Human colon cancer cell lines show a diverse pattern of nitric oxide synthase gene expression and nitric oxide generation.

A panel of human colonic adenocarcinoma cell lines was examined both for expression of mRNAs of the nitric oxide synthase (NOS) gene family and for evidence of enzymic activity based on citrulline and nitrite (NO2-) formation. Reverse transcription-polymerase chain reaction (RT-PCR), revealed that all lines (SW480, SW620, DLD-1 and WiDr) expressed mRNA for the Ca(2+)-dependent endothelial (e)NOS, while SW480 cells also expressed the Ca(2+)-dependent neuronal (n)NOS. The mRNA for the Ca(2+)-independent inducible (i)NOS was expressed both by cytokine-stimulated and by unstimulated SW480, SW620 and DLD-1 cells, but none was seen at any time in the WiDr cells. There was, however, little correlation between mRNA expression and enzymic activity based on citrulline and NO2- formation. Thus none of the cell lines exhibited measurable Ca(2+)-dependent NOS activity, while Ca(2+)-independent NOS activity was seen in all but the WiDr cells. Furthermore, DLD-1 cells generated citrulline with resultant NO2- formation only after stimulation with lipopolysaccharide (LPS) and/or cytokines, while SW480 and SW620 did so constitutively. Thus RT-PCR studies indicate that tumour cells of similar epithelial origin display a diverse pattern of NOS gene family expression, and parallel biochemical studies clearly indicate that such expression does not always result in measurable enzymic activity leading to the generation of NO

A Cognitive Model of an Epistemic Community: Mapping the Dynamics of Shallow Lake Ecosystems

We used fuzzy cognitive mapping (FCM) to develop a generic shallow lake ecosystem model by augmenting the individual cognitive maps drawn by 8 scientists working in the area of shallow lake ecology. We calculated graph theoretical indices of the individual cognitive maps and the collective cognitive map produced by augmentation. The graph theoretical indices revealed internal cycles showing non-linear dynamics in the shallow lake ecosystem. The ecological processes were organized democratically without a top-down hierarchical structure. The steady state condition of the generic model was a characteristic turbid shallow lake ecosystem since there were no dynamic environmental changes that could cause shifts between a turbid and a clearwater state, and the generic model indicated that only a dynamic disturbance regime could maintain the clearwater state. The model developed herein captured the empirical behavior of shallow lakes, and contained the basic model of the Alternative Stable States Theory. In addition, our model expanded the basic model by quantifying the relative effects of connections and by extending it. In our expanded model we ran 4 simulations: harvesting submerged plants, nutrient reduction, fish removal without nutrient reduction, and biomanipulation. Only biomanipulation, which included fish removal and nutrient reduction, had the potential to shift the turbid state into clearwater state. The structure and relationships in the generic model as well as the outcomes of the management simulations were supported by actual field studies in shallow lake ecosystems. Thus, fuzzy cognitive mapping methodology enabled us to understand the complex structure of shallow lake ecosystems as a whole and obtain a valid generic model based on tacit knowledge of experts in the field.Comment: 24 pages, 5 Figure

    Gamma Rays from Centaurus A

Centaurus A, the cosmic ray accelerator a few Mpc away from us is possibly one of the nearest sources of extremely high energy cosmic rays. We investigate whether the gamma ray data currently available from Centaurus A in the GeV-TeV energy band can be explained with only proton proton interactions. We show that for a single power law proton spectrum, mechanisms of γ\gamma-ray production other than proton proton interactions are needed inside this radio-galaxy to explain the gamma ray flux observed by EGRET, upper limits by H.E.S.S./CANGAROO-III and the correlated extremely energetic cosmic ray events observed by the Pierre Auger experiment. In future with better γ\gamma-ray data, simultaneous observation with γ\gamma-ray and cosmic ray detectors, it would be possible to carry out such studies on different sources in more detail.Comment: 7 pages, JCAP in pres

The STAR Silicon Strip Detector (SSD)

The STAR Silicon Strip Detector (SSD) completes the three layers of the Silicon Vertex Tracker (SVT) to make an inner tracking system located inside the Time Projection Chamber (TPC). This additional fourth layer provides two dimensional hit position and energy loss measurements for charged particles, improving the extrapolation of TPC tracks through SVT hits. To match the high multiplicity of central Au+Au collisions at RHIC the double sided silicon strip technology was chosen which makes the SSD a half million channels detector. Dedicated electronics have been designed for both readout and control. Also a novel technique of bonding, the Tape Automated Bonding (TAB), was used to fullfill the large number of bounds to be done. All aspects of the SSD are shortly described here and test performances of produced detection modules as well as simulated results on hit reconstruction are given.Comment: 11 pages, 8 figures, 1 tabl

Parsec-scale dust distributions in Seyfert galaxies - Results of the MIDI AGN snapshot survey

The emission of warm dust dominates the mid-infrared spectra of active galactic nuclei (AGN). Only interferometric observations provide the necessary angular resolution to resolve the nuclear dust and to study its distribution and properties. The investigation of dust in AGN cores is hence one of the main science goals for the MID-infrared Interferometric instrument MIDI at the VLTI. As the first step, the feasibility of AGN observations was verified and the most promising sources for detailed studies were identified. This was carried out in a "snapshot survey" with MIDI using Guaranteed Time Observations. In the survey, observations were attempted for 13 of the brightest AGN in the mid-infrared which are visible from Paranal. The results of the three brightest, best studied sources have been published in separate papers. Here we present the interferometric observations for the remaining 10, fainter AGN. For 8 of these, interferometric measurements could be carried out. Size estimates or limits on the spatial extent of the AGN-heated dust were derived from the interferometric data of 7 AGN. These indicate that the dust distributions are compact, with sizes on the order of a few parsec. The derived sizes roughly scale with the square root of the luminosity in the mid-infrared, s ~ sqrt(L), with no clear distinction between type 1 and type 2 objects. This is in agreement with a model of nearly optically thick dust structures heated to T ~ 300 K. For three sources, the 10 micron feature due to silicates is tentatively detected either in emission or in absorption. Based on the results for all AGN studied with MIDI so far, we conclude that in the mid-infrared the differences between individual galactic nuclei are greater than the generic differences between type 1 and type 2 objects.Comment: 18 pages, 8 figures, updated to version published in A&A 502, 67-8

Observations of Massive Star Forming Regions with Water Masers: Mid-Infrared Imaging

We present here a mid-infrared imaging survey of 26 sites of water maser emission. Observations were obtained at the InfraRed Telescope Facility 3-m telescope with the University of Florida mid-infrared imager/spectrometer OSCIR, and the JPL mid-infrared camera MIRLIN. The main purpose of the survey was to explore the relationship between water masers and the massive star formation process. It is generally believed that water masers predominantly trace outflows and embedded massive stellar objects, but may also exist in circumstellar disks around young stars. We investigate each of these possibilities in light of our mid-infrared imaging. We find that mid-infrared emission seems to be more closely associated with water and OH maser emission than cm radio continuum emission from UC HII regions. We also find from the sample of sources in our survey that, like groups of methanol masers, both water and OH masers have a proclivity for grouping into linear or elongated distributions. We conclude that the vast majority of linearly distributed masers are not tracing circumstellar disks, but outflows and shocks instead.Comment: 49 pages; 23 figures; To appear in February 2005 ApJS; To download a version with better quality figures, go to http://www.ctio.noao.edu/~debuizer
