    Biometric traits of Xylotrechus arvicola adults from laboratory and grape fields

    Xylotrechus arvicola is a pest in vineyards on the Iberian Peninsula. The action of the larvae, associated to the spread of wood fungi, causes a direct and indirect damage in the crop. This article presents biometric traits of X. arvicola population adults captured in three grape fields with Protected Designation of Origin (PDO) from the Iberian Peninsula and one population of this beetle obtained in the laboratory. The aim of the study was to observe the influence of sex and environment on the size of adults. The adults showed intersexual differences for the length and width of the pronotum, the length and width of the elytra and total length. The wild females were larger than wild males and the males from grape fields were slender than wild females. The environment affected the size of the adults. The laboratory adults reached the greatest body size. The size of the adult reflects the volume of material that had been used as host where the insect had developed. The captured adults in PDO Toro, with Bush Vines Training System (BVTS) were larger than the other PDOs, leading to bigger galleries and exit holes, which could affect the structural resistance of the plant and increase pathogenic fungi infection. However, BVTS facilitates the renovation of attacked branches, which is more difficult and slower in the Bilateral Cordon Training System (BCTS)

    Identification and functional analysis of novel phosphorylation sites in the RNA surveillance protein Upf1.

    One third of inherited genetic diseases are caused by mRNAs harboring premature termination codons as a result of nonsense mutations. These aberrant mRNAs are degraded by the Nonsense-Mediated mRNA Decay (NMD) pathway. A central component of the NMD pathway is Upf1, an RNA-dependent ATPase and helicase. Upf1 is a known phosphorylated protein, but only portions of this large protein have been examined for phosphorylation sites and the functional relevance of its phosphorylation has not been elucidated in Saccharomyces cerevisiae. Using tandem mass spectrometry analyses, we report the identification of 11 putative phosphorylated sites in S. cerevisiae Upf1. Five of these phosphorylated residues are located within the ATPase and helicase domains and are conserved in higher eukaryotes, suggesting a biological significance for their phosphorylation. Indeed, functional analysis demonstrated that a small carboxy-terminal motif harboring at least three phosphorylated amino acids is important for three Upf1 functions: ATPase activity, NMD activity and the ability to promote translation termination efficiency. We provide evidence that two tyrosines within this phospho-motif (Y-738 and Y-742) act redundantly to promote ATP hydrolysis, NMD efficiency and translation termination fidelity

    Artificial intelligence assisted Mid-infrared laser spectroscopy in situ detection of petroleum in soils

    A simple, remote-sensed method of detection of traces of petroleum in soil combining artificial intelligence (AI) with mid-infrared (MIR) laser spectroscopy is presented. A portable MIR quantum cascade laser (QCL) was used as an excitation source, making the technique amenable to field applications. The MIR spectral region is more informative and useful than the near IR region for the detection of pollutants in soil. Remote sensing, coupled with a support vector machine (SVM) algorithm, was used to accurately identify the presence/absence of traces of petroleum in soil mixtures. Chemometrics tools such as principal component analysis (PCA), partial least square-discriminant analysis (PLS-DA), and SVM demonstrated the e ectiveness of rapidly di erentiating between di erent soil types and detecting the presence of petroleum traces in di erent soil matrices such as sea sand, red soil, and brown soil. Comparisons between results of PLS-DA and SVM were based on sensitivity, selectivity, and areas under receiver-operator curves (ROC). An innovative statistical analysis method of calculating limits of detection (LOD) and limits of decision (LD) from fits of the probability of detection was developed. Results for QCL/PLS-DA models achieved LOD and LD of 0.2% and 0.01% for petroleum/soil, respectively. The superior performance of QCL/SVM models improved these values to 0.04% and 0.003%, respectively, providing better identification probability of soils contaminated with petroleum

    Model Predictive Control-Based Optimized Operation of a Hybrid Charging Station for Electric Vehicles

    This paper presents an energy management system (EMS) based on a novel approach using model predictive control (MPC) for the optimized operation of power sources in a hybrid charging station for electric vehicles (EVs). The hybrid charging station is composed of a photovoltaic (PV) system, a battery, a complete hydrogen system based on a fuel cell (FC), electrolyzer (EZ), and tank as an energy storage system (ESS), grid connection, and six fast charging units, all of which are connected to a common MVDC bus through Z-source converters (ZSC). The MPC-based EMS is designed to control the power flow among the energy sources of the hybrid charging station and reduce the utilization costs of the ESS and the dependency on the grid. The viability of the EMS was proved under a long-term simulation of 25 years in Simulink, using real data for the sun irradiance and a European load profile for EVs. Furthermore, this EMS is compared with a simpler alternative that is used as a benchmark, which pursues the same objectives, although using a states-based strategy. The results prove the suitability of the EMS, achieving a lower utilization cost (-25.3%), a notable reduction in grid use (-60% approximately) and an improvement in efficiency

    Differential expression of dnaA and dosR genes among members of the Mycobacterium tuberculosis complex under oxic and hypoxic conditions

    Major differences regarding the pathology and host immune response of the Beijing and Canettii genotypes of Mycobacterium tuberculosis have been reported; however, studies on the genetic expression of these genotypes during in vitro dormancy are scarce. This study examined the expression of five cell-cycle-related genes and two dormancy-related genes in M. canettii, M. tuberculosis H37Rv, and M. tuberculosis Beijing during the Wayne model of dormancy. The results showed that under hypoxic conditions the three tuberculosis genotypes were able to transcribe genes involved in DNA replication and cellular division. In addition, dosR was found to be up-regulated in M. tuberculosis Beijing during the exponential growth phase but down-regulated under hypoxic conditions. In this genotype, the replication-related gene dnaA was also strongly down-regulated. These latter two findings suggest that, compared to M. tuberculosis H37Rv and M. canettii, the Beijing genotype has a lower capacity to synthesize dosR, hspX, and dnaA mRNAs during in vitro dormancy.     The Galactic evolution of phosphorus

