    South African Dyslipidaemia Guideline Consensus Statement

    Prepared on behalf of the South African Heart Association and Lipid and Atherosclerosis Society of Southern Africa. The European Society of Cardiology together with the European Atherosclerosis Society published updated dyslipidaemia guidelines in 2011. SA Heart and the Lipid and Atherosclerosis Society of Southern Africa officially adopt these guidelines. This statement adapts aspects of the guidelines to the South African situation. Using the updated Framingham risk charts, interventional strategies are based according to the cardiovascular risk score and low-density lipoprotein cholesterol (LDL-C) levels. The Framingham risk score refers to the 10-year risk of any cardiovascular event, and includes four categories of risk. Treatment targets are those of the European guidelines. The LDL-C goal is 1.8 mmol/l for the very high-risk group (>30%), 2.5 mmol/l for the high-risk group (15 - 30%), and 3 mmol/l for those below 15% risk. Intensive management of dyslipidaemia in South Africa will significantly reduce the cardiovascular disease health burden

    Developing global pedotransfer functions to estimate available soil phosphorus

    There are a large number of investigations that estimate available soil phosphorous (P), but a paucity of global data on available soil P. One significant modern challenge is developing low cost, accurate approaches to predict available soil P that are useful to scientists around the world. We conducted a global meta-analysis using data on available soil P from 738 sites, 640 in the USA and 149 in 14 other countries. Four different methods of determining available soil P, New Zealand (NZ), acid oxalate, Bray and Mehlich 3 were represented in the dataset. Inputs evaluated for inclusion in the pedotransfer functions to predict available soil P were clay (C), fine silt, (FSi) coarse silt (CSi), very fine sand (VFS), fine sand (FS), medium sand (MS), coarse sand (CS), very coarse sand (VCS), organic carbon (OC), pH, calcium (Ca), magnesium (Mg), potassium (K), iron (Fe), aluminum (Al), and manganese (Mn). Available soil P was estimated for: 1) the entire dataset, 2) only the USA, and 3) the non-USA dataset. The best models to estimate available soil P were obtained for the NZ method (using the co-variates C, FSi, CSi, VFS, MS, CS, OC, Fe, Al, Mn, Ca, Mg, and pH) and for the acid oxalate method (using the co-variates C, FSi, Fe, Al, Mn, Ca, and Mg). Although estimation of available soil P determined with the acid oxalate method was poor for the entire dataset, good estimates were obtained for the USA and non-USA datasets separately. Models for the Bray and Mehlich 3 methods only predicted available soil P well for the non-USA dataset. Using pedotransfer function models to estimate available soil P could provide an efficient and cost effective way to estimate global distributions of a soil property that is important for a number of agricultural and environmental reasons

    Ensuring traceability of field experimental data or laboratory sampling data to conduct reproducible research is a challenge at the present time. Between the time when geolocalized specimens (biotic or abiotic) are taken, and the time the resulting data ends up in analysis published with a study, many manual operations take place and may generate errors. The French LTSER have joined forces at the national level to propose a solution as generic as possible to this problem of monitoring of the samples and the data associated with them. Compared to existing solutions (such as Laboratory Information Management Systems), we target a robust labeling solution adapted to outdoor working conditions, with the management of stocks and movements of samples. We designed and realized a prototype tested from end to end, using an open source software, cheap Zebra printers (mobile or not) and raspberries as devices. This solution provides sufficient flexibility for the wide variety of existing protocols. In strength is the record of all contextual data associated with the samples, which constitute important parameters of the subsequent analyses. At last, not only traceability is guaranteed, but also a gain of time and a rationalization of the storage of samples that will induce a return on investment

