    Safer Tripping: Serotonergic Psychedelics and Drug Checking. Submission and Detection Rates, Potential Harms, and Challenges for Drug Analysis

    Purpose of Review With the continuous emergence of new psychoactive substances, drug checking (DC) services are challenged by an increasingly complex drug market. Considering the resumed scientific and public interest in serotonergic psychedelics (SPs) like LSD, psilocybin, and 2C-B, we present the results of a literature search investigating the presence and proportion of SPs in DC samples. Recent Findings: In 15 identified reports, submission and detection rates of SPs were comparably low, but increasing. Samples contained considerable amounts of adulterations or analogues, mostly novel SPs with unknown toxicological profiles and in some cases potentially life-threatening effects. The detection of SPs, however, requires advanced analysis techniques currently not available to most DC services. Summary: Given the substantial proportion of novel SPs in DC samples and the associated risks, DC can be a valuable harm reduction and monitoring tool for SPs if analysis techniques with high sensitivity are employed

    Breadboard model of the LISA phasemeter

The breadboard is build in the frame of an ESA technology development activity to demonstrate the feasibility and readiness of the LISA metrology baseline architecture.     Readout for intersatellite laser interferometry: Measuring low frequency phase fluctuations of HF signals with microradian precision

    Precision phase readout of optical beat note signals is one of the core techniques required for intersatellite laser interferometry. Future space based gravitational wave detectors like eLISA require such a readout over a wide range of MHz frequencies, due to orbit induced Doppler shifts, with a precision in the order of μrad/Hz\mu \textrm{rad}/\sqrt{\textrm{Hz}} at frequencies between 0.1 mHz0.1\,\textrm{mHz} and 1 Hz1\,\textrm{Hz}. In this paper, we present phase readout systems, so-called phasemeters, that are able to achieve such precisions and we discuss various means that have been employed to reduce noise in the analogue circuit domain and during digitisation. We also discuss the influence of some non-linear noise sources in the analogue domain of such phasemeters. And finally, we present the performance that was achieved during testing of the elegant breadboard model of the LISA phasemeter, that was developed in the scope of an ESA technology development activity.Comment: submitted to Review of Scientific Instruments on April 30th 201

    Localized actuation of temperature responsive hydrogel-layers with a PCB-based micro-heater array

    Space-resolved stimulation of active hydrogel layers can be achieved for example by using a micro-heater array. In the current work, we present the interaction of (i) such a rigid array of heating elements that can be selectively activated and (ii) an active thermo-responsive hydrogel layer that responds to the local stimulus change. Due to the respective local actuation, (iii) the surface form of a passive top-layer can be manipulated. We present continuum-based simulative predictions based on the Temperature Expansion Model and compare them to experimental outcomes for the system

    Psychedelic Experiences During the Early COVID-19 Pandemic: Findings From an International Online Survey

    Introduction: The current corona virus disease (COVID-19) pandemic has caused a serious global health crisis that has affected large parts of the public and private life worldwide, including the use of psychoactive substances. In this study, we investigated the effect of the COVID-19 pandemic on the use of serotonergic psychedelics, i.e., the settings in which people use psychedelics, the motives of usage, and the subjective quality of psychedelic experiences. Methods: The study was part of an international, cross-sectional, internet-based survey (N = 5,049) available in five languages (English, German, Spanish, Italian, and Korean) carried out during the early phase of the pandemic from April to August 2020. Participants were asked to retrospectively rate settings and motives of psychedelic substance use before the pandemic and in the last 4 weeks during the pandemic, as well as changes in psychedelic experiences. Results: Of n = 1,375 participants that reported the use psychedelics in 2019 or 2020, n = 642 (46.6%) also took psychedelics during the pandemic. During the pandemic, participants used psychedelics significantly less often in settings that were outside their home. Top motives to use psychedelics were comparable before and during the pandemic, but participants consumed less out of curiosity, to celebrate, or because friends took it, and more out of boredom. An increase in positively connoted, often pro-social experiences was observed. Two thirds of participants who used psychedelics during the pandemic claimed that psychedelics had helped them to deal better with the corona pandemic at least slightly. Discussion: Changes in setting and motives were mostly in line with restrictions caused by control measures to contain the spread of the virus. The unexpected increase in positively connoted experiences possibly reflects a favorable interaction of environmental macro- and individual micro-contexts during the pandemic (e.g., by reducing the use in more uncontrolled recreational settings or by encouraging a strong self-selection of substance users due to the expectation of "bad trips"). Increased pro-social feelings under psychedelics might reflect a desire for social interactions in times of social distancing and pandemic-related stress and anxiety

    Identifying predictors of attitudes towards local onshore wind development with reference to an English case study

    The threats posed by climate change are placing governments under increasing pressure to meet electricity demand from low-carbon sources. In many countries, including the UK, legislation is in place to ensure the continued expansion of renewable energy capacity. Onshore wind turbines are expected to play a key role in achieving these aims. However, despite high levels of public support for onshore wind development in principle, specific projects often experience local opposition. Traditionally this difference in general and specific attitudes has been attributed to NIMBYism (not in my back yard), but evidence is increasingly calling this assumption into question. This study used multiple regression analysis to identify what factors might predict attitudes towards mooted wind development in Sheffield, England. We report on the attitudes of two groups; one group (target) living close to four sites earmarked for development and an unaffected comparison group (comparison). We found little evidence of NIMBYism amongst members of the target group; instead, differences between general and specific attitudes appeared attributable to uncertainty regarding the proposals. The results are discussed with respect to literature highlighting the importance of early, continued and responsive community involvement in combating local opposition and facilitating the deployment of onshore wind turbines.     Predictors of Alcohol Consumption Among Younger Adults During the First Phase of the COVID-19 Pandemic

