annotated genome, we explore the spatio-temporal regulation of gene expression. We reveal gene expression clusters and predicted functions that accompany the major developmental transitions. Focusing on effectors, we identify a putative cis-regulatory motif associated with expression in the dorsal glands, providing an insight into effector regulation. We combine the presence of this motif with several other criteria to predict a novel set of putative dorsal gland effectors. Finally, we show this motif, and thereby its utility, is broadly conserved across the Meloidogyne genus, and we name it Mel-DOG. Taken together, we provide the first genome-wide analysis of spatio-temporal gene expression in a root-knot nematode and identify a new set of candidate effector genes that will guide future functional analyses

    ECG periodic components as a promising tool for complexity assessment during stepwise ablation of atrial fibrillation

    Introduction: Stepwise radiofrequency catheter ablation (step-CA) has become a treatment of choice for the restoration of sinus rhythm (SR) in patients (pts) with long-standing persistent atrial fibrillation (pers-AF). Its success rate appears limited as the amount of ablation to achieve long term SR is unknown. Recently, intracardiac organization indices (OI) of AF have been used to track the efficiency of step-CA, with limited success. Our study is aimed at developing new OIs based on the relationships between harmonic components of atrial activity from the surface ECG as a global assessment of AF complexity and organization during step-CA. Methods: 3 pts with pers-AF (age 62, AF duration 17 months) underwent a step-CA. An adaptive tracking algorithm was developed for estimating the instantaneous frequency of atrial activity on chest lead V1 (after QRST subtraction) and for extracting its fundamental and harmonic components. An adaptive organization index (AOI) was computed as the ratio between the power of the extracted components and the total power of the signal to evaluate the temporal evolution of AF oscillations. The variance of the phase difference (PD) between the fundamental and harmonic components was used for measuring AF regularity. Results: Step-CA terminated 2/3 pers-AF into flutter. Importantly, in the 2 terminated pts, the AOI did not show any significant change during the step-CA (from pre-ablation to CFAE), while the PD showed a gradual reduction suggestive of increased coupling between the fundamental and the 1st harmonic. See figure. Conclusions: The PD and the AOI as measurements of complexity from the surface ECG appear as promising methods for tracking the effect of step-CA on global AF organization. This, however, needs to be validated on a larger population

Parte-se de uma visão individualista sobre competência, onde o indivíduo é exclusivamente responsável pelo desenvolvimento da mesma, para uma visão amparada na perspectiva histórico-cultural. Nesta última, a competência é desenvolvida e avaliada quando utilizada em contexto real de trabalho, na maneira como o indivíduo a enfrenta e como mobiliza a rede de atores sociais para resolver as situações. Dessa forma, apresenta-se possíveis consequências que a perspectiva histórico-cultural traz tanto para a abordagem do desempenho quanto para o desenvolvimento da qualificação profissional, principalmente no que se refere à transmissão escolar e extra-escolar das competências

    Using global existing fiber networks for environmental sensing

    We review recent advances in distributed fiber optic sensing (DFOS) and their applications. The scattering mechanisms in glass, which are exploited for reflectometry-based DFOS, are Rayleigh, Brillouin, and Raman scatterings. These are sensitive to either strain and/or temperature, allowing optical fiber cables to monitor their ambient environment in addition to their conventional role as a medium for telecommunications. Recently, DFOS leveraged technologies developed for telecommunications, such as coherent detection, digital signal processing, coding, and spatial/frequency diversity, to achieve improved performance in terms of measurand resolution, reach, spatial resolution, and bandwidth. We review the theory and architecture of commonly used DFOS methods. We provide recent experimental and field trial results where DFOS was used in wide-ranging applications, such as geohazard monitoring, seismic monitoring, traffic monitoring, and infrastructure health monitoring. Events of interest often have unique signatures either in the spatial, temporal, frequency, or wavenumber domains. Based on the temperature and strain raw data obtained from DFOS, downstream postprocessing allows the detection, classification, and localization of events. Combining DFOS with machine learning methods, it is possible to realize complete sensor systems that are compact, low cost, and can operate in harsh environments and difficult-to-access locations, facilitating increased public safety and smarter cities

    A New Method for ECG Tracking of Persistent Atrial Fibrillation Termination during Stepwise Ablation

Its success rate appears limited as the amount of ablation to achieve long term SR is unknown. Multiple organization indexes (OIs) have been previously developed to track the organization of AF during step-CA, however, with limited success. We report an adaptive method for tracking AF termination (AF-term) based on OIs characterizing the relationship between harmonic components of atrial activity from the surface ECG of AF activity. By computing their relative evolution during the last two steps preceding AF-term, we found that the performance of our OIs was superior to classical indices to track the efficiency of step-CA “en route” to AF-term. Our preliminary results suggest that the gradual synchronization between the fundamental and its first harmonic of AF activity appears as a promising parameter for predicting AF-term during step-CA

    Boulder deposition during major tsunami events

    A remarkable accumulation of marine boulders located above the present spring tide level has occurred in two coastal lowlands of the Algarve (Portugal). The size-interval of the particles studied here is seldom reported in the literature in association with extreme events of coastal inundation, thus making this study of relevance to many other coasts worldwide. The spreads of boulders extend several hundred meters inland and well beyond the present landward limit of storm activity. The marine origin of the boulders is demonstrated by well-developed macro-bioerosion sculpturing and in situ skeletal remains of endolithic shallow marine bivalves. The good state preservation of the fossils within the boulders indicates that abrasion duringtransport and redeposition was not significant. We envisage boulder deposition as having taken place during the Lisbon tsunami of ad 1755 through the simultaneous landward entrainment of coarse particles from nearshore followed by rapid shoreward suspended-dominated transport and non-graded redeposition that excluded significant sorting by weight or boulder dimensions. We use numerical hydrodynamic modeling of tsunami (and storm) waves to test the observational data on boulder dimensions (density, size, distribution) on the most likely processes of sediment deposition.     Exploring the mobility of mobile phone users

    Mobile phone datasets allow for the analysis of human behavior on an unprecedented scale. The social network, temporal dynamics and mobile behavior of mobile phone users have often been analyzed independently from each other using mobile phone datasets. In this article, we explore the connections between various features of human behavior extracted from a large mobile phone dataset. Our observations are based on the analysis of communication data of 100000 anonymized and randomly chosen individuals in a dataset of communications in Portugal. We show that clustering and principal component analysis allow for a significant dimension reduction with limited loss of information. The most important features are related to geographical location. In particular, we observe that most people spend most of their time at only a few locations. With the help of clustering methods, we then robustly identify home and office locations and compare the results with official census data. Finally, we analyze the geographic spread of users' frequent locations and show that commuting distances can be reasonably well explained by a gravity model.Comment: 16 pages, 12 figure