    Functionalized Positive Nanoparticles Reduce Mucin Swelling and Dispersion

    Multi-functionalized nanoparticles (NPs) have been extensively investigated for their potential in household and commercial products, and biomedical applications. Previous reports have confirmed the cellular nanotoxicity and adverse inflammatory effects on pulmonary systems induced by NPs. However, possible health hazards resulting from mucus rheological disturbances induced by NPs are underexplored. Accumulation of viscous, poorly dispersed, and less transportable mucus leading to improper mucus rheology and dysfunctional mucociliary clearance are typically found to associate with many respiratory diseases such as asthma, cystic fibrosis (CF), and COPD (Chronic Obstructive Pulmonary Disease). Whether functionalized NPs can alter mucus rheology and its operational mechanisms have not been resolved. Herein, we report that positively charged functionalized NPs can hinder mucin gel hydration and effectively induce mucin aggregation. The positively charged NPs can significantly reduce the rate of mucin matrix swelling by a maximum of 7.5 folds. These NPs significantly increase the size of aggregated mucin by approximately 30 times within 24 hrs. EGTA chelation of indigenous mucin crosslinkers (Ca2+ ions) was unable to effectively disperse NP-induced aggregated mucins. Our results have demonstrated that positively charged functionalized NPs can impede mucin gel swelling by crosslinking the matrix. This report also highlights the unexpected health risk of NP-induced change in mucus rheological properties resulting in possible mucociliary transport impairment on epithelial mucosa and related health problems. In addition, our data can serve as a prospective guideline for designing nanocarriers for airway drug delivery applications

    Systems Biology Approach Identifies Prognostic Signatures of Poor Overall Survival and Guides the Prioritization of Novel BET-CHK1 Combination Therapy for Osteosarcoma

    Osteosarcoma (OS) patients exhibit poor overall survival, partly due to copy number variations (CNVs) resulting in dysregulated gene expression and therapeutic resistance. To identify actionable prognostic signatures of poor overall survival, we employed a systems biology approach using public databases to integrate CNVs, gene expression, and survival outcomes in pediatric, adolescent, and young adult OS patients. Chromosome 8 was a hotspot for poor prognostic signatures. The MYC-RAD21 copy number gain (8q24) correlated with increased gene expression and poor overall survival in 90% of the patients (n = 85). MYC and RAD21 play a role in replication-stress, which is a therapeutically actionable network. We prioritized replication-stress regulators, bromodomain and extra-terminal proteins (BETs), and CHK1, in order to test the hypothesis that the inhibition of BET + CHK1 in MYC-RAD21+ pediatric OS models would be efficacious and safe. We demonstrate that MYC-RAD21+ pediatric OS cell lines were sensitive to the inhibition of BET (BETi) and CHK1 (CHK1i) at clinically achievable concentrations. While the potentiation of CHK1i-mediated effects by BETi was BET-BRD4-dependent, MYC expression was BET-BRD4-independent. In MYC-RAD21+ pediatric OS xenografts, BETi + CHK1i significantly decreased tumor growth, increased survival, and was well tolerated. Therefore, targeting replication stress is a promising strategy to pursue as a therapeutic option for this devastating disease

    The same neuron may play different functional roles in the neural circuits to which it belongs. For example, neurons in the Tritonia pedal ganglia may participate in variable phases of the swim motor rhythms [1]. While such neuronal functional variability is likely to play a major role the delivery of the functionality of neural systems, it is difficult to study it in most nervous systems. We work on the pyloric rhythm network of the crustacean stomatogastric ganglion (STG) [2]. Typically network models of the STG treat neurons of the same functional type as a single model neuron (e.g. PD neurons), assuming the same conductance parameters for these neurons and implying their synchronous firing [3, 4]. However, simultaneous recording of PD neurons shows differences between the timings of spikes of these neurons. This may indicate functional variability of these neurons. Here we modelled separately the two PD neurons of the STG in a multi-neuron model of the pyloric network. Our neuron models comply with known correlations between conductance parameters of ionic currents. Our results reproduce the experimental finding of increasing spike time distance between spikes originating from the two model PD neurons during their synchronised burst phase. The PD neuron with the larger calcium conductance generates its spikes before the other PD neuron. Larger potassium conductance values in the follower neuron imply longer delays between spikes, see Fig. 17.Neuromodulators change the conductance parameters of neurons and maintain the ratios of these parameters [5]. Our results show that such changes may shift the individual contribution of two PD neurons to the PD-phase of the pyloric rhythm altering their functionality within this rhythm. Our work paves the way towards an accessible experimental and computational framework for the analysis of the mechanisms and impact of functional variability of neurons within the neural circuits to which they belong