    As a galaxy evolves, its chemical composition changes and the abundance ratios of different elements are powerful probes of the underlying evolutionary processes. Phosphorous is an element whose evolution has remained quite elusive until now, because it is difficult to detect in cool stars. The infrared weak P I lines of the multiplet 1, at 1050-1082 nm, are the most reliable indicators of the presence of phosphorus. The availability of CRIRES at VLT has permitted access to this wavelength range in stellar spectra.We attempt to measure the phosphorus abundance of twenty cool stars in the Galactic disk. The spectra are analysed with one-dimensional model-atmospheres computed in Local Thermodynamic Equilibrium (LTE). The line formation computations are performed assuming LTE. The ratio of phosphorus to iron behaves similarly to sulphur, increasing towards lower metallicity stars. Its ratio with respect to sulphur is roughly constant and slightly larger than solar, [P/S]=0.10+- 0.10. We succeed in taking an important step towards the understanding of the chemical evolution of phosphorus in the Galaxy. However, the observed rise in the P/Fe abundance ratio is steeper than predicted by Galactic chemical evolution model model developed by Kobayashi and collaborators. Phosphorus appears to evolve differently from the light odd-Z elements sodium and aluminium. The constant value of [P/S] with metallicity implies that P production is insensitive to the neutron excess, thus processes other than neutron captures operate. We suggest that proton captures on 30Si and alpha captures on $27Al are possibilities to investigate. We see no clear distinction between our results for stars with planets and stars without any detected planet.Comment: To be published on A&

    Resistance training modulates reticulum endoplasmic stress, independent of oxidative and inflammatory responses, in elderly people

    [EN] Aging is related to changes in the redox status, low-grade inflammation, and decreased endoplasmic reticulum unfolded protein response (UPR). Exercise has been shown to regulate the inflammatory response, balance redox homeostasis, and ameliorate the UPR. This work aimed to investigate the effects of resistance training on changes in the UPR, oxidative status, and inflammatory responses in peripheral blood mononuclear cells of elderly subjects. Thirty elderly subjects volunteered to participate in an 8-week resistance training program, and 11 youth subjects were included for basal assessments. Klotho, heat shock protein 60 (HSP60), oxidative marker expression (catalase, glutathione, lipid peroxidation, nuclear factor erythroid 2-related factor 2, protein carbonyls, reactive oxygen species, and superoxide dismutase 1 and 2), the IRE1 arm of UPR, and TLR4/TRAF6/pIRAK1 pathway activation were evaluated before and following training. No changes in the HSP60 and Klotho protein content, oxidative status markers, and TLR4/TRAF6/pIRAK1 pathway activation were found with exercise. However, an attenuation of the reduced pIRE1/IRE1 ratio was observed following training. Systems biology analysis showed that a low number of proteins (RPS27A, SYVN1, HSPA5, and XBP1) are associated with IRE1, where XBP1 and RPS27A are essential nodes according to the centrality analysis. Additionally, a gene ontology analysis confirms that endoplasmic reticulum stress is a key mechanism modulated by IRE1.     Comparative Performance of Four Single Extreme Outlier Discordancy Tests from Monte Carlo Simulations

    Using highly precise and accurate Monte Carlo simulations of 20,000,000 replications and 102 independent simulation experiments with extremely low simulation errors and total uncertainties, we evaluated the performance of four single outlier discordancy tests (Grubbs test N2, Dixon test N8, skewness test N14, and kurtosis test N15) for normal samples of sizes 5 to 20. Statistical contaminations of a single observation resulting from parameters called δ from ±0.1 up to ±20 for modeling the slippage of central tendency or ε from ±1.1 up to ±200 for slippage of dispersion, as well as no contamination (δ=0 and ε=±1), were simulated. Because of the use of precise and accurate random and normally distributed simulated data, very large replications, and a large number of independent experiments, this paper presents a novel approach for precise and accurate estimations of power functions of four popular discordancy tests and, therefore, should not be considered as a simple simulation exercise unrelated to probability and statistics. From both criteria of the Power of Test proposed by Hayes and Kinsella and the Test Performance Criterion of Barnett and Lewis, Dixon test N8 performs less well than the other three tests.     Simplified model of battery energy-stored quasi-Z-source inverter-based photovoltaic power plant with Twofold energy management system

    The use of a battery energy-stored quasi-Z-source inverter (BES-qZSI) for large-scale PV power plants exhibits promising features due to the combination of qZSI and battery as energy storage system, such as single-stage power conversion (without additional DC/DC boost converter), improvements in the output waveform quality (due to the elimination of switching dead time), and continuous and smooth delivery of energy to the grid (through the battery energy storage system). This paper presents a new simplified model of a BES-qZSI to represent the converter dynamics with sufficient accuracy while using a less complex model than the detailed model (including the modelling of all switches and switching pulses). It is based on averaged values of the variables, voltage/current sources, and the same control circuit than the detailed model, except for the switching pulses generation. The simplified model enables faster time-domain simulation and is useful for control design and dynamic analysis purposes. Additionally, an energy management system has been developed to govern the power supply to grid under two possible scenarios: 1) System operator command following; or 2) economic dispatch of the stored energy.
