    Dwarf mistletoe-pandora moth interaction and its contribution to ponderosa pine mortality in Arizona

    The interaction between Southwestern dwarf mistletoe, Arceuthobtum vaginalum subspecies cryptopodum, infestation and defoliation by the pandora moth, Coloradia pandora pandora , on the Kaibab Plateau in Arizona was evaluated. Heavy defoliation of ponderosa pine, Pinus ponderosa, in 1979 and 1981 resulted in mortality of individual trees in areas of heavy dwarf mistletoe infestation. Postmortem evaluation of ponderosa pines indicated that dead trees had a significantly higher dwarf mistletoe rating than did nearby paired live trees. Of 25 tree pairs evaluated, only two live trees had higher dwarf mistletoe ratings than the paired dead tree. Mean dwarf mistletoe ratings were:live trees 2.9, dead trees 4.6 (6 class dwarf mistletoe rating system). Implications for management of the pandora moth are discussed

    The marine reservoir age of Greenland coastal waters

    Knowledge of the marine reservoir age is fundamental for creating reliable chronologies of marine sediment archives based on radiocarbon dating. This age difference between the 14C age of a marine sample and that of its contemporaneous atmosphere is dependent on several factors (among others, ocean circulation, water mass distribution, terrestrial runoff, upwelling, and sea-ice cover) and is therefore spatially heterogeneous. Anthropogenic influence on the global isotopic carbon system, mostly through atmospheric nuclear tests, has complicated the determination of the regional reservoir age correction ΔR, which therefore can only be measured in historic samples of known age. In this study we expand on the few existing measurements of ΔR for the coastal waters around Greenland, by adding 92 new radiocarbon dates on mollusks from museum collections. All studied mollusk samples were collected during historic expeditions of the late 19th and early 20th centuries, and besides coastal sites around Greenland, the new measurements also include localities from the western Labrador Sea, Baffin Bay, and the Iceland Sea. Together with existing measurements, the new results are used to calculate average ΔR values for different regions around Greenland, all in relation to Marine20, the most recent marine radiocarbon calibration curve. To support further discussions and comparison with previous datasets, we use the term ΔR13, where the suffix 13 refers to the previous calibration curve Marine13. Our study explores the links between the marine reservoir age and oceanography, sea-ice cover, water depth, mollusk feeding habits, and the presence of carbonate bedrock. Although we provide regional averages, we encourage people to consult the full catalogue of measurements and determine a suitable ΔR for each case individually, based on the exact location including water depth. Despite this significant expansion of the regional reservoir age database around Greenland, data from the northern coast, directly bordering the Arctic Ocean, remain missing.</p

All three systems have orbital obliquity measurements, WASP-24 and WASP-26 have been observed with Spitzer, and WASP-25 was previously comparatively neglected. Our light curves were obtained using the telescope-defocussing method and have scatters of 0.5–1.2 mmag relative to their best-fitting geometric models. We use these data to measure the physical properties and orbital ephemerides of the systems to high precision, finding that our improved measurements are in good agreement with previous studies. High-resolution Lucky Imaging observations of all three targets show no evidence for faint stars close enough to contaminate our photometry. We confirm the eclipsing nature of the star closest to WASP-24 and present the detection of a detached eclipsing binary within 4.25 arcmin of WASP-26.Publisher PDFPeer reviewe

    Stepwise emergence of azole, echinocandin and amphotericin B multidrug resistance in vivo in Candida albicans orchestrated by multiple genetic alterations.

    OBJECTIVES: The objective of this study was to characterize the underlying molecular mechanisms in consecutive clinical Candida albicans isolates from a single patient displaying stepwise-acquired multidrug resistance. METHODS: Nine clinical isolates (P-1 to P-9) were susceptibility tested by EUCAST EDef 7.2 and Etest. P-4, P-5, P-7, P-8 and P-9 were available for further studies. Relatedness was evaluated by MLST. Additional genes were analysed by sequencing (including FKS1, ERG11, ERG2 and TAC1) and gene expression by quantitative PCR (CDR1, CDR2 and ERG11). UV-spectrophotometry and GC-MS were used for sterol analyses. In vivo virulence was determined in the insect model Galleria mellonella and evaluated by log-rank Mantel-Cox tests. RESULTS: P-1 + P-2 were susceptible, P-3 + P-4 fluconazole resistant, P-5 pan-azole resistant, P-6 + P-7 pan-azole and echinocandin resistant and P-8 + P-9 MDR. MLST supported genetic relatedness among clinical isolates. P-4 harboured four changes in Erg11 (E266D, G307S, G450E and V488I), increased expression of ERG11 and CDR2 and a change in Tac1 (R688Q). P-5, P-7, P-8 and P-9 had an additional change in Erg11 (A61E), increased expression of CDR1, CDR2 and ERG11 (except for P-7) and a different amino acid change in Tac1 (R673L). Echinocandin-resistant isolates harboured the Fks1 S645P alteration. Polyene-resistant P-8 + P-9 lacked ergosterol and harboured a frameshift mutation in ERG2 (F105SfsX23). Virulence was attenuated (but equivalent) in the clinical isolates, but higher than in the azole- and echinocandin-resistant unrelated control strain. CONCLUSIONS: C. albicans demonstrates a diverse capacity to adapt to antifungal exposure. Potentially novel resistance-inducing mutations in TAC1, ERG11 and ERG2 require independent validation

    Online consultations in mental healthcare: Modelling determinants of use and experience based on an international survey study at the onset of the pandemic

