Genome‑wide analysis of CCHC‑type zinc finger (ZCCHC) proteins in yeast, Arabidopsis, and humans

The diverse eukaryotic proteins that contain zinc fingers participate in many aspects of nucleic acid metabolism, from DNA transcription to RNA degradation, post-transcriptional gene silencing, and small RNA biogenesis. These proteins can be classified into at least 30 types based on structure. In this review, we focus on the CCHC-type zinc fingers (ZCCHC), which contain an 18-residue domain with the CX2CX4HX4C sequence, where C is cysteine, H is histidine, and X is any amino acid. This motif, also named the “zinc knuckle”, is characteristic of the retroviral Group Antigen protein and occurs alone or with other motifs. Many proteins containing zinc knuckles have been identified in eukaryotes, but only a few have been studied. Here, we review the available information on ZCCHC-containing factors from three evolutionarily distant eukaryotes—Saccharomyces cerevisiae, Arabidopsis thaliana, and Homo sapiens—representing fungi, plants, and metazoans, respectively. We performed systematic searches for proteins containing the CX2CX4HX4C sequence in organism-specific and generalist databases. Next, we analyzed the structural and functional information for all such proteins stored in UniProtKB. Excluding retrotransposon-encoded proteins and proteins harboring uncertain ZCCHC motifs, we found seven ZCCHC-containing proteins in yeast, 69 in Arabidopsis, and 34 in humans. ZCCHC-containing proteins mainly localize to the nucleus, but some are nuclear and cytoplasmic, or exclusively cytoplasmic, and one localizes to the chloroplast. Most of these factors participate in RNA metabolism, including transcriptional elongation, polyadenylation, translation, pre-messenger RNA splicing, RNA export, RNA degradation, microRNA and ribosomal RNA biogenesis, and post-transcriptional gene silencing. Several human ZCCHC-containing factors are derived from neofunctionalized retrotransposons and act as proto-oncogenes in diverse neoplastic processes. The conservation of ZCCHCs in orthologs of these three phylogenetically distant eukaryotes suggests that these domains have biologically relevant functions that are not well known at present

Missplicing suppressor alleles of Arabidopsis PRE-MRNA Processing factor 8 increase splicing fidelity by reducing the use of novel splice sites

El empalme eficiente requiere un equilibrio entre la selección del sitio de empalme (SS) de alta fidelidad y la velocidad. En Saccharomyces cerevisiae , el factor de procesamiento pre-ARNm 8 (Prp8) ayuda a equilibrar la selección precisa de SS y la escisión de intrones y la unión de exones rápida y eficiente. argonaute1-52 ( ago1-52 ) e incurvata13 ( icu13 ) son alelos hipomórficos de los genes de Arabidopsis thaliana ARGONAUTE1 ( AGO1 ) y AUXIN RESISTANT6 ( AXR6 ) que albergan mutaciones puntuales que crean nuevos 3′SS y 5′SS, respectivamente. El espliceosoma reconoce estos nuevos SS, así como los SS genuinos intactos, produciendo una mezcla de ARNm maduros aberrantes y de tipo salvaje. Aquí, caracterizamos cinco nuevos alelos mutantes de PRP8 (uno de los dos coortólogos de levadura Prp8 de Arabidopsis ), nombrando la morfología de estos alelos de ago1-52 suprimido5 ( mas5 ). En el fondo mas5-1 , el espliceosoma reconoce preferentemente el 3′SS genuino intacto de ago1-52 y el 5′SS de icu13 . Dado que las mutaciones puntuales que dañan los SS genuinos hacen que el espliceosoma sea propenso a reconocer SS crípticos, también analizamos los alelos de cuatro genes que portan SS genuinos dañados y descubrimos que mas5-1 no suprimió su error de empalme. Las mutaciones mas5-1 y mas5-3 representan una nueva clase de supresores de empalme incorrecto que aumentan la fidelidad del empalme al dificultar el uso de nuevos SS, pero no alteran el empalme general del pre-ARNm

Compromiso organizacional y desempeño laboral de los profesionales de enfermería del Hospital Regional Hermilio Valdizán de Huánuco, 2022

Objetivo: Determinar la relación que existe entre el compromiso organizacional y el desempeño laboral de los profesionales de enfermería del Hospital Regional Hermilio Valdizán de Huánuco, 2022. Material y método: La investigación fue básica, descriptivo, no experimental, corte trasversal y correlacional. La población estuvo conformada por los profesionales de enfermería (N = 140), y la muestra de (n = 103). La técnica fue la encuesta y como instrumentos dos cuestionarios acerca del compromiso organizacional y desempeño laboral. Resultados: El compromiso organizacional de los profesionales de enfermería; el 61.2% es regular, el 19.4% es malo y el 19.4% es bueno; asimismo, el desempeño laboral de los profesionales de enfermería; el 60.2% es medio, el 21.4% es deficiente y el 18.4% es eficiente. Conclusiones: La Prueba de Correlación de Spearman, Rho=0.789, p-valor=0.011 (<0.05); por lo tanto, existe relación significativa entre el compromiso organizacional y el desempeño laboral de los profesionales de enfermería del Hospital Regional Hermilio Valdizán de Huánuco, 2022

