    Magnificent Distance: Five Site-Specific Installations Washington DC 2012

    5x5, Washington DCā€™s inaugural public art festival, was conceived as a flagship biennial in which five curators would each be invited to curate new site-specific artworks by five artists ā€“ leading to the simultaneous installation of twenty-five artworks across Washington DC. The primary research question explored in the curation of the five Magnificent Distance artworks was the slippage between the symbolic DC of the worldwide public imagination and the ā€˜domesticā€™, human DC with its complex histories and communities. Many of the exhibition sites, selected as part of my curatorial role, were at the interstices of these two DC realities ā€“ at the meeting point between federal and community environments, in locations undergoing transformation from one use to another, and at points where differing scales of architecture meet

    The inverse relationship between serum albumin concentration and its half-life suggested to early workers that albumin would be protected from a catabolic fate by a receptor-mediated mechanism much like that proposed for IgG. We show here that albumin binds FcRn in a pH dependent fashion, that the lifespan of albumin is shortened in FcRn-deficient mice, and that the plasma albumin concentration of FcRn-deficient mice is less than half that of wild-type mice. These results affirm the hypothesis that the major histocompatibility complexā€“related Fc receptor protects albumin from degradation just as it does IgG, prolonging the half-lives of both

    Structure of an Hsp90-Cdc37-Cdk4 complex

    Activation of many protein kinases depends on their interaction with the Hsp90 molecular chaperone system. Recruitment of protein kinase clients to the Hsp90 chaperone system is mediated by the cochaperone adaptor protein Cdc37, which acts as a scaffold, simultaneously binding protein kinases and Hsp90. We have now expressed and purified an Hsp90-Cdc37-Cdk4 complex, defined its stoichiometry, and determined its 3D structure by single-particle electron microscopy. Comparison with the crystal structure of Hsp90 allows us to identify the locations of Cdc37 and Cdk4 in the complex and suggests a mechanism by which conformational changes in the kinase are coupled to the Hsp90 ATPase cycle

    Corrected score methods for estimating Bayesian networks with error-prone nodes

    Motivated by inferring cellular signaling networks using noisy flow cytometry data, we develop procedures to draw inference for Bayesian networks based on error-prone data. Two methods for inferring causal relationships between nodes in a network are proposed based on penalized estimation methods that account for measurement error and encourage sparsity. We discuss consistency of the proposed network estimators and develop an approach for selecting the tuning parameter in the penalized estimation methods. Empirical studies are carried out to compare the proposed methods and a naive method that ignores measurement error with applications to synthetic data and to single cell flow cytometry data

    Estimating Intervention Effects in a Complex Multi-Level Smoking Prevention Study

    This paper illustrates how to estimate cumulative and non-cumulative treatment effects in a complex school-based smoking intervention study. The Instrumental Variable method is used to tackle non-compliance and measurement error for a range of treatment exposure measures (binary, ordinal and continuous) in the presence of clustering and dropout. The results are compared to more routine analyses. The empirical findings from this study provide little encouragement for believing that poorly resourced school-based interventions can bring about substantial long-lasting reductions in smoking behaviour but that novel components such as a computer game might have some short-term effect

    How to Educate Entrepreneurs?

    Entrepreneurship education has two purposes: To improve studentsā€™ entrepreneurial skills and to provide impetus to those suited to entrepreneurship while discouraging the rest. While entrepreneurship education helps students to make a vocational decision its effects may conflict for those not suited to entrepreneurship. This study shows that vocational and the skill formation effects of entrepreneurship education can be identified empirically by drawing on the Theory of Planned Behavior. This is embedded in a structural equation model which we estimate and test using a robust 2SLS estimator. We find that the attitudinal factors posited by the Theory of Planned Behavior are positively correlated with studentsā€™ entrepreneurial intentions. While conflicting effects of vocational and skill directed course content are observed in some individuals, overall these types of content are complements. This finding contradicts previous results in the literature. We reconcile the conflicting findings and discuss implications for the design of entrepreneurship courses

    La proliferaciĆ³n de rankings de universidades en las Ćŗltimas dĆ©cadas y su papel emergente en la evaluaciĆ³n de los sistemas universitarios justifican el estudio de estos productos en los medios de masas. El presente trabajo pretende estudiar la repercusiĆ³n que han tenido los ranking de universidades en la prensa espaƱola durante 160 meses, periodo comprendido entre el 1 de enero de 1998 hasta el 30 de abril de 2012, para ello se ha utilizado MyNewsOnlineĀ®. Se analiza la producciĆ³n temporal, territorial y por cabeceras. Igualmente se estudian las noticias que han generado los principales rankings de universidades. Los resultados demuestran el creciente interĆ©s de los medios por estas herramientas cientĆ­ficas, especialmente por parte de la prensa de Ć”mbito nacional. En las cabeceras de cobertura nacional destacan los rankings ARWU y Times, mientras que en la prensa local lo hace el Scimago. Las cabeceras mĆ”s productivas son El PaĆ­s, La Vanguardia y ABC.The proliferation of university rankings in the last few decades and their increasing role in the evaluation of university systems justify the study of their impact on the media. The present work aims to measure the impact of university rankings in the Spanish press over 160 months, between January 1, 1998 and April 30, 2012. Using the news database MyNewsOnline as the main source of information, we analysed the production of news according to the following variables: timing, geographical distribution, newspaper and rankings. Results show an increasing interest in these tools. The Shanghai Ranking and Times Higher Education University Ranking stand out in the national press, while in the local press the Scimago Institutions Ranking draws the most attention from media. The newspapers with the most news about rankings are ā€œEl PaĆ­sā€, ā€œLa Vanguardiaā€ and ā€œABCā€

    Mycobacterium tuberculosis infection induces il12rb1 splicing to generate a novel IL-12RĪ²1 isoform that enhances DC migration

    RNA splicing is an increasingly recognized regulator of immunity. Here, we demonstrate that after Mycobacterium tuberculosis infection (mRNA) il12rb1 is spliced by dendritic cells (DCs) to form an alternative (mRNA) il12rb1Deltatm that encodes the protein IL-12Rbeta1DeltaTM. Compared with IL-12Rbeta1, IL-12Rbeta1DeltaTM contains an altered C-terminal sequence and lacks a transmembrane domain. Expression of IL-12Rbeta1DeltaTM occurs in CD11c(+) cells in the lungs during M. tuberculosis infection. Selective reconstitution of il12rb1(-/-) DCs with (mRNA) il12rb1 and/or (mRNA) il12rb1Deltatm demonstrates that IL-12Rbeta1DeltaTM augments IL-12Rbeta1-dependent DC migration and activation of M. tuberculosis-specific T cells. It cannot mediate these activities independently of IL12Rbeta1. We hypothesize that M. tuberculosis-exposed DCs express IL-12Rbeta1DeltaTM to enhance IL-12Rbeta1-dependent migration and promote M. tuberculosis-specific T cell activation. IL-12Rbeta1DeltaTM thus represents a novel positive-regulator of IL12Rbeta1-dependent DC function and of the immune response to M. tuberculosis.This work was supported by the Trudeau Institute and the National Institutes of Health (AI067723 to A.