    Introduction: While online consultations have shown promise to be a means for the effective delivery of high -quality mental healthcare and the first implementations of these digital therapeutic contacts go back nearly two decades, uptake has remained limited over the years. The onset of the COVID-19 pandemic dramatically altered this relative standstill and created a unique turning point, with a massive amount of both professionals and clients having first hands-on experiences with technology in mental healthcare.Objective: The current study aimed to document the uptake of online consultations and explore if specific characteristics of mental health professionals across and beyond Europe could predict this.Methods: An international survey was designed to assess mental health professionals' (initial) experiences with online consultations at the onset of the pandemic: their willingness to make use of them and their prior and current experiences, alongside several personal characteristics. Logistic mixed-effects models were used to identify predictors of the use of online consultations, personal experience with this modality, and the sense of telepresence.Results: A total of 9115 healthcare professionals from 73 countries participated of which about two-thirds used online consultations during the initial COVID-19 outbreak. The current study identifies multiple determinants relating to the use and experience of online consultations, including the professionals' age, experience with the technology before the outbreak, the professional context, and training.Conclusions: Despite strong evidence supporting the relevance of training in digital mental health, this is clearly still lacking. Nevertheless, the COVID-19 pandemic presented a first, and potentially transformative, experience with online consultations for many healthcare professionals. The insights from this study can help supportprofessionals and, importantly, (mental) healthcare organisations to create optimal circumstances for selective and high-quality continued use of online consultations

    Accuracy of qPCR and bacterial culture for the diagnosis of bovine intramammary infections and teat skin colonisation with Streptococcus agalactiae and Staphylococcus aureus using Bayesian analysis

    Streptococcus agalactiae (Strep. agalactiae) and Staphylococcus aureus (Staph. aureus) are originally regarded as contagious mastitis pathogens, however, both pathogens have recently been isolated from extramammary and environmental sites, indicating that other sites than the udder might contribute to the spread of these pathogens potentially causing intramammary infections. Diagnostic tools to identify pathogens at extramammary sites are available but still needs to be validated. The objective of this cross-sectional field study was to estimate the diagnostic sensitivity (Se) and specificity (Sp) of the commercially available Mastit4 qPCR assay and bacterial culture (BC) in identifying Strep. agalactiae and Staph. aureus from milk and teat skin samples. We randomly selected 30-40 cows with high somatic cell counts from eight Danish Strep. agalactiae-positive dairy herds with automatic milking systems. Teat skin samples and aseptic milk samples were collected from right rear quarters (n = 287) for BC and PCR analysis. Se and Sp were estimated in a Bayesian latent class analysis. For milk samples, the Se and Sp of qPCR for Strep. agalactiae were estimated to 0.97 and 0.99, respectively, whereas the Se and Sp of BC were 0.41 and 1.00, respectively. The Se and Sp of qPCR for Staph. aureus were estimated to 0.95 and 0.99, respectively, whereas the Se and Sp of BC were 0.54 and 0.77, respectively. For teat skin samples, the Se and Sp of qPCR for Strep. agalactiae were estimated to be 0.97 and 0.96, respectively, whereas the Se and Sp of BC were 0.33 and 1.00, respectively. The Se and Sp of qPCR for Staph. aureus were estimated to 0.94 and 0.98, respectively, whereas the Se and Sp of BC were 0.44 and 0.74, respectively. In conclusion, the Se for diagnosing Strep. agalactiae and Staph. aureus IMI was higher for qPCR than BC, suggesting that qPCR is a valuable method for detecting both pathogens from quarter-level milk samples. The performance of BC in the detection of Strep. agalactiae and Staph. aureus on teat skin was poor compared to qPCR, indicating that differences in the target condition of the two methods should be considered when implementing them as routine diagnostic tests for detecting teat skin colonisers. The low Se of BC may preclude the use of BC for skin testing, and qPCR is better for this task.info:eu-repo/semantics/publishedVersio

    MOA-2010-BLG-477Lb: constraining the mass of a microlensing planet from microlensing parallax, orbital motion and detection of blended light

    Microlensing detections of cool planets are important for the construction of an unbiased sample to estimate the frequency of planets beyond the snow line, which is where giant planets are thought to form according to the core accretion theory of planet formation. In this paper, we report the discovery of a giant planet detected from the analysis of the light curve of a high-magnification microlensing event MOA-2010-BLG-477. The measured planet-star mass ratio is q=(2.181±0.004)×103q=(2.181\pm0.004)\times 10^{-3} and the projected separation is s=1.1228±0.0006s=1.1228\pm0.0006 in units of the Einstein radius. The angular Einstein radius is unusually large θE=1.38±0.11\theta_{\rm E}=1.38\pm 0.11 mas. Combining this measurement with constraints on the "microlens parallax" and the lens flux, we can only limit the host mass to the range 0.13<M/M<1.00.13<M/M_\odot<1.0. In this particular case, the strong degeneracy between microlensing parallax and planet orbital motion prevents us from measuring more accurate host and planet masses. However, we find that adding Bayesian priors from two effects (Galactic model and Keplerian orbit) each independently favors the upper end of this mass range, yielding star and planet masses of M=0.670.13+0.33 MM_*=0.67^{+0.33}_{-0.13}\ M_\odot and mp=1.50.3+0.8 MJUPm_p=1.5^{+0.8}_{-0.3}\ M_{\rm JUP} at a distance of D=2.3±0.6D=2.3\pm0.6 kpc, and with a semi-major axis of a=21+3a=2^{+3}_{-1} AU. Finally, we show that the lens mass can be determined from future high-resolution near-IR adaptive optics observations independently from two effects, photometric and astrometric.Comment: 3 Tables, 12 Figures, accepted in Ap