SMALL ORGAN4 Is a Ribosome Biogenesis Factor Involved in 5.8S Ribosomal RNA Maturation

Ribosome biogenesis is crucial for cellular metabolism and has important implications for disease and aging. Human (Homo sapiens) glioma tumor-suppressor candidate region gene2 (GLTSCR2) and yeast (Saccharomyces cerevisiae) Nucleolar protein53 (Nop53) are orthologous proteins with demonstrated roles as ribosome biogenesis factors; knockdown of GLTSCR2 impairs maturation of 18S and 5.8S ribosomal RNAs (rRNAs), and Nop53 is required for maturation of 5.8S and 25S rRNAs. Here, we characterized SMALL ORGAN4 (SMO4), the most likely ortholog of human GLTSCR2 and yeast Nop53 in Arabidopsis (Arabidopsis thaliana). Loss of function of SMO4 results in a mild morphological phenotype; however, we found that smo4 mutants exhibit strong cytological and molecular phenotypes: nucleolar hypertrophy and disorganization, overaccumulation of 5.8S and 18S rRNA precursors, and an imbalanced 40S:60S ribosome subunit ratio. Like yeast Nop53 and human GLTSCR2, Arabidopsis SMO4 participates in 5.8S rRNA maturation. In yeast, Nop53 cooperates with mRNA transport4 (Mtr4) for 5.8S rRNA maturation. In Arabidopsis, we found that SMO4 plays similar roles in the 5.8S rRNA maturation pathway than those described for MTR4. However, SMO4 seems not to participate in the degradation of by-products derived from the 5′-external transcribed spacer (ETS) of 45S pre-rRNA, as MTR4 does

Hábitos de lectura y consumo de información de los adolescentes en el ámbito digital

The present work tries to echo how the communicative metamorphoses occurred in the last years, inferring in the reading habits and in the consumption of information in a general way and more specifically in adolescents. The analysis will start from the report prepared by the writers' guild 2018 (2019), since it allows us to monitor the state of reading habits and book purchases in a generalized way in Spain. Then, apply what has been poured out to young people extensively and, finally, materializing by extrapolating all the data analyzed to the geographical area of Cadiz adolescents and their position in relation to the rest of Spanish adolescents. Gender variables will also be taken into account in a very special way, to see if there is a certain preference depending on the sex of the reader in the choice of medium (digital or analogue) and if within the digital medium there are discrepancies in the type of text read. We will also check whether digital reading by young people is reduced exclusively to short texts and whether this choice can be derived from the very nature of digital. In the same way, we will pay special attention to the preferences of analogical reading versus digital reading, when we talk about long texts, again due to the personal nature of paper reading. We will analyze the reading frequency of the different digital media, as well as the choice of other shorter texts, categorized within the taxonomy of digital reading. Finally, we will take into account what types of readers with respect to reading frequency become from digital media. The idea is to draw up a panorama that shows how reading occurs among adolescents in the digital world, by definition, closer to their thoughts and feelings

Delitos ambientales en la minería ilegal aurífera en el distrito de Suyo - Ayabaca - Piura 2019