    Background: The COVID-19 pandemic may lead to negative mental health effects but the effect on alcohol consumption among younger adults is unclear. We assess predictors of change in alcohol consumption during the first phase of the COVID-19 pandemic among younger adults. Methods: This cross-sectional internet-based survey was part of an overarching project, the Corona Drug Survey, which was conducted from April 30 to August 4, 2020. Participants of any sex and >= 18 years old were included. The primary outcome measure was change in alcohol consumption during the early COVID-19 pandemic. We implemented an ordinal logistic regression to assess the effect (odds ratio [OR] and 95% confidence interval [CI]) of the following predictors: quarantine restrictions on leaving the residence, number of individuals in the household, problematic alcohol consumption before the pandemic (CAGE [cutting down, annoyance by criticism, guilty feeling, and eye-opener] score), personal concern regarding the pandemic, age, and sex. Results: 3,321 participants with a mean age of 32 (SD: 13) years were included in this study. 70.4% of participants reported less or unchanged alcohol consumption in the recent 4 weeks of the pandemic compared to before the pandemic. A higher number of individuals in the household was associated with a reduced alcohol consumption (OR = 0.869; 95% CI = 0.815-0.927). No quarantine restrictions on leaving the residence (OR = 1.593; 95% CI = 1.397-1.817), a higher age (1.006; 1.001-1.011), and female sex (compared to males: 1.206; 1.062-1.371) were associated with an increase in alcohol consumption. The CAGE score before the pandemic (OR = 0.983; 95% CI = 0.931-1.037) and the pandemic concern (0.927; 0.857-1.003) were not associated with a significant change in alcohol consumption. Celebrations were no longer frequent drinking occasions during the pandemic compared to before the pandemic. The majority of participants (60.9%) did not use alcohol drinking as a coping mechanism to mitigate negative effects of the pandemic. Interpretation: In this cohort of younger adults with fewer celebratory drinking occasions, restrictions on leaving the residence and the number of persons in the household were the strongest predictors of reduced alcohol consumption during the early phase of the pandemic.</p

    Die Aktivität der akuten Atemwegserkrankungen (ARE-Raten) in der Bevölkerung (GrippeWeb) ist in der 30. KW 2022 im Vergleich zur Vorwoche gesunken. Die Werte liegen weiterhin auf einem deutlich höheren Niveau als in den Vorjahren. Im ambulanten Bereich (Arbeitsgemeinschaft Influenza) wurden in der 30. KW bundesweit weniger Arztbesuche wegen ARE registriert als in der Vorwoche. Die Zahl der Arztbesuche liegt ebenfalls noch deutlich über dem Niveau der Vorjahre (seit 2006) um diese Zeit. Kalenderwoche 30 (25.7. bis 31.7.2022) Im NRZ für Influenzaviren wurden in der 30. KW 2022 in insgesamt 51 (55 %) der 92 eingesandten Sentinelproben respiratorische Viren identifiziert, darunter 17 (18 %) Proben mit Parainfluenzaviren (PIV), 16 (17 %) mit SARS-CoV-2, elf (12 %) mit Rhinoviren, drei (3 %) mit Respiratorischen Synzytialviren (RSV) und jeweils zwei (3 %) mit humanen saisonalen Coronaviren (hCoV), humanen Metapneumoviren (hMPV) und Influenzaviren. Im Rahmen der ICD-10-Code basierten Krankenhaussurveillance (ICOSARI) ist die Zahl schwerer akuter respiratorischer Infektionen (SARI) in der 30. KW 2022 insgesamt gesunken, nachdem die Werte seit der 16. KW 2022 trotz leichter Schwankungen weitestgehend stabil waren. In den Altersgruppen unter 60 Jahren liegen die SARI-Fallzahlen auf einem während der Sommermonate üblichen Niveau, insbeson-dere bei den ab 80-Jährigen werden allerdings noch höhere Fallzahlen beobachtet als in den Vorsaisons. Die im Vergleich mit den Vorjahren aktuell noch deutlich höhere ARE-Aktivität ist auf die Ko-Zirkulation verschiedener Atemwegserreger zurückzuführen. Es werden SARS-CoV-2 hauptsächlich bei Erwachsenen und Parainfluenza- sowie Rhinoviren bei Kindern nachgewiesen. In den letzten Wochen wurde wieder sporadisch RSV in Sentinelproben identifiziert

    Impact of SARS-CoV-2 ORF6 and its variant polymorphisms on host responses and viral pathogenesis

    : Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) encodes several proteins that inhibit host interferon responses. Among these, ORF6 antagonizes interferon signaling by disrupting nucleocytoplasmic trafficking through interactions with the nuclear pore complex components Nup98-Rae1. However, the roles and contributions of ORF6 during physiological infection remain unexplored. We assessed the role of ORF6 during infection using recombinant viruses carrying a deletion or loss-of-function (LoF) mutation in ORF6. ORF6 plays key roles in interferon antagonism and viral pathogenesis by interfering with nuclear import and specifically the translocation of IRF and STAT transcription factors. Additionally, ORF6 inhibits cellular mRNA export, resulting in the remodeling of the host cell proteome, and regulates viral protein expression. Interestingly, the ORF6:D61L mutation that emerged in the Omicron BA.2 and BA.4 variants exhibits reduced interactions with Nup98-Rae1 and consequently impairs immune evasion. Our findings highlight the role of ORF6 in antagonizing innate immunity and emphasize the importance of studying the immune evasion strategies of SARS-CoV-2