Los “Delitos Ambientales en la Minería Ilegal en el Distrito de Suyo – Ayabaca – Piura 2019”, permite determinar los delitos ambientales generados por la actividad minera ilegal aurífera que se práctica en nuestro país desde hace muchos años atrás, con limitada regulación socioambiental de parte del Estado. Los microextractores ilegales auríferos en el Distrito de Suyo, expolian concesiones, terrenos de Comunidades Campesinas o del Estado; la misma, genera grandes daños ambientales que impacta directamente en la calidad ambiental y salud ambiental, en el entorno social, las mismas que pueden ser considerados como delitos ambientales, enmarcada en el “Título XIII Delitos Ambientales, Capítulo I Delitos de Contaminación (Artículos 304-.307). Delitos de Minería Ilegal (Artículos 307-A-307- F) (Código Penal, 2016, pp. 190 - 192). “Capítulo II Delitos contra los recursos naturales (Artículos 310, 311-313). Capítulo III Responsabilidad funcional e información falsa (Artículo 314). Capítulo IV Medidas cautelares y exclusión o reducción de pena (Artículos 314-C - 314-D)” (Código Penal, 2016, pp.196, 198, 200). Para el desarrollo de la Tesis se utilizó el método general científico y como método específico el “método descriptivo correlacional” (Hernández, Fernández, 2007, p. 63), ya que ello, permite evaluar la relación entre dos variables en un contexto particular; el método jurídico, permite obtener una visión lo más aproximada posible a la verdad de la materia jurídica y la: diferentes líneas metodológicas” (Sánchez, 2018)., el diseño general aplicado de investigación fue “descriptivo correlacional” (Hernández y Baptista, 2007, pp 143 – 146). Concluyendo, que esta actividad ilícita, contraviene a uno de los derechos fundamentales de la persona, “gozar de un ambiente equilibrado y adecuado para el desarrollo de su vida” (Constitución Política del Perú, 1993, p.3), a lo que se señala “toda persona tiene el derecho irrenunciable a vivir en un ambiente saludable, equilibrado y adecuado para el pleno desarrollo de la vida” (Ley General del Ambiente, 2005, p.302291).The “2019 Environmental crimes from illegal mining in the district of Suyo – Ayabaca – Piura”, allows to determine the environmental crimes generated by the illegal gold mining activity that has been practiced in our country for many years without control nor social and environmental state regulation. The illegal gold small scale extractors in the District of Suyo, usurp concessions, which belong to Peasant Communities or the State. This situation generates great environmental damages that directly impact the environmental quality and environmental health, the social environment; which are categorized as environmental crimes, framed in the Penal Code, Title XIII “Environmental Crimes”, Chapter I “Pollution Crimes”(Articles 304 to 307),“ Illegal Mining Crimes ”(Articles 307-A to 307-F); Chapter II “Crimes against natural resources” (Articles 310, 311 and 313); Chapter III "Functional responsibility and false information" (Article 314); Chapter IV "Precautionary measures and exclusion or reduction of penalty" (Articles 314-C and 314-D). For the development of this thesis, it was used the general scientific method, and as a specific method, it was used the descriptive correlational method based on Hernández, Fernández (2007) page 63. This, because it allowed us to measure the relationship between two variables in a particular context; the legal method, which will allow obtaining a closer vision to the truth of the legal matter in accordance with what was stated by Sánchez Hidalgo, Adolfo (2018). El presente expediente versa sobre un procedimiento penal por el presunto delito de violación sexual a una persona en incapacidad de resistir, tipificado en el artículo 172° del Código Penal. En dicho proceso la agraviada es una menor de edad, quien manifestó haber mantenido relaciones sexuales consentidas con su enamorado de iniciales M.T.C (25); sin embargo, durante las investigaciones se dio a conocer que la menor fue diagnosticada con retardo mental leve. Por su parte, el imputado refirió que no tenía conocimiento de la discapacidad mental de la agraviada, puesto que, a simple vista parece una persona normal. Y que, en caso de haberlo sabido, no hubiera mantenido relaciones sexuales con ella. Teniendo en cuenta lo expuesto, se infiere que los principales temas jurídicos del presente proceso son los siguientes: i) violación sexual de persona en incapacidad de resistir; ii) libertad sexual; iii) indemnidad sexual y iv) error de tipo.This file deals with criminal proceedings for the alleged crime of rape of a person incapable of resisting, typified in article 172 of the Penal Code. In said process, the aggrieved party is a minor, who stated that she had had consensual sexual relations with her boyfriend with the initials M.T.C (25); however, during the investigations it was revealed that the minor suffered from mild mental retardation. For his part, the defendant stated that he was unaware of the mental deficiency of the aggrieved, since, at first glance, he appears to be a normal person. And that, if he had known, he would not have had sex with her. Taking into account the foregoing, it is inferred that the main legal issues in this proceeding are the following: i) sexual rape of a minor; ii) mentally disabled person; iii) sexual indemnity and iv) type error.Trabajo de suficiencia profesiona

Funcionamiento familiar y dependencia emocional en estudiantes de la Facultad de Psicología de una universidad nacional de Lima

El objetivo de este estudio fue establecer la relación entre Funcionamiento familiar y Dependencia emocional en estudiantes de la Facultad de Psicología de una Universidad Nacional de Lima. La metodología utilizada, fue la investigación no experimental, transversal y diseño correlacional. La muestra estuvo formada por 250 alumnos (58.4% mujeres y 41.6% hombres), su edad era de 20 a 27 años. Las pruebas utilizadas fueron la Escala de evaluación del Funcionamiento Familiar FACES-III y Dependencia Emocional (ACCA, 2013). Los resultados obtenidos reflejan que existe una relación de tipo indirecta entre el funcionamiento familiar y la dependencia emocional (rho=-.322, p=.000), mostrando que a menor funcionamiento familiar mayor dependencia emocional. Se descubrió la relación indirecta, entre el funcionamiento familiar y ansiedad por la separación (rho=-.202, p=.001), búsqueda de aceptación (rho=-.199, p=.002), percepción de su autoestima (rho=-.201, p=.001), apego a la seguridad (rho=-.125, p=.049), percepción de su autoeficacia (rho=-.270, p=.000), y el abandono de planes (rho=-.223, p=.000), explicando que a mayor apoyo, afecto, unión, expresión, organización y control dentro de la relación familiar menor características de la dependencia emocional. Se revelo la relación entre la dependencia emocional y la cohesión familiar (rho=-.280, p=.000) y la adaptación familiar (rho=-.278, p=.000)
